Abstract
Description
도 3은 Rk1 유사체인 sac0504 및 sac0601이 VEGF에 의해 유도되는 액틴 스트레스 파이버 형성을 억제함을 보여주는 결과이다. 컨플루언트 HUVECs를 20 ng/ml VEGF (1 hr)로 처리하기 전에 상기 화합물(10 /ml)로 60분 동안 전처리하였다. 세포를 로다민 팔리오딘으로 염색하였다.
도 4a-4b 및 도 5는 Sac0601 및 Sac0504가 혈청결여-유도 세포사멸로부터 망막 내피세포를 보호함을 보여주는 실험 결과이다. HRECs(3 x 104cells/well)를 20% 우태아혈청을 포함하는 M199 배지가 있는 24-웰 플레이트에 플레이팅 하였다. 다음 날, 0.1-10 /ml Sac0601(도 4a) 또는 0.1-10 /ml Sac050(도 4b)가 포함되어 있는 배지로 옮겼다. 48시간 후 세포 생존도를 MTT 분석으로 결정하였다. 도 5는 세포 형태를 관찰한 결과이다.
도 6a-6b는 Sac0601 및 Sac0504가 혈청결여-유도 세포사멸로부터 망막 내피세포를 보호함을 보여주는 실험 결과이다. HRECs(3 x 104cells/well)를 20% 우태아혈청을 포함하는 M199 배지가 있는 24-웰 플레이트에 플레이팅 하였다. 다음 날, 0.1-10 /ml Sac0601(도 6a) 또는 0.1-10 /ml Sac050(도 6b)가 포함되어 있는 배지로 옮겼다. 48시간 후 DNA 단편화를 TUNEL 분석으로 관찰 하였다.
도 7a-7b는 Rk1 유사체인 sac0504 및 sac0601이 VEGF에 의해 유도되는 액틴 스트레스 파이버 형성을 억제하고 외피 액틴 링 구조의 형성을 유도하는 것을 보여주는 결과이다. 컨플루언트 HUVECs를 20 ng/ml VEGF (1 hr)로 처리하기 전에 상기 화합물(10 /ml)로 60분 동안 전처리 하였다. 세포를 로다민 팔리오딘으로 염색하였다.
도 8a는 컨플루언트 HRECs를 20 ng/ml VEGF (1 hr)로 처리하기 전에 sac0601(도 8a의 패널 A) 및 sac0504(도 8a의 패널 B)로 60분 동안 전처리하였다. 이어, 세포를 오클루딘 항체로 염색하였다.
도 8b는 컨플루언트 HRECs를 20 ng/ml VEGF (1 hr)로 처리하기 전에 sac0601(10 ug/ml)로 60분 동안 전처리 하였다. 세포 파쇄물에 대하여 항-오클루딘 항체로 웨스턴 블롯팅을 하였다.
도 9a-9b는 컨플루언트 HRECs를 sac0504 및 sac0601로 처리하기 전에 20 ng/ml VEGF (1 hr)로 자극시켰다. 세포를 항-오클루딘 항체(도 9a) 및 로다민 팔리오딘(도 9b)으로 염색하였다.
도 10a-10b는 Sac0601(도 10a) 및 Sac0504(도 10b)가 VEGF-유도 망막 내피세포 투과성을 억제함을 보여주는 결과이다. HRECs를 트랜스웰 필터에 플레이팅 하였다. 컨플루언시에 도달 후, 20 ng/ml VEGF (1 hr)로 처리하기 전에 sac0601(도 10a) 및 sac0504(도 10b)로 60분 동안 전처리하였다. HREC 투과도는 트랜스웰의 하부에 확산되는 [14C]sucrose(1/)의 방사성 양을 액상씬틸레이션 카운터로 측정하여 결정하였다.
도 11a-11b는 당뇨 마우스 모델에서 망막의 혈관누출을 sac0601이 완벽하게 억제하며 sac0504는 부분적으로 억제함을 보여주는 결과이다. 당뇨병 마우스에 10 sac0601 또는 10 sac0504를 한쪽 눈의 유리체에 주입하였다. 주입 24시간 후, 100 FITC-Dextran (30 mg/ml in filtered DW)를 좌심실에 주입하였다. 망막을 형광현미경(도 11a)으로 관찰하고, 형광 세기를 정량화 하였다(도 11b).
도 12a-12b는 마우스에서 VEGF-유도 혈관 누출을 sac0601이 크게 감소시키는 것을 보여주는 결과이다. 01은 sac0601을 나타낸다.
Claims (16)
- 진세노사이드 Rk1 또는 Rg3 유사체로서 다음 화학식 1로 표시되는 화합물:

상기 화학식에서, X는 산소이고; R1은 수소; 헤테로원자로서 산소를 포함하는 비치환된 C3-8 헤테로사이클로알킬; C2-8 알킬카르복실, C3-8 알킬카르복실알킬 또는 이들의 조합에 의하여 치환된 헤테로원자로서 산소를 포함하는 C3-8 헤테로사이클로알케닐; 또는 C1-5 알콕시, C2-6 알콕시알킬 또는 이들의 조합에 의하여 치환된 헤테로원자로서 산소를 포함하는 C3-8 헤테로사이클로알케닐이고; R21은 C4-8 알킬, C4-10 카르복시알킬, 또는 C4-10 알킬카르복시알킬이고; R22는 수소, 또는 히드록시이고; R23은 C1-5 알킬, 또는 R21 및 R22와 함께 결합하는 탄소에 대하여 이중결합을 형성하고; R21은 R22 및 R23과 함께 결합하는 탄소에 대하여 이중결합을 형성할 수 있으며; R21 또는 R23이 상기 탄소에 이중결합을 형성하는 경우 R22는 원자를 포함하지 않으며; R3 및 R4는 메틸이고; 는 이중결합을 나타낸다.
- 제 1 항에 있어서, 상기 진세노사이드 Rk1 또는 Rg3 유사체는 다음 화학식 2로 표시되는 것을 특징으로 하는 화합물:

상기 화학식에서, X는 산소이고; R1은 수소; 헤테로원자로서 산소를 포함하는 비치환된 C3-8 헤테로사이클로알킬; C2-8 알킬카르복실, C3-8 알킬카르복실알킬 또는 이들의 조합에 의하여 치환된 헤테로원자로서 산소를 포함하는 C3-8 헤테로사이클로알케닐; 또는 C1-5 알콕시, C2-6 알콕시알킬 또는 이들의 조합에 의하여 치환된 헤테로원자로서 산소를 포함하는 C3-8 헤테로사이클로알케닐이고; R21은 C4-8 알킬, C4-10 카르복시알킬, 또는 C4-10 알킬카르복시알킬이고; R22는 수소, 또는 히드록시이고; R23은 C1-5 알킬, 또는 R21 및 R22와 함께 결합하는 탄소에 대하여 이중결합을 형성하고; R21은 R22 및 R23과 함께 결합하는 탄소에 대하여 이중결합을 형성할 수 있으며; R21 또는 R23이 상기 탄소에 이중결합을 형성하는 경우 R22는 원자를 포함하지 않으며; R3 및 R4는 메틸이고; 는 이중결합을 나타낸다.

- (a) 제 1 항 또는 제 2 항의 진세노사이드 Rk1 또는 Rg3 유사체의 약제학적 유효량; 및 (b) 약제학적으로 허용되는 염을 포함하는 혈관누출(vascular leakage) 질환 예방 또는 치료용 약제학적 조성물.
- 제 13 항에 있어서, 상기 혈관누출 질환은 당뇨병, 염증, 망막증, 당뇨병성 망막증, 황반변성, 녹내장, 협착, 재협착, 동맥경화증, 아테롬성 동맥경화증, 뇌부종, 관절염, 관절병증(arthropathy), 포도막염, 염증성 장질환, 황반부종, 암, 고지혈증, 허혈성질환, 당뇨병성 족부궤양, 폐성고혈압, 급성 폐손상, 심근허혈, 심부전, 급성하지허혈, 심근경색, 뇌졸중, 허혈 또는 재관류 손상, VLS(vascular leakage syndrome), 부종, 이식거부, 화상, 급성 또는 성인 호흡곤란증후군(ARDS), 패혈증 또는 자가면역질환인 것을 특징으로 하는 약제학적 조성물.
- 제 1 항 또는 제 2 항의 진세노사이드 Rk1 또는 Rg3 유사체를 유효성분으로 포함하는 혈관누출(vascular leakage) 질환 예방 또는 개선용 식품 조성물.
- 제 15 항에 있어서, 상기 혈관누출 질환은 당뇨병, 염증, 망막증, 당뇨병성 망막증, 황반변성, 녹내장, 협착, 재협착, 동맥경화증, 아테롬성 동맥경화증, 뇌부종, 관절염, 관절병증(arthropathy), 포도막염, 염증성 장질환, 황반부종, 암, 고지혈증, 허혈성질환, 당뇨병성 족부궤양, 폐성고혈압, 급성 폐손상, 심근허혈, 심부전, 급성하지허혈, 심근경색, 뇌졸중, 허혈 또는 재관류 손상, VLS(vascular leakage syndrome), 부종, 이식거부, 화상, 급성 또는 성인 호흡곤란증후군(ARDS), 패혈증 또는 자가면역질환인 것을 특징으로 하는 식품 조성물.
