Abstract
The invention comprises 1 - methyl - 3 - piperidylmethyl - 2 - cycloalkyl - 4 - methyl - pentanoates of the formula <FORM:0720628/IV (b)/1> wherein R is a cyclohexyl or cyclopentyl radical, or water-soluble acid-addition or quaternary ammonium salts thereof. The compounds are formed by reacting the acid chloride of the corresponding 2 - cycloalkyl - 4 - methyl - pentanoic acid with 1-methyl-3-piperidylmethanol, and reacting the resulting hydrochloride with a base. Acid addition salts are then prepared by reaction with appropriate acids while quaternary ammonium salts are prepared by reaction with an alkyl or aralkyl ester of an inorganic acid (e.g. methyl bromide) or organic sulphonic acid respectively. The example describes the preparation of the hydrochloride, hydrobromide and methobromide of 1-methyl-3-piperidylmethyl-2-(cyclopentyl or cyclohexyl)-4-methylpentanoates. Ethyl 1 - methylpiperidine - 3 - carboxylate is prepared by catalytic hydrogenation of ethyl nicotinate followed by further hydrogenation of a mixture of the hydrogenation product and formaldehyde. 1 - Methyl - 3 - piperidylmethanol is prepared by reducing ethyl 1-methylpiperidine-3-carboxylate with a mixture of 2-methylpentanol-4 and sodium in toluene followed by hydrolysis of the resulting sodium derivative with water.
