Abstract

질병에 의한 야기된 당단백질의 당 사슬중 구조적 변화도는 측정될 하나 이상의 당단백질 분해물의 특수한 당 사술구조를 인지할 수 있는 렉틴과, 모든 단백질 분해물에 대한 결합 특성을 가지지만 렉틴이 부착된 당단백질 분해물에는 결합되지 않는 제1항체의 배합물을 사용하고, 제1항체가 부착된 당단백질 분해물과 제1항체가 부착되어 있지 않은 당단백질을 이들간 성질의 차이를 이용하여 분리 및 측정함으로써, 당사슬 구조가 다르지만 실질적으로는 동일한 단백질 구조를 하는 두가지 이상의 형태를 가지는 당단백질을 분리 및 측정함에 의해 고 정밀도로 신속하게 측정할 수 있다.

제3도는 실시예 1에서 사용한 HPLC장치의 개요를 나타낸 개요도이다.

Claims (7)

  1. 두개 이상의 형태를 가지는 당단백질을 포함하는 시료를 측정될 하나 이상의 당단백질의 특수한 당 사슬 구조를 인지할 수 있는 렉틴 및 모든 당단백질에 대한 결함특성을 가지지만 렉틴이 부착된 당 단백질에 대해서는 결합하지 않는 제1항체와 반응시킨 후, 제1항체가 부착된 당단백질과 제1항체가 부착되지 않은 당단백질을 이들 두 당단백질 간의 특성의 차이를 이용함으로써 분리 및 측정하는 것을 특징으로 하며, 당 사슬 구조가 다르지만 실질적으로는 동일한 단백질 구조를 가지는 분석물로서 둘 이상의 형태를 가지는 당단백질의 분리 및 측정방법.
  2. 제1항에 있어서, 측정될 당단백질과 제1항체의 복합물의 성질을 변화시킬 수 있는 물질 또는 표지(labeling)물질을 제1항체가 부착물질로서 가지고 있는 분리 및 측정방법.
  3. 제1항에 있어서, 렉틴이 부착된 당단백질을 포함하여 측정될 모든 당단백질에 대한 결합능력을 가지며, 표지 물질이 부착되어 있는 제2항체를 렉틴과 제1항체와 함께 사용하는 분리 및 측정방법.
  4. 제1항에 있어서, 측정될 당단백질을 글리코시다제로 예비 처리하거나, 당단백질을 렉틴 및 제1항체와 반응시킬 때 글리코시다제가 존재하는 분리 및 측정방법.
  5. 제3항에 있어서, 측정될 당단백질을 글리코시다제로 예비 처리하거나, 당단백질을 렉틴, 제1항체 및 제2항체와 반응시킬 때 글리코시다제가 존재하는 분리 및 측정방법.
  6. 제1항에 있어서, 성질 차이가 분자량, 등전점, 소수성 또는 전하량의 차이인 분리 및 측정방법.
  7. 제1항에 있어서, 렉틴을 당단백질의 고정 검색 제한 농도의 10배 이상인 농도로 사용하고, 제1항체의 농도가 (렉틴의 농도)×(렉틴의 결할 상수)/(제1항체의 결합 상수)이하인 분리 및 측정방법.
