Description

Claims (15)
- 各染色剤が生物試料内の特定のバイオマーカーを識別する1つまたは複数の染色剤で染色される生物試料を撮像する方法であって、
所定のパワー値に達するように複数の画像チャネルの各画像チャネルについて光源のパワー出力を設定すること(500)であって、前記所定のパワー値は前記複数の画像チャネルの各画像チャネルについて実質的に同じであり、前記複数の画像チャネルの各画像チャネルについて独立に前記パワー出力を設定すること、
前記所定のパワー値で標的強度をもたらすためにカメラの露出時間を調整すること(501)であって、前記複数の画像チャネルの各画像チャネルの前記露出時間を、特定の画像チャネルについて前記標的強度をもたらすために独立に調整すること、
前記調整された露出時間に基づいて、前記複数の画像チャネルの各画像チャネルにおいて前記生物試料の一連の画像を取得すること(502)、および、
前記1つまたは複数の染色剤の各染色剤に特異的な導出された画像補正係数に基づいて前記取得された一連の画像の各画像内のピクセルデータを調整すること(504)を含む、方法。 - 前記光源についての前記パワー出力のレベルは、前記複数の画像チャネルの各画像チャネルについて既存の較正データから導出される、請求項1に記載の方法。
- 前記既存の較正データは、パワー出力対測定パワーの較正曲線である、請求項2に記載の方法。
- 前記既存の較正データは、前記複数の画像チャネルの各画像チャネルについて、そのチャネルの中心波長情報を、パワーメーターに入力することによって少なくとも部分的に決定される、請求項3に記載の方法。
- 前記所定のパワー値は、特定のアッセイに特異的である、請求項1から4のいずれか一項に記載の方法。
- 前記所定のパワー値は、前記複数の画像チャネルの各画像チャネルについて同じである、請求項5に記載の方法。
- 特定のチャネルについての標的強度は、前記チャネルについての較正試料の標準化標的強度の所定のパーセンテージである、請求項1から6のいずれか一項に記載の方法。
- 前記所定のパーセンテージは、前記チャネルについての前記標準化標的強度の約20%から約500%に及ぶ、請求項7に記載の方法。
- 第1のチャネルについての前記標準化標的強度は、(i)複数の画像チャネルの各画像チャネルについての前記較正標準の較正強度値を決定し、(ii)前記複数の画像チャネルの各画像チャネルについての前記較正標準の前記較正強度値を、前記正規化較正強度値の各正規化較正強度値の最大強度値に対して正規化し、(iii)前記複数の画像チャネルにわたって正規化強度の比を導出し、(iv)前記複数の画像チャネル内の最大輝度画像チャネルのために標的強度値を割り当て、(v)残りの画像チャネルについて前記標準強度値を計算することによって導出される、請求項7に記載の方法。
- 前記導出された画像補正係数は、(i)複数の染色剤について輝度値を正規化し、(ii)各染色剤についての前記正規化された輝度値を整数1から減算することによって計算される、請求項1から9のいずれか一項に記載の方法。
- 各取得された画像内の前記ピクセルデータは、各取得された画像内の各ピクセル値にそれぞれの前記画像補正係数を乗算することによって調整される、請求項1から10のいずれか一項に記載の方法。
- ピクセルデータを調整する前に、前記取得された画像をアンミックスすることをさらに含む、請求項1から11のいずれか一項に記載の方法。
- 前記光源のパワー出力が、光パワーの絶対単位で前記所定のパワー値に達するように較正される、請求項1から12のいずれか一項に記載の方法。
- コンピュータ実行可能命令を記憶する非一時的コンピュータ可読媒体であって、前記コンピュータ実行可能命令は、1つまたは複数のプロセッサによって実行されると、前記1つまたは複数のプロセッサに請求項1から13のいずれか一項に記載の方法を実行させる、非一時的コンピュータ可読媒体。
- 1つまたは複数のプロセッサおよび前記1つまたは複数のプロセッサに結合されたメモリを備えるシステムであって、前記メモリは、コンピュータ実行可能命令を記憶するためのものであり、前記コンピュータ実行可能命令は、前記1つまたは複数のプロセッサによって実行されると、前記1つまたは複数のプロセッサに請求項1から13のいずれか一項に記載の方法を実行させる、システム。
