1. Safety test
Referring to cosmetic technical Specification 2015, the safety of the mild shampoo and bath foam for infants prepared in examples 1-3 and comparative examples 1-5 was verified by using a human skin closed patch test.
The test method comprises the steps of selecting 80 healthy subjects with age distribution of 18-55 years old, dividing the healthy subjects into 8 groups, dividing the healthy subjects into 10 persons in each group, respectively placing the infant mild shampoo and the bath shampoo prepared in the examples 1-3 and the comparative examples 1-5 into different groups of spot testers according to a closed spot paste test method, using the shampoo and the bath shampoo in an amount of about 0.020-0.025 mL, applying the spot testers added with the test objects on the curved sides of the forearms by using 3M adhesive tapes, uniformly applying the spot testers on the skin by using palm light pressure, continuing to record the reaction results according to skin reaction score grades after removing the spot testers for 30min, 24h and 48h, and taking average values of the score grade results as shown in Table 2.
Skin response scores were rated as follows:
0, negative reaction;
1, suspicious reaction, only weak erythema;
2, weak positive reaction (erythema reaction) erythema, infiltration, edema, and possibly pimple;
A strong positive response (herpes response) of erythema, infiltration, edema, papule, herpes, a response beyond the test area;
4, extremely strong positive reaction (fusion herpes reaction) with obvious erythema, severe infiltration, edema and fusion herpes, and reaction beyond the tested area.
Table 2:
according to the analysis of the skin response scoring grade test results of the examples 1-3, the comparative examples 1-5 and the table 2, the infant soothing and mild shampoo and bath lotion prepared by the examples 1-3 and the comparative examples 1-5 has no irritation, and has good mildness and safety.
2. Promoting hyaluronic acid production assay
Taking human skin keratinocytes (HaCat cells) cultured to logarithmic phase, regulating the cell concentration to be 1 x 10 5 cells/mL cell suspension by adopting a DMEM liquid culture medium, inoculating 500 mu L cell suspension into a 96-well plate, culturing for 24 hours under the conditions of 37 ℃ and 5% CO 2, sucking the culture medium, adding 100 mu L of the DMEM culture medium into a negative control group, respectively containing 5wt% of the mild shampoo for infants and infants of examples 1-3 and comparative examples 1-5 into 100 mu L of the DMEM culture medium, continuously culturing for 24 hours under the conditions of 37 ℃ and 5% CO 2, collecting cell culture medium supernatants of each group, detecting the synthesis condition of hyaluronic acid of each group by referring to a human hyaluronic acid ELISA kit, calculating the relative concentration (%) of hyaluronic acid of the experiment group by taking the synthesis condition of hyaluronic acid of the negative control group as a benchmark (100%), and recording the detection results in Table 3.
