Therefore, the invention aims to provide the essence containing the osmanthus fragrans extract with the functions of relieving the tightening and resisting the wrinkle and the preparation method, and the essence containing the osmanthus fragrans extract with the functions of relieving the tightening and resisting the wrinkle and the preparation method can not only promote cells to secrete collagen, but also obviously reduce the content of MMP-1, thereby reducing the degradation of the collagen, being beneficial to increasing the skin elasticity, improving the skin tightening degree, repairing damaged skin, promoting healing, and having the functions of resisting aging and repairing.
In order to achieve the above purpose, the present invention adopts the following technical scheme:
the essence containing the osmanthus fragrans extract comprises, by mass, 10-30 parts of the osmanthus fragrans extract, 2-10 parts of the boswellia serrata seed extract, 2-10 parts of betaine, 2-10 parts of pentapeptide, 1-5 parts of hydrogenated soybean lecithin, 1-5 parts of xylitol and 60-80 parts of water, wherein the solid content of the osmanthus fragrans extract is 3-5 mg/mL.
The Olibanum seed is seed of Olibanum (Schinus terebinthifolius). The Chinese zodiac Boswellia is also called Brazil pepper or Brazil pepper tree, is a common plant, is native to Brazil and other areas of south america, is a evergreen shrub or small arbor, and has bright red fruits and dark green leaves. Seeds and leaves of mastic gum contain some beneficial ingredients, such as volatile oils, tannins, phenolic compounds, etc., which impart some medicinal and flavoring uses to mastic gum. The Chinese zodiac olibanum seed can be used as medicinal plant, and has antioxidant effect.
Betaine (Betaine) is a natural compound commonly used in cosmetics as a moisturizer and anti-irritant. It can help regulate the osmotic pressure of cosmetics, thus having the function of moisturizing, keeping the skin moisture and improving the softness and smoothness of the skin. In addition, the betaine has the characteristics of comfort and anti-irritation, is beneficial to relieving the uncomfortable feeling of skin, and has a certain protection effect on sensitive skin.
Five peptides with CAS number 214047-00-4. The pentapeptide can stimulate the body to generate specific immune response, inhibit neurotransmitter release, reduce facial muscle contraction, play a good role in resisting oxidization and wrinkle, promote collagen synthesis and regeneration, and have a good effect in improving skin.
Preferably, the osmanthus flower is golden osmanthus flower (Laurus nobilis l.).

Preferably, the preparation method of the osmanthus fragrans flower extract comprises the following steps:
crushing osmanthus fragrans, mixing with water, distilling under reduced pressure to obtain distillate, and concentrating the distillate until the solid content is 3-5 mg/mL to obtain osmanthus fragrans extract;
The feed liquid ratio of the osmanthus fragrans to the water is 1 (10-20) g/mL, the temperature of reduced pressure distillation is 80-90 ℃, the time is 2-3 h, and the pressure is-0.09 MPa to-0.1 MPa.
Preferably, the preparation method of the osmanthus fragrans flower extract comprises the following steps of:
(1) Fresh flos Osmanthi Fragrantis (Laurus nobilis L.) flowers are taken, the surface ash of petals is cleaned, crushed, and placed in a steam generator, and heated for 10min under 90 ℃ steam.
(2) Placing the osmanthus powder obtained in the step (1) into a distillation flask of a rotary evaporator, adding deionized water into the distillation flask, wherein the feed liquid ratio of the osmanthus powder to the deionized water is 1:10g/mL, then carrying out reduced pressure distillation for 3 hours at 80 ℃ and minus 0.09MPa, controlling the rotating speed of the rotary evaporator to be 70r/min, repeating the operation, carrying out secondary distillation, mixing the distilled liquid obtained by the secondary distillation to obtain a mixed liquid, sterilizing, concentrating, and controlling the solid content of the mixed liquid to be 5mg/mL to obtain the osmanthus extract.
Preferably, the preparation method of the extract of the Chinese zodiac olibanum seed comprises the following steps:
mixing the Chinese zodiac seed with ethanol, extracting for 1-2 hours at 50-60 ℃, concentrating under reduced pressure, and freeze-drying the obtained product to obtain the Chinese zodiac seed extract.
The preparation method of the Chinese zodiac olibanum seed extract comprises the steps of taking Chinese zodiac olibanum seeds, crushing, adding 45% v/v ethanol solution, extracting for 2 times at 50-60 ℃ with the feed liquid ratio of Chinese zodiac olibanum seed powder to ethanol solution being 1 (8-10) g/ml, concentrating under reduced pressure for 30min each time, freezing the product at-18 ℃ for 24 hours, and sublimating and resolving drying at-10 ℃, 50Pa,30 ℃ and 20Pa respectively to obtain the Chinese zodiac olibanum seed extract.
Preferably, the essence containing the osmanthus fragrans extract with the functions of relieving tightening and resisting wrinkle further comprises a preservative and/or a stabilizer, wherein the preservative comprises phenoxyethanol, and the stabilizer comprises EDTA disodium.
Preferably, the essence containing the osmanthus fragrans extract with the functions of relieving and tightening and resisting wrinkle comprises the following components in parts by mass:
10 parts of osmanthus fragrans extract, 6 parts of Chinese zodiac olibanum seed extract, 4 parts of betaine, 5 parts of pentapeptide, 2 parts of hydrogenated soybean lecithin, 2 parts of xylitol, 0.5 part of EDTA disodium, 1 part of phenoxyethanol and 69.5 parts of water;
or the essence containing the osmanthus fragrans extract with the functions of relieving and tightening and resisting wrinkle and light wrinkles comprises the following components in parts by mass:
20 parts of osmanthus fragrans extract, 5 parts of Chinese zodiac olibanum seed extract, 3 parts of betaine, 4 parts of pentapeptide, 2 parts of hydrogenated soybean lecithin, 2 parts of xylitol, 0.5 part of EDTA disodium, 1 part of phenoxyethanol and 62.5 parts of water;
or the essence containing the osmanthus fragrans extract with the functions of relieving and tightening and resisting wrinkle and light wrinkles comprises the following components in parts by mass:
15 parts of osmanthus fragrans extract, 4 parts of Chinese zodiac olibanum seed extract, 3 parts of betaine, 6 parts of pentapeptide, 2 parts of hydrogenated soybean lecithin, 2 parts of xylitol, 0.5 part of EDTA disodium, 1 part of phenoxyethanol and 66.5 parts of water.
The invention also provides a preparation method of the essence containing the osmanthus fragrans extract with the functions of relieving and tightening and resisting wrinkle, which comprises the following steps:
(1) Mixing flos Osmanthi Fragrantis extract, olibanum seed extract, betaine, pentapeptide and water, and heating to 50deg.C to obtain water phase;
mixing hydrogenated soybean phospholipid, EDTA disodium, xylitol and phenoxyethanol, and heating to 50deg.C to obtain oil phase;
(2) Mixing the oil phase and the water phase, and shearing for 5-15 min under the condition of 10000-12000 rpm to obtain liposome colostrum;
(3) Homogenizing the liposome colostrum under high pressure of 500-600 bar to obtain the essence containing the osmanthus fragrans extract with the functions of relieving and tightening and resisting wrinkle.
Preferably, the preparation method of the essence containing the osmanthus fragrans extract with the functions of relieving and tightening and resisting wrinkle comprises the following steps of:
(1) Weighing flos Osmanthi Fragrantis extract, olibanum seed extract, betaine and pentapeptide, respectively, adding into water, heating to 50deg.C, and stirring to thoroughly mix the system;
(2) Respectively weighing hydrogenated soybean phospholipid, EDTA disodium, xylitol and phenoxyethanol, mixing, heating the system to 50 ℃, and stirring to uniformly mix the system;
(3) Adding the oil phase into the water phase, and shearing for 9min under 10000rpm to obtain liposome colostrum;
(4) Homogenizing liposome colostrum under 500bar for 3 times to obtain essence containing flos Osmanthi Fragrantis extract with effects of relieving tightening and removing wrinkle.
The invention also provides application of the essence containing the osmanthus fragrans extract with the functions of relieving and tightening and resisting wrinkle and reducing the wrinkle in anti-inflammatory.
Preferably, the inflammatory factor comprises TNF- α.
Preferably, the concentration of the essence containing the osmanthus fragrans extract with the functions of relieving and tightening and resisting wrinkle is 10-20wt%.
The invention also provides application of the essence containing the osmanthus fragrans extract with the functions of relieving and tightening and resisting wrinkle and reducing wrinkles in anti-aging.
The invention also provides application of the essence containing the osmanthus fragrans extract with the functions of relieving and tightening and resisting wrinkle and the essence containing the osmanthus fragrans extract with the functions of reducing the content of matrix metalloproteinase and/or promoting collagen secretion.
Preferably, the collagen comprises type I collagen.
Preferably, the concentration of the essence containing the osmanthus fragrans extract with the functions of relieving and tightening and resisting wrinkle is 10-20wt%.
Compared with the prior art, the essence containing the osmanthus fragrans extract with the functions of relieving and tightening and resisting wrinkle and the preparation method thereof comprise 4 active ingredients, namely the osmanthus fragrans extract, the frankincense seed extract, the pentapeptide and the betaine, and the active ingredients are wrapped by liposome, so that the essence has a good anti-inflammatory effect, and the content of MMP-1 can be obviously reduced, thereby reducing the degradation of collagen, being beneficial to increasing the skin elasticity, improving the skin tightness, repairing damaged skin, promoting healing and having the effects of resisting aging and repairing.

The technical solutions of the present invention will be clearly and completely described in conjunction with the embodiments of the present invention, and it is apparent that the described embodiments are only some embodiments of the present invention, but not all embodiments. All other embodiments, which can be made by those skilled in the art based on the embodiments of the invention without making any inventive effort, are intended to be within the scope of the invention.
In order to further illustrate the present invention, the following examples are provided. The raw materials used in the following examples and comparative examples of the present invention are commercially available, and the raw material companies listed below are one of the routes for purchasing the raw materials.
The betaines used in the examples and comparative examples were purchased from Shanghai Seiyaku Biotechnology Co., ltd. Under the designation S18046, and the pentapeptides were purchased from Shanghai Seiyaku Biotechnology Co., ltd. Under the designation B28580.
The preparation method of the osmanthus fragrans extract used in the embodiment and the comparative example comprises the following steps:
(1) Fresh flos Osmanthi Fragrantis (Laurus nobilis L.) flowers are taken, the surface ash of petals is cleaned, crushed, and placed in a steam generator, and heated for 10min under 90 ℃ steam.

The preparation method of the extract of the Chinese zodiac olibanum seeds used in the examples and the comparative examples comprises the following steps:
Pulverizing Olibanum seed, adding 45% v/v ethanol solution, extracting at 60deg.C for 2 times (30 min each time), concentrating under reduced pressure, freezing at-18deg.C for 24 hr, sublimating, resolving and drying under-10deg.C, 50Pa,30 deg.C and 20Pa respectively to obtain Olibanum seed extract.

The raw materials and contents of the osmanthus fragrans extract-containing essence with the effects of relieving tightening and wrinkle reducing in examples 1-3 of the present invention are shown in Table 1.

The preparation method of the osmanthus fragrans extract-containing essence with the functions of relieving tightening and wrinkle reducing in examples 1-3 comprises the following steps:
(1) Weighing flos Osmanthi Fragrantis extract, olibanum seed extract, betaine and pentapeptide according to the ratio in table 1, respectively, adding into water, heating to 50deg.C, and stirring to thoroughly mix the system;
(2) Respectively weighing hydrogenated soybean phospholipid, EDTA disodium, xylitol and phenoxyethanol, mixing, heating the system to 50 ℃, and stirring to uniformly mix the system;
(3) Adding the oil phase into the water phase, and shearing for 9min under 10000rpm to obtain liposome colostrum;
(4) Homogenizing liposome colostrum under 500bar for 3 times to obtain essence containing flos Osmanthi Fragrantis extract with effects of relieving tightening and removing wrinkle.
Comparative example 1
The comparative example was different from example 1 only in that the extract of boswellia serrata seeds was omitted and the lacking amount of boswellia serrata seed extract was distributed to the corresponding components in proportion of osmanthus fragrans extract, betaine and pentapeptide, with reference to the components and preparation method of example 1. The remaining steps were carried out as described in example 1.
Comparative example 2
The comparative example was different from example 1 only in that betaine was omitted and the amount of betaine missing was distributed to the corresponding components in proportion to osmanthus fragrans extract, boswellia serrata seed extract, and pentapeptide, with reference to the components and preparation method in example 1. The remaining steps were carried out as described in example 1.
Comparative example 3
The comparative example was different from example 1 only in that the comparative example omits the pentapeptide, and the amount of the pentapeptide missing was distributed to the corresponding components in proportion to the osmanthus fragrans extract, betaine, and boswellia serrata seed extract, with reference to the components and the preparation method in example 1. The remaining steps were carried out as described in example 1.
Comparative example 4
The comparative example is different from example 1 only in the components of example 1, and the preparation method of the essence of the comparative example comprises the following steps:
The components were weighed according to the ratios in Table 1, mixed, heated to 50℃and sheared for 20min at 1000rpm to obtain the essence.
Test example 1
The anti-inflammatory effect of the essence obtained in the examples and the comparative examples is explored in the test example, and the test result is shown in Table 2 with specific reference to T/SHRH 033-2020, "cosmetic soothing efficacy test-in vitro TNF-alpha inflammatory factor content determination lipopolysaccharide-induced macrophage RAW 264.7 test method", wherein the addition concentration of LPS is 1mg/mL, the test content of the essence is 10 wt%.

As shown in Table 2, compared with the comparative example, the essence provided by the example of the invention can obviously inhibit the inflammatory factor TNF-alpha.
Test example 2
The test example explores the promotion effect of the essence obtained in the examples and the comparative examples on collagen secretion, and specifically refers to T/SHRH 031 2020 'cosmetic tightening and anti-wrinkle efficacy test in vitro fibroblast type I collagen content determination', the test content of the essence is 10wt%, and the test results are shown in Table 3.

As shown in table 3, compared with the comparative example, the essence provided by the example of the invention can obviously promote fibroblasts to secrete type I collagen.
Test example 3
Matrix Metalloproteinases (MMPs) are a class of proteolytic enzymes capable of degrading extracellular matrix (such as collagen, elastin, proteoglycans, etc.), and matrix metalloproteinase 1 (MMP-1) is one of the MMP families, which mainly degrades type I collagen. MMP-1 content in normal dormant tissues is extremely low, but MMP-1 expression is promoted after ultraviolet radiation, abnormal collagen degradation is increased, and strength and elasticity of skin are reduced, so that skin aging is caused.
The test example explores the inhibition effect of the essence obtained in the examples and the comparative examples on matrix metalloproteinase, and specifically comprises the following steps:
(1) Human Skin Fibroblasts (HSF) in an exponential growth phase are taken, added with 0.25% trypsin digestion solution, digested to enable adherent cells to fall off, counted to be 1 multiplied by 10 6/mL, and made into cell suspension.
(2) 10 5 Cell suspensions per well were appropriately diluted and inoculated onto 6-well plates until the cells were grown to about 80% full, and a UV photoaging model was established. A blank control group of 200. Mu.L PBS was set, medium was supplemented to 800. Mu.L without UV irradiation, a UV group of UV irradiation, PBS was added, and an experimental group of the essence (1 wt%) obtained in the examples and comparative examples was set.
(3) The UV group and the experimental group were repeatedly washed to colorless by adding an appropriate amount of PBS, adding 200. Mu.L of PBS, and irradiating under a UV lamp of 80mJ/cm 2 with a lamp-to-flask spacing of 15cm. After irradiation, PBS was discarded, the UV group was added to the PBS solution and medium to 800. Mu.L, the experimental group was added to the medium and the sample to be tested to 800. Mu.L, and incubation was continued in the incubator for 48h for the blank, UV group, and experimental group.
(4) Scraping cells, beating and mixing uniformly, extracting cellular protein by using RIPA, quantifying BCA to 1mg/mL, and operating according to the operation instruction of matrix metalloproteinase 1 ELISA. The OD of each well was measured at 450nm with an ELISA reader over 15min and the experimental results are shown in Table 4.
