Claims (21)
- (a) 반응버퍼에서 돼지시료와 항-스카톨 항체를 합쳐 반응혼합물을 형성하고; (b) 이 반응 혼합물을, 반응 혼합물에 존재하는 스카톨을 가지는 항체-스카톨 착물이 형성되기가 충분한 시간동안 배양하고; 그리고 (c) 돈육중의 숫돼지 오염 지표로서 항체-스타톨의 양을 측정하는 것으로 이루어지는, 돈육중의 숫돼지 오염을 측정하기 위한 방법.
- 제1항에 있어서, 시료가 전혈, 혈청 또는프라즈마로 이루어짐을 특징으로 하는 방법.
- 제1항에 있어서, 시료가 등 지방추출물로 이루어짐을 특징으로 하는 방법.
- 제1항에 있어서, 항체가 고체기질에 부착됨을 특징으로 하는 방법.
- 제4항에 있어서, 아는 양의 라벨된 스카톨 동족물을 시료와 함께 반응 혼합물을 가하고 고체 기질에 결합된 라벨의 양을 검출함으로써 측정함을 특징으로 하는 방법.
- 제1항에 있어서, 시료중에 존재하는 스카톨을 가지는 항체-스카톨 착물이 형성되기에 충분한 시간동안 시료를 아는양의 항-스카톨 항체와 반응시켜 반응 혼합물을 형성시키고, 반응 혼합물을 고체기질-부착된 스카톨 동족물과 함께 배양하고 기질에 결합되었거나 반응 혼합물중에 있는 항체의 양을 측정함을 특징으로 하는 방법.
- (a) 반응 버퍼에서 돼지의 혈액시료와 고체기질의 부착된 항-스카톨 항체를 합쳐 반응 혼합물을 형성하고; (b) 이 반응 혼합물을, 반응 혼합물에 존재하는 스카톨을 가지는 항체-스카톨 착물이 형성되기가 충분한 시간동안 배양하고; (c) 고체기질을 씻고; 그리고 (d) 돈육중의 숫퇘지 오염지표로서 고체기질에 부착된항체-스타톨 착물의 양을 측정하는 것으로 이루어지는, 돈육중의 숫돼지 오염을 검출하기 위한 방법.
- (a) 반응 버퍼에서, 라벨돈 스카톨 동족물, 돼지의 혈액시료 및 고체기질에 부착돈 항-스타톨 항체를 합쳐 반응 혼합물을 형성하고; (b) 이 반응 혼합물을, 반응 혼합물에 존재하는 스카톨을 가지는 항체-스타톨 착물이 형성되기에 충분한 시간동안 배양하고; (c) 고체기질을 씻고; 그리고 (d) 돈육중의 숫퇘지 오염의 지표로서 아는 양의 스카톨을 가지는 시료와 비교하여 고체기질에 결합된 라벨의 양을 측정하는 것으로 이루어지는, 돈육중의 숫돼지 오염을 검출하기 위한 방법.
- 제8항에 있어서, 라벨이 효소임을 특징으로 하는 방법.
- 제9항에 있어서, 효소가 가수분해 효소임을 특징으로 하는 방법.
- 제9항에 있어서, 효소가 산화환원효소임을 특징으로 하는 방법.
- 제8항에 있어서, 스카톨 동족물이 인돌-3-브티르산, 3-카르복시메틸인돌 또는 3-메틸인돌임을 특징으로 하는 방법.
- 제12항에 있어서, 접합물이 인돌-3-브티르산/알칼리성 포스파타아제 접합물임을 특징으로 하는 방법.
- 제8항에 있어서, 측정단계(d)가 (i) 효소기질을 고체기질에 가하여 제2의 반응 혼합물을 형성하고; (ⅱ) 이 제2반응 혼합물을, 검출성 기질숙성 생성물이 형성되기에 충분한 시간동안 배양하고; (ⅲ) 제2반응 혼합물의 강학적 밀도를 측정하는 것으로 이루어짐을 특징으로 하는 방법.
- (a) 반응 버퍼에서 스카톨 동족물/효소접합물, 돼지의 혈액시료 및 고체기질에 부착된 항-스타톨 항체를 합쳐 제1의 반응 혼합물을 형성하고; (b) 제1반응 혼합물을, 항체-스카톨 착물이 형성되기에 충분한 시간동안 배양하고; (c) 고체기질을 세척버퍼로 세척하고; (d) 효소기질 용액을 고체기질과 함께 배양하여 제2의 반응 혼합물을 형성하고; 그리고 (e) 돈육중의 숫돼지 오염의 지표로서 아는 양의 스카톨을 가지는 시료의 광학적 밀도와 비교하여 제2반응 혼합물의 밀도를 측정하는 것으로 이루어지는, 돈육중의 숫돼지 오염을 검출하기 위한 방법.
- (a) 돼지의 혈액시료, 트리스-버퍼화된 식염수중의 인돌-3-부티르산/알칼리성 포스파타아제 접합물, 및 마이크로 타이터 플레이트에 부착된 항-스카톨 항체를 합쳐 제1반응 혼합물을 형성하고; (b) 이 반응 혼합물을 약 4℃에서 약 4시간동안 배양하고; (c) 고체기질을 식염수-포릴옥실렌솔비탄 버퍼로 세척하고; (d) 약 37℃에서 약 1시간동안 마이크로타이터 플레이트로 P-니트로페닐포스페이트용액을 배양하고; (e) 효소활성을 중지시키기 위해 제2반응 혼합물을 pH를 변경시켜 제3의 반응혼합물을 형성하고; 그리고 (f) 시료의 스카톨 농도를 측정하기 위해 아느 양의 스카톨을 가지는 시료의 광학적밀도와 비교하여 제3반응 혼합물의 광학적 밀도를 측정하여, 시료가 약 0.3ppm이상의 스카톨을 가질 때 돈육을 숫돼지 오염된 것으로 판정하는 것으로 이루어지는, 돈육중의 숫돼지 오염을 검출하기 위한 방법.
- 제16항에 있어서, 돼지의 혈액시료가 혈청 또는 플라즈마로 이루어짐을 특징으로 하는 방법.
- 분리된 용기에 항-스카톨 항체와 라벨된 스카톨 동족물을 포함하는, 돼지 혈액시료중의 숫돼지 오염을 측정하기 취한 킷.
- 제18항에 있어서, 항-스카톨 항체가 마이크로타이터 플레이트에 부착됨을 특징으로 하는 킷.
- 제18항에 있어서, 스카톨 동족물이 효소로 라벨됨을 특징으로 하는 킷.
