Description
(式中、Rはメチル基を示し、Xはフッ素原子を示す。)
で表される3−ニトロベンゼンスルホニルクロリド化合物のニトロ基及びクロロスルホニル基を、塩酸触媒存在下、亜鉛を用いて還元することを特徴とする、一般式(2)
(式中、R及びXは前記と同じ意味を示す。)
で表される3−メルカプトアニリン化合物の製造方法。
(式中、R及びXは前記と同じ意味を示す。)
で表されるベンゼンスルホニルクロリド化合物(原料化合物)をニトロ化することにより製造される、一般式(1)で表される3−ニトロベンゼンスルホニルクロリド化合物を、還元することにより製造することができる。
2−フルオロ−4−メチル−5−メルカプトアニリンの製造
メカニカルスターラー、温度計、滴下ロートを備えた2lのナス型フラスコに、亜鉛156.9g(2.4mol)、エタノール500ml、次いで4−フルオロ−2−メチル−5−ニトロベンゼンスルホニルクロリド77g(0.3mol)を加えた後、水浴下、塩酸500g(5mol)を反応系の温度が50℃を超えないように滴下した後、75℃で6時間攪拌した。減圧下、エタノールを留出させた後、室温まで冷却し、重曹を加え、反応系を中和した後、酢酸エチルで2回抽出し、酢酸エチル層を水、次いで飽和食塩水で洗浄した。酢酸エチル層を無水硫酸ナトリウムで乾燥した後、減圧下、酢酸エチルを留去し、41gの標題化合物を得た。収率87%。
1H NMR(300MHz,CDCl3)σ値:6.81(d,J=10.8Hz,1H),6.75(d,J=8.7Hz,1H),3.14(s,1H),2.22(s,3H)ppm.
