Claims (18)
- 유전자 보수 올리고핵염기(gene repair oligonucleobase: GRON) 매개 돌연변이를 세포 내 표적 데옥시리보핵산(deoxyribonucleic acid: DNA) 서열로 도입하는 방법으로서,
상기 식물 세포로의 GRON의 전달 전에 및/또는 해당 전달과 동시에 하나 이상의 세포 DNA 보수 과정을 증가시키는 조건 하에 세포를 배양하는 단계를 포함하는 방법. - 제1항에 있어서, GRON의 존재 하에 상기 하나 이상의 세포 DNA 보수 과정을 증가시키는 조건은 염기 삭제 보수를 표적화하는 식물 세포 DNA로의 하나 이상의 부위의 도입, 비상동성 말단 연결(non-homologous end joining)을 표적화하는 식물 세포 DNA로의 하나 이상의 부위의 도입, 마이크로상동성 매개 말단 연결(microhomology-mediated end joining)을 표적화하는 식물 세포 DNA로의 하나 이상의 부위의 도입, 상동성 재조합을 표적화하는 식물 세포 DNA로의 하나 이상의 부위의 도입, 및 보수의 가이드 및 증가를 표적화하는 식물 세포 DNA로의 하나 이상의 부위의 도입 중 하나 이상을 포함하는 방법.
- 제1항 또는 제2항에 있어서, 상기 하나 이상의 세포 DNA 보수 과정을 증가시키는 조건은, 상기 식물 세포로의 GRON의 전달 전에 또는 해당 전달과 동시에, 상기 식물 세포로 단일 가닥 닉(nick) 또는 이중 DNA 가닥 파괴를 유도하는 하나 이상의 화합물을 도입하는 단계를 포함하는 방법.
- 제3항에 있어서, 상기 단일 가닥 닉 또는 이중 DNA 가닥 파괴를 유도하는 하나 이상의 화합물은 하나 이상의 뉴클레아제 및/또는 하나 이상의 블레오마이신 패밀리 화합물을 포함하는 방법.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 상기 단일 가닥 닉 또는 이중 DNA 가닥 파괴를 유도하는 하나 이상의 화합물은 상기 GRON에 공유 커플링된 방법.
- 제1항 내지 제5항 중 어느 한 항에 있어서, 상기 세포는 카눌라, 해바라기, 옥수수(corn), 담배, 사탕무, 면, 메이즈(maize), 밀, 보리, 쌀, 알팔파, 보리, 수수, 토마토, 망고, 복숭아, 사과, 배, 딸기, 바나나, 멜론, 감자, 당근, 상추, 양파, 대두, 콩 종(soya spp), 사탕수수, 완두콩, 병아리콩, 경협종완두(field pea), 잠두(faba bean), 렌틸콩(lentils), 순무, 루타바가(rutabaga), 싹양배추, 루핀, 꽃양배추, 케일, 강남콩, 포플러, 소나무, 유칼립투스, 포도, 감귤류, 라이밀, 알팔파, 호밀, 귀리, 잔디(turf) 및 벼과 작물(forage grass), 아마, 유채, 겨자, 오이, 나팔꽃, 발삼, 후추, 가지, 메리골드, 연꽃(lotus), 양배추, 데이지, 카네이션, 튤립, 아이리스 및 백합으로 이루어진 군으로부터 선택된 종의 식물 세포인 방법.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 상기 세포는 형질전환된 방법.
- 제7항에 있어서, 상기 표적 DNA 서열은 상기 세포의 내인성 유전자인 방법.
- 제1항 내지 제8항 중 어느 한 항에 있어서, 상기 세포는 식물 세포이고, 상기 방법은 상기 식물 세포로부터 상기 GRON에 의해 도입된 돌연변이를 갖는 식물을 재생하는 단계를 추가로 포함하는 방법.
- 제9항에 있어서, 상기 식물로부터 종자를 수집하는 단계를 추가로 포함하는 방법.
- 제1항 내지 제80항 중 어느 한 항의 방법에 따라 GRON에 의해 도입된 게놈 변형을 포함하는 식물 세포, 또는 제9항의 방법에 따라 GRON에 의해 도입된 게놈 변형을 포함하는 식물, 또는 제10항의 방법에 따라 GRON에 의해 도입된 게놈 변형을 포함하는 종자.
- 세포에서, 도너로서의 유전자 보수 올리고뉴클레오타이드(GRON)에 의해 돌연변이 전환을 개선하기 위해 단일 가닥 닉 또는 이중 가닥 파괴를 도입하는 하나 이상의 화합물을 도입하는 방법.

- 제13항에 있어서, 상기 단일 가닥 닉 또는 이중 가닥 파괴를 도입하는 하나 이상의 화합물은 하나 이상의 블레오마이신 패밀리 화합물을 포함하는 방법.
- 제13항 내지 제15항 중 어느 한 항에 있어서, 상기 표적 DNA 서열은 상기 프로토플라스트의 염색체 DNA에 존재하는 방법.
- 제13항 내지 제15항 중 어느 한 항에 있어서, 상기 표적 DNA 서열은 플라스미드에 존재하는 방법.
- 제13항 내지 제15항 중 어느 한 항에 있어서, 상기 단일 가닥 닉 또는 이중 가닥 파괴를 도입하는 하나 이상의 화합물은 상기 단일 가닥 닉 또는 이중 가닥 파괴를 도입하는 하나 이상의 화합물의 부재 하에 상기 GRON에 대해 결정된 효율에 비해 증가된 전환 효율을 제공하는 방법.
