Abstract
높은 당질 농도의 영양 배지를 특정의 저점도 범위로 유지하여 연속 배양함으로써 보다 생산성 높은 공업적으로 유리한 풀룰란 고함유물과 그 제조 방법 및 용도를 확립하였다.
본 발명은 풀룰란 생성능을 가진 미생물을 당질 농도 10∼20 w/v% 함유하는 영양 배지에서 이 배지의 점도를 30 센티포이즈 이하로 유지하면서 연속 배양하여 얻어진 평균 분자량 250,000 이하의 풀룰란 고함유물과 그 제조 방법 및 이것을 함유시킨 증점제, 코우팅제, 접착제 및 성형물, 혹은 음식물, 화장품, 의약품, 농림 수축산 자재, 종이 가공 자재 및 광공업 자재 등에의 용도를 구성으로 한다.
Description
본 발명은 풀룰란(pullulan) 고함유물과 그 제조 방법 및 용도에 관한 것으로서, 더욱 상세하게는 풀룰란 생성능(生成能)을 가진 미생물을 당질(糖質) 농도 10∼20 w/v% 함유하는 영양 배지에서 이 배지의 점도를 30 센티포이즈(centipoise : 이하, 간단히 "cp"로 표기함) 이하로 유지하면서 연속 배양하여 얻어지는 평균 분자량 250,000 이하의 풀룰란 고함유물과 그 제조 방법 및 용도에 관한 것이다.
풀룰란은 풀룰란 생성능을 가진 미생물을 단당류, 올리고당류를 함유하는 영양 배지에서 호기적(好氣的)으로 배양하여 얻어지는 점성의 글루칸(glucan)으로서 공업적으로 제조되고 있다.
그 제조 방법으로서는, 예컨대 일본국 특허 공개 소 49-42894 호에 개시되어 있는 바와 같이 영양 배지의 초기 pH를 5.5∼8.0으로 조성하고, 특히 높은 pH를 조정함으로써 풀룰란의 생산성을 증대시키는 방법과, 일본국 특허 공개 소 52-130993 호에 개시되어 있는 바와 같이 풀룰란 생성능을 가진 미생물을 아밀라아제 활성 저해 물질 공존하에 영양 배지의 점도 저하를 방지하면서 배양하여 풀룰란의 생산성을 증대시키는 방법이 제안되어 있다. 그러나, 이들 방법은 어느 것이나 뱃치식(batchwise : 즉, 回分式) 배양 방법이어서 풀룰란의 공업 생산적 견지에서 반드시 만족할 수 있는 것은 아님이 판명되었다.
한편, 풀룰란 생성능을 가진 미생물을 연속 배양하여 풀룰란을 제조하는 방법이 아래의 간행물에서 개시되어 있다.

그러나, 이들 간행물도 연속 배양에 의한 풀룰란 생산에 대하여 생물 화학 공학적으로 검토가 되어 있지 않고, 글루코오스, 수크로오스, 이탄 가수 분해물(peat hydrolyzate) 등의 당질의 농도가 2∼5 w/v%로서 낮으며, 영양 배지의 점도에 대해서도 검토가 되어 있지 않다. 그리고, 영양 배지의 pH에 대해서는 Boa 등의 문헌에서는 명확히 되어 있지 않고, Bulmer 등의 문헌에서는 pH 5.5로 유지하고 있고, McNeil 등의 문헌에서는 pH 4.5가 적당한 pH라고 보고되어 있다. 배양액 1L 당 및 배양 1 시간 당의 풀룰란의 생산성 (gL-1h-1)(이하, 본 발명에서는 간단히 "풀룰란 생산성"이라 함)에 대해서는 최고가 Boa등의 문헌에 기재되어 있는 0.48로서 낮은 값이고, 그 연속 배양의 기간에 대해서도 불명이다. 따라서, 이들 보고를 이용하여 공업적 생산에 유리한 풀룰란 고함유물을 생산한다는 것은 극히 곤란하다.
위와 같은 종래 방법의 결점을 해소하여 공업적으로 유리한 신규의 연속 배양 방법에 의한 풀룰란 고함유물의 제조 방법과 그 용도의 확립이 요망되고 있다.
본 발명자등은 연속 배양에 의한 풀룰란 고함유물의 제조 방법, 특히 고농도의 당질을 함유시킨 영양 배지를 사용하여 그 배양액의 점도와 그 풀룰란 생산성에 착안하여 공업적 생산에 유리한 풀룰란 고함유물과 그 제조 방법 및 용도의 확립을 목표로 하여 예의 연구를 계속하였다.
그 결과, 종래의 기술과는 전혀 다르게 풀룰란 생성능을 가진 미생물을 당질 농도 10∼20 w/v% 함유하는 영양 배지에서 점도 30 cp 이하로 유지하면서 연속 배양하여 얻어지는 평균 분자량 250,000 이하의 풀룰란 고함유물은 연속 배양에 의한 풀룰란 생산성이 높고 그 배양 기간도 장시간에 걸쳐 안정하게 실시할 수 있으며, 더욱이 그 공업적 생산이 극히 유리하다는 것을 발견하였다. 따라서, 연속 배양에 의한 제조 방법 및 제조되는 풀룰란 고함유물을 함유시켜 증점제, 코우팅제, 접착제, 성형물 등에, 그리고 음식물, 화장품, 의약품, 농림 수산 자재, 종이 가공 자재, 광공업 자재 등 다방면에 그 용도를 확립하여 본 발명을 완성하였다.
이하 본 발명을 더욱 상세하게 설명한다.
본 발명에 사용되는 풀룰란 생성능을 가진 미생물은 풀룰란을 생성할 수 있는 균주이면 어떠한 균주라도 좋다. 예를 들자면, 오레오바시듐 퍼어멘탄스 바르퍼어멘타스(Aureobasidium fermentans var fermentans) IFO 6410, 오레오바시듐 퍼어멘타스 바르 푸스카(Aureobasidium fermentans var fusca) IFO 6402, 오레오바시듐 풀룰란스(Aureobasidium pulluans) IFO 6353 및 오레오바시듐 풀룰란스(Aureobasidium pulluans) IFO 4464 등의 오레오바시듐속(Aureobasidium 層)에 속하는 미생물 또는 이들의 돌연변이주 등을 적절히 사용할 수 있다.
영양 배지로서는 탄소원, 질소원, 유기 영양원, 무기 물질 등을 적절히 함유시킨 것이 사용된다. 탄소원으로서는 통상 예를 들자면, 글루코오스, 말토오스, 말토올리고당, 이소말토올리고당, 전분 시럽 또는 전분 가수 분해물, 수크로오스, 푸럭토오스, 당화 전분, 전화당, 이성화당, 당밀 등에서 선택되는 한가지 또는 두가지 이상의 당질을 농도 10∼20 w/v%, 보다 바람직하게는 10∼18 w/v% 의 범위에서 사용된다. 질소원으로서는 암모늄 염, 질산 염 등의 무기 질소원 또는 글루탐산 염, 펩톤, 효모엑스, 코온 스티입 리커(corn steep liquor)등의 유기 질소원 등으로 부터 선택되는 한가지 또는 두가지 이상이 사용된다. 기타 무기 물질로서는 인산 염, 마그네슘 염, 철 염 등이 적절히 사용된다.
본 발명에서 사용되는 배양은 연속 방법으로 영양 배지의 점도를 30 cp 이하로 유지하면서 호기적(好氣的)으로 실시한다. 바람직하게는 pH 4.0 이하, 보다 바람직하게는 pH 2.0 이상, 4.0 이하에서 온도 약 25∼30℃의 범위에서 선택되는 조건하에서 배양한다.
본 발명의 방법에 있어서 사용되는 영양 배지의 점도를 30 cp 이하로 유지하는 방법은, 예컨대 간헐적으로 또는 연속적으로 영양 배지 일부를 배출하면서 추가 공급하는 영양 배지의 공급 속도에 의하여 영양 배지의 희석 속도를 통상 0.01∼0.1 h-1의 범위로 조정하거나 또는 풀룰란을 부분적으로 가수 분해하여 영양 배지의 점도 저하를 일으키는 아밀라아제, 예컨대 액화형 α-아밀라아제, 이소아밀라아제 등의 전분질을 부분적으로 가수 분해하는 효소를 적당량 첨가하여 실시할 수 있다. 더욱이, 필요하다면 영양 배지로서 예비 배양한 것을 사용하는 것도 유리하게 실시할 수 있다.
이와 같이 하여 수득한 풀룰란 고함유 배양액은 통상의 방법에 따라 제균, 농축하고, 필요에 따라 탈색, 탈염하여 용액상의 풀룰란 고함유물 또는 이것을 다시 분말화 하여 분말상의 풀룰란 고함유물을 회수한다. 이와 같이 하여 제조된 풀룰란 고함유물은 풀룰란을 고형물 당 약 50 w/w% 이상, 바람직하게는 약 55∼90 w/w%의 범위에서 함유하며, 그 평균 분자량은 통상 300,000 이하, 바람직하게는 약 40,000∼250,000의 범위이다. 필요하다면 분자량 분획, 용기 용매 침전 분리 등에 의하여 이 생성물을 보다 고순도의 풀룰란 고함유물로 할 수도 있다.
본 발명의 풀룰란 고함유물은 수용성, 증점성, 조막성(造膜性), 광택성, 투명성, 가스 배리어성(gas-barrier 性), 내유성, 내염성, 접착성, 성형성, 가식성(可食性), 난소화성 등의 특성을 가지고 있으므로, 그 용도로서는 예컨대 증점제, 코우팅제, 접착제, 성형물 등에 유리하게 이용할 수 있다.
또한, 본 발명 풀룰란 고함유물은 음식물[예 : 비피도박테리움(Bifidobacterium)미생물 증식 촉진제, 다이어터리 파이버(dietary fiber) 식품, 저칼로리 식품 등], 화장품[예 : 치마제(dentifrice), 유액, 크리임, 팩, 정발료, 삼푸우, 입욕제 등], 의약품 [예 : 정제, 캡슐제, 대용 혈장 등의 의약품 등], 농림 수축산(農林 水蓄産) 자재 [예 : 코우팅 종자, 조립(造粒)]농약, 성형 비료, 성형 사이료 등], 종이 가공 자재 [예 : 호제, 사이징제], 광공업 자재 [예 : 폐수 처리제, 용접봉, 주형 등]등 다방면의 산업 분야에 광범위하게 이용할 수 있다.
본 발명의 풀룰란 고함유물을 위해 나온 각종 용도에 이용할 경우에 있어서는 목적에 따라 본 발명의 풀룰란 고함유물을 단독으로 함유시키든지 본 발명의 풀룰란 고함유물과 더불어 글리세린, 소르비톨, 말티톨, 락티톨 등의 다가 알코올을 유리하게 함유시킬 수도 있다.
필요에 따라서는 위에 나온 제품중에 본 발명의 풀룰란 고함유물과 더불어 기타의 재료, 예컨대 풀룰란 이외의 다당류, 가소제, 증량제, 부형제, 충전제, 증점제, 계면 활성제, 발포제, 소포제, pH 조정제, 안정제, 난연제, 이형제, 항균제, 착색제, 착향제, 영양물, 기호물, 증미물, 약효 물질, 생리 활성 물질등으로 부터 선택되는 한가지 또는 두가지 이상을 적절히 함유시킬 수도 있다. 본 발명의 풀룰란 고함유물을 함유시킨다는 것은 그 용도 제품의 제조가 완료하기 까지의 공정에서 풀룰란 고함유물을 함유시킬 수 있으면 좋고, 그 방법으로서는 예컨대 혼화, 반죽, 용해, 침지, 도포, 산포, 분무, 주입 등의 공지의 방법을 적절히 선택한다.
구체적으로는, 예컨대 증점제 제조의 경우에 있어서는 일본국 특허 공개 소 51-51543호, 특허 공개 소 52-41252호, 특허 공개 소 58-213076호 등에 개시된 방법을 이용하고, 코우팅제 제조의 경우에 있어서는 일본국 특허 공개 소 52-56603호, 특허 공개 소 53-19416호, 특허 공개 소 53-119741호 등에 개시된 방법을 이용하며, 접착제 제조의 경우에 있어서는 일본국 특허 공개 소 51-119321호, 특허 공개 소 53-54593호, 특허 공개 소 54-108828호 등에 개시된 방법을 이용할 수 있다. 그리고, 성형물 제조의 경우에 있어서는 일본국 특허 공개 소 50-116545호, 특허 공개 소 51-4272호, 특허 공개 소 51-100470호 등에 개시된 방법을 이용하고, 음식물 제조의 경우에 있어서는 일본국 특허 공개 소 53-79045호, 특허 공개 소 60-219238호, 특허 공개 평 2-289520호 등에 개시된 방법을 이용하며, 화장품 제조의 경우에 있어서는 일본국 특허 공개 소 50-6441호, 특허 공개 소 63-28369호 등에 개시된 방법을 이용하고, 의약품 제조의 경우에 있어서는 일본국 특허 공개 소 53-12417호, 특허 공개 소 57-67602호, 특허 공개 소 63-122739호에 개시된 방법을 이용할 수 있다.
이하 본 발명의 연속 배양에 의한 풀룰란 고함유물의 제조 방법에 대하여 아래의 실험에 따라 설명한다.
[실험 1]
[풀룰란 생산성에 미치는 당질 농도와 점도 제어의 영향]
[실험 1-A]
[영양 배지의 공급에 의한 점도 제어]
자아 퍼어멘터(jar fermentor)에 표 1에 나와 있는 영양 배지를 넣고, 통상의 방법에 따라 120℃에서 20분간 가압 멸균하고 약 27℃로 냉각한 후 미생물의 종배양액(種培養液)을 무균적으로 식균(植菌)한 다음, 염산 또는 수산화 나트륨을 첨가하여 pH 3.6∼3.8로 유지하면서 통기 교반함으로써 배양하였다.
종배양액은 동일한 조성의 영양 배지의 균주인 오레오바시듐 풀룰란스 IFO 6353을 약 27℃에서 24 시간 동안 호기적으로 배양한 것을 사용하였다.

영양 배지의 점도 제어 방법은 영양 배지를 간헐적 또는 연속적으로 배출하면서 영양 배지를 추가 공급하는 희석 속도의 조정에 의하여 실시하였다. 영양 배지의 공급은 영양 배지가 정상 상태에 도달한 후부터 개시하였다. 영양 배지의 점도 측정은 연속 배양 도중 영양 배지의 일부를 샘플링하여 균을 제거한 후, 회전식 E 형 점도계 (30℃)를 사용하여 샘플의 점도를 측정하였다.
풀룰란 생산성의 측정은 연속 배양 300∼320시간의 사이에 채취한 영양 배지 시료에 대하여 영양 배지 1L 당 및 배양 1시간 당의 풀룰란의 약(g)을 측정함으로써 구하였다. 여기서 말하는 "풀룰란의 양"이라 함은 75 v/v% 메탄올에 불용성이고 풀룰라나아제(pullulanase)(EC 3.2.1.41)에 의하여 가수 분해를 받아 주로 말토트리오스를 생성하는 고분자 물질의 양을 뜻하는 것이다.
대조로서 영양 배지의 추가 공급을 하지 않고 점도를 제어하지 않는 조건에서 동일한 연속 배양을 하였다.
그 결과는 표 2에 나와 있다.

표 2의 결과로 부터 명백한 바와 같이 여러가지 농도의 당질을 함유하는 영양 배지에서 영양 배지를 배출하면서 영양 배지를 추가 공급하여 그 점도를 제어해서 연속 배양을 했을 경우 당질 농도 10∼20 w/v%, 바람직하게는 10∼18 w/v%의 범위에서 영양 배지의 점도를 30cp 이하로 유지했을 때에 0.88 gL-1h-1이상의 높은 생산성을 얻을 수 있음이 판명되었다.

[이소아밀라아제 공존에 의한 점도 제어]
실험 1-A의 경우와 동일한 조성의 영양 배지를 사용하여 영양 배지의 점도를 60cp 이상, 90 cp 이하로 제어하도록 영양 배지를 배출하면서 영양 배지를 추가 공급하고, 더욱이 이소아밀라아제 (일본국의 (株) 林原 생물 화학 연구소 판매)의 첨가에 의하여 표 3에 있는 각종 점도 범위로 유지하면서 pH 2.6∼3.0의 범위로 유지하여 배양하였다. 그 외의 조건은 실험 1-A와 마찬가지로 하였다.
그 결과는 표 3에 나와 있다.

표 3의 결과로 부터 명백한 바와 같이 이소아밀라아제 첨가에 의하여 영양 배지의 점도를 제어하면서 연속 배양을 했을 경우 당질 농도 10∼20 w/v%, 바람직하게는 10∼18 w/v%의 범위에서 영양 배지의 점도를 30cp 이하로 유지했을 때 0.92 gL-1h-1 이상의 극히 높은 풀룰란 생산성을 얻을 수 있음이 판명되었다.
[실험 2]
[점도 제어된 연속 배양에서의 풀룰란 생산성에 미치는 pH 제어의 영향]
당질 농도 16 w/v%의 영양 배지를 사용하고 실험 1-B에서와 마찬가지로 이소아밀라아제 공존하에 영양 배지의 점도를 30cp 이하로 유지하면서 각종 pH에서 연속 배양하여 풀룰란 생산성을 조사하였다.
그 결과는 표 4에 나와 있다.

표 4의 결과로 부터 명백한 바와 같이 연속 배양시의 pH 제어를 pH 4.0 이하, 바람직하게는 pH 2.0 이상, 4.0 이하로 하였을 때 0.92 gL-1h-1이상의 극히 높은 풀룰란 생산성을 얻을 수 있음이 판명되었다.
이하 실시예 A 에서는 풀룰란 고함유물의 제조예를, 그리고 실시예 B 에서는 풀룰란 고함유물의 용도예를 각각 설명한다.

[풀룰란 고함유물]
당질로서 고 DE의 전분 시럽(일본국 (株) 林原 상사 판매, 등록상표 "SUN ROSE") 농도 14 w/v%을 사용한 것 외에는 표 1과 동일한 조성으로 된 영양 배지를 사용하여 오레오바시듐 풀룰란스 IFO 6365의 종배양액을 접종하여 연속 배양하였다. 배양액을 배출하면서 여기에 영양 배지를 0.01∼0.04 h-1의 희석 속도로 추가공급 하면서 영양 배지의 점도를 30cp 이하로 유지하고 pH를 3.6∼3.8의 범위로 유지하여 27℃에서 1 개월간 연속 배양을 하였다. 배양 개시후 4 일째 되는 날부터 영양 배지를 회수하여 통상의 방법에 따라 제균 탈색, 농축, 건조하여 분말상의 풀룰란 고함유물을 얻었다.
이 제품의 분석 결과는 표 5에 나와 있다.

[실시예 A-2]
[풀룰란 고함유물]
실시예 A-1과 동일한 성분으로 된 영양 배지에서 오레오바시듐 풀룰란스 IFO 6353의 종배양액을 접종하고 연속 배양을 하였다. 배양액을 배출하면서 여기에 영양 배지를 0.01∼0.02h-1의 희석 속도로 추가 공급하면서 액화형 α-아밀라아제(일본국의 上田化學工業 (株) 판매, 상품명 "Hi-Maltosin S")의 첨가에 의하여 점도를 30cp 이하로 유지하고 pH를 3.2∼3.4로 유지하면서 27℃에서 2개월간 연속 배양을 하였다. 실시예 A-1과 마찬가지로 영양 배지를 회수하고 처리하여 분말상의 풀룰란 고함유물을 얻었다.
이 제품의 분석 결과는 표 6에 나와 있다.

[실시예 A-3]
[풀룰란 고함유물]
당질로서 말토오스 고함유 전분 시럽 (일본국의 (株) 林原 상사 판매, 등록 상표 "MALTO-RUP")을 농도 11, 12, 13, 14, 15, 16 또는 18 w/v%을 사용한 것 외에는 표 1과 동일한 조성의 영양 배지에서 오레오바시듐 풀룰란스 IFO 4464의 종배양액을 접종하여 연속 배양하였다. 배양액을 배출하면서 여기에 영양 배지를 0.01∼0.02 h-1의 희석 속도로 추가 공급하면서 이소 아밀라아제 (일본국의 (株) 林原 생물 화학 연구소 판매)의 첨가에 의하여 영양 배지의 점도를 30cp 이하로 유지하고 pH를 2.4, 2.5, 2.6 또는 2.8로 유지하여 27℃에서 2개월간 연속 배양을 가하였다. 실시예 A-1과 마찬가지로 영양 배지를 회수하고 통상의 방법에 따라 멤브레인 여과 및 농축하여 액상의 풀룰란 고함유물을 얻었다.
이들 제품의 분석 결과는 표 7에 나와 있다.

[실시예 B-1]
[슬라이딩 접촉부 시일용 조성물]
글리세린 56 중량부에 실시예 A-1의 방법으로 제조한 분말상 풀룰란 고함유물 6 중량부를 가하고 110℃로 가열하면서 교반 혼합하여 균일히 용해시킨 후 글리코실 수크로오스 [일본국의 (株) 林原 상사 판매, 등록상표 "COUPLING SUGAR") 137 중량부를 가한 다음, 수분 증발을 방지하면서 탈포(脫泡), 교반 혼합하였다.
이 조성물의 점도는 약 300 cp이었다. 이 조성물을 가스, 특히 액화 가스를 추진제로 하는 에어로졸 용기용 슬라이딩 접촉부의 시일용 조성물로 사용했을 경우 45℃에서 6 개월간, -5℃에서 1년간 방치하였으나 가스 배리어(gas-barrier) 성능에 이상이 없이 만족한 성능을 나타내었다.
또한, 이 조성물을 사람과 가족에 무독, 무해한 것이므로 슬라이딩 접촉부에 압력차를 가진 기체나 액화 가스가 접해 있는 각종 가정용품, 식료품, 화장품, 의약품 등을 충전하는 에어로졸 용기의 슬라이딩 접촉부 시일제로서 유리하게 이용할 수 있다.
[실시예 B-2]
[무우절임(신-즈께]
무우 30kg을 통상의 방법에 따라 절임(pickling)하고 사카린 10g과 더불어 계속해서 절임한 다음, 겨 1.7 kg, 사카린 10g, 글리신 30g, 글루탐산 나트륨 200g, 복합 조미료 70g, 소르비톨 500g, 식염 360g, 소주(35도) 250㎖ 및 A-2의 방법으로 제조한 분말상 풀룰란 고함유물 340g으로 된 혼합물에서 마무리 절임을 하였다.
이 제품은 제조 12일 후에 있어서도 점도 저하가 거의 없고, 겨가 제품에 잘 부착해 있어 광택과 텍스쳐(texture)가 모두 양호하였다.
[실시예 B-3]
[보냉재(保冷材)]
실시예 A-1의 방법으로 제조한 분말상 풀룰란 고함유물 16 중량부를 글리세린 16중량부와 물 68 중량부에 가열 용해하고, 여기에 적당량의 항균제를 혼합하여 패킹하므로써 보냉재를 제조하였다.
이 제품을 냉각한 것은 보냉 효과가 높기 때문에 사람이 열이 날 때 머리를 차게 한다거나 야채류 등 식물의 선도 유지 등에 유리하게 이용할 수 있다.
[실시예 B-4]
[피복막]
실시예 A-3의 No.4의 방법으로 제조한 액상의 풀룰란 고함유물을 농도 1.0 w/w% 수용액으로 하여, 여기에 산란 후 10시간 이내의 신선 란을 30초 동안 침지한 다음 30℃의 온도에서 2시간 건조하여 란 껍질 표면에 피복막을 형성하였다.
이렇게 하여 얻은 란을 실온(15∼25℃)에서 보존하여 그 가식(可食) 기간을 대조인 무처리 란과 비교하였다. 그 결과, 피복막을 형성시킨 란의 보존 기간은 약 5∼10배나 더 연장되었다.
[실시예 B-5]
[추출용 포장재]
시판의 비교적 망목(網目)이 큰 여과지로 제조된 홍차용의 작은 포장체에 실시예 A-3의 No.6의 방법으로 제조한 액상 풀룰란 고함유물을 농도 10 w/w% 수용액으로 하여, 이것을 칼렌더법(Calender method)으로 코우팅하고 60℃에서 열풍으로 건조하였다.
제조된 소포장체에 1회 사용량 만큼의 홍차를 넣고 히이트 시일(heat seal)하여 밀봉하였다. 대조로서 동일 품질의 홍차를 넣은 시판의 소포장체를 사용하였다. 상대 습도를 60%, 온도를 30℃로 유지하고 1 개월간 방치한 후 여기에 열탕을 부어 넣어 홍차를 추출하여 얻은 추출액의 우열로써 포장체를 비교하였다.
본 발명의 풀룰란 고함유물로 코우팅된 것도 열탕을 부어주면 대조와 마찬가지로 용이하게 추출되었다. 추출액은 어느것이나 투명하였다. 그러나, 추출액의 향기, 색, 맛의 어느 점에 있어서도 풀룰란 고함유물로 코우팅된 것이 우수하였다.
[실시예 B-6]
[골판지용 접착 호료]
실시예 A-3의 No.5의 방법으로 제조한 액상 풀룰란 고함유물을 농도 3 w/w% 수용액으로 한 것 100 중량부에 10 w/v%의 수산화 나트륨을 10 중량부 첨가하고 20 분간 교반하여 캐리어부(carrier 部)로 하였다. 따로 물 100 중량부에 옥수수 전분 40 중량부를 가해 슬러리로 하고, 다시 붕사 1 중량부를 첨가하여 메인부(main 부)를 제조하였다. 이 메인부 중에 캐리어부를 서서히 첨가 혼합한 후 5분간 교반하여 호료(糊料)를 제조하였다.
이 호료(糊料)는 종래의 전분 호료에 비하여 점도 변화가 작고, 또한 양면 코루게이터(corrugator)를 사용하여 B 급 라이너 240 g/㎡와 세미 중심(中芯) 125 g/㎡을 서로 첩합(貼合) 시키는 첩합 실험을 한 결과, 종래의 전분 호료에서는 120 m/min 이상에서는 첩합성 불량을 일으키는 경향이 나타난 반면, 본 발명의 호료 사용시에는 160 m/min 에서도 문제가 없이 첩합이 가능하였고 접착성도 양호하였다.
[실시예 B-7]
[고형상 접착제]
디메틸술폭시드 30 중량부, 물 25 중량부, 에루시난 5 중량부, 실시예 A-2의 방법으로 제조한 분말상 풀룰란 고함유물 5 중량부 및 벤질리덴 크실리톨 2 중량부로 된 혼합물을 온도 90℃에서 1시간 교반하여 용해시킨 후, 이것을 직경 14 mm, 높이 50 mm의 원통상의 반복 상하 이동이 가능한 기구(mechanism)를 가진 립스틱형 용기속에 주입하여 실온에서 방냉함으로써 고형상 접착제를 제조하였다.
이 접착제를 크라프트지에 도포한 결과, 얇고 균일하게 도포할 수 있었고 초기 접착력도 충분하였다. 이 접착제는 온도 변화에 따른 경도 변화가 비교적 작고 극히 양호한 도포성과 접착성을 나타내었다.
[실시예 B-8]
[필름]
실시예 A-3의 No.4의 방법으로 제조한 액상 풀룰란 고함유물을 농도 15 w/w% 수용액으로 하고, 여기에 고형물당 카라게난 1 w/w%와 수크로오스 모노라우레이트 0.1 w/w%을 용해하여 이 액을 폴리에스테르 필름 위에 부어 흘려 인취(引取) 속도 3 m/min으로 두께 0.03㎜ 필름으로 한 후 90℃의 열풍으로 건조시켜 제품으로 하였다.
이 제품은 풀룰란 단독 필름과는 달리 수성계에서는 신속히 용해하지 않으나 서서히 용해하여 붕괴하며 먹을 수 있는 필름이다.
따라서, 의약용 웨이퍼와 마찬가지로 먹기 어려운 가루 모양 의약 등의 포장제로서, 그리고 용해, 붕괴했을 때 점착성을 가지므로 인공치아 고정용 필름 등으로서도 유리하게 이용할 수 있다.
[실시예 B-9]
[섬유]
실시예 A-3의 No.1의 방법으로 제조한 액상 풀룰란 고함유물을 농도 40 w/w% 용액으로 하고, 여기에 알긴산을 용해하여 고형물당 2 w/w%의 농도가 되게 하였다. 수득한 용액을 방사(紡絲) 원액으로 하고 이것을 60℃로 하여 직경 0.3 mm, 길이 1 mm의 원통상 노즐로 부터 압력을 3 kg/㎠으로 하여 실온의 공기중으로 압출하여 스트랜드를 만든 다음, 수분을 증발 건조시키면서 권취기에서 권취하였다.
제조된 섬유의 굵기는 약 20㎛ 이었고 충분한 강도를 가졌으며, 이 섬유는 연산, 편직, 제직 등으로 가공할 수 있다. 더욱이, 친수성이고 무독하며 피부에 대해 자극이 없다는 특징을 가지고 있기 때문에, 예를 들자면 탈지면, 생리면, 거어즈, 수술용 실(絲) 등에 적절히 사용할 수 있다.
그리고, 기타 섬유와 혼방하면 그 흡습성, 비대전성, 염색성을 살려 내의나 기타 옷감으로서도 사용할 수 있다.
[실시예 B-10]
[종이]
실시예 B-9의 방법으로 제조한 풀룰란 고함유물의 섬유를 5∼10 cm로 절단한 것과, 이 섬유의 절반량의 목재 펄프로 된 혼합물을 제조하고, 이 혼합물의 약 50 배량 되는 10℃의 75 v/v% 메탄올에 이 혼합물을 균일히 현탁하여 초지기(抄紙機)에서 처리하였다.
건조 로울은 50∼80℃로 조절하여 건조한 후 가볍게 칼렌더 로울을 통과시켜 종이를 제조하였다.
이 종이는 표면이 평활하며 광택이 비교적 작고 일본지의 지합(地合)과 비슷하다. 필기용 잉크와 잘 조화되었고 잉크의 얼룩이 없었다.
이 종이는 뜨거운 물에 쉽게 용해하기 때문에 비밀 문서 등의 특수 용지에 적합하다. 그리고, 먹을 수 있으므로 의약품의 내포장재, 조미료, 커피, 코코아 등의 식품 포장재 등으로서도 적합하다.
[실시예 B-11]
[발포 시이트]
폴리염화비닐 수지 100 중량부에 대하여 가소제로서 디옥틸 프탈레이트 60 중량부를 가하고, 이 혼합물에 실시예 A-2의 방법으로 제조한 분말상 풀룰란 고함유물의 50 w/w% 수용액을 전체량에 대하여 30 w/w% 가한 다음, 이 혼합물을 혼합기에서 충분히 혼합한 후 애플리케이터(applicator)를 사용하여 알루미늄판 위에 3 mm 두께의 시이트로 하고, 이것을 190℃의 열풍로에서 10 분간 열처리함으로써 발포 배율이 약 5배인 균일한 셀(cell)을 가진 발포 시이트를 제조하였다.
이 제품은 방음재, 단열재, 포장용기, 완충재 등에 적합하다. 이 제품을 하천물에 침지하여 방치하면 1 개월 이내에 붕괴한 반면, 본 발명의 풀룰란 고함유물 무첨가의 것은 12 개월 후에도 원형을 그대로 가지고 있었다.
[실시예 B-12]
[골프 티이]
실시예 A-2의 방법으로 제조한 분말상 풀룰란 고함유물 10 중량부와 산성 백토 4 중량부를 교반하면서 물을 분무하여 수분 함량 약 30 w/w% 되게 하고, 이것을 사출성형기에서 120℃에서 골프 티이를 성형한 다음, 쉘락-알코올 용액에 침지한 후 공기 건조하여 제품을 얻었다.
이 제품은 쇼트(shot)에 의하여 조그만 덩어리로 파괴되어 빗물에 의하여 서서히 붕괴하여 생분해 된다.
따라서, 이 제품은 골프장의 미관을 해치는 일이 없고 환경도 파괴하지 않는다.
[실시예 B-13]
[화분]
실시예 A-1의 방법으로 제조한 분말상 풀룰란 고함유물 100 중량부와 글리세린 15 중량부 및 소르비톨 5 중량부의 혼합물을 사출성형기를 사용하여 135℃에서 화분을 성형하고 용융 왁스에 침지한 다음 방냉하여 제품을 얻었다.
이 제품은 서서히 붕괴하고 생분해성이기 때문에 이식용 화분으로써 적합하다. 이 화분에서 성장한 식물을 화분으로 부터 끄집어 내는 일이 없이 이식할 수 있기 때문에 식물이 손상을 입을 우려가 없다.
[실시예 B-14]
[캡슐]
실시예 A-1의 방법으로 제조한 분말상 풀룰란 고함유물 40 중량부, 젤라틴 60 중량부, 글리세린 30 중량부를 혼합하고, 여기에 물 80 중량부를 가하여 70℃에서 가열 용해하고 탈기하여 피막용 용액을 제조한 다음, 이것을 사용하여 통상의 방법에 따라 비타민 E 고함유 기름을 내용물로 한 연질 캡슐을 제조하였다.
이 제품은 젤라틴 단독 캡슐과는 달리 가스 배리어성이 높고, 비타민 E를 안정하게 유지할 수 있을 뿐만 아니라 수성계에서 신속히 용해하는 특성을 가지고 있다.
[실시예 B-15]
[막대상 비료]
배합 비료(N=14 w/w%, P2O5=8 w/w%, K2O=12 w/w%)와 와 실시예 A-1의 방법으로 제조한 분말상 풀룰란 고함유물, 황산 칼슘, 물의 비율을 중량부로 각각 70, 10, 15, 5로 하여 충분히 혼합한 후, 압출기 [L/D비=20, 압축비=1.8, 다이(die)의 구경=30mm]에서 80℃로 가열하여 표제의 제품을 얻었다.
이 제품은 비료용 용기가 불필요하고 취급이 용이하며, 전층 시비(全層施肥)에 적합한 강도를 가지고 있고, 더욱이 배합 비율을 변화시킴으로써 비료 성분의 용출 속도를 조절할 수 있는 것이다. 그리고, 필요한 경우, 이 막대상 비료에 식물 호르몬, 농업용 약제 및 토양 개량제 등의 혼합도 용이하다.
[실시예 B-16]
[담배 성형물]
담황색 담배 식물로 부터 제조한 분말 담배 원료 50 중량부에 실시예 A-2의 방법으로 제조한 분말상 풀룰란 고함유물의 2 w/w% 수용액 200 중량부, 락티톨 0.1 중량부를 혼합하고, 0.2 mm 슬리트(slit)로 부터 스테인레스-스틸제 엔드레스 벨트 위에 사출하여, 이것을 적외선 건조함으로써 수분 13 w/w%의 시이트상 담배 65 중량부를 얻었다.
이 제품은 궐련(cigarette) 용의 담배 또는 충전제 및 시가(cigar)와 시가릴로(cigarilro) 용의 결합제로서 적합하다. 그리고, 이 제품은 담배의 각종 성분의 열화(劣化)를 억제함과 아울러 보향성(保香性)이 크며, 끽연시에 이취(異臭), 이미(異味)를 발생하지 않으므로 향기와 맛이 양호하였다. 이 제품중의 풀룰란의 배합비를 변화시킴으로써 니코틴 함량과 연소 속도를 조절할 수 있었다.
[실시예 B-17]
[비피도박테리움균 증식 촉진 락트산 음료]
탈지유 10,000 중량부를 80℃에서 20 분간 가열 살균한 후 40℃로 냉각하고, 여기에 스타아터(starter)(락트산균) 300 중량부를 가하여 35∼37℃에서 10시간 발효시켰다. 이어서, 이것을 균질화한 후 이성화당 시럽 4,000 중량부, 수크로오스 2,000 중량부, 실시예 A-1의 방법으로 제조한 분말상 풀룰란 고함유물 170 중량부를 혼합 용해하고 70℃로 유지하여 살균 하였다. 이것을 냉각한 후 소량의 향료를 가하고 병에 채워 종래의 락트산 음료 타입의 비피도박테리움균 증식 촉진 락트산 음료를 제조하였다.
이 제품은 비피도박테리움균 증식 촉진 효과 뿐만 아니라 식물 섬유의 효과도 발휘할 수 있고, 장내의 pH를 저하시키며 부패 세균의 증식을 억제하고 변비를 예방할 수 있다.
[실시예 B-18]
[저칼로리 빵]
주재료의 일부로서 강략분 6 중량부와 아밀로오스 12 중량부에 물을 가하여 혼합기에서 반죽하였다. 이 혼합물에 부재료(마아가린 6 중량부, 수크로오스 2.5 중량부, 란 6 중량부, 식엽 0.4 중량부 및 물 6.5 중량부로 된 것) 21.4 중량부와 발효 물질(드라이 이스트 0.8 중량부, 수크로오스 0.4 중량부 및 물 4 중량부로 된 것) 5.2 중량부를 가하고 적당한 굳기의, 반죽을 만들었다. 이어서, 30℃에서 60 분간 제 1 발효를 시켜 도중 생성된 가스를 제거하였다. 다시, 주재료의 나머지 12 중량부(강략분 3 중량부 및 실시예 A-2의 방법으로 제조한 분말상 풀룰란 고함유물 9 중량부)를 가하고 반죽하여 균일히 혼합하고, 필요량의 물을 가하여 반죽의 굳기를 조절하였다. 냉장고에 60 분간 두어 제 2 발효를 하였다. 제 3 발효는 35℃에서 30분간 하여 생성된 가스를 제거하고 성형하였다. 다시, 35℃에서 30 분간 제 4 발효를 한 후 210℃의 오븐속에 넣고 20 분간 구웠다. 대조용의 빵은 주재료로서 강력분 30 중량부만을 사용하고, 또한 부재료인 물을 8 중량부로 한 것 외에는 위와 마찬가지로 하였다.
빵의 외관은 대조인 통상의 빵에 비하여 표면의 색조가 진하고, 내부 조직은 균일하였다. 식미는 대조인 통상의 빵과 거의 차가 없고, 이취, 의미가 없는 것은 물론이고 다소 담백한 감이 있었다. 치아에 대한 부착은 거의 느껴지지 않고 깨물기도 좋고 표피는 거의 굳은 감이 있었다.
이 제품은 저칼로리 식품으로서의 효과 뿐만 아니라 비피도박테리움균 증식 촉진 효과, 다이어터리 파이버로서의 효과도 발휘할 수 있으므로 미용, 건강의 유지 증진에 유리하게 이용할 수 있다.
[실시예 B-19]
[연치마(練齒魔)]
제 2 인산 칼슘 45.0 중량부, 실시예 A-3의 No.4의 방법으로 제조한 액상의 풀룰란 고함유물을 농도 10 w/w% 수용액으로 한 것 30.0 중량부, 폴리에틸렌 글리콜(평균 분자량 1,500) 30.0 중량부, 라우릴 황산 나트륨 1.5 중량부, 글리세린 17.0 중량부, 폴리옥시에틸렌 소르비탄 모노라우레이트 0.5 중량부, α-글리코실 스테비오시드 감미료(일본국의 東洋精糖 (株) 제조, 상품명 "αG Sweet") 0.2 중량부, 방부제 0.05 중량부를 통상의 방법에 따라 혼합하여 연치마를 제조하였다.
이 제품은 적당한 감미를 가지고 있고 어린이용 연치마로서 적합하다. 그리고, 이 제품은 풀룰란과 폴리에틸렌 글리콜의 회합물(會合物)을 함유하고 있으므로 풀룰란 단독 사용의 경우와 비교하여 점착성이 없는 사용하기 용이한 연치마이다.
[실시예 B-20]
[화장 팩]
리놀렌산 0.5 중량부를 통상의 방법에 따라 스콰알렌 1.5 중량부, 폴리옥시에틸렌 경화 피마자유 0.5 중량부, L-락트산 나트륨 5.5 중량부, 글리세린 4.0 중량부, 실시예 A-3의 No.2의 방법으로 제조한 액상의 풀룰란 고함유물을 농도 40 w/w% 수용액으로 한 것 50.0 중량부, 에틸 알코올 10.0 중량부 및 정제수 33.0 중량부에 균일하게 혼합하여 화장 팩을 제조하였다.,
이 제품은 색백제로서 적합하며, 갈색반점, 주근깨, 햇볕에 그을음 등의 국소성 및 아디소니즘 등의 전신성 색소 침착증의 치료용 및 예방용 등의 경우에 유리하게 이용할 수 있다.
[실시예 B-21]
[샴푸우]
실시예 A-3의 No.4의 방법으로 제조한 액상의 풀룰란 고함유물을 2.5 w/w% 수용액으로 한 것 80.0 중량부에 에틸 알코올 13.0 중량부, 글리세린 2.0 중량부, 향료 0.3 중량부, 폴리옥시에틸렌 소르비탄 모노라우레이트 1.5 중량부를 첨가하고 혼합 용해하여 샴푸우를 제조하였다.
이 제품은 손가락으로 머리칼을 쉽사리 헤집을 수 있고 머리를 씻은 후의 모발의 촉감이 부드러우며, 사용감, 상쾌감이 극히 우수한 샴푸우이다.

중량 150 mg되는 소정(素錠)을 심제(芯劑)로 하고, 여기에 결정 말티톨 40 중량부, 실시예 A-3의 No.2의 방법으로 제조한 액상의 풀룰란 고함유물을 농도 10 w/w% 수용액으로 한 것 20 중량부, 물 12 중량부, 탈크 25 중량부 및 산화 티탄 3 중량부로 된 제1코우팅액을 사용하여 정제 중량이 약 230 mg 되기 까지 당의(糖衣)하고, 이어서 마찬가지 결정 말티톨 65 중량부, 마찬가지 10 w/w% 농도의 풀룰란 수용액 10 중량부 및 물 25 중량부로 된 제2코우팅액을 사용하여 당의하고, 왁스액으로 다시 코우팅하여 우수한 광택이 있는 외면을 가진 당의정을 제조하였다.
이 제품은 당의(糖衣)작업이 용이하고 내충격성도 우수하며 고품질을 장시간 유지한다.
[실시예 B-23]
[대용 혈장]
실시예 A-3의 No.7의 방법으로 제조한 액상의 풀룰란 고함유물을 농도 10 w/w% 수용액으로 하고, 여기에 메탄올을 40 v/v%가 되도록 가하고, 생성된 하층부를 제거한 후 수득한 상층부에 다시 메탄올을 55 v/v% 가 되도록 가하여 방치한 다음 하층부를 분별 채취하였다. 수득한 하층부중의 메탄올을 증류하여 제거하고, 잔존한 풀룰란 수용액에 활성탄을 가하고 탈색하여 H+형 및 OH-형 이온 교환 수지로 탈염한 다음, 다시 멤브레인 필터로 여과하여 얻는 정제 풀룰란을 농축, 건조 분쇄하여 ()이 1.4 이고 ()가 50,000인 파이로겐 무함유의 풀룰란 백색 분말을 약 45%의 수율로 얻었다. 이렇게 하여 제조한 풀룰란을 4∼10 w/v%의 수용액으로 하여 여기에 염류, 당류 등의 등장화제(等張化劑)를 가하고 멸균하여 주사제를 제조하였다.
이 제품은 혈장 증량제, 혈류 개선제 등으로 적합하다.
위에서 설명한 바와 같이, 본 발명은 당질 온도 10∼20 w/v%로 한 영양 배지를 30 cp 이하로 유지하면서 연속 배양함으로써 보다 높은 풀룰란 생산성을 얻을 수 있음을 발견하여 공업적 생산에 유리한 분자량 250,000 이하의 풀룰란 고함유물과 그 제조 방법 및 용도를 확립한 것이다.
본 발명의 연속 배양 방법에 의하여 풀룰란 고함유물의 대량 및 저렴한 공급을 용이하게 하며, 이것을 함유시켜 증점제, 코우팅제, 접착제, 성형물 등에, 그리고 음식물, 화장품, 의약품, 나아가서는 농림 수축산 자재, 종이 가공 자재, 광공업 자재 등에 그 이용이 길을 개척한 것이어서 그 공업적 의의는 크다.
Claims (5)
- 풀룰란 생성능을 가진 오레오바시듐속 미생물을 당질 온도 10∼20 w/v% 함유하는 영양 배지에서 이 배지의 점도를 30 센티포이즈 이하로 유지하면서 pH 2.0 이상 4.0 이하에서 연속 배양하고, 수득한 배양물을 농축한 후, 농축된 풀룰란 고함유물의 배양물을 회수함을 특징으로 하는 풀룰란 고함유물의 제조 방법.
- 제1항에 있어서, 당질은 글루코오스, 푸럭토오스, 수크로오스, 말토오스, 말토올리고당류, 이소말토올리고당류, 전분 가수 분해물, 당화 전분, 전화당, 이성화당 및 당밀로 된 군으로 부터 선택되는 것인 제조 방법.
- 제1항에 있어서, 연속 배양을 호기적 조건하에서 실시하는 제조 방법.
- 제1항에 있어서, 연속 배양은 풀룰란을 부분적으로 가수 분해하는 아밀라아제의 공존하에 실시하는 제조 방법.
- 제4항에 있어서, 아밀라아제는 액화형 α-아밀라아제와 이소아밀라아제로된 군으로 부터 선택되는 것인 제조 방법.
