Abstract

3 : 4-Dihydroxyphenyl-monoalkyl- or monocycloalkyl - aminobutanols are prepared by brominating 3 : 4 - dihydroxybutyrophenone, preferably after preliminary aralkylation, treating the brominated product with an alkyl- or cycloalkyl-amine, and reducing the resulting alkyl- or cycloalkyl-aminoketone. The products, which possess therapeutic properties, may be separated into their optical isomers by means of optically-active acids. Examples are given of the preparation of (1) 3 : 4-dihydroxyphenyl - methylaminobutanol by brominating 3 : 4-dibenzyloxy-butyrophenone, treating the brombutyrophenone with methylbenzylamine and reducing the resulting 3 : 4-dibenzyloxy-phenylmethylbenzylaminobutanone as its hydrochloride in alcohol solution with hydrogen in the presence of palladium; (2) 3 : 4 - dihydroxyphenyl - cyclopentylaminobutanol by treating brominated 3 : 4-dibenzyloxy-butyrophenone with cyclopentylamine and the product first reduced in alcohol solution using palladium and hydrogen and the resulting 3 : 4-dihydroxycyclopentylaminobutyrophenone then hydrogenated in aqueous hydrochloric acid solution in the presence of palladium. The Specification as open to inspection under Sect. 91 also describes the preparation of 3 : 4-dihydroxyphenyl-ethylaminobutanol by reducing with hydrogen the reaction product of 3 : 4 - dibenzyloxy - bromobutyrophenone with ethylbenzylamine in presence of palladium. This subject-matter does not appear in the Specification as accepted. 3 : 4-Dibenzyloxybutyrophenone is obtained by heating butyropyrocatechin in alcohol solution with benzyl chloride in presence of potassium carbonate and sodium iodide.
