Description
温度計、Arパージ用ガラス管(Φ5mm)、還流冷却器、および磁気攪拌子を備えた50ml四口平底フラスコに、メタントリスルホン酸(下記表1参照)および10.324g(66mmol)のケトイソホロンを仕込んだ。2分間以内で、無水酢酸を高速攪拌下で滴下した(下記表1参照)。添加中、混合物は、暗黄色に変化して最終的には暗褐色になり、内部温度が上昇した。所望の反応温度まで冷却させた後、油浴を利用してその温度に保持した。サンプルを採取して定性GC分析にかけた。反応時間(下記表1参照)の後、反応混合物を室温まで冷却させ、そして3.7g(70mmol)の無水炭酸ナトリウムを添加することにより触媒を不活性化させた。40℃/10mbarで反応混合物を濃縮することにより、酢酸および未反応無水物を留去した。内部標準としてスクアランを用いてGCにより粗生成物を分析した。結果および反応条件を以下の表1に与える。
230mlフラスコを使用し、ケトイソホロンと無水酢酸との比を変化させて、実施例1の手順を反復した。反応条件および結果を以下の表2に与える。
実施例1のときと類似の手順を踏んで、統計モデル(STAVEX)を用いて反応条件を最適化した。20.32g(200mmol)の無水酢酸を10分間以内で10.324g(66mmol)のケトイソホロンに添加した。結果を以下の表3にまとめる。
実施例3の結果に基づいて、最適化条件下で実験を行った。結果を表4に要約する。
Claims (9)
- メタントリスルホン酸の存在下でケトイソホロンをアシル化剤と反応させることにより2,3,5−トリメチル−1,4−ヒドロキノンジアシレートを調製する方法。
- 前記反応が、ケトイソホロンを基準にして0.01mol%〜2mol%のメタントリスルホン酸の存在下で行われる、請求項1に記載の方法。
- 前記反応が、ケトイソホロンを基準にして0.1mol%〜1mol%のメタントリスルホン酸の存在下で行われる、請求項2に記載の方法。
- アシル化剤とケトイソホロンとのモル比が、10対1である、請求項1〜3のいずれか一項に記載の方法。
- アシル化剤とケトイソホロンとのモル比が、3対1である、請求項1〜3のいずれか一項に記載の方法。
- 前記アシル化剤は無水酢酸である、請求項1〜5のいずれか一項に記載の方法。
- 前記反応が、0℃〜140℃で行われる、請求項1〜6のいずれか一項に記載の方法。
- 前記反応が、20℃〜70℃で行われる、請求項1〜6のいずれか一項に記載の方法。
- 得られた2,3,5−トリメチル−1,4−ヒドロキノンジアシレートが、α−トコフェロールに転化される、請求項1〜8のいずれか一項に記載の方法。
