Description
以下において、本発明のすべての態様に適用される定義を述べる。
発明人は、驚くべきことに、被験者において体液性免疫応答を誘導する、すなわち被験者の免疫系に腫瘍関連抗原に対する抗体、特に自己抗体を生じさせることが可能であり、生成された抗体は、細胞の表面のその天然形態の腫瘍関連抗原を認識することができ、前記腫瘍関連抗原を担持する細胞に対して免疫エフェクター機能を及ぼすことができることを見出した。特に、本発明は、B型肝炎ウイルスコア抗原タンパク質から成るウイルス様粒子を腫瘍関連抗原に由来するエピトープのための担体として利用する。
(i)配列表の配列番号:9〜23および45〜78に示すアミノ酸配列から成る群より選択されるアミノ酸配列、
(ii)好ましくはエピトープ配列の全長にわたって(i)の下のアミノ酸配列と少なくとも60%、65%、70%、80%、81%、82%、83%、84%、84%、85%、86%、87%、88%、89%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、好ましくは少なくとも80%同一であり、(i)の下のアミノ酸配列と免疫学的に等価であるアミノ酸配列、または
(iii)切断型であり、(i)の下のアミノ酸配列と免疫学的に等価である、(i)もしくは(ii)の下のアミノ酸配列
を含み、好ましくは基本的に前記アミノ酸配列から成り、好ましくは前記アミノ酸配列から成る。前記切断はアミノ末端またはカルボキシ末端においてであり得、また、エピトープ配列の全長の好ましくは40%以下、好ましくは30%以下、好ましくは20%以下、より好ましくは10%以下である。
(i)配列番号:1に示すアミノ酸配列もしくは少なくとも50アミノ酸、好ましくは少なくとも60アミノ酸、好ましくは少なくとも70アミノ酸、好ましくは少なくとも80アミノ酸、好ましくは少なくとも90アミノ酸、好ましくは少なくとも100アミノ酸、好ましくは少なくとも110アミノ酸、好ましくは少なくとも120アミノ酸、好ましくは少なくとも130アミノ酸、好ましくは少なくとも140アミノ酸のその部分、または
(ii)好ましくは(i)の下のアミノ酸配列もしくはその部分の全長にわたって、(i)の下のアミノ酸配列もしくはその部分と少なくとも60%、65%、70%、80%、81%、82%、83%、84%、84%、85%、86%、87%、88%、89%、90%、91%、92%、93%、94%、95%、96%、97%、98%または99%同一である、好ましくは少なくとも80%同一であるアミノ酸配列
から成る群より選択されるアミノ酸配列を含み、好ましくは基本的に前記アミノ酸配列から成り、好ましくは前記アミノ酸配列から成る。好ましくは、本発明で使用されるB型肝炎ウイルスコア抗原タンパク質は、ウイルス様粒子への、好ましくはB型肝炎ウイルスコア抗原ウイルス様粒子の通常の形状、すなわち180または240サブユニットから成り、正二十面体構造を有する形状への組織化に関して、天然に生じるB型肝炎ウイルスコア抗原タンパク質と機能的に等価である。
(i)B型肝炎ウイルスコア抗原タンパク質のアミノ末端アミノ酸に結合している、B型肝炎ウイルスコア抗原タンパク質の30個のアミノ末端アミノ酸残基、好ましくは20個のアミノ末端アミノ酸残基、好ましくは10個のアミノ末端アミノ酸残基、好ましくは5個のアミノ末端アミノ酸残基内に挿入されている、またはB型肝炎ウイルスコア抗原タンパク質の30個のアミノ末端アミノ酸残基、好ましくは20個のアミノ末端アミノ酸残基、好ましくは10個のアミノ末端アミノ酸残基のうちの1もしくはそれ以上、例えば1、2、3、4、5、6、7、8、9もしくは10アミノ酸を置換している、および/または
(ii)B型肝炎ウイルスコア抗原タンパク質のカルボキシ末端アミノ酸に結合している、またはB型肝炎ウイルスコア抗原タンパク質の30個のカルボキシ末端アミノ酸残基、好ましくは20個のカルボキシ末端アミノ酸残基、好ましくは10個のカルボキシ末端アミノ酸残基内に挿入されている、またはB型肝炎ウイルスコア抗原タンパク質の30個のカルボキシ末端アミノ酸残基、好ましくは20個のカルボキシ末端アミノ酸残基、好ましくは10個のカルボキシ末端アミノ酸残基のうちの1もしくはそれ以上、例えば1、2、3、4、5、6、7、8、9もしくは10アミノ酸を置換している、および/または
(iii)B型肝炎ウイルスコア抗原タンパク質のMIRに対応するアミノ酸配列内に挿入されている、またはB型肝炎ウイルスコア抗原タンパク質のMIRに対応するアミノ酸配列内の1もしくはそれ以上のアミノ酸、例えば例えば1、2、3、4、5、6、7、8、9、10、11、12、13、14、15もしくは16アミノ酸を置換している、および/または
(iv)B型肝炎ウイルスコア抗原タンパク質のMIRに対応するアミノ酸配列内に位置する1もしくはそれ以上のアミノ酸、例えば配列番号:1のアミノ酸74、75、76、77、78、79、80、81、82、83、84、85、86、87、88または89の1もしくはそれ以上もしくは対応するアミノ酸に結合している。
(i)配列表の配列番号:1に示すアミノ酸配列の77位と78位のアミノ酸の間もしくは対応する位置でB型肝炎ウイルスコア抗原タンパク質に挿入されている、または
(ii)配列表の配列番号:1に示すアミノ酸配列の74〜81位、76〜81位、76〜79位もしくは79〜80位もしくは対応する位置のアミノ酸を置換している。
(i)配列番号:37〜40に示すアミノ酸配列、または
(ii)(i)の下のアミノ酸配列の全長の好ましくは60%にわたって、より好ましくは少なくとも70%にわたって、より好ましくは少なくとも80%にわたって、より好ましくは少なくとも90%にわたって、最も好ましくは少なくとも95%にわたって、(i)の下のアミノ酸配列と少なくとも60%、65%、70%、80%、81%、82%、83%、84%、84%、85%、86%、87%、88%、89%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、好ましくは少なくとも80%同一であり、および(i)の下のアミノ酸配列と機能的に、好ましくは免疫学的に等価であるアミノ酸配列
から成る群より選択されるアミノ酸配列を含む、好ましくは基本的に前記アミノ酸から成る、好ましくは前記アミノ酸から成るタンパク質である。例えば、(ii)の下のアミノ酸配列は、好ましくはウイルス様粒子に組織化することができ、好ましくはウイルス様粒子の形態でおよびアジュバントなしで被験者に投与された場合、好ましくは、前記被験者においてエピトープが由来する腫瘍関連抗原に対する免疫応答、好ましくは体液性免疫応答を誘発することができる。上記で特定したように、生成される抗体は、好ましくは細胞の表面上の、好ましくは腫瘍細胞の表面上のその天然立体配座の腫瘍関連抗原を、好ましくは認識し、それに結合することができる。好ましくは、前記免疫反応はまた、腫瘍関連抗原が被験者における自己タンパク質である場合にも誘発される。
キメラHBcAg融合タンパク質の組換え生成のために、HBcAg配列内のエピトープ挿入部位が異なる様々な細菌発現ベクター(HBcAg骨格)を生成した。生成したHBcAg骨格のいずれにおいても、エピトープをHBcAg MIRの特定の領域に挿入し得る。HBcAg骨格を除き、HBcAg Del 79〜80リンカーおよびHBcAg Del 79〜80エピトープを付加的にHBcAgタンパク質のアミノ末端に結合し得るかまたはアミノ末端部分、例えばSalI制限部位とSpeI制限部位の間に挿入し得る(図1)。HBc遺伝子サブタイプawyのN末端部分の野生型配列はMDIDPYKである。制限部位SalIおよびSpeIの挿入により、配列M VDAATS DIDPYKが導かれるが、制限部位の分離のためにアラニンが必要である。制限部位SalIとSpeIの間のエピトープ(xxx)の挿入の結果、配列M VE xxx SS DIDPYKとなる。融合タンパク質のDNA配列を、大腸菌における発現のために配列最適化した。制限酵素のための適切な認識配列を融合タンパク質の発現カセットに組み込んでおり、これは、大きな労力を必要とせずに挿入するカセットまたはエピトープの様々な領域を置換するまたは組み込むことを可能にする(図1および7参照)。
精製キメラHBcAg VLPの免疫原性および抗原性にわたる全般的な様々な種の検査のために、それらをNZWウサギおよびBalb/cマウス(VLPを担持するCLDN18.2エピトープのみ)にそれぞれ適用した。アジュバントを添加した免疫試験とアジュバントなしでの免疫試験を実施した。
挿入したCLDN18.2エピトープに対する抗体応答を誘導するキメラVLPの潜在能をマウスおよびウサギの免疫によって分析した。重要な点として、選択したエピトープおよび短命胃細胞に厳密に限定されるオルソログタンパク質の組織分布は、3つの種すべてにおいて保存されている。
HBcAg Del 79〜80リンカーCLDN18.2−EC1短縮型VLPの予防的インビボ効果を評価するため、マウスCLDN18.2で安定に形質導入したCT26結腸癌細胞の静脈内適用によって肺転移形成を誘導した免疫応答性BALB/cマウスにおける同系腫瘍モデルを使用した。
以下、参考形態の例を付記する。
1.B型肝炎ウイルスコア抗原タンパク質のアミノ酸配列の全部または一部とその中に挿入されたまたはそれに結合されたエピトープを含むアミノ酸配列を含むタンパク質であって、前記エピトープが腫瘍細胞の表面と結合した腫瘍関連抗原の細胞外部分に由来する、タンパク質。
2.アジュバントなしにウイルス様粒子の形態で被験者に投与した場合、細胞の表面と結合した前記腫瘍関連抗原に対する体液性免疫応答を誘発することができ、前記腫瘍関連抗原が前記被験者における自己タンパク質である、1に記載のタンパク質。
3.前記腫瘍関連抗原がクローディンファミリーのタンパク質またはPLAC1であり、前記クローディンファミリーのタンパク質が、好ましくはCLDN18.2およびCLDN6から成る群より選択される、請求項1または2に記載のタンパク質。
4.前記エピトープが、
(i)配列表の配列番号:9〜23および45〜78に示すアミノ酸配列から成る群より選択されるアミノ酸配列、
(ii)(i)の下のアミノ酸配列と少なくとも80%同一であり、免疫学的に等価であるアミノ酸配列、または
(iii)切断型であり、(i)の下のアミノ酸配列と免疫学的に等価である、(i)もしくは(ii)の下のアミノ酸配列
を含む、1乃至3のいずれかに記載のタンパク質。
5.前記B型肝炎ウイルスコア抗原タンパク質が、
(i)配列番号:1に示すアミノ酸配列もしくは少なくとも50アミノ酸のその部分、または
(ii)(i)の下のアミノ酸配列もしくはその部分と少なくとも80%同一であるアミノ酸配列
から成る群より選択されるアミノ酸配列を含む、1乃至4のいずれかに記載のタンパク質。
6.エピトープを含む前記アミノ酸配列が、
(i)前記B型肝炎ウイルスコア抗原タンパク質のアミノ末端アミノ酸に結合している、および/または
(ii)前記B型肝炎ウイルスコア抗原タンパク質のカルボキシ末端アミノ酸に結合している、および/または
(iii)前記B型肝炎ウイルスコア抗原タンパク質のMIRに対応するアミノ酸配列内に挿入されているもしくはアミノ酸配列内のアミノ酸の1もしくはそれ以上を置換している、および/または
(iv)前記B型肝炎ウイルスコア抗原タンパク質のMIRに対応するアミノ酸配列内に位置する1もしくはそれ以上のアミノ酸に結合している、
1乃至5のいずれかに記載のタンパク質。
7.エピトープを含む前記アミノ酸配列が、
(i)配列表の配列番号:1に示す前記アミノ酸配列の77位と78位のアミノ酸の間もしくは対応する位置で前記B型肝炎ウイルスコア抗原タンパク質に挿入されている、または
(ii)配列表の配列番号:1に示す前記アミノ酸配列の74〜81位、76〜81位、76〜79位もしくは79〜80位のもしくは対応する位置のアミノ酸を置換している、
1乃至6のいずれかに記載のタンパク質。
8.(i)配列番号:37〜40に示すアミノ酸配列、または
(ii)(i)の下のアミノ酸配列と少なくとも80%同一であり、免疫学的に等価であるアミノ酸配列
から成る群より選択されるアミノ酸配列を含む、1乃至7のいずれかに記載のタンパク質。
9.1乃至8のいずれかに記載のタンパク質をエンコードする核酸。
10.9に記載の核酸を含むベクター。

13.アジュバントなしで被験者に投与した場合、細胞の表面と結合した腫瘍関連抗原に対する体液性免疫応答を誘発することができ、前記腫瘍関連抗原が前記被験者における自己タンパク質である、12に記載のウイルス様粒子。
14.1乃至8のいずれかに記載のタンパク質、9に記載の核酸、10に記載のベクター、11に記載の宿主細胞、または12もしくは13に記載のウイルス様粒子ならびに医薬的に許容される希釈剤、担体および/または賦形剤を含有し、好ましくはアジュバントを含まない免疫原性組成物。
15.被験者において細胞の表面と結合した腫瘍関連抗原に対する体液性免疫応答を誘発するための免疫原性組成物であって、腫瘍関連抗原が前記被験者における自己タンパク質である、14に記載の免疫原性組成物。
16.腫瘍の予防的および/または治療的処置のための、1乃至8のいずれかに記載のタンパク質、9に記載の核酸、10に記載のベクター、11に記載の宿主細胞、12もしくは13に記載のウイルス様粒子、または14もしくは15に記載の免疫原性組成物。
17.被験者において腫瘍関連抗原に対する体液性免疫応答を誘発するための方法であって、腫瘍関連抗原が前記被験者における自己タンパク質であり、1乃至8のいずれかに記載のタンパク質、9に記載の核酸、10に記載のベクター、11に記載の宿主細胞、12もしくは13に記載のウイルス様粒子、または14もしくは15に記載の免疫原性組成物を前記被験者に投与することを含み、前記被験者が腫瘍に罹患しているかまたは腫瘍を発症する危険性があり、前記腫瘍が腫瘍細胞の表面と腫瘍関連抗原の結合を特徴とし、投与が、好ましくはアジュバントを伴わない方法。
18.被験者において腫瘍関連抗原に対する自己寛容を打破するためまたは被験者において腫瘍を治療するおよび/もしくは予防するための方法であって、1乃至8のいずれかに記載のタンパク質、9に記載の核酸、10に記載のベクター、11に記載の宿主細胞、12もしくは13に記載のウイルス様粒子、または14もしくは15に記載の免疫原性組成物を前記被験者に投与することを含み、投与が、好ましくはアジュバントを伴わない方法。
Claims (12)
- 配列番号:38に示すアミノ酸配列を有するタンパク質であって、
当該タンパク質が、B型肝炎ウイルスコア抗原タンパク質のアミノ酸配列とその中に挿入されたエピトープを含むアミノ酸配列を含み、
前記エピトープが腫瘍細胞の表面と結合した腫瘍関連抗原の細胞外部分に由来し、配列表の配列番号:21に示すアミノ酸配列を有する、
タンパク質。 - アジュバントなしにウイルス様粒子の形態で被験者に投与した場合、細胞の表面と結合した前記腫瘍関連抗原に対する体液性免疫応答を誘発することができ、前記腫瘍関連抗原が前記被験者における自己タンパク質である、請求項1に記載のタンパク質。
- 請求項1または2に記載のタンパク質をエンコードする核酸。
- 請求項3に記載の核酸を含むベクター。
- 請求項3に記載の核酸または請求項4に記載のベクターを含む宿主細胞。
- 請求項1または2に記載のタンパク質の複数のコピーを含むウイルス様粒子。
- アジュバントなしで被験者に投与した場合、細胞の表面と結合した腫瘍関連抗原に対する体液性免疫応答を誘発することができ、前記腫瘍関連抗原が前記被験者における自己タンパク質である、請求項6に記載のウイルス様粒子。
- 請求項1もしくは2に記載のタンパク質、請求項3に記載の核酸、請求項4に記載のベクター、請求項5に記載の宿主細胞、または請求項6もしくは7に記載のウイルス様粒子ならびに医薬的に許容される希釈剤、担体および/または賦形剤を含有し、アジュバントを含まない免疫原性組成物。
- 被験者において細胞の表面と結合した腫瘍関連抗原に対する体液性免疫応答を誘発するための免疫原性組成物であって、前記腫瘍関連抗原が前記被験者における自己タンパク質である、請求項8に記載の免疫原性組成物。
- 腫瘍の予防的および/または治療的処置のための、請求項1もしくは2に記載のタンパク質、請求項3に記載の核酸、請求項4に記載のベクター、請求項5に記載の宿主細胞、請求項6もしくは7に記載のウイルス様粒子、または請求項8もしくは9に記載の免疫原性組成物。
- 被験者において腫瘍関連抗原に対する体液性免疫応答を誘発するための方法に使用するための医薬の調製における、請求項1もしくは2に記載のタンパク質、請求項3に記載の核酸、請求項4に記載のベクター、請求項5に記載の宿主細胞、請求項6もしくは7に記載のウイルス様粒子、または請求項8もしくは9に記載の免疫原性組成物の使用であって、前記被験者が腫瘍に罹患しているかまたは腫瘍を発症する危険性があり、前記腫瘍が腫瘍細胞の表面と前記腫瘍関連抗原の結合を特徴とする使用。
- 被験者において腫瘍関連抗原に対する自己寛容を打破するためまたは被験者において腫瘍を治療するおよび/もしくは予防するための方法に使用するための医薬の調製における、請求項1もしくは2に記載のタンパク質、請求項3に記載の核酸、請求項4に記載のベクター、請求項5に記載の宿主細胞、請求項6もしくは7に記載のウイルス様粒子、または請求項8もしくは9に記載の免疫原性組成物の使用。
