Description
도 2는, 스크리닝을 통해 선별된 GFP 표적 cp-asiRNA인 cp-asiGFP13을 세포전달체 이용 없이 세포에 처리하고 배양했을 때의 GFP 유전자 침묵 효과를 qRT-PCR로 확인한 결과이다.
도 3은, GFP 표적 cp-asiRNA인 cp-asiGFP13을 세포전달체 이용 없이 세포에 처리하고 배양했을 때의 GFP 유전자 침묵 효과를 Image cytometry 및 형광 현미경으로 확인한 결과이다.
도 4는, 유전자 침묵 효과를 증진시키는 화합물의 스크리닝 과정을 그림으로 도시한 것이다.

도12a 및 도 12b는, Amlodipine(암로디핀) 화합물에 대해 cp-asiRNA의 농도를 바꿔가며 상대적인 mRNA 발현수준을 측정한 것으로, 각각 cp-asiGFP 및 cp-asisurvivin를 처리하여 GFP(도 12a) 및 survivin(도 12b)의 발현수준을 qRT-PCR로 측정한 결과이다.
도 13은, 본 발명에 따른 화학적으로 변형시킨 cp-lasiRNA의 구조를 그림으로 도시한 것이다.
도 14a 및 도 14b는, Amlodipine(암로디핀)과 Cilnidipine(실니디핀) 화합물에 대해 cp-lasiCTGF의 농도를 달리하여 상대적인 mRNA 발현수준을 측정한 것으로, 상기 cp-lasiCTGF와 함께 각각 Amlodipine(도 14a)및 Cilnidipine(도 14b)을 농도별로 처리한 후 CTGF mRNA의 발현수준을 qRT-PCR로 측정한 결과이다.

상기 작은 간섭 RNA는 화학적으로 변형된 것을 특징으로 하는, 조성물.
- 제3항에 있어서,
상기 작은 간섭 RNA는 2' O-메틸(Methyl) 변형, 콜레스테롤(cholesterol) 접합, 및 포스포로티오에이트 결합(Phosphorothioate bond)으로 구성된 군에서 선택되는 하나 이상으로 변형된 것을 특징으로 하는, 조성물.
- 제1항에 있어서,
상기 작은 간섭 RNA는 세포전달체 없이 세포 내로 도입되는 것을 특징으로 하는, 조성물.
- 제1항의 조성물을 인체를 제외한 세포에 처리하는 단계를 포함하는, 작은 간섭 RNA(small interfering RNA)의 유전자 발현 억제 효과 증진 방법.
