도 7은 선행하는 사이클로부터 첨가된 단편을 사용하는 신규한 사이클의 개시;
를 모식적으로 나타내고
도 8은 효소적 첨가의 상 및 정확한 시퀀스의 단편의 정제의 상을 수행하기 위한 "반응기(reactor)"의 가능한 배치의 모식도를 나타낸다.
Claims (19)
- n이 3 내지 1012의 범위 내인, n 개의 뉴클레오티드를 포함하는, 초기 단편으로 알려진, 핵산 단편의 적어도 하나의 연장(elongation) 사이클을 포함하는, 핵산의 합성 방법으로,
각 사이클이 하기의 방법으로 세분되고:
a) 하기의 단계를 포함하는, 제1 상으로 알려지는, 상기 단편의 하나의 말단에 X 개의 뉴클레오티드(상기 X는 1 내지 5)를 효소적으로 첨가하여 n + X 개의 뉴클레오티드를 포함하는 단편을 수득할 수 있는, 효소적 첨가 상(phase of enzymatic addition):
- n 개의 뉴클레오티드를 포함하는, 초기 핵산 단편 또는 연장 과정 내 핵산 단편의 제1 말단을 제1 지지체에 부착하는 제1 부착 단계로, 상기 제1 지지체가 상기 단편의 5'말단과 가역적 상호작용을 허용하는 부착 표면을 가지는, 제1 부착 단계,
- 효소적 첨가에 필요한 시약의 첨가 단계로, 상기 초기 핵산 단편의 제2 말단으로의 X개의 뉴클레오티드의 효소적 첨가 단계로, 단편에 첨가되는 뉴클레오티드 또는 뉴클레오티드들의 유리 단부(free end)가, 하나의 그리고 동일한 단편에의 X 개의 뉴클레오티드의 다중 첨가를 방지하도록 하기 위한 보호성 화학기를 포함하는, 첨가 단계,
- 반응 매질(medium)로부터의 원치 않는 시약을 제거하는 임의적 단계,
- 상기 제1 지지체로부터 n + X 개의 뉴클레오티드를 포함하는 단편을 탈착하는 단계, 및
- 상기 n + X 개의 뉴클레오티드를 포함하는 단편을 이동 유체 스트림(stream)의 생성에 의하여, 비공유 상호작용에 의해 n + X개의 뉴클레오티드를 포함하는 단편의 3' 말단의 보호기와 상호작용할 수 있는 제2 지지체로 이송하는 제1 이송 단계;
b) 하기의 연속적인 단계를 포함하고, 제2 상으로 알려지는, n + X 개의 뉴클레오티드를 포함하는 정확한 시퀀스를 갖는 단편의 정제 상(phase of purification):
- 3' 말단에 의하여 상기 n + X 개의 뉴클레오티드를 포함하는 단편을 제2 지지체에 부착하는 제2 부착 단계,
- 첨가되지 않은 단편 및 제2 지지체에 부착되지 않은 단편의 제거 단계,
- 상기 제2 지지체로부터의, 상기 n + X 개의 뉴클레오티드를 포함하는 단편의 탈착 단계,
- 반응 매질로부터 원치 않는 잔류 시약을 제거하는 임의적 단계;
c) 하기의 연속적인 단계를 포함하는, 제3 상으로 알려지는, n + X개의 뉴클레오티드를 포함하는 정확한 시퀀스를 갖는 단편을 증폭하는, 임의적 증폭 상(phase of amplification):
- 증폭에 필요한 시약의 첨가 단계,
- n + X 개의 뉴클레오티드를 포함하는 단편을, 증배 인자(multiplication factor) Y에 의해 증배하는(multiplication) 단계로서, 상기 Y는 1 내지 4 X 1010의 범위 내인, 단계,
- n + X개의 뉴클레오티드를 포함하는 단편의 이송 단계;
이 때, 상기 각 사이클은, 수성 매질과 같은, 효소적 첨가 및 효소 증폭과 호환될 수 있는 반응 매질 중에서 수행되며,
합성 방법은 또한 연장 사이클의 모두의 말단에서 증배 인자(multiplication factor) Y에 의한 최종 증폭 단계를 포함하는, 방법. - 제1항에 있어서,
n + X 개의 뉴클레오티드를 포함하는 정확한 시퀀스의 단편의 정제 상이, 임의의 원치 않는 잔류 시약의 제거를 포함함을 특징으로 하는 방법. - 제1항 또는 제2항에 있어서,
증폭 상이, 반응 매질 중에서, 부정확한 시퀀스 단편으로 알려진 다른 단편의 증배 인자에 비하여, 정확한 시퀀스를 갖는 단편을 10 배 이상 더 큰 증배 인자로 증배하는, 정확한 시퀀스 단편으로 알려진 n + X개의 뉴클레오티드를 포함하는 시퀀스를 갖는 단편의 선택적 증폭임을 특징으로 하는 방법. - 제1항 또는 제2항에 있어서,
제1 지지체 및 제2 지지체가 서로 다른 속성의 부착 표면을 나타냄(exhibit)을 특징으로 하는 방법. - 제1항 또는 제2항에 있어서,
제1 지지체가, 적어도 3개의 뉴클레오티드를 포함하는 핵산 가닥을 비공유적으로 고정하는 표면을 나타내고, 이들 핵산 가닥의 상기 뉴클레오티드는 초기 단편의 제1 말단에 존재하는 뉴클레오티드에 상보적이어서, 그들 개개의 염기들 간의 수소 결합에 의해, 단편의 상기 제1 말단을 부동화(immobilize)하는 것을 특징으로 하는 방법. - 제1항 또는 제2항에 있어서,
상기 제1 및 제2 지지체가 유리판(glass sheet), 폴리머 물질의 판(sheet of polymer material) 또는 비드의 형태임을 특징으로 하는 방법. - 제1항 또는 제2항에 있어서,
X 개의 뉴클레오티드의 첨가가, 기질 가닥(matrix strand)의 존재 없이 변성된(modified) 뉴클레오티드를 중합할 수 있는 효소를 사용하여, 효소적 경로에 의해 수행됨을 특징으로 하는 방법. - 제7항에 있어서,
효소가 텔로머라아제(telomerase), 번역 효소(translation enzyme), 주형-비의존성 RNA 폴리머라아제 족의 효소 또는 말단 디옥시뉴클레오티딜 트랜스퍼라아제 족의 효소로부터 선택됨을 특징으로 하는 방법. - 제1항 또는 제2항에 있어서,
제2 지지체가 단백질과 같은 분자로 피복되어, 상기 분자와 상기 보호성 화학기 간의 비공유 결합, 및 이에 따른, 제2 부착 단계 동안의, 첨가된 단편의 부동화를 가능하게 함을 특징으로 하는 방법. - 제1항 또는 제2항에 있어서,
상기 제2 지지체로부터 상기 n + X 개의 뉴클레오티드를 포함하는 단편을 탈착하는 단계가, pH 또는 온도의 변경과 같은 반응 매질 조건의 변경(modification)에 의해 수행되거나 또는 상기 제2 지지체가 자석 지지체인 경우 전자기 방사(electromagnetic radiation)의 작용 하에서 수행됨을 특징으로 하는 방법. - 제1항 또는 제2항에 있어서,
효소적 첨가가 5'에서 3' 방향으로 수행됨을 특징으로 하는 방법. - 제1항에 있어서,
X가 1 내지 3임을 특징으로 하는 방법. - 제1항에 있어서,
Y가 1 내지 1 x 1010임을 특징으로 하는 방법. - 제1항에 있어서,
증폭 상이 효소적인 것을 특징으로 하는 방법. - 제4항에 있어서,
제1 지지체와 제2 지지체가 별개인 것을 특징으로 하는 방법. - 제6항에 있어서,
상기 제1 및 제2 지지체 중 하나가 자기적 특성을 나타내는 것을 특징으로 하는 방법. - 제1항 또는 제2항에 있어서
유전자의 생산, 핵산의 합성 시퀀스 생산, DNA의 생산 또는 RNA의 생산을 위한 방법. - 하기를 포함하는, 제1항 또는 제2항에 따른 방법을 시행하기 위한 키트:
- n 개의 뉴클레오티드를 포함하는 핵산 단편을 포함하는 반응 매질
- 뉴클레오티드의 첨가를 위한 효소 시약
- 상기 효소적 첨가 시약에 의해 첨가될 수 있는, 3' 말단에 화학적 보호기를 포함하는 뉴클레오티드 또는 뉴클레오티드들의 조합
- 정제 상을 위한, 세척(13, 14) 또는 완충제 용액 또는 이들의 조합

- 사용을 위한 지시서(instruction). - 제18항에 있어서,
- 핵산 단편의 제1 부착 지지체로의 부착에 선호되는 완충제 매질, 핵산 단편의 제2 부착 지지체로의 부착에 선호되는 완충제 매질, 및
- 제1 부착 지지체로부터 핵산 단편을 탈착하는데 선호되는 완충제 매질, 제2 부착 지지체로부터 핵산 단편을 탈착하는데 선호되는 완충제 매질
을 추가로 포함하는 키트.
