Abstract
本发明提供了一种能有效治疗肾阳虚衰，气虚血亏引起的腰膝酸软，畏寒肢冷等症所致以上症状的中药组合物。该中药组合物是由人参、枸杞子、杜仲、肉桂、淫羊藿、锁阳、肉苁蓉、附子、菟丝子、熟地黄、山茱萸、仙茅、韭菜子、黄芪、当归、鹿鞭、鱼鳔、五味子、海龙、海马、蛤蚧、阿胶、龟甲胶以及冬虫夏草按一定的比例配制而成。本发明从扶正祛邪、调整阴阳、调理气血三个方面进行了药效学考察，表明本发明的药物具有显著的作用；临床试验表明：发明本品对肾阳虚衰，气虚血亏引起的腰膝酸软，畏寒肢冷等症状疗效确切，总有效率95.08％。

本发明属于中药领域，涉及一种中药组合物及其制备方法，尤其涉及一种治疗肾阳虚衰，气虚血亏的中药组合物及该中药组合物的制备工艺。
背景技术
中医认为，任何疾病的发生发展过程都是由于致病因素作用于人体，引起机体阴阳偏盛偏衰，脏腑经络机能失常的结果，自古有“邪气盛则实，精气夺则虚”之说。药物防病治病的基本作用，是驱除病邪，消除病因，扶正固本，恢复脏腑功能的协调，纠正阴阳偏盛偏衰，使机体恢复到阴平阳秘的正常状态。中药及其复方功效基本作用是“扶正祛邪，调节平衡”。通过扶正祛邪增强机体抗病能力，驱除病邪。通过调节平衡来调整阴阳失调，调和气血运行，从而使正胜邪去，阴阳和合，气血盈畅，达到病去人安的目的。

本发明药物的有效服用量：口服，一次5粒，一日3次，温开水送服。
下面以本发明药物的胶囊剂(即参龙虫草益肾胶囊)为样品，通过以下实验说明本发明药物治疗肾阳虚衰，气虚血亏等症的效果。
1、药效学实验：实验从壮阳方面、免疫方面、抗缺氧作用三个方面进行了药效学考察：
1.1壮阳方面
共做了“对大鼠交配能力的影响”，对大鼠阴茎勃起时间的影响及“对去势大鼠生殖器官脏器系数的影响”三个试验。结果证实参龙虫草益肾胶囊能明显缩短大鼠的捕捉及射精潜伏期；提高去势大鼠阴茎对外部刺激的兴奋性，缩短阴茎勃起时间；参龙虫草益肾胶囊对去势大鼠的主要性器官没有影响，初步认为无激素样作用，但能提高体肛及重量，而有同化激素作用。
1.2免疫方面
共做了“对正常及免疫抑制小鼠碳粒廊清速率”、“对小鼠腹腔巨噬细胞吞噬功能的影响”、“对溶血素形成的影响”、“对迟发型超敏反应的影响”、“对刀豆素A诱导的淋巴细胞增殖反应的影响”和“对正常空斑形成的影响”六个试验。结果证实参龙虫草益肾胶囊能显著提高正常及免疫抑制小鼠碳粒廊清速率，可提高因环磷酰胺所致实验小鼠吞噬功能低下恢复正常，可使小鼠因环磷酰胺所致溶血素形成抑制状态恢复正常，可使小鼠因环磷酰胺导致的皮肤迟发型超敏反应的抑制状态恢复正常，可显著提高刀豆素A诱导的小鼠脾淋巴细胞的增殖反应，可提高正常小鼠的抗体形成细胞数。以上结果提示参龙虫草益肾胶囊能提高机体的免疫功能。
1.3抗缺氧作用
做了在常压下PO或ip给药对小鼠的抗缺氧作用，结果证实参龙虫草益肾胶囊PO给药有显著的抗缺氧作用，ip给药有极显著的抗缺氧作用。
2.急性毒性试验：由于给药浓度、给药体积等诸多因素限制，经预试无法测出半数致死量(LD50)值。在小鼠可接受的最大容积下进行最大耐受量(MTD)测定试验，结果测得小鼠LD50大于72g/公斤，已相当于人临床用量的144倍，说明毒性极小，安全性高。
3.长期毒性试验：分四个组：临床用量的100倍、60倍、30倍及对照组比较，经60天长期毒性试验，结果表明无论从宏观观察的症状，或从微观上的组织病理形态学检查及血常规肝功能、肾功能化验均说明该药长期毒性小。
4.临床试验：由甘肃省中医院进行临床实验，观察《参龙虫草益肾胶囊》治疗肾阳虚衰，气虚血亏导致的阳萎60例，其中治疗组与对照组各30例，并在治疗组中观察治疗前后的免疫试验变化30例。
4.1受试者：所观察60例患者。选定患者时均按《参龙虫草益肾胶囊》临床试验计划规定的：(1)中医诊断阳萎标准及肾阳虚型者；(2)西医诊断为功能性阳痿患者的标准条件确诊。
4.2观察时间及投药方法：
观察时间定两周为一疗程，两疗程为一治疗周期。投药方法为治疗组口服《参龙虫草益肾胶囊》，每日三次，每次五粒，连服两周，四周后分别填写观察变化。对照组口服《男宝》，每日二次，每次三粒，其填写观察记录同治疗组；两组药物观察期间均停服一切相关药物及能促进性功能低下的药物。并要求忌用烟酒和辛辣刺激性食物。
4.3观察方法、项目及疗效标准：
4.3.1在选择病例同时要剔除一些不适应证，其排除的标准为：①确诊为器质性阳萎者；②某些药物导致的阳萎者；③配偶有全身严重器质性疾病者；④合并有心血管、肝、肾和造血系统等严重原发疾病、精神病患者等；⑤对本药服后发生过敏反应者；⑥未按规定用药或兼服其它药物，无法判断疗效或资料不全等影响疗效和安全性判断者等。
4.3.2疗效性观测：包括中医临床表现，如机体状况，阳萎程度、阴茎是否寒凉、腰膝是否畏寒，有无精冷滑泄、舌诊脉诊等西医性功能表现情况：像性欲、阴茎勃起程度、性交持续时间、有无遗精、滑精，早泄情况等。
4.3.3安全性观测：包括一般体检项目、血尿常规化验、肝、肾功能化验等。
4.3.4免疫性观测：对治疗组病例要在治疗前后分别检测血清补体C3T细胞计数、免疫球蛋白IgA、IgG、IgH的变化，并做出统计、分析和评定。
4.3.5对疗效判断的标准定位：①近期治愈：治疗三个月内阴茎勃起大于90°，性交机会的75％以上能成功；②显效；治疗后勃起＞90°，性交机会的50％以上能成功；③有效：治疗后勃起有改善，性交机会的25％以上能成功；④无效：用药前后各项指标均为改善。

通过上表可以看出，治疗组明显优于对照组，经两周治疗后，治疗组就开始出现疗效，其数字高于对照组，当治疗四周后其优势更明显于对照组，尤其在肾阳衰、命火不足所致的阳茎寒凉，腰膝畏寒、精冷滑泻诸证方面优于对照组。对于身体一般状况治疗组也有一定改善，治疗前经常感冒的患者，经治疗后就感冒减少，身体虚弱状况也开始好转和改善。

通过上表可看出，治疗组在性功能的阴茎勃起角度、性交能力上均优于对照组，而且对早些的疗效尤为明显。

从以上表可看出，治疗组在近期治愈和显效方面均高于对照组。在从有效率方面治疗组更明显高于对照组。
结果表明：
1、本发明胶囊剂治疗肾阳虚衰，气虚血亏具有明显疗效，临床有效率94.08，对照组75.66％，明显高于对照组，P均小于0.01。
2、主要症状体征起效和消失时间，主要症状体征平均起效和消失时间明显短于对照组，P小于0.05，主要症状体征平均下降分明显高于对照组。
3、腰部病理检查：用药前后病理检查，治疗组30例，对照组20例，综合判断各项指标治疗后平均降分，治疗组明显高于对照组，P小于0.01，说明参龙虫草益肾胶囊对肾阳虚衰，气虚血亏病灶有明显改善作用。

Claims (8)
1、一种治疗肾阳虚衰，气虚血亏的中药组合物，是由以下重量比份的原料采用药剂学常用的辅料和工艺制备而成：
人 参10～30份 枸杞子12.5～37.5份 杜 仲9～27
肉 桂2～6份 淫羊藿15～45份 锁 阳9～27份
肉苁蓉12.5～37.5份 附 子5～15份 菟丝子12.5～37.5份
熟地黄12.5～37.5份 山茱萸12.5～37.5份 仙 茅12.5～37.5份
韭菜子12.5～37.5份 黄 芪15～45份 当 归12.5～37.5份
鹿 鞭10～30份 鱼 鳔10～30份 五味子12.5～37.5份
海 龙10～30份 海 马10～30份 蛤 蚧10～30份
阿 胶9～27份 龟甲胶9～27份 冬虫夏草9～27份。
2、如权利要求1所述的一种治疗肾阳虚衰，气虚血亏的中药组合物，其特征在于：采用中药药剂学的常规工艺及辅料，制备成任何一种中药口服制剂。
3、如权利要求2所述治疗肾阳虚衰，气虚血亏的中药组合物，其特征在于：采用制备水煎剂的常规工艺制备成水煎剂。
4、如权利要求2所述治疗肾阳虚衰，气虚血亏的中药组合物，其特征在于：采用制备颗粒剂的常规工艺及辅料制备成颗粒剂。
5、如权利要求2所述治疗肾阳虚衰，气虚血亏的中药组合物，其特征在于：采用制备片剂的常规工艺及辅料制备成片剂。
6、如权利要求2所述治疗肾阳虚衰，气虚血亏的中药组合物，其特征在于：采用制备散剂的常规工艺制备成散剂。
7、如权利要求2所述治疗肾阳虚衰，气虚血亏的中药组合物，其特征在于：采用制备胶囊剂的常规工艺及辅料制备成胶囊剂。
8、如权利要求2所述一种治疗肾阳虚衰，气虚血亏的中药组合物，其特征在于：采用制备丸剂的常规工艺及辅料制备成丸剂。
