Abstract
Description
Claims (1)
- 【特許請求の範囲】 1.ホスト細胞において、当該ホスト細胞に対し外因性の芳香族アミノ酸生合成 共通経路を経由して芳香族化合物の生合成を向上させる方法であって、当該方法 は、炭素源が中間体基質に作用する酵素種によって特徴付けられる多段階反応シ ーケンスによって芳香族化合物に転換されるものであって、かつ当該方法は、 当該ホスト細胞の培養上清を分析して蓄積した共通経路中間体基質を同定し、 これによって共通経路中の律速段階を同定する段階と、 当該ホスト細胞中に、前記経路における律速反応段階での前記同定された蓄積 中間体基質に作用する酵素種をコードするDNAを含むリコンビナントDNAを 導入し、当該ホスト細胞中の前記酵素種の発現を向上させる段階、 とを包含する、芳香族化合物の生合成向上方法。 2.ホスト細胞が大腸菌株である、請求項1に記載の方法。 3.同定された共通経路中間体が3−デオキシ−D−アラビノ−ヘプツロソネー ト−7−リン酸(DAHP)、3−デヒドロシキメート、シキメート、およびシ キメート−3−フォスフェートである、請求項2に記載の方法。 4.ホスト細胞が、次の酵素種、3−デヒドロキネート・シンターゼ、シキメー ト・キナーゼ、および5−エノールピルヴォイルシキメート−3−リン酸シンタ ーゼ(EPSPシンターゼ)をコードするDNAを包含するリコンビナントDN Aで形質転換されるものである、請求項3に記載の方法。 5.ホスト細胞が、さらにコリスミ酸シンターゼをコードするDNAを含有する リコンビナントDNAで形質転換され、ホスト細胞中でコリスミ酸シンターゼの 発現を向上させるものである、請求項4に記載の方法。 6.ホスト細胞が、次の酵素、トランスケトラーゼ(tkt)、3−デオキシ− D−アラビノ−ヘプツロソネート−7−リン酸シンターゼ(DAHPシンターゼ )、3−デヒドロキネートシンターゼ(DHQシンターゼ)、シキメート・キナ ーゼ、5−エノールピルヴォイル−シキメート−3−リン酸シンターゼおよびコ リスミ酸シンターゼをコードするDNAを包含するリコンビナントDNAで形質 転換され、当該酵素のホスト細胞中における発現を向上させるものである、請求 項3に 記載の方法。 7.ホスト細胞が前記酵素種をコードするDNAを包含する一以上のリコンビナ ント・プラスミド・ベクターで形質転換される、請求項1に記載の方法。 8.ホスト細胞が前記酵素種をコードするDNAを包含する一以上のリコンビナ ント・プラスミド・ベクターで形質転換される、請求項6に記載の方法。 9.請求項2に記載の方法によって調製した細胞形質転換体。 10.請求項7に記載の方法によって調製した大腸菌形質転換体。 11.次の酵素種、トランスケトラーゼ(tkt)、3−デオキシ−D−アラビ ノ−ヘプツロソネート−7−リン酸シンターゼ(DAHPシンターゼ)、3−デ ヒドロキネートシンターゼ(DHQシンターゼ)、シキメート・キナーゼ、5− エノールピルヴォイル−シキメート−3−リン酸シンターゼおよびコリスミ酸シ ンターゼに対する外因性構造遺伝子の発現向上を特徴とする、大腸菌形質転換体 。 12.炭素源から生物触媒的に芳香族化合物を生産する方法であって、当該方法 は、前記経路において前記炭素源の使用を促す条件の下、かつ前記炭素源の存在 下で、前記炭素源をその共通経路で利用することができる請求項12の細胞形質 転換体を培養する段階を包含するものである方法。 13.シキメート・キナーゼおよびEPSPシンターゼに対する構造遺伝子を包 含するプラスミド構造体。 14.さらにコリスミ酸シンターゼに対する構造遺伝子を包含する、請求項13 に記載のプラスミド構造体。 15.プラスミドpKAD50。 16.大腸菌株D2704/pKD136/pKAD50。 17.ホスト細胞に対し外因的である芳香族アミノ酸生合成の共通経路から得ら れる化合物の生合成を当該経路中の反応を触媒するタンパクの発現を高めること によって向上させる方法において、次の段階: 当該ホスト細胞の培養上清中の蓄積された共通経路中間体を同定することによ って当該経路における律速段階を同定する段階と、 当該経路中の、前記同定された律速反応段階を触媒するタンパクの発現を向上 させる段階、 とを包含する改良がなされたものである方法。 18.律速反応におけるタンパク触媒の発現が、ホスト細胞を形質転換して当該 タンパク触媒をコードする外因性遺伝子を発現させ、構造的にホスト細胞中での 当該タンパクの濃度を上昇させるものである、請求項17に記載の改良。 19.ホスト細胞が大腸菌株であって、当該ホスト細胞はシキメート・キナーゼ 、EPSPシンターゼ、およびコリスミ酸・シンターゼに対する遺伝子を包含す る外因性構造遺伝子を発現するように形質転換されるものである、請求項18に 記載の改良。 20.多段階の酵素によって仲介された多段階の反応段階を有する公知の生合成 経路を経由してホスト細胞中で化合物の生合成を向上させる方法であって、当該 方法は、 蓄積された経路中間体を同定することによって当該経路における律速段階を同 定する段階と、 当該ホスト細胞中にリコンビナントDNAを導入し、前記律速反応段階を仲介 する酵素種の発現を向上させる段階、 とを包含するものである、化合物の生合成向上方法。
