Abstract
본 발명은 대두 추출물을 유효성분으로 포함하는 백반증 또는 백모의 예방, 치료 또는 개선용 조성물에 관한 것이다. 본 발명의 대두 추출물은 세포내 멜라닌 색소 합성의 핵심 효소인 티로시나아제의 활성을 증가시킴과 동시에 티로시나아제, TRP-1, 및 TRP-2의 mRNA 발현을 촉진함으로써, 멜라닌 합성을 증진시킨다. 또한, 본 발명의 대두 추출물은 천연소재로서 세포에 대한 독성도 거의 없어, 피부와 모발의 색소저침작증인 백반증과 백모의 치료제로 개발될 가능성이 매우 크다.
Description
본 발명은 대두 추출물을 유효성분으로 포함하는 백반증 또는 백모의 치료, 예방 또는 개선 용도의 조성물에 관한 것이다.
멜라닌은 진피의 기저층에 분포된 멜라닌세포에 의해 분비된다(Kim and Uyama 2005). 신경능에서 분화된 멜라닌세포에 의해 멜라닌이 생합성 되면 각질형성세포를 통해 표피로 이동한다(Yaar et al. 2006). 멜라닌세포는 멜라닌의 생산을 조절하고 여러 효소를 포함하는 멜라노좀이라고 하는 리소좀과 같은 특수화된 소기관을 가지고 있다(Tiedtke et al. 2004). 멜라닌은 자외선 흡수에 의한 자외선의 해로운 효과로부터 피부를 보호하고 활성산소종을 제거하며 독성 약물과 화학 물질을 소거시키는데 있어서 중요한 역할을 한다(Yaar et al. 2006). 멜라닌 합성은 티로시나아제(tyrosinase), TRP1(tyrosinase-related protein 1), TRP2/DCT(tyrosinase-related protein 2)를 포함하는 단백질의 티로시나아제(tyrosinase) 부류의 참여로 일어난다(Huang et al. 2008). 자외선 조사에 의해 노출된 피부에서 멜라닌합성은 티로시나아제 효소를 통해 시작된다(Parvez et al. 2006). 멜라닌 세포에서 멜라닌의 합성을 촉매 하는데 중요한 효소로 알려진 티로시나아제(Sturm et al. 2000)는 티로신(tyrosine)을 도파(dopa)로, 도파(dopa)를 도파퀴논(dopaquinone)으로 산화시키는 반응에 있어서 중요한 촉매 작용을 한다(Tripath et al. 1992). 도파퀴논(Dopaquinone)은 자연적으로 도파크롬(dopachrome)으로 변화된다. TRP2(DCT)는 도파크롬(dopachrome)을 DHICA로 변환시키는 촉매 작용을 하고, TRP1는 DHICA의 산화로 인돌-5,6-퀴논-2-카르복시산(indole-5,6-quinone-2-carboxylic acid)로의 촉매 작용을 한다(Kim and Uyama 2005).
백반증을 가진 많은 사람에 있어서 하얀 반점은 삶의 질에 있어서 큰 영향을 미친다(Ongenae et al. 2006). 처음에는 백반증이 작은 장애로 보일지라도, 특히 정도가 심한 색소침착을 가진 사람에게는 미용적으로 인한 이성 교제의 어려움(Papadopoulos et al. 1999)과 심리학적으로 인한 자아 존중감 및 사회 활동에 영향을 줄 수 있다(Kent et al. 1996). 따라서, 미백제와 착색제로 인한 피부 색소침착의 변화는 약리학 뿐만 아니라 약용 화장품에 많은 관심을 불러 일으켰다(Michaela and Vincent 2008).
백반증은 피부 표피로부터의 멜라닌과 기능적인 멜라닌세포의 손실의 결과로 국한된 탈색소 반점을 특징으로 하는 색소결핍증이다(Helen et al. 2007). 표피에서 멜라닌 세포의 기능부전을 설명하기 위해서 자가면역 기전, 자가-세포독성 기전 또는 각질형성세포 주위에 있는 비정상적인 멜라닌세포가 멜라닌세포 기능 감소를 초래한다고 제시한다는 가설들이 제시되고 있다(Ongenae 2003; Moretti 2002). 백반증의 다른 원인으로는 스트레스, 감염, 유전 인자, 멜라토닌 수용체, 그리고 손상된 멜라닌세포 이주와 증식이 있다(Helen et al. 2007).
멜라닌합성의 강한 자극제인 IBMX(3-isobutyl-1-methylxanthine)(David 2001)는 cAMP의 포스포디에스테라제를 억제하여 세포내의 cAMP 함량을 증가시키고(Im et al. 1998), dibutyryl cAMP는 티로시나아제 활성과 mRNA 발현을 증가시킨다(Hoganson et al 1989). MITF는 멜라닌세포 발전과 멜라닌합성의 주요 조절자(Levy et al. 2006)이고, 주 색소침착 효소인 티로시나아제, TRP-1과 TRP-2의 전사를 조절한다(Koo et al. 2008).
현재, 백반증의 치료에 있어서 국부의 코르티코스테로이드, 칼시뉴린 억제제, 비타민 D 파생물, 광선치료(UVA, 단파장 UVB, 광화학치료), 수술, 국부적인 치료와 광선치료의 조합을 포함한 다양한 치료들이 과잉되고 있으며(Maxin et al. 2008), 광화학 치료를 꺼리는 의사와 환자에게 비흑색종 피부암과 흑색종의 발생정도를 증가시키는 부작용을 가지고 있다(Rajatanavin et al. 2003). 따라서, 새로운 피부 관리 약품의 발전에 있어서 천연 물질을 찾는데 노력을 하고 있으며, 피부 관리 화장품에 있어 천연 물질 사용은 최근에 더욱 강조되고 있다(Kiken et al. 2002).
백모(Canities, Hair graying)는 멜라닌 생합성의 세포 독성적 산화특성 및 멜라노사이트가 모근 상부의 외부에 위치한 저장소로부터 진피 돌기(dermal papilla)에 근접한 색소 방출 미세환경으로 이동하는 것이 원활하지 않은 것에 기인하는 모발구근(hair bulbar) 멜라노사이트의 티로시나아제 활성이 감소되는 것이 원인이다 (Neste and Tobin 2004; Tobin et al. 2001). 또한, 백모는 인간 노화의 전형적인 타입의 하나이고, 모발 색소화의 유지는 멜라노사이트가 지속적으로 존재하고 그 기능을 잃지 않아야 하는 것에 달려있다(Kerscher et al. 2007; Lin and Fisher 2007; Sarin et al. 2007). 멜라노사이트 줄기세포(Melanocyte stem cells, MSCs)이 모낭에서 발견되었다. 상피 멜라노사이트와는 다르게, 매우 고정적이어서, 모낭 멜라노사이트는 각 모발 사이클의 초기에 형성되어 모발 사이클의 끝에 아폽토시스에 의해 사멸한다(Robinson and Fisher 2009). 이러한 사이클이 45년 이상에 걸쳐 7-15회 반복되면, 모낭은 더 이상 멜라닌 색소를 만들 수 없게 된다(Veis et al. 1993). 모발은 일생동안 일정한 사이클링/재색소화 과정을 진행하기 때문에, 인간과 마우스와 같은 동물에서 노화와 더불어 모발의 색이 점차 소실되는 것은 백모가 MSCs의 자기 유지능이 손상된 것에 기인한 것일 수 있다는 가능성을 암시해 준다(Richard and Leonard 2008).
대두(Glycine maxim)는 콩과(Leguminosae)에 속하는 일년생 초본식물로서, 대단히 다양하고 많은 영양성분을 갖는 작물이다. 대두는 단백질, 지방, 탄수화물과 같은 영양물질 이외에도 생물학적 활성에 관심이 집중되고 있는 이소플라본, 사포닌, 식이섬유, 올리고당, 레시틴 등 다양한 생리활성물질들을 다량 함유하고 있어, 항산화, 항돌연변이, 골다공증 및 고지혈증 예방, 비만 방지, 혈당 및 콜레스테롤 조절 등의 효능, 효과가 우수하여 각종 성인병 및 항암 예방 효과가 있음이 많은 연구자들에 의해 증명되고 있다. 그 중에서도 이소플라본은 여성 호르몬인 에스트로겐과 같은 유사한 특성을 지니고 있어 폐경기 여성들의 각종 질환에 관여하여 폐경기 이후에 나타나는 각종 증후군을 예방하고 완화하는 효과가 있는 것으로도 알려져 있다.
본 명세서 전체에 걸쳐 다수의 논문 및 특허문헌이 참조되고 그 인용이 표시되어 있다. 인용된 논문 및 특허문헌의 개시 내용은 그 전체로서 본 명세서에 참조로 삽입되어 본 발명이 속하는 기술 분야의 수준 및 본 발명의 내용이 보다 명확하게 설명된다.
본 발명자들은 백반증(vitiligo), 백모(Canities)와 같은 색소저침착 질환의 치료 물질을 천연물로부터 개발하기 위해 예의 연구 노력하였다. 그 결과, 대두(Glycine maxim)으로부터 얻은 추출물이 멜라닌 합성의 핵심 효소인 티로시나아제(tyrosinase)의 활성을 증가시키고, 티로시나아제, TRT-1 및 TRP-2의 mRNA 발현을 촉진함으로써, 최종적으로 멜라닌 합성을 증진시킨다는 것을 실험적으로 확인하여 본 발명을 완성하였다.
따라서, 본 발명의 목적은 대두 추출물을 유효성분으로 포함하는 백반증 또는 백모의 치료, 예방 또는 개선 용도의 조성물을 제공하는 것에 있다.
본 발명의 목적 및 장점은 하기의 발명의 상세한 설명, 청구의 범위 및 도면에 의해 보다 명확하게 된다.
본 발명의 일 양태에 따르면, 본 발명은 대두(Glycine maxim) 추출물을 유효성분으로 포함하는 백반증(vitiligo) 또는 백모(canities) 개선용 화장품학적 조성물을 제공한다.
본 발명의 다른 일 양태에 따르면, 본 발명은 대두(Glycine maxim) 추출물을 유효성분으로 포함하는 백반증(vitiligo) 또는 백모(canities)의 치료 또는 예방용 약제학적 조성물을 제공한다.
본 발명에서 대두 추출물은 천연물로부터 추출물을 추출하는 당업계에 공지된 통상적인 방법에 따라, 즉, 통상적인 온도, 압력의 조건 하에서 통상적인 용매를 사용하여 분리할 수 있다. 본 발명에서 대두 추출물은 대두(Glycine maxim)의 잎, 줄기, 열매, 뿌리 등 모든 부분으로부터 얻은 추출물을 의미하며, 추출 부위가 특정 부분으로 제한되지 않는다.
본 발명의 대두 추출물을 추출하기 위한 추출 용매로는 추출공정에서 일반적으로 사용할 수 있는 용매를 사용할 수 있으며, 둘 이상의 서로 다른 용매를 순차적으로 사용하여 추출할 수도 있다. 바람직하게는, 본 발명의 추출용매는 물, 탄소수 1-4개의 무수 또는 함수 저급 알코올(메탄올, 에탄올, 프로판올, 부탄올), 아세톤, 에틸아세테이트, 부틸아세테이트, 디클로로메탄(CH2Cl2), 클로로포름, 헥산(Hexane) 및 1,3-부틸렌 글리콜로 구성된 군으로부터 선택되는 용매를 사용할 수 있으며, 보다 바람직하게는, 메탄올, n-헥산, 디클로로메탄, 에틸아세테이트, 또는 물이며, 가장 바람직하게는 메탄올이다.
본 발명의 바람직한 일 구현예에 의하면, 본 발명에서 상기 대두 추출물은 총 조성물 중량에 대해 0.001 - 30중량％의 농도로 포함되어 있다.
본 발명의 개선, 치료 또는 예방의 대상인 "백반증(vitiligo)"은 피부 표피로부터의 멜라닌과 기능적인 멜라닌세포의 손실의 결과로 국한된 탈색소 반점을 특징으로 하는 색소 결핍증이다. 또한, "백모(canities, Hair graying)"는 "모발의 회색화 또는 백발화"라고도 하는 증상으로서, 개개 모발의 색조가 점차 약해져, 정상 색조로부터 백색 모발 까지 다양한 색조의 털이 혼재하는 증상을 의미한다. 상기 백모는 멜라닌 생합성 세포인 멜라노사이트(melanocyte)의 독성적 산화 손상에 의해 모발구근(hair bulbar) 멜라노사이트에서의 티로시나아제 활성이 감소되는 것이 원인인 것으로 알려져 있다.
하기 일 실시예에서 입증되는 바와 같이, 본 발명 조성물의 유효성분인 대두 추출물은 세포내에서 멜라닌 색소 합성에 핵심 효소인 티로시나아제 활성을 크게 증가시킴으로써 멜라닌의 형성을 촉진한다. 따라서, 본 발명의 대두 추출물은 백반증과 백모의 치료에 유용하게 사용될 수 있다.
본 발명은 백반증 또는 백모 개선 용도의 화장품학적 조성물로 제공될 수 있다.
본 발명의 화장품학적 조성물은 당업계에서 통상적으로 제조되는 어떠한 제형으로도 제조될 수 있으며, 예를 들어, 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클린싱, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션 및 스프레이 등으로 제형화될 수 있으나, 이에 한정되는 것은 아니다. 보다 상세하게는, 유연 화장수, 영양 화장수, 로션, 영양 크림, 마사지 크림, 에센스, 아이 크림, 클렌징 크림, 클렌징 포옴, 클렌징 워터, 팩, 스프레이 또는 파우더의 제형으로 제조될 수 있다.
본 발명의 제형이 페이스트, 크림, 로션, 또는 겔인 경우에는 담체 성분으로서 동물성유, 식물성유, 왁스, 파라핀, 전분, 트라칸트, 셀룰로오스 유도체, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다.
본 발명의 제형이 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토스, 탈크, 실리카, 알루미늄 히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 이용될 수 있고, 특히 스프레이인 경우에는 추가적으로 클로로플루오로히드로카본, 프로판/부탄 또는 디메틸 에테르와 같은 추진체를 포함할 수 있다.
본 발명의 제형이 용액 또는 유탁액인 경우에는 담체 성분으로서 용매, 용해화제 또는 유탁화제가 이용되고, 예컨대 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌 글리콜, 1,3-부틸글리콜 오일, 글리세롤 지방족 에스테르, 폴리에틸렌 글리콜 또는 소르비탄의 지방산 에스테르가 있다.
본 발명의 제형이 현탁액인 경우에는 담체 성분으로서 물, 에탄올 또는 프로필렌 글리콜과 같은 액상의 희석제, 에톡실화 이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있다.
본 발명의 제형이 계면-활성제 함유 클린징인 경우에는 담체 성분으로서 지방족 알코올 설페이트, 지방족 알코올 에테르 설페이트, 설포숙신산 모노에스테르, 이세티오네이트, 이미다졸리늄 유도체, 메틸타우레이트, 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄올아미드, 식물성 유, 라놀린 유도체 또는 에톡실화 글리세롤 지방산 에스테르 등이 이용될 수 있다.
본 발명의 화장품학적 조성물에 포함되는 성분은 유효 성분과 담체 성분 이외에, 화장품학적 조성물에 통상적으로 이용되는 성분들을 포함하며, 예컨대 항산화제, 안정화제, 용해화제, 비타민, 안료 및 향료와 같은 통상적인 보조제를 포함할 수 있다.
본 발명의 다른 일 양태에 따르면, 본 발명은 대두 추출물을 유효성분으로 포함하는 백반증 또는 백모 치료 또는 예방용 약제학적 조성물 형태로 제공될 수 있다.
본 발명의 약제학적 조성물은 유효성분으로서 상술한 대두 추출물 이외에 약제학적으로 허용되는 담체를 포함할 수 있으며, 이러한 담체는 제제시에 통상적으로 이용되는 것으로서, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 약제학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다. 적합한 약제학적으로 허용되는 담체 및 제제는 Remington's Pharmaceutical Sciences (19th ed., 1995)에 상세히 기재되어 있다.
본 발명의 약제학적 조성물의 적합한 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있다. 한편, 본 발명의 약제학적 조성물의 경구 투여량은 바람직하게는 1일 당 0.0001-100 mg/kg(체중)이다.
본 발명의 약제학적 조성물은 경구 또는 비경구로 투여할 수 있고, 비경구로 투여되는 경우, 피부에 국소적으로 도포, 정맥내 주입, 피하 주입, 근육 주입, 복강 주입, 경피 투여 등으로 투여할 수 있다.
본 발명의 약제학적 조성물이 멜라닌 색소의 결손으로 인한 백반증 또는 백모의 치료 또는 예방을 위해 적용되는 점을 감안하면, 본 발명 조성물의 투여는 피부에 국소적으로 도포되어 이루어지는 것이 바람직하다.
본 발명의 조성물에 포함되는 유효성분의 농도는 치료 목적, 환자의 상태, 필요기간 등을 고려하여 결정할 수 있으며 특정 범위의 농도로 한정되지 않는다.
본 발명의 약제학적 조성물은 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화 함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질중의 용액, 현탁액 또는 유화액 형태이거나 엑스제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 또는 안정화제를 추가적으로 포함할 수 있다.
본 발명의 바람직한 구현예에 의하면, 본 발명의 약제학적 조성물은 피부외용 제형을 갖는다. 피부외용 제형은 특별히 한정되지 않으며 바람직하게는 파우더, 젤, 연고, 크림, 로션, 액제 또는 에어로졸 제형이다.
상술한 바와 같이, 본 발명은 대두 추출물을 유효성분으로 포함하는 백반증 또는 백모의 예방, 치료 또는 개선용 조성물에 관한 것이다. 본 발명의 대두 추출물은 세포내 멜라닌 색소 합성의 핵심 효소인 티로시나아제의 활성을 증가시킴과 동시에 티로시나아제, TRP-1, 및 TRP-2의 mRNA 발현을 촉진함으로써, 멜라닌 합성을 증진시킨다. 또한, 본 발명의 대두 추출물은 천연소재로서 세포에 대한 독성도 거의 없어, 피부와 모발의 색소저침작증인 백반증과 백모의 치료제로 개발될 가능성이 매우 크다.

이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예
실험방법 및 재료
1. 시약 및 기기
L-DOPA(3,4-dihydroxy-L-phenyl-alanine), DMSO(dimethyl sulfoxide), BHT (2, 6-di-tert-butylated hydroxytoluene), β-액틴, Glyceraldehyde-3-phosphate dehydrogenase, DPPH(1,1-diphenyl-2-icryl hydrazyl), 탄닌산(tannic acid), L-티로신(L-tyrosine), 아스코르브산(ascorbic acid), Folin-ciocalteu's phenol reagent, MTT[3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyl-tetrazolium bromide], IBMX, MITF-M, 및 디(에틸렌글리콜) 시약들은 Sigma Chemical Company(USA)으로부터 구입하였다. 프로필렌글리콜(Propylene glycol)은 OCI (Korea)으로부터 구입하였다. 도림현미경(Inverted microscope)(CKX41, Olympus, Japan)은 세포주 관찰에 사용하였다.
2. 식물재료
대두(Glycine maxim Leguminosae)의 잎, 콩, 줄기 및 뿌리의 메탄올 추출물은 한국식물추출물 은행(Korea Plant Extract Bank) (Daejeon, Korea)으로부터 구입하였고 DMSO에 녹여 사용하였다.
3. 항산화능 실험
3-1. 총 폴리페놀 함량
총 폴리페놀 함량은 Folin-Denis법(Folin and Denis 1912)에 의해 비색 정량하였다. 적절한 배율로 희석한 시료 1 ㎖에 folin-시약 1 ㎖를 가하여 3분간 정치한 후 10％ Na2CO3 1 ㎖를 혼합하고, 1 시간 실온에서 방치하여 760 ㎚에서 흡광도를 측정하였다. 검량곡선은 탄닌산(tannic acid)를 이용하여 작성하였다.
3-2. 총 플라보노이드 함량
총 플라보노이드 함량은 Davis 변법(AOAC 1995)을 이용하였다. 적절한 배율로 희석한 시료 용액 1 ㎖에 디(에틸렌글리콜)[di(ethylene glycol)] 시약 10 ㎖ 및 1N NaOH 1 ㎖를 가하고 잘 혼합한 다음 37℃ 수욕상에서 1시간 반응시킨 후 420㎚에서 흡광도를 측정하였다. 검량곡선은 루틴(rutin)을 이용하여 작성하였다.
3-3. 전자공여능
전자공여능은 Blois의 방법(Blois 1958)으로 측정하였다. 동결 건조시킨 대두 메탄올 추출물의 분말을 100, 500, 1,000 ㎍/㎖의 농도로 DMSO에 녹여 제조하고, 1 ㎖를 테스트 튜브(test tube)에 취하여, 4X10-4M의 DPPH 용액 4 ㎖를 가하여 60℃ 수욕상에서 10초간 진탕하고, 실온에서 20분 동안 방치한 후에 525㎚에서 흡광도를 측정하였다. 추출물 무 첨가군에는 시료 대신 메탄올 1 ㎖를 첨가하여 동일하게 실험하고 추출물 첨가군에 대한 흡광도의 감소비율로 전자공여능을 나타내었다. 양성대조군으로 합성 항산화제인 BHT를 동일한 방법으로 실험하였으며, 다음의 식에 의해 전자공여능(％)을 구하였다.

4. 세포 실험
4-1. 세포 배양
C57BL/6 마우스에서 유래한 불사멸화된 세포주(immortalized cell line)인 Melan-a 세포를 10％ FBS(fetal bovine serum)와 1％ PS(penicillin/streptomycin), 200nM TPA(12-O-tetradecanoylphorbol-13-acetate)가 함유된 RPMI-1640 배지를 사용하여 37℃, 10％ CO2 조건의 인큐베이터에서 배양하였다.
4-2. MTT 분석
MTT 분석법은 MTT[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide] 시약이 세포내로 흡수된 후 미토콘드리아의 석시네이트 디하이드로게나아제(succinate dehydronase)에 의해 포르마잔(formazan)을 형성하는데 이 물질의 세포내 축적은 미토콘드리아의 활성, 넓게는 세포의 활성을 의미하는 것으로써 세포의 생장율을 측정하는 대표적인 방법이다. Melan-a 세포를 10％ FBS, 1％ P/S, 200 nM TPA가 함유된 RPMI-1640 배지에 풀어 37℃, 10％ CO2 인큐베이터에서 48 시간 동안 안정화시킨 후에 실험에 사용하였다. Melan-a 세포를 96 웰 플레이트에 적정 세포수(0.5 X 104 세포/웰)로 분주하고, 37℃, 10％ CO2 인큐베이터에서 24 시간 배양한 다음 대두 메탄올 추출물을 RPMI-1640 배지에 농도별(6.25, 12.5, 25, 50 ㎍/㎖)로 희석시켜, 각 웰에 분주한 후, 37℃, 10％ CO2 인큐베이터에서 48 시간 배양하였다. 이어서, MTT가 0.5 ㎎/㎖ 함유된 배지를 넣은 후 37℃, 10％ CO2 인큐베이터에서 3 시간 배양하였다. 플레이트를 1,000 rpm에서 10분간 원심분리하여 세포가 바닥에 가라앉게 한 다음 배지를 버리고, DMSO를 200㎕씩 넣고 플레이트 진탕기(plate shaker)에서 15분간 세포를 녹여낸 다음 ELISA reader로 540㎚ 파장에서 흡광도를 측정하였다. 세포생존율은 다음 식에 의해 산출하였다.

4-3. 세포형태 관찰
세포의 형태를 관찰하기 위해, 대두메탄올 추출물(GMME)을 1.563, 3.125, 6.25, 12.5 ㎍/㎖의 농도로 Melan-a 세포에 첨가하여 37℃, 10％ CO2의 인큐베이터에서 72 시간 동안 배양하였다. 세포의 형태 변화를 관찰하기 위하여 배양이 끝난 후 새 배지로 교환한 후 도립현미경으로 관찰하였다.
4-4. 멜라닌 생성능 분석
Melan-a 세포를 96-웰 플레이트에 적정세포수(2 X 104 세포/웰)로 분주하고 37℃, 10％ CO2 인큐베이터에서 24시간 배양한 다음, RPMI-1640 배지로 농도별(1.563, 3.125, 6.25, 12.5㎍/㎖)로 희석시킨 대두 메탄올 추출물 200 ㎕를 웰에 넣고, 37℃, 10％ CO2 인큐베이터에서 72시간 배양하였다. 이어서 세정하고 동일한 처리를 1회 반복하였다. 다음, 멜라닌을 1N NaOH 용액으로 용해시켜 490 nm에서 ELISA reader를 사용하여 흡광도를 측정하였다. 멜라닌 정량은 다음 식에 의해 산출하였다.

4-6. 세포 추출된 티로시나아제 활성능

4-7. 웨스턴 블로팅 분석
세포 용해물은 Melan-a 세포를 1％ Nonidet P-40, 0.01％ SDS 및 프로테아제 억제자 칵테일(Roche, Mannheim, Germany)을 포함하는 0.1 M Tris-HCl(pH 7.2) 완충액내에서 초음파분쇄(sonication)하여 준비하였다. 세포용해물의 단백질 농도는 Pierce Protein Assay Kit (Pierce Biotechnology, Inc., Rockford, USA)를 사용하여 BSA를 표준물질로 하여 측정하였다. 동일한 양의 단백질(10 ㎍)을 각각의 레인(lane)에 로딩하고 10％ 폴리아크릴아마이드젤상에서 전기영동하여 분리하였다. 니트로셀룰로오스막상에 트랜스블로팅한 후, 막을 PEP7 (1:10000 희석, 항-티로시나아제 항체), PEP1 (1:10000 희석, 항-TRP1 항체) 및 PEP8 (1:10000 희석, 항-TRP2 항체)와 인큐베이션하였다. 다음에, 막을 호오스래디쉬 퍼옥시다아제 컨쥬게이트된 항-래빗 IgG (1:1000 희석; Amersham, Bucks, UK)와 인큐베이션하였다. 면역 활성 밴드는 ECL 웨스턴 블로팅 검출기(Amersham, Bucks, UK)을 사용하여 화학발광에 의해 검출하였다. 밴드 강도는 ImageJ program (NIH, Bethesda, USA)을 사용하여 측정하였다. β-액틴은 면역블로팅에 대한 내부 대조군으로 사용하였다.
4-8. RT-PCR
총 RNA는 제조사에서 제공된 지시에 따라 Trizol-Reagent (Invitrogen, Caylsbad, CA)를 이용해 분리하였다. 총 RNA 5㎍을 40 ㎕ 용량에서 M-MLV RT 5X 배지의 8 ㎕, 10 mM dNTPs의 3 ㎕, 10,000 U RNase 억제자 0.45 ㎕, 50,000 U M-MLV 역전사효소(Promega, Madison, USA) 0.3 ㎕ 및 50 pmol/㎕ 올리고 dT (Bioneer, Chungbuk, Korea) 1.5 ㎕를 이용해 역전사 하였다. Single stranded cDNA는 5 X green Go Taq flexi buffer 4㎕, 10 mM dNTPs 0.4 ㎕, 500 U Taq polymerase 0.1 ㎕, 25 mM MgCl2 (Promega, Madison, USA) 1.2 ㎕와 티로시나아제, TRP-1, TRP-2, MITF-M, β-액틴의 특이 센스 및 안티 센스 프라이머 각각 20 pmol/L 0.4 ㎕를 사용하여 PCR로 증폭하였다.

티로시나아제, TRP-1, TRP-2, MITF-M 및 β-액틴에 대한 PCR 산물의 예상 사이즈는 1192, 268, 1044, 326 및 528 bp이다.
각 PCR 조건은 다음과 같았다: 티로시나아제 및 TRP-1은 94℃에서 60초간 변성, 56℃에서 60초간 어닐링 및 72℃에서 60초간 연장의 28 싸이클; TRP-2은 94℃에서 60초간 변성, 64℃에서 60초간 어닐링 및 72℃에서 60초간 연장의 28 싸이클; MITE-M은 94℃에서 30초간 변성, 54℃에서 30초간 어닐링 및 72℃에서 30초간 연장의 30 싸이클; β-액틴은 94℃에서 30초간 변성, 51℃에서 30초간 어닐링 및 72℃에서 60초간 연장의 30 싸이클. PCR 산물은 1.2％ 아가로즈젤상에서 분석하였다. β-액틴은 티로시나아제, TRP-1 및 TRP-2 및 MITE-M의 상대 발현수준과 비교 평가하기 위해 내부 대조군으로 사용하였다.
5. 통계학적 분석
각 군간의 측정값들의 비교는 일원배치 분산분석(one-way ANOVA)을 이용하여 분석하였고, SPSS(version 17.0) 통계프로그램을 이용한 Duncan's multiple range test로 사후 분석을 실시하였다. 통계학적 유의성은 α=0.001, α=0.01, α=0.05인 경우로 하였다.
실험결과
1. 항산화능
1-1. 총 폴리페놀 함량
탄닌산(tannic acid)을 사용한 표준곡선에 대비하여 측정한 대두 추출물의 총 폴리페놀 함량은 54.8 ㎎/g이었다(도 1 참조).
1-2. 총 플라보노이드 함량
루틴(rutin)을 사용한 표준곡선과 대비하여 측정한 대두 추출물의 총 플라보노이드 함량은 62.7㎎/g이었다(도 1 참조).
1-3. 전자공여능
대두 추출물의 전자공여능은 100, 500, 및 1,000 ㎍/㎖의 농도에서 각각 1.2％, 6.8％, 및 14.4％이었다. BHT의 전자공여능은 동일한 농도에서 각각 16.3％, 44.1％, 및 68.4％ 이었다. BHT의 수치와 비교하면, 대두 추출물의 전자공여능은 비교적 낮았으나, 대두추출물은 만족할만한 항산화능을 갖으며, 용량의존적 방식의 반응을 보여주었다(도 2 참조).
2. 세포실험
2-1. 세포 생존율에 미치는 대두 추출물의 영향
MTT 분석을 행하여 Melan-a 세포에 대한 대두추출물의 최대 허용 농도(maximum permissible level, MPL)를 측정하였다. 대두추출물에 의해 세포 생존율은 50 ㎍/㎖의 농도에서 27.6％의 감소율을 나타내어, MPL은 25 ㎍/㎖으로 계산되었다. IBMX 처리군의 MPL은 12.5 ㎍/㎖이었다(도 3 참조).
2-2. Melan-a 세포에 대한 형태학적 관찰
대조군과 비교하여, 대두추출물로 처리한 군에서 용량의존적 방식으로, 멜라닌 합성이 증가하고, 수지상돌기(dendricity)가 발달하였다(도 4 참조).
2-3. 멜라닌 합성에 대한 대두 추출물의 효과
대조군과 비교하여, 대두 추출물 처리군의 멜라닌 함량은 1.563, 3.125, 6.25, 및 12.5 ㎍/㎖의 농도에서 각각 16.1, 31.0, 45.9, 및 51.2％ (p<0.00l) 크게 증가하였다. IBMX 12.5 ㎍/㎖ 처리군에서, 멜라닌 함량은 31.6％ 크게 증가하였다(p<0.00l)(도 5 참조).
2-4. 세포내 티로시나아제 활성에 대한 대두 추출물의 영향
대조군과 비교하여, 대두 추출물 처리군의 티로시나아제 활성은 1.563, 3.125, 6.25, 및 12.5 ㎍/㎖의 농도에서, 각각 56.6, 57.3, 75.0, 및 90.0％ 으로 유의성(p<0.00l) 있게 증가하였다. IBMX 처리군은 12.5 ㎍/㎖의 농도에서 54.6％ 증가하였다(p<0.001)(도 6 참조).
2-5. 세포 추출물의 티로시나아제 활성에 대한 대두 추출물의 영향
대조군과 비교하여, 대두 추출물 처리군의 티로시나아제 활성은 3.125, 6.25 및 12.5 ㎍/㎖의 농도에서 각각 10.0, 10.2, 및 12.5％으로 유의성 있게 증가하였다(p<0.00l). IBMX 처리군은 12.5 ㎍/㎖의 농도에서 27.2％ 증가하였다(p<0.001)(도 7 참조).
2-6. 티로시나아제 단백질 발현에 대한 대두 추출물의 영향
대조군과 비교하여, 대두 추출물 처리군의 티로시나아제 단백질 발현은 3.125, 6.25 및 12.5 ㎍/㎖의 농도에서 각각 36.1, 41.8, 및 43.6％으로 유의성 있게 증가하였다(p<0.00l). IBMX 처리군은 12.5 ㎍/㎖의 농도에서 31.7％ 증가하였다(p<0.001)(도 8 참조).
2-7. TRP-1 단백질 발현에 대한 대두 추출물의 영향
대조군과 비교하여, 대두 추출물 처리군의 TRP-1 단백질 발현은 3.125, 6.25 및 12.5 ㎍/㎖의 농도에서 각각 8.6 (p<0.05), 9.8 (p<0.05), 및 19.6％ (p<0.00l) 증가하였다. IBMX 처리군은 12.5 ㎍/㎖의 농도에서 2.2％ 증가하였다(p<0.001)(도 9 참조).
2-8. TRP-2 단백질 발현에 대한 대두 추출물의 영향
대조군과 비교하여, 대두 추출물 처리군의 TRP-2 단백질 발현은 6.25 및 12.5 ㎍/㎖의 농도에서 각각 40.7 및 82.7％ 증가하였다(도 10 참조). 반면에, 3.125 ㎍/㎖의 농도에서 27.4％ 감소하였다. IBMX 12.5 ㎍/㎖ 처리군은 25.1％ 유의성 있게 증가하였다(도 10 참조).
2-9. 티로시나아제 mRNA 발현에 대한 대두 추출물의 영향
대조군과 비교하여, 대두 추출물 처리군의 티로시나아제 mRNA 발현은 1.563, 3.125, 6.25, 및 12.5 ㎍/㎖의 농도에서 28.0, 28.6, 28.6, 및 46.8％ 유의성 있게 증가하였다. IBMX 12.5 ㎍/㎖ 처리군은 40.3％으로 유의성 있게 증가하였다(도 11 참조).
2-10. TRP-1 mRNA 발현에 대한 대두 추출물의 영향
대조군과 비교하여, 대두 추출물 처리군의 TRP-1 mRNA 발현은 3.125, 6.25, 및 12.5㎍/㎖의 농도에서 각각 9.0 (p<0.05), 11.4 (p<0.01), 및 17.0％ (p<0.001) 유의성 있게 증가하였다. IBMX 12.5 ㎍/㎖ 처리군은 14.8％으로 유의성 있게 증가하였다(도 12 참조).
2-11. TRP-2 mRNA 발현에 대한 대두 추출물의 영향
대조군과 비교하여, 대두 추출물 처리군의 TRP-2 mRNA 발현은 3.125, 6.25, 및 12.5㎍/㎖의 농도에서 14.2, 11.0, 및 14.3％ 유의성 있게 증가하였다(p<0.01). IBMX 12.5 ㎍/㎖ 처리군은 4.3％으로 증가하였다(도 13 참조).
2-12. MITF-M mRNA 발현에 대한 대두 추출물의 영향
대조군과 비교하여, IBMX 처리군의 MITF-M mRNA 발현은 12.5 ㎍/㎖의 농도에서 172.7％ 유의성 있게 증가하였다(p<0.001). 반면에, 대두 추출물 처리군은 1.563 및 3.125 ㎍/㎖의 농도에서 각각 79.5 및 59.1％ 유의성 있게 감소하였고(p<0.001), 6.25 및 12.5 ㎍/㎖의 농도에서는 대조군과 비교하여 유사한 수준을 보였다(도 14 참조).
이상으로 본 발명의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현 예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.

- 대두(Glycine maxim) 알코올 추출물을 유효성분으로 포함하는 백모(Canities)의 개선용 화장품학적 조성물.
- 제 1 항에 있어서, 상기 대두 알코올 추출물은 탄소수 1-4개의 무수 또는 함수 저급 알코올을 용매로 사용한 추출물인 것을 특징으로 하는 조성물.
- 대두(Glycine maxim) 알코올 추출물을 유효성분으로 포함하는 백모의 치료 또는 예방용 약제학적 조성물.
- 제 3 항에 있어서, 상기 대두 알코올 추출물은 탄소수 1-4개의 무수 또는 함수 저급 알코올을 용매로 사용한 추출물인 것을 특징으로 하는 조성물.
- 제 3 항에 있어서, 상기 조성물은 파우더, 젤, 연고, 크림, 로션, 액제 및 에어로졸로 이루어지는 군에서 선택되는 피부외형 제형을 갖는 것을 특징으로 하는 조성물.
