Abstract

PURPOSE: To carry out the quantitative determination of free fatty acids, by treating the free fatty acids with an acyl CoA synthetase in the presence of ATP and CoA, oxidizing the produced acyl CoA with an acyl CoA oxidase in the presence of an SH reagent, and determining the produced hydrogen peroxide.
CONSTITUTION: An aqueous solution or dispersion of an optionally diluted specimen is treated with an acyl coenzyme A synthetase, adenosine triphosphate, and coenzyme A at pH7W8 and 37°C for 5W60min. An acyl coenzyme A oxidase, an SH reagent such as a maleimide derivative, iodoacetamide, etc., peroxidase or catalase, and color-developing reagents such as 4-aminoantipyrin and phenol, etc. are added simultaneously or separately to the above treated product, and heated at 37°C for 5W60min. The density of the developed color is determined by the colorimetry using a blank test as a contrast.
