Description
Claims (25)
- 핵산 및 적어도 하나의 형질감염제 외에, 핵으로의 DNA 전달능과 DNA-결합성을 겸비하는 적어도 하나의 화합물을 함유함을 특징으로 하는, 적어도 하나의 핵산의 형질감염용으로 유용한 약학 조성물.
- 핵산 및 적어도 하나의 형질감염제 외에, HMG 계열에 속하는 적어도 하나의 화합물 또는 이의 유도체 중 하나를 함유함을 특징으로 하는, 적어도 하나의 핵산의 형질감염용으로 유용한 약학 조성물.
- 제 1 항 또는 제 2 항에 있어서, 화합물이 HMG 1, 2, I, Y, 14 및 17형 단백질 또는 이들의 유도체 중에서 선택됨을 특징으로 하는 약학 조성물.
- 제 1 항 내지 3 항 중 어느 한 항에 있어서, 화합물이 인간 HMG1 단백질, 이의 유도체 중 하나 또는 동족체의 전부 또는 일부로 표시됨을 특징으로 하는 약학 조성물.
- 제 1 항 내지 4 항 중 어느 한 항에 있어서, 화합물이 또한 폴리글리코실화, 설폰화, 포스포릴화 및/또는 복합당에 또는 친유제에 그래프팅됨을 특징으로 하는 약학 조성물.
- 제 1 항 내지 5 항 중 어느 한 항에 있어서, 화합물이 또한 핵- 또는 세포 수용체 리간드와 결합됨을 특징으로 하는 약학 조성물.
- 제 1 항 내지 6 항 중 어느 한 항에 있어서, 형질감염제가 양이온 중합체 또는 리포펙턴트임을 특징으로 하는 약학 조성물.
- 제 7 항에 있어서, 양이온 중합체가 바람직하게는 화학식 1의 화합물임을 특징으로 하는 약학 조성물.화학식 1상기식에서,R은 수소원자 또는 화학식의 그룹이고n은 2 내지 10의 정수이며,p 및 q는 정수이며, p와 q의 합은 중합체의 평균 분자량이 100 내지 107이 되도록 하는 값이다.
- 제 7 항 또는 제 8 항에 있어서, 양이온 중합체가 폴리에틸렌이민(PEI) 및 폴리프로필렌이민(PPI) 중에서 선택됨을 특징으로 하는 약학 조성물.
- 제 9 항에 있어서, 중합체가 평균분자량이 50,000인 폴리에틸렌이민(PEI50K) 및 평균분자량이 800,000인 폴리에틸렌이민(PEI800K) 중에서 선택됨을 특징으로 하는 약학 조성물.
- 제 7 항에 있어서, 리포펙턴트가 화학식 2의 적어도 하나의 친수성 폴리아민 영역을 포함함을 특징으로 하는 약학 조성물.화학식 2상기식에서,m은 2 이상의 정수이고,n은 1 이상의 정수이며,m은 포화 또는 불포화 탄화수소쇄, 콜레스테롤 또는 라멜라 상 또는 육변형 상을 형성할 수 있는 천연 또는 합성 지질을 포함하는 유형의 친유성 영역과 공유결합되어 있는 두 아민 사이에 놓인 상이한 탄소그룹 간에 다양할 수 있다.
- 제 11 항에 있어서, 폴리아민 영역이 스퍼민, 써민 또는 핵산결합성을 보유한 이들의 동족체 중 하나임을 특징으로 하는 약학 조성물.
- 제 11 항에 있어서, 친유성 영역이 화학식 4로 표현됨을 특징으로 하는 약학 조성물.화학식 4상기식에서,X 및 X′는 상호 독립적으로 산소원자, 메틸렌 그룹 -(CH2)q-(여기에서, q는 0, 1, 2 또는 3이다), 또는 아미노 그룹 -NH- 또는 -NR′-(여기에서, R′은 C1-C4알킬 그룹이다)이고,Y 및 Y′는 상호 독립적으로 메틸렌 그룹, 카보닐 그룹 또는 C=S 그룹이며,R3, R4및 R5는 상호 독립적으로 수소원자 또는 치환되거나 비치환된 C1-C4알킬 라디칼이며, p는 0 내지 5 범위일 수 있으며,R6는 콜레스테롤 유도체 또는 알킬아미노 그룹 -NR1R2이며 R1및 R2는 상호 독립적으로 포화 또는 불포화된 직쇄 또는 측쇄 C12-C22지방족 라디칼이다.
- 제 11 항 내지 13 항 중 어느 한 항에 있어서, 바람직하게는 디옥타데실아미도글리실스퍼민 (DOGS), 팔미토일포스파티딜에탄올아민 5-카복시스퍼밀아미드 (DPPES), 2,5-비스(3-아미노프로필아미노)펜틸(디옥타데실카바모일메톡시)아세테이트 또는 1,3-비스(3-아미노프로필아미노)-2-프로필(디옥타데실카바모일메톡시)아세테이트 중에서 선택된 리포폴리아민임을 특징으로 하는 약학 조성물.
- 제 1 항 내지 7 항, 11 항, 12 항 및 14 항 중 어느 한 항에 있어서, 형질감염제가 디옥타데실아미도글리실스퍼민(DOGS)임을 특징으로 하는 약학 조성물.
- 제 1 항 내지 15 항 중 어느 한 항에 있어서, 핵산이 디옥시리보핵산임을 특징으로 하는 약학 조성물.
- 제 1 항 내지 16 항 중 어느 한 항에 있어서, 핵산이 리보핵산임을 특징으로 하는 약학 조성물.
- 제 16 항 또는 제 17 항에 있어서, 핵산이 화학적으로 변형됨을 특징으로 하는 약학 조성물.
- 제 16 항 내지 18 항 중 어느 한 항에 있어서, 핵산이 안티센스 서열임을 특징으로 하는 약학 조성물.
- 제 16 항 내지 19 항 중 어느 한 항에 있어서, 핵산이 치료 유전자를 함유함을 특징으로 하는 약학 조성물.
- 제 1 항 내지 20 항 중 어느 한 항에 있어서, 하나 이상의 중성지질을 또한 포함함을 특징으로 하는 약학 조성물.
- 제 21 항에 있어서, 중성지질(들)이 합성지질 또는 천연지질 중에서 선택되고 생리학적 조건에서 쯔비터이온성이거나 이온 전하를 전혀 띠지 않을 수 있음을 특징으로 하는 약학 조성물.
- 제 21 항 또는 제 22 항에 있어서, 중성지질(들)이 디올레오일포스파티딜에탄올아민(DOPE), 올레오일팔미토일포스파티딜에탄올아민(POPE), 디스테아로일, -팔미토일 및 -미리스토일-포스파티딜에탄올아민 및 1 내지 3회 N-메틸화된 이들의 유도체; 포스파티딜글리세롤, 디아실글리세롤, 글리코실디아실글리세롤, 세레브로시드(특히, 예를 들면, 갈락토세레브로시드), 스핑고리피드(특히, 예를 들면, 스핑고마이엘린) 및 아시알로강글리오시드(특히, 예를 들면, 아시알로GM1 및 GM2) 중에서 선택됨을 특징으로 하는 약학 조성물.
- 핵산의 시험관내, 생체외 및/또는 생체내 전달을 위한 제 1 항 내지 23 항 중 어느 한 항에 따른 약학 조성물의 용도.
- 세포 수용체 리간드, 항체 또는 항체 유도체와 커플링되어 있고, 상응하는 수용체 또는 항원을 발현하는 세포 쪽으로 핵산을 표적화하는 제 1 항 또는 제 2 항 에 따른 화합물의 용도.
