Abstract
p-Aminosalicylic acid (2-hydroxy-4-amino-benzoic acid) is prepared by heating 6-nitro-benzisoxazole carboxylic acid (4-nitro-2-oxyphenyl-glyoxime), or a derivative thereof resulting from substitution in the carboxylic group, e.g. an alkyl ester or amide, in the presence of sulphuric acid and reducing the resultant 4-nitrosalicylic acid. In examples: (1) the methyl ester of 6-nitrobenzisoxazole-3-carboxylic acid is heated with sulphuric acid to give 4-nitrosalicylic acid, which is reduced to the amino compound, either by hydrogenation in the presence of a platinum catalyst or by means of tin and hydrochloric acid; (2) the free acid is used in place of the methyl ester in (1) and (3) the amide is used in place of the methyl ester in (1). It is stated in the Specification to be known that nitrobenzisoxazole-3-carboxylic acid methyl ester is hydrolysed by mineral acids such as hydrochloric acid, to give 4-nitrosalicylic acid at temperatures considerably above 100 DEG C. The Specification as open to inspection under Sect. 91 comprises also the preparation of derivatives p-aminosalicylic acid arising from further substitution in the benzene ring thereof. This subject-matter does not appear in the Specification as accepted. Reference has been directed by the Comptroller to Liebigs Annalen der Chemie, Volume 390, page 3.
