- 【特許請求の範囲】 1.インチミン、またはインチミンの一部を含む富化タンパク質であって、該一 部が、野生型結合活性を遮断する抗体を誘導する、富化タンパク質。 2.インチミン、またはインチミンの一部を含む精製タンパク質であって、該一 部が、野生型結合活性を遮断する抗体を誘導する、精製タンパク質。 3.インチミン、インチミン様タンパク質、またはそれらの一部を含むタンパク 質であって、該インチミン、インチミン様タンパク質、またはそれらの一部がヒ スチジンタグを有する、タンパク質。 4.前記インチミン、インチミン様タンパク質、またはそれらの一部が富化され ている、請求項3に記載のタンパク質。 5.前記インチミン、インチミン様タンパク質、またはそれらの一部が精製され ている、請求項3に記載のタンパク質。 6.前記インチミン、インチミン様タンパク質、またはそれらの一部が、該イン チミン、インチミン様タンパク質、またはそれらの一部と化学的、物理的または 組換え的に結合する少なくとも1つの抗原、少なくとも1つの薬剤、またはそれ らの組合わせ物をさらに含む、請求項1、2、3、4、または5に記載のタンパ ク質。 7.精製されたインチミンまたは精製されたインチミンの一部を作製する方法で あって、 −ヒスチジンタグを有するインチミンまたはインチミンの一部を含むタンパ ク質を発現させ、 −該インチミンまたはインチミンの一部を精製する、 ことを含んでなる方法。 8.前記精製の前または後に、前記インチミンまたはインチミンの一部から前記 ヒスチジンタグを除去することをさらに含む、請求項7に記載の方法。 9.精製されたインチミン様タンパク質またはその一部を作製する方法であって 、 ヒスチジンタグを有するインチミン様タンパク質またはその一部を含むタン パク質を発現させ、 該インチミン様タンパク質またはその一部を精製する、 ことを含んでなる方法。 10.前記精製の前または後に、前記インチミン様タンパク質またはその一部から 前記ヒスチジンタグを除去することをさらに含む、請求項9に記載の方法。 11.富化されたインチミンまたは富化されたインチミンの一部を作製する方法で あって、 −ヒスチジンタグを有するインチミンまたはインチミンの一部を含むタンパ ク質を発現させ、 −該インチミンまたはインチミンの一部を富化する、 ことを含んでなる方法。 12.前記富化の後に、前記インチミンまたはインチミンの一部から前記ヒスチジ ンタグを除去することをさらに含む、請求項11に記載の方法。 13.富化されたインチミン様タンパク質またはその一部を作製する方法であって 、 ヒスチジンタグを有するインチミン様タンパク質またはその一部を含むタン パク質を発現させ、該インチミン様タンパク質またはその一部を富化する、 ことを含んでなる方法。 14.前記富化されたインチミン様タンパク質またはその一部から、前記ヒスチジ ンタグを除去することをさらに含む、請求項13に記載の方法。 15.インチミンまたはインチミン様タンパク質を発現する細菌に対して防御免疫 応答を増強する方法であって、結合活性を遮断する抗体を誘導するインチミン、 インチミン様タンパク質、またはそれらの一部を、患者に投与することを含んで なる方法。 16.前記インチミン、インチミン様タンパク質、またはそれらの一部が精製され ている、請求項15に記載の方法。 17.前記インチミン、インチミン様タンパク質、またはそれらの一部が富化され ている、請求項15に記載の方法。 18.前記インチミン、インチミン様タンパク質、またはそれらの一部がヒスチジ ンタグ付けされている、請求項15に記載の方法。 19.少なくとも1つの抗原に対する防御免疫応答を増強する方法であって、 インチミン、インチミン様タンパク質、または結合機能を保持するそれらの 一部 と、化学的、物理的または組換え的に結合した少なくとも1つの抗原を含む組成 物を患者に投与することを含んでなる方法。 20.前記インチミン、インチミン様タンパク質、またはそれらの一部が精製され ている、請求項19に記載の方法。 21.前記インチミン、インチミン様タンパク質、またはそれらの一部が富化され ている、請求項19に記載の方法。 22.前記インチミン、インチミン様タンパク質、またはそれらの一部がヒスチジ ンタグ付けされている、請求項19に記載の方法。 23.少なくとも1つの抗原、少なくとも1つの薬剤、またはそれらの組合わせ物 の送達を、上皮細胞を標的として行う方法であって、インチミン、インチミン様 タンパク質、または結合機能を保持するそれらの一部に結合した、少なくとも1 つの抗原、少なくとも1つの薬剤、またはそれらの組合わせ物を含む組成物を患 者に投与することを含んでなる方法。 24.前記インチミン、インチミン様タンパク質、またはそれらの一部が精製され ている、請求項23に記載の方法。 25.前記インチミン、インチミン様タンパク質、またはそれらの一部が富化され ている、請求項23に記載の方法。 26.前記インチミン、インチミン様タンパク質、またはそれらの一部がヒスチジ ンタグ付けされている、請求項23に記載の方法。 27.抗−インチミン抗体を、それを必要とする患者に投与することを含んでなる 、受動免疫防御を提供する方法。 28.抗−インチミン抗体を含む組成物であって、インチミンを発現する宿主細菌 に対して特異的な他の抗体を含有しない、組成物。 29.前記組成物が、EHECに特異的な他の抗体を含有しない、請求項28に記載 の組成物。 30.前記抗体がモノクローナル抗体である、請求項28に記載の組成物。 31.前記抗体がポリクローナル抗体である、請求項28に記載の組成物。 32.前記抗体がアフィニティー精製されている、請求項28に記載の組成物。 33.抗−インチミン抗体の調製方法であって、 −ヒスチジンタグを有するインチミンまたはヒスチジンタグを有するインチ ミンの一部を発現させ、 該インチミンまたはインチミンの一部を患者に投与し、および 抗−インチミン抗体を回収する、 ことを含んでなる方法。 34.前記投与前に、前記ヒスチジンタグを除去することをさらに含む、請求項33 に記載の方法。 35.前記投与前に、前記インチミンまたはインチミンの一部を富化することをさ らに含む、請求項33に記載の方法。 36.富化後に、ヒスチジンタグを除去することをさらに含む、請求項35に記載の 方法。 37.投与前に、前記インチミンまたはインチミンの一部を精製することをさらに 含む、請求項35に記載の方法。 38.前記精製の前または後に、ヒスチジンタグを除去することをさらに含む、請 求項37に記載の方法。 39.抗−インチミン抗体の調製方法であって、 −ヒスチジンタグを有するインチミン様タンパク質またはその一部を発現し 、 −該インチミン様タンパク質またはその一部を患者に投与し、および −抗−インチミン抗体を回収する、 ことを含んでなる方法。 40.前記投与前に、前記ヒスチジンタグを除去することをさらに含む、請求項39 に記載の方法。 41.前記投与前に、前記インチミン様タンパク質またはその一部を富化すること をさらに含む、請求項39に記載の方法。 42.前記富化後に、前記ヒスチジンタグを除去することをさらに含む、請求項41 に記載の方法。 43.前記投与前に、前記インチミン様タンパク質またはその一部を精製すること をさらに含む、請求項41に記載の方法。 44.前記精製の前または後に、前記ヒスチジンタグを除去することをさらに含む 、請求項43に記載の方法。 45.インチミンまたはインチミンの一部を含む富化タンパク質であって、該一部 が野生型結合活性を保持する、富化タンパク質。 46.インチミンまたはインチミンの一部を含む精製タンパク質であって、該一部 が野生型結合活性を保持する、精製タンパク質。 47.インチミンまたはインチミン様タンパク質を発現する細菌に対する防御免疫 応答を増強する方法であって、インチミン、インチミン様タンパク質、または結 合活性を保持するそれらの一部を患者に投与することを含んでなる方法。 48.前記インチミン、インチミン様タンパク質、またはそらの一部が精製されて いる、請求項47に記載の方法。 49、前記インチミン、インチミン様タンパク質、またはそらの一部が富化されて いる、請求項47に記載の方法。 50.前記インチミン、インチミン様タンパク質、またはそらの一部がヒスチジン タグ付けされている、請求項47に記載の方法。
