Abstract
Description
본 발명에서, 용어 "압타머", 또는 "핵산 압타머"는 "Chemical Antibody"로서 특정 화합물부터 단백질까지 다양한 종류의 표적 리간드에 높은 특이도와 친화도로 결합할 수 있는 특성을 가지는 짧은 길이(20~80개 염기)의 단일 가닥 핵산 분자를 의미한다. 압타머는 SELEX(Systematic evolution of ligands by exponential enrichment)를 통해 생체 외(In Vitro)에서 제조할 수 있다. 상기 "압타머", 또는 "핵산 압타머"는 혼용되어 사용될 수 있다.

Claims (15)
- 글리피칸-3(Glypican-3, GPC3)에 특이적으로 결합하는 핵산 압타머 및 항암제를 포함하는, 변형 압타머로서,
상기 핵산 압타머는 서열번호 6의 염기서열 또는 이의 변이체를 포함하는 압타머이며,
상기 변이체는 서열번호 6의 염기서열에서 하나 이상의 뉴클레오티드 내 염기 구조의 피리미딘 기의 5번 탄소 위치에서 나프틸기로의 치환; 하나 이상의 뉴클레오티드가 당과 염기를 가지지 않는 링커로의 치환; 하나 이상의 뉴클레오티드 내 염기 구조의 2번 탄소 위치에서 -OH 기가 -Methoxy(메톡시), -F(불소) 또는 Methoxyethyl(메톡시에틸) 중 어느 하나 이상으로의 치환으로 이루어지는 군으로부터 선택되는 어느 하나 이상의 변형을 포함하는 것이고,
상기 항암제는 젬시타빈(Gemcitabine) 또는 사이타라빈(cytarabine)인 것인, 변형 압타머.

- 제1항에 있어서, 상기 핵산 압타머는 서열번호 6 내지 서열번호 25로 이루어지는 어느 하나의 염기서열을 포함하는 것인, 변형 압타머.
- 제1항에 있어서, 상기 항암제는 상기 핵산 압타머에 1개 이상 결합되는 것인, 변형 압타머.
- 제5항에 있어서, 상기 항암제는 상기 핵산 압타머의 염기서열 내 어느 하나 이상의 염기가 치환되어 결합된 것인, 변형 압타머.
- 제6항에 있어서, 상기 염기는 시토신 또는 구아닌인 것인, 변형 압타머.
- 삭제
- 제1항에 있어서, 상기 항암제는 젬시타빈인 것인, 변형 압타머.
- 제1항, 제4항 내지 제7항 및 제9항 중 어느 한 항의 변형 압타머를 유효성분으로 포함하는, 간암 예방 또는 치료용 약학 조성물.
- 서열번호 6 내지 서열번호 25로 이루어지는 군으로부터 선택되는 어느 하나의 염기서열을 포함하는, 글리피칸-3에 특이적으로 결합하는 핵산 압타머.
- 서열번호 6으로 이루어진 염기서열의 변이체를 포함하는, 글리피칸-3에 특이적으로 결합하는 핵산 압타머로서,
상기 변이체는 서열번호 6의 염기서열에서 하나 이상의 뉴클레오티드 내 염기 구조의 피리미딘 기의 5번 탄소 위치에서 나프틸기로의 치환; 하나 이상의 뉴클레오티드가 당과 염기를 가지지 않는 링커로의 치환; 하나 이상의 뉴클레오티드 내 염기 구조의 2번 탄소 위치에서 -OH 기가 -Methoxy(메톡시), -F(불소) 또는 Methoxyethyl(메톡시에틸) 중 어느 하나 이상으로의 치환으로 이루어지는 군으로부터 선택되는 어느 하나 이상의 변형을 포함하는 것인, 핵산 압타머.
- 제11항 또는 제12항의 핵산 압타머를 유효성분으로 포함하는, 글리피칸-3 검출용 조성물.
- 간암의 진단 또는 예후에 필요한 정도를 제공하기 위하여,
제13항의 검출용 조성물을 이용하여 검사 대상체 유래의 생물학적 시료로부터 글리피칸-3의 함량을 검출하는 단계;
상기 함량 검출결과를 대조군 시료의 글리피칸-3의 함량 검출결과와 비교하는 단계; 및
상기 대조군 시료와 비교하여, 상기 대상체 유래 시료의 함량 수준에 변화가 있는 경우, 상기 대상체를 간암의 진단 또는 예후와 연관시키는 단계를 포함하는,
글리피칸-3을 검출하는 방법.
- 제13항의 검출용 조성물을 포함하는, 간암 진단용 키트.
