Abstract
PURPOSE: To determine the freshness of foods, especially that of fish meat, easily and rapidly, by reacting a food extract liquid with an enzyme having selectivity to inosinic acid, inosine, and hypoxanthine, and detecting the concentration changes of the compounds caused by the enzymatic reaction.
CONSTITUTION: A liquid specimen obtained by the extraction of a food is treated with an enzyme which reacts specifically with each of inosinic acid (IMP), inosine (HxR) and hypoxanthine (Hx), and the change of the concentration of each component caused by the enzymatic reaction is determined by an electrochemical means. The freshness of the food can be obtained as the L-value or the M-value given by the formulas. Examples of the enzyme are : IMP dehydrogenase for IMP, N-ribosylpurine ribohydrolase for HxR, and xanthine oxidase for Hx. The electrode for the electrochemical determination is, e.g. a thin-layer electrode composed of immobilized inosinase and xanthine oxidase, and the one composed of immobilized 5'-nucleotidase, inosinase and xanthine oxidase.
