Claims (4)
- 라세미화합물 또는 거울상이성질체 형태의 구조식(Ⅰ)의 [(1-아릴피롤리딘-2-일)메틸]피페라진 유도체 및 그것의 약제학적으로 허용되는 산 부가염.여기서 Z는 구조식 N-R1기를 나타내고 R1은 수소원자 또는 C1-3알킬기 또는 구조식 Ar-CnH2n기(N은 0, 1또는 2를 나타내고, Ar은 하나 또 그 이상의 할로겐원자 및/또는 C1-3알킬 또는 C1-3알콕시라디칼로 임의로 치환도니 페닐기를 나타낸다.) 또는 구조식 COR2기(R2는 CH3,C6H5,CH2C6H5또는 OC2H5기를 나타낸다.)이다.
- 제1항에 있어서, Z가 N-(CH2)2-Ar 라디칼(Ar은 하나 또는 그 이상의 플루오르 또는 염소원자 및/또는 하나 또는 그 이상의 메틸 또는 메톡시 라디칼에 의해 임의로 치환된 페닐기를 나타낸다.)를 나타내는 것을 특징으로 하는 유도체.
- 적당한 부형제와 함께 제1항 또는 제2항의 화합물을 함유하는 것을 특징으로 하는 대뇌 허혈 또는 경련상태를 치료하기 위한 약제학적 조성물.
- 제1항의 화합물의 제조방법에 있어서, 구조식(Ⅱ)의 프롤린으로부터 유도된 에스테르와, 톨루엔과 같은 불활성 용매중 -20 내지 20℃의 온도범위에서 미리 제조한 구조식(Ⅲ)의 트리메틸알루미늄-아민 착체를, 톨루엔과 같은 불활성 용매중 80 내지 110℃의 온도범위에서 반응시켜 화합물(Ⅳ)을 얻고또는 프롤린(Ⅴ)을 DMF 또는 N-메틸피롤리돈과 같은 비양성자성 용매중 100 내지 150℃의 온도범위에서, 염기성 매질, 예를 들면 K2CO3중의 금속구리 또는 Cu0형태의 Cu++염의 존재하에 할로겐화물 C6H5Hal과 반응시켜 구조식(Ⅵ)의 산을 얻고, 그 다음 이 산(Ⅵ)을 테트라히드로푸란(THF) 또는 염화메틸렌( CH2Cl2)과 같은 용매중 20 내지 40℃의 온도범위에서 카르보닐디이미다졸로 처리하여 중간체 이미다졸리드를 형성하고 이것을 Z가 상기 주어진 의미를 갖는 구조식(Ⅲ)의 아민으로 원위치 처리함으로써 산(Ⅵ)을 아미드(Ⅳ)로 전환시키고; 최종적으로, 얻어진 구조식(Ⅳ)의 아미드를 에테르 또는 테트라히드로푸란(THF)과 같은 에테르용매중 20 내지 50℃의 온도범위에서 수소화알루미늄리튬으로 환원시켜 구조식(Ⅰ)의 화합물을 얻는 것을 특징으로 하는 제조방법.
