ヒトの角化細胞を24穴プレートに播種し、37℃、5%CO2−95%airの下にて、各試料添加培地(試料濃度:50μg/mL)で24時間培養後、上清100μLをエライザプレートに移し換え一晩4℃でプレートに吸着させたのち、溶液を捨て、0.05質量%Tween20を含むリン酸生理緩衝液(PBS−T)にて、洗浄を行った。その後、1質量%ウシ血清アルブミンを含むリン酸生理緩衝液で、ブロッキング操作を行った。溶液を捨て、0.05質量%Tween20を含むリン酸生理緩衝液(PBS−T)にて洗浄を行い、抗ヒトラミニン5抗体(マウスIgG、ケミコン社製)を反応させた。溶液を捨て、0.05質量%Tween20を含むリン酸生理緩衝液(PBS−T)にて、洗浄を行い、ビオチン標識抗体マウスIgG(アマシャムバイオサイエンス社製)を反応させた。溶液を捨て、0.05質量%Tween20を含むリン酸生理緩衝液(PBS−T)にて洗浄を行い、アビジン−ビオチン化ペルオキシダーゼ複合体と反応させたのち、同様の洗浄操作を行い、発色反応を行った。
ラミニン5産生促進率は、試料無添加時における吸光度を100%として算出した。各
試料のラミニン5産生促進率(%)を表4に示す。
