Abstract

본 발명은 효소 활성을 지닌 미생물 세포를 이의 현택액으로서 또는 입자중의 고정화 세포의 현탁액으로서, 100mM 내지 무기 염의 포화 농도 범위의 몰 농도를 갖는, 무기 염의 중성 또는 약염기성 수용액인 수성 배지중에서, 상기 효소 활성을 지닌 세포 및 이의 효소 활성을 장시간 동안 안정하게 보존시키는 방법에 관한 것이다.
본 발명은 실온에서 조차도 세포 용해도 또는 효소 열화없이도 장시간(예 : 300일)동안 니트릴 하이드라타제 또는 니트릴라제 효소 활성을 갖는 다량의 세포 또는 입자중의 고정화 세포를 보존시키는 산업상 유용한 방법을 제공한다. 또한, 본 발명은 통상의 보존 방법에 필요한 노동력과 냉각비용을 상당히 감소시킬수 있게 한다.

Claims (14)

  1. 효소 활성을 지닌 미생물 세포를 이의 현탁액으로서 또는 입자중에 고정화된 세포의 현탁액으로서, 100mM 내지 무기염의 포화 농도 범위의 몰 농도를 갖는, 무기 염의 중성 또는 약 염기성 수용액인 수성 배지내에 보존시킴을 포함하여, 상기 효소 활성을 지닌 미생물 세포 및 이의 효소 활성을 장시간 동안 안정하게 보존시키는 방법.
  2. 제1항에 있어서, 무기염의 수용액이 인산염, 붕산염, 황산염, 아황산염 및 염산염으로 이루어진 그룹중에서 선택된 하나 이상의 염의 수용액인 방법.
  3. 제1항에 있어서, 무기 염이 나트륨염, 칼륨염 및 암모늄염으로 이루어진 그룹 중에서 선택된 하나 이상의 것인 방법.
  4. 제1항에 있어서, 무기 염의 수용액의 pH 값이 pH 7 내지 10의 범위인 방법.
  5. 제1항에 있어서, 효소 활성이 니트릴 하이드라타제 활성 또는 니트릴라제 활성인 방법.
  6. 제1항에 있어서, 미생물 세포가 고르도나 테라에 MA-1(FERM BP-4535)이 방법.
  7. 제1항에 있어서, 미생물 세포가 로도코코스 로도크로우스종 HT40-6 (FERM BP-5231)인 방법.
  8. 제1항에 있어서, 미생물 세포가 로도코커스 로도크로우스 J-1 (FERM BP-1478)인 방법.
  9. 제1항에 있어서, 미생물 세포가 로도코커스 로도크로우스 ATCC 33278인 방법.
  10. 제1항에 있어서, 입자중에 고정화된 세포의 농도가 건조 세포를 기준으로 하여 0.1 내지 30중량%의 범위인 방법.
  11. 제1항에 있어서, 보존 온도 범위가 0 내지 40℃인 방법.
  12. 제1항에 있어서, 무기 염의 수용액이 300 내지 700mM 범위의 몰 농도를 갖는 인산염 완충액을 포함하는 방법.
  13. 제1항에 있어서, 무기 염의 수용액이 50mM의 몰 농도를 갖는 인산염의 완충액 및 300mM의 몰 농도를 갖는 황산나트륨 용액을 포함하는 방법.
  14. 제1항에 있어서, 입자중에 고정화된 세포를 황산암모늄 수용액중에 현탁시키는 방법.
