Claims (40)
- 연질 폴리사카라이드 하이드로겔의 제조에 사용하기 위한 조성물로서, 연질 폴리사카라이드 하이드로겔은 연질 폴리사카라이드 하이드로겔에 이온성 용액을 첨가함으로써 경질 폴리사카라이드 하이드로겔로 전환가능하고,
상기 조성물은,
하나 이상의 수용성 고 아실 젤란 검 중합체;
하나 이상의 수용성 저 아실 젤란 검 중합체; 및
하나 이상의 수용성 화학적으로 변형된 젤란 검 중합체 또는 하나 이상의 펩티드 변형된 젤란 검 중합체
를 포함하며,
하나 이상의 고 아실 젤란 검 중합체는 반복 단위 당 1개의 글리세레이트 및 2개의 반복 단위마다 1개의 아세테이트를 가지고,
하나 이상의 저 아실 젤란 검 중합체는 상기 고 아실 젤란 검 중합체에서 상기 글리세레이트 및 아세테이트 기가 제거된 중합체이고,
연질 폴리사카라이드 하이드로겔은 균일한 매트릭스 구조를 나타내고, 경질 폴리사카라이드 하이드로겔은 응집된 매트릭스 네트워크 구조를 나타내는, 조성물. - 제1항에 있어서, 연질 폴리사카라이드 하이드로겔이 전단력에 의해 액체 상태로 전환되거나, 또는 전단력이 중단되면 그의 하이드로겔 상태로 회복하는 것을 허용함으로써, 상기 연질 폴리사카라이드 하이드로겔이 전단 박하 및 자기 치유의 유동학 특성을 나타내는, 조성물.
- 제2항에 있어서, 상기 전단력이 피펫팅, 주사기 주입, 또는 펌프 관류 또는 그의 조합에 의해 가해지는, 조성물.
- 제1항에 있어서, 상기 경질 폴리사카라이드 하이드로겔이 유동학적 특성을 갖는 3-D 겔 구조를 나타내어 상기 경질 폴리사카라이드 하이드로겔이 피펫팅 또는 전단에 의해 파괴될 때, 상기 경질 폴리사카라이드 하이드로겔이 더 작은 겔 입자로 부서지고, 하나 이상의 생체활성 분자에 대한 친화성을 갖도록 하는, 조성물.
- 제1항에 있어서, 상기 경질 폴리사카라이드 하이드로겔이 10 Pa 초과의 저장 모듈러스 값을 갖는, 조성물.
- 제1항에 있어서, 상기 경질 폴리사카라이드 하이드로겔이 그의 겔 형성을 80℃ 이하의 온도에서 유지하지만, 외력으로 흐트러뜨렸을 때 더 작은 겔 입자로 부서질 수 있는, 조성물.
- 제1항에 있어서, 상기 하나 이상의 화학적으로 변형된 젤란 검 중합체가 펩티드 변형된 젤란 검 중합체인, 조성물.
- 제4항에 있어서, 가열 전에 또는 후에 상기 젤란 검 용액 내에 첨가되는 상기 하나 이상의 생체활성 분자가 세포, 펩티드, 단백질, 지질, 폴리사카라이드, 성장 인자, 성장 호르몬, 항체, 효소, 세포 수용체, 세포 리간드, 항생제, 항균제, 항진균제, 항곰팡이제, 및 트리펩티드 Arg-Gly-Asp 모티프, Ile-Lys-Val-Ala-Val 펩티드, Arg-Glu-Asp-Val 펩티드, 라미닌 서열 Tyr-lle-Gly-Ser-Arg, 및 폴리 리신을 포함하는 NH2, COOH 및 CONH2 기를 갖는 기능성 펩티드 분자로 이루어진 군으로부터 선택되는, 조성물.
- 제1항에 있어서, 상기 연질 폴리사카라이드 하이드로겔을 여분의 포스페이트 완충액, 세포 배양 배지, 또는 이온성 용액의 수용액, 또는 그의 조합에 침지함으로써 상기 연질 폴리사카라이드 하이드로겔이 경질 폴리사카라이드 하이드로겔로 전환되는, 조성물.
- 제4항에 있어서, 하나 이상의 생체활성 분자가 그의 생체활성을 유지하면서 연질 및 경질 폴리사카라이드 하이드로겔과 접촉하거나, 그에 부착되거나, 또는 매립되는, 조성물.
- 제4항에 있어서, 상기 하나 이상의 생체활성 분자 각각이 그의 생체활성을 유지하면서 연질 및 경질 폴리사카라이드 하이드로겔에 현탁되거나 또는 포획되는, 조성물.
- 제1항에 있어서, 펩티드에 의해 변형된 젤란 검의 용액이 펩티드를 상기 젤란 검 용액 내에 혼합물로서 첨가하고 이어서 100℃ 이상의 온도 및 1 내지 40 psi의 압력에서 3 내지 30분의 기간 동안 가열함으로써 형성되는, 조성물.
- 제1항에 있어서, 변형된 젤란 검의 용액이 젤란 검 중합체 용액을 형성하는 수용성 젤란 검 중합체에 나트륨 시트레이트를 첨가하고, 젤란 검 중합체 용액의 pH를 중성 pH로 조정함으로써 형성되는, 조성물.
- 제1항에 있어서, 0.001% 내지 20%의 상기 하나 이상의 고 아실 젤란 검 중합체, 0.001% 내지 20%의 상기 하나 이상의 저 아실 젤란 검 중합체, 0.001% 내지 20%의 상기 하나 이상의 변형된 젤란 검 중합체를 포함하고, 0.00001% 내지 30%의 상기 하나 이상의 생체활성 분자를 추가로 포함하는, 조성물.
- 제14항에 있어서, 10 내지 20000 Pa의 저장 모듈러스 값을 갖는, 조성물.
