Abstract
Se proveen métodos de modulación de la expresión de un ácido nucleico objetivo en una célula, que incluyen introducir en la célula un primer ácido nucleico ajeno que codifica uno o más ARN complementarios al ADN, en donde el ADN incluye el ácido nucleico objetivo, introducir en la célula un segundo ácido nucleico ajeno que codifica una proteína Cas9 nula de nucleasa que se une al ADN y es guiado por los uno o más ARN, introducir en la célula un tercer ácido nucleico ajeno que codifica una proteína o dominio regulador de transcripción, en donde son expresados los uno o más ARN, la proteína Cas9 nula de nucleasa y la proteína o dominio regulador de transcripción, en donde los uno o más ARN, la proteína Cas9 nula de nucleasa y la proteína o dominio regulador de transcripción se colocalizan en el ADN, y en donde la proteína o dominio regulador de transcripción regula la expresión del ácido nucleico objetivo.
