図6に示す電気培養装置1により実験を行った。培養槽7としての容器20は250mL容のガラスバイアル瓶(Duran製)とした。培養液4は容器20の八分目程度まで入れた。容器20には蓋30をした。蓋30の上面30aにはシリコーンゴム栓を設けて、配線や電極、管と通した際の密閉性を確保した。また、シリコーンゴム栓を設けることにより注射針の突き刺しを可能とし、且つ注射針の差し込みにより生じた孔が注射針を抜いた際に塞がるようにした。
微生物2として、Clostridium acetobutylicum NBRC13948(以下、単にクロストリジウムと呼ぶ)を用いた。この微生物は、無酸素条件下でグルコースからエタノール、ブタノール、アセトン、酢酸、酪酸を生成することのできる偏性嫌気性微生物である。培養温度は30℃とした。また、酸化還元物質3としてFe(III)−EDTAを用い、培養液4の酸化還元物質濃度は2mMとした。実験に使用した培養液4の構成成分を表1に示す。また、電解液4aの組成は、表1の組成からFe(III)−EDTAを除いたものとした。尚、培養液4は攪拌子34で攪拌し、蓋30に備えられたシリコーンゴム栓に注射針を2本刺し、一方の注射針からN2ガスを1時間通気することにより、ヘッドスペースをN2ガスで置換した。

Claims (10)
- イオン交換膜によって仕切られた二つの槽と、作用電極及び対電極と、前記作用電極と前記対電極との間に電位差を与える電源とを有し、前記二つの槽のうちの一方の槽を培養槽として酸化還元物質を含む培養液が収容されると共に前記作用電極が前記培養液に浸され、前記二つの槽のうちの他方の槽を対電極槽として電解液が収容されると共に前記対電極が前記電解液に浸され、前記培養液に培養対象たる微生物が添加されて、前記電源により前記作用電極と前記対電極との間に電位差を与えながら前記微生物を培養する電気培養装置において、前記培養槽を密閉構造の容器とし、前記容器に収容可能な密閉構造の小容器を前記対電極槽とし、前記小容器は少なくとも一部に前記イオン交換膜を備えると共に前記対電極槽から発生するガスを前記容器の外に排出するガス排出管を備え、前記培養液が前記イオン交換膜を介して前記電解液と接触していること、前記培養液が前記イオン交換膜を介して前記対電極と接触していること、及び/又は、前記作用電極が前記イオン交換膜を介して前記電解液と接触していることにより、前記作用電極と前記対電極の間でのイオン電流の発生を可能としていることを特徴とする電気培養装置。
- 多孔性の前記作用電極が前記イオン交換膜に接触し、前記作用電極と前記イオン交換膜との接触面にて前記培養液が前記イオン交換膜と接触し、前記培養液及び前記作用電極が前記イオン交換膜を介して前記電解液及び/又は前記対電極と接触している請求項1に記載の電気培養装置。
- 多孔性の前記対電極が前記イオン交換膜に接触し、多孔性の前記対電極と前記イオン交換膜との接触面にて前記電解液が前記イオン交換膜と接触し、前記電解液及び前記対電極が前記イオン交換膜を介して前記培養液及び/又は前記作用電極と接触している請求項1または2に記載の電気培養装置。
- 前記対電極槽に前記電解液が収容されずに多孔性の前記対電極が前記イオン交換膜と接触している請求項1または2に記載の電気培養装置。
- 前記小容器は、その全体が前記イオン交換膜で形成された袋状の容器である、請求項1〜4のいずれか1項に記載の電気培養装置。
- 前記培養槽に収容されている前記培養液の液面よりも上部の空間に滞留するガス状物質を回収するガス回収手段を備える請求項1〜5のいずれか1項に記載の電気培養装置。
- 前記培養槽に収容されている前記培養液を採取する培養液採取手段を備える請求項1〜6のいずれか1項に記載の電気培養装置。
- 請求項6に記載の電気培養装置によりガス状物質を生産する微生物または揮発性物質を生産する微生物を培養し、前記ガス状物質または前記揮発性物質が揮発して発生したガス成分を前記培養槽内の前記培養液の液面よりも上部の空間に滞留させ、これを前記電気培養装置のガス回収手段で回収することを特徴とする物質生産方法。
- 請求項7に記載の電気培養装置により揮発性物質を生産する微生物を培養し、前記微生物が生産する前記揮発性物質を前記電気培養装置の培養液採取手段から採取した前記培養液から回収することを特徴とする物質生産方法。
- 請求項1〜7のいずれか1項に記載の電気培養装置により揮発性の有害物質を分解する微生物を培養し、前記培養槽内の前記培養液に添加された前記有害物質を分解処理することを特徴とする物質分解処理方法。
