Abstract
Description
Claims (15)
- 하기 단계를 포함하는 식 I-a의 화합물의 제조 방법으로서,
(i) 알코올레이트 염기의 존재 하에 에틸 니코티네이트와 N-비닐피롤리돈을 반응시켜 3-니코티노일-1-비닐피롤리딘-2-온을 얻는 단계;
(ii) 3-니코티노일-1-비닐피롤리딘-2-온을 산과 반응시켜 미오스민을 얻는 단계;
(iii) 환원제를 사용하여 미오스민을 노르니코틴으로 환원시키는 단계; 및
(iv) 노르니코틴을 메틸화하여 식 Ia의 화합물을 얻는 단계,
상기 단계 (i)에서의 알코올레이트 염기는 나트륨 에탄올레이트, 칼륨 에탄올레이트 및 이의 혼합물로부터 선택되는 알칼리 금속 알코올레이트이고,
1-포트 공정으로 수행하는 제조 방법. - 제1항에 있어서, 단계 (i)을 방향족 용매의 존재 하에 수행하는 것인 제조 방법.
- 제2항에 있어서, 방향족 용매가 벤젠, 톨루엔 또는 이의 혼합물인 제조 방법.
- 제1항 내지 제3항 중 어느 한 항에 있어서, 단계 (i)에서 1 당량의 에틸 니코티네이트를 기준으로 알코올레이트 염기를 1.4 내지 2.0 당량의 양으로 첨가하는 것인 제조 방법.
- 제1항 내지 제3항 중 어느 한 항에 있어서, 단계 (ii)를, 90 내지 115℃의 온도에서 무기산을 사용하여 수행하는 것인 제조 방법.
- 제5항에 있어서, 무기산이 HCl인 제조 방법.
- 제1항 내지 제3항 중 어느 한 항에 있어서, 단계 (iv)를 포름산 및 파라포름알데히드, 또는 포름산 및 포름알데히드를 사용하여 수행하는 것인 제조 방법.
- 제7항에 있어서, 단계 (iv)를 40 내지 95℃의 온도에서 수행하는 것인 제조 방법.
- 제1항에 있어서, 단계 (iii)을, 환원제로서 NaBH4를 사용하여 수행하는 것인 제조 방법.
- 제1항 내지 제3항 중 어느 한 항에 있어서, 140 내지 160℃의 온도에서 식 I-a의 화합물을 유기 염기와 반응시키는 단계를 추가로 포함하는 제조 방법.
- 제1항 내지 제3항 중 어느 한 항에 있어서, 키랄 O,O'-이치환된 타르타르산의 첨가에 의해 식 I-a의 화합물의 거울상 이성질체를 분리하는 단계를 추가로 포함하는 제조 방법.
- 제11항에 있어서, O,O'-이치환된 타르타르산은 디벤조일 타르타르산인 제조 방법.
- 제12항에 있어서, 디벤조일 타르타르산이 O,O'-디벤조일-L-타르타르산인 제조 방법.
- 제12항에 있어서, 디벤조일 타르타르산은 O,O'-디벤조일-L-타르타르산(L-DBTA) 대 O,O'-디벤조일-D-타르타르산(D-DBTA)의 몰비가 80:20 이상인 L-DBTA와 D-DBTA의 혼합물인 제조 방법.
