Description
本発明は、ヒト抗Cath−D中和モノクローナル抗体及びその使用に関する。より具体的には、本発明は、Cath−Dの触媒活性と、そのLRP1(LDLレセプター関連タンパク質−1)レセプターへの結合との両方を阻害するヒトモノクローナル抗体フラグメントに関する。
乳ガンは、先進国における女性の主要な死亡原因の1つである[1]。発生率の高いトリプルネガティブ乳ガン(ER−及びPR−、HER2非増幅)の治療は不十分な状態である。加えて、ER+乳ガンは、ホルモン療法に対しても耐性になった。したがって、乳ガンの新規処置が緊急に必要とされている。腫瘍進行は、腫瘍細胞と、腫瘍間質として知られている周囲の支持組織との間のクロストークの進化の結果として認識されている[2]。ガン細胞は、細胞外マトリックス内のいくつかの正常細胞型、例えば線維芽細胞、浸潤性免疫細胞、内皮細胞及び脂肪細胞などと動的に相互作用する。間質細胞と腫瘍細胞とは、局所の細胞外マトリックスを改変し、遊走及び浸潤を刺激し、間質細胞及び腫瘍細胞の増殖及び生存を促進する酵素、成長因子及びサイトカインをやり取りする。過去10年間において、メタロプロテイナーゼ、セリンプロテアーゼ、システイン及びアスパラギン酸カテプシンなどの分子が相互作用して腫瘍形成促進性タンパク質分解ネットワークを形成する細胞周囲の微小環境を腫瘍細胞が作り出すことが次第に明らかになっている[3]。したがって、細胞外プロテアーゼは、ガンにおいてディファレンシャルに発現するので、主要な創薬ターゲットである[4〜6]。
本発明者らは、ファージディスプレイによる、Cath−Dに特異的なヒトモノクローナル抗体scFvフラグメントの単離及び特性評価について記載した。Cath−Dのタンパク質分解活性と、線維芽細胞LRP1レセプターへの結合との両方を阻害する能力について、Cath−D結合scFvを機能的にスクリーニングした。IgG1、λフォーマットでクローニングした2つのscFv(抗Cath−D IgG1 F1及びE2)は、トリプルネガティブ及びER+乳ガン細胞の創傷治癒、コロニー形成及びマトリゲルにおける三次元伸長を阻害した。抗Cath−D IgG1 F1及びE2は、ヌードマウスにおけるトリプルネガティブMDA−MB−231乳ガン細胞の腫瘍成長を有意に減少させた。これらの知見は、乳房腫瘍微小環境内のCath−Dの抗体ベースのターゲティングが乳ガン処置に治療上有効であり得ることを強く示唆している。
本明細書を通じて、いくつかの用語が用いられ、以下の段落で定義される。
本発明は、単離された抗Cath−D中和モノクローナル抗体又はそのフラグメントを提供する。
−H−CDR1領域に配列番号:2、H−CDR2領域に配列番号:3及びH−CDR3領域に配列番号:4を含む重鎖可変領域と、
−L−CDR1領域に配列番号:6、L−CDR2領域に配列番号:7及びL−CDR3領域に配列番号:8を含む軽鎖可変領域とを含む抗Cath−D抗体を提供する。
−H−CDR1領域に配列番号:10、H−CDR2領域に配列番号:11及びH−CDR3領域に配列番号:12を含む重鎖可変領域と、
−L−CDR1領域に配列番号:14、L−CDR2領域に配列番号:15及びL−CDR3領域に配列番号:16を含む軽鎖可変領域とを含む抗Cath−D抗体を提供する。
したがって、本発明は、Cath−Dに対する結合について、上記で定義される本発明の抗Cath−D IgG(特に、IgG1)F1及びE2と競合する単離されたモノクローナルscFcフラグメントであって、Cath−Dの触媒活性と、LRP1線維芽細胞レセプターに対するその結合との両方を阻害する単離されたモノクローナルscFcフラグメントに関する。
本発明の抗Cath−D抗体は、当技術分野で公知の任意の技術、例えば限定されないが、任意の化学的技術、生物学的技術、遺伝的技術又は酵素学的技術(単独又は組み合わせのいずれか)によって生産され得る。

本発明の抗体は、抗Cath−Dイムノコンジュゲートを形成するために、検出可能な標識とコンジュゲートされ得る。適切な検出可能な標識としては、例えば放射性同位体、蛍光標識、化学発光標識、酵素標識、生物発光標識又は金コロイドが挙げられる。このような検出可能に標識されたイムノコンジュゲートを作製及び検出する方法は、当業者に周知であり、以下により詳細に記載する。
本発明のさらなる態様は、ガン疾患、及びCath−Dレベルが改変(増加又は減少)している他の疾患を診断及び/又はモニタリングするための本発明の抗Cath−D抗体に関する。
本発明の抗体、フラグメント又はイムノコンジュゲートは、Cath−D過剰発現に関連する任意の疾患(優先的には、ガン)を処置するために有用であり得る。本発明の抗体は単独で、又は任意の適切な薬剤と組み合わせて使用され得る。
投与のために、抗Cath−D抗体又は抗体−薬物コンジュゲートは、医薬組成物として製剤化される。抗Cath−D抗体又は抗体−薬物コンジュゲートを含む医薬組成物は、薬学的に有用な組成物を調製するための公知の方法にしたがって製剤化され得、それにより、治療分子は、薬学的に許容し得る担体との混合物で組み合わされる。組成物は、その投与がレシピエント患者に許容し得る場合、「薬学的に許容し得る担体」と言われる。滅菌リン酸緩衝生理食塩水は、薬学的に許容し得る担体の一例である。他の適切な担体は、当業者に周知である(例えば、Gennaro (ed.), Remington’s Pharmaceutical Sciences (Mack Publishing Company, 19th ed. 1995)を参照のこと)。製剤は、1つ以上の賦形剤、保存剤、可溶化剤、緩衝剤、バイアル表面のタンパク質のロスを防止するためのアルブミンなどをさらに含み得る。
最後に、本発明はまた、本発明の少なくとも1つの抗体を含むキットを提供する。本発明の抗体を含有するキットは、Cath−Dの発現(増加又は減少)を検出する際に、又は治療的アッセイ若しくは診断的アッセイにおいて使用される。例えば、IgG1 E2及びF1抗体は、免疫蛍光によって細胞カテプシンDを認識する。本発明のキットは、例えば組織培養プレート又はビーズ(例えば、セファロースビーズ)などの固体支持体にカップリングされた抗体を含有し得る。例えば、ELISAでは、Cath−Dをin vitroで検出及び定量するための抗体を含有するキットが提供され得る。検出に有用なこのような抗体は、蛍光又は放射性標識などの標識と共に提供され得る。
材料及び方法
材料:酵母ツーハイブリッドアッセイによって同定したpYESTrp2−LRP1β(307〜479)由来のLRP1β(307〜479)をコードするPCR増幅cDNAを、EcoRIによって以前に設計されたpGEX−4T−1に挿入することによって、LRP1β(307〜479)を作製した[29]。52−、48−、34−、14−又は4kDaのヒトCath−D鎖をコードするPCR増幅cDNAを、EcoRIによって以前に設計されたpGEX−4T−1に挿入することによって、pGEX−4T−1−Cath−D構築物を得た[29]。サンドイッチELISAに使用したマウス抗ヒトCath−Dモノクローナル抗体M2E8は、52kDaのプロCath−Dとのみ相互作用し、48kDa又は34kDaのCath−Dと相互作用しない[46]。52−、48−及び34kDa型のCath−Dを認識する抗ヒトCath−Dモノクローナル抗体(BD Biosciences)、14kDaのCath−D軽鎖を認識する抗ヒトCath−D抗体(ABCAM; ab75811)及び4kDaのCath−Dプロドメインを認識する抗ヒトCath−D抗体(Pr. M. Fusek (Oklahoma Medical Research Foundation, Oklahoma City, OK, U.S.A.)[18]の厚意による提供)をイムノブロッティングに使用した。HRPにコンジュゲートしたヤギポリクローナル抗ヒトFc抗体(A0170)、マウスモノクローナルHRP抗his(A7058)、細胞Cath−D(ヒト肝臓由来)、D−エリスロ−セラミドC81−リン酸(C8355)、ペプスタチンA、BSA及びクリスタルバイオレットは、(Sigma Aldrich)から購入した。ポリクローナル抗マウスcathD(sc−6486)、及びHRPにコンジュゲートしたモノクローナル抗c−Myc抗体(9E10)は(Santa Cruz Biotechnology)から購入し、リツキシマブ(抗CD20抗体)はRocheから購入し、HRPにコンジュゲートしたモノクローナル抗M13抗体(27942101)は(GE Heathcare)から購入した。シュードCath−Dの蛍光発生ペプチド基質(M2295:Ac−Glu−Asp(EDANS)−Lys−Pro−Ile−Leu−Phe−Phe−Arg−Leu−Gly−Lys(DABCYL)−Glu−NH2)は(Sigma Aldrich)から購入し、成熟Cath−Dの蛍光発生ペプチド基質(M0938:MCA−Gly−Lys−Pro−Ile−Leu−Phe−Phe−Arg−Leu−Lys−DNP−Arg−NH2)は(BACHEM)から購入した。52kDaのリコンビナントヒトプロCath−Dは、(R&D Systems)から購入した。マトリゲルは、(BD Biosciences)から購入した。
ファージディスプレイによる抗Cath−D scFvの選択及び機能的スクリーニング:オープンな触媒部位を有する51kDaのヒトリコンビナントシュードCath−D、及び34+14kDaの精製ヒト肝臓成熟Cath−Dを、ファージ抗体HuscIライブラリーからヒト抗体を単離するめの免疫原として使用した[37]。このライブラリーは、高レベルの発現に最適化された単一のフレームワーク使用する[37]。多様性を5つのアミノ酸(Y、N、D、G、S)に限定し、パラトープに最も寄与する残基に対応する位置の6つのCDRに導入した[38]。固定化したシュードCath−D又は成熟Cath−Dに対してELISAで特異的結合を示すscFvフォーマットのポリクローナル抗体を単離し、4ラウンドのバイオパニングによってライブラリーから濃縮した[37]。本発明者らは、ELISAによって、プロCath−D、シュードCath−D及び成熟Cath−Dの全てに結合する8個のモノクローナルscFvを選択した(図1A)。これらのモノクローナルscFvを精製した。ELISA分析により、精製scFvが、トリプルネガティブMDA−MB−231 BCC(図1B)及びER+MCF−7 BCC(図2A)由来のヒト分泌型プロCath−D、シュードCath−D及び細胞Cath−Dに結合し、MEF細胞由来の成熟Cath−D(2つの成熟タンパク質間で80%の同一性)と交差反応したことが明らかになった。pH6におけるシュードCath−D(図3;図5)及び成熟Cath−D(図4;図5)のタンパク質分解活性の阻害について、8個のscFvをそれらのKi値について特性評価した。シュードCath−D及び成熟Cath−Dのタンパク質分解活性を阻害するより強力なscFvは、F1、E2、E12及びH2であった(表3)。また、GSTプルダウンアッセイにおいて、LRP1線維芽細胞レセプターに対するプロCath−Dの相互作用を阻害する能力について、8個のscFvをスクリーニングした[29](図6)。H7を除く全てのscFvが、GST−LRP1βフラグメントに対するプロCath−Dの結合を(50〜79%)阻害した(図6;表3)。これら2つの機能的スクリーニングに基づいて、本発明者らは、pH6におけるCath−Dのタンパク質分解活性と、LRP1に対するその結合との両方を阻害したF1、E2及びE12 scFvを用いて、本発明者らの実験を行った(表3)。
設定実験では、マトリゲルと混合(1:1)し、Balb/cヌードマウスの皮下に樹立した1又は2 106個のMDA−MB−231 BCC異種移植片は、指数関数的な腫瘍成長を示し、100%の取り込み及び十分な腫瘍サイズの均一性を伴って、45日間で1000mm3の腫瘍体積に達した(図S8)。次に、Balb/cヌードマウスにおけるMDA−MB−231異種移植片(マトリゲルと混合(1:1)した1.5 106個の細胞)を皮下に樹立し、IgG1 F1及びE2(15mg/kg、週3回)で45日間にわたって直接腹腔内処置した。腫瘍体積が2000mm3に達したら、マウスを屠殺した。IgGF1及びE2処置は両方とも、腫瘍成長を有意に阻害し(図8A)、全生存を改善した(図8B)。
本報告は、抗Cath−DヒトIgG1 F1及びE2を使用して、乳ガン腫の微小環境において高レベルで発現されるCath−Dをターゲティング及び遮断する能力を示している。in vitroでは、これらの抗体は、BCCの創傷治癒、コロニー形成及び三次元伸長をin vitroで阻害し、トリプルネガティブBCCの腫瘍成長をin vivoで減少させた。Cath−Dが、リソソーム内タンパク質の異化及び細胞ホメオスタシスの維持に不可欠なカテプシンの遍在する重要な必須メンバーであることは注目に値する。Cath−Dノックアウトマウスは、神経セロイドリポフスチン症(NCL)表現型で出生後直ぐに死亡する[42]。ヒトにおける先天的なCath−D突然変異は、その合成の減少又は酵素的に不活性なタンパク質の産生につながり、NCLをもたらす[43]。したがって、(細胞内Cath−Dを超える)病的な細胞外Cath−Dを優先的にターゲティングするヒト抗体は、特異的細胞透過性小分子阻害剤と比較して、オフターゲット効果を回避するために重要である。
