Abstract
The invention relates to the technical field of traditional Chinese medicines, in particular to a traditional Chinese medicine composition with detoxifying and blood cooling effects, a detoxifying and blood cooling mixture, and a preparation method and application thereof. The traditional Chinese medicine composition takes dyers woad leaf, radix isatidis and bunge corydalis herb as monarch drugs, plays the roles of detoxification and cooling blood, takes radix scutellariae, honeysuckle, fructus forsythiae, lithospermum, schizonepeta and radix angelicae as ministerial drugs for dispelling wind and clearing heat, takes radix scrophulariae, sanguisorba, wild chrysanthemum and radix paeoniae rubra as ministerial drugs, and takes liquorice as messenger to harmonize the drug properties, and finally achieves the effect of treating acne. According to the specific embodiment, the traditional Chinese medicine composition can effectively treat acne, and the treatment course is short; meanwhile, the traditional Chinese medicine composition is a common traditional Chinese medicine, and has very little side effect.
Description
Technical Field
The invention relates to the technical field of traditional Chinese medicines, in particular to a traditional Chinese medicine composition with detoxifying and blood cooling effects, a detoxifying and blood cooling mixture, and a preparation method and application thereof.
Background
Acne, also known as comedo, is a chronic inflammatory skin condition that occurs well in puberty and primarily involves the pilosebaceous glands of the face. Meanwhile, acne is the most common skin chronic inflammatory disease in the world, and is often caused at the position with vigorous sebum secretion of hair follicles, and the quality of life of many people is influenced by large-area skin lesions. At present, the pathogenesis of acne is not completely elucidated, for example, hormone-induced sebaceous gland transitional bleeding lipid, abnormal keratosis of a pilosebaceous canal, abnormal proliferation of hair follicle microorganisms such as propionibacterium acnes and staphylococcus epidermidis, inflammation, immune response and the like can be related to the pathogenesis of acne. Acne is frequently found in puberty, but not puberty, and is often found in sebaceous gland-rich parts such as face, back and chest, and is classified clinically according to skin loss properties in a three-degree four-level manner, namely: mild (grade i): only acne, manifested by open comedones (blackheads), closed comedones (whiteheads); moderate (grade ii): inflammatory papules; moderate (grade iii): pustules appear; severe (grade iv): has tubercles and cysts. Inflammatory papules, pustules, and abscesses may cause severe pigmentation and even scarring after healing.
At present, the acne is mostly treated by antibacterial drugs. However, the long-term use of antimicrobial drugs has serious effects on skin micro-ecology, such as damage to cell integrity, and hair follicle damage. Therefore, it is necessary to find a medicament for treating acne which has a short course of treatment, is effective and has no toxic or adverse effects.

Mixing Scutellariae radix, herba corydalis Bungeanae, fructus forsythiae, radix Arnebiae, flos Chrysanthemi Indici, radix Paeoniae Rubra, glycyrrhrizae radix, herba Schizonepetae and radix Angelicae Dahuricae, and performing first water extraction to obtain residue and first filtrate; carrying out second water extraction on the filter residues to obtain second filtrate; combining the first filtrate and the second filtrate to obtain an aqueous extract;
Concentrating the water extract, precipitating with ethanol, filtering, collecting filtrate, and removing ethanol to obtain fluid extract;

The preparation method specifically comprises the following steps:
Step 1: mixing radix Scutellariae, herba Violae, fructus forsythiae, radix Arnebiae, flos Chrysanthemi Indici, radix Paeoniae Rubra, radix Glycyrrhizae, herba Schizonepetae and radix Angelicae Dahuricae, extracting with water for 1.5 hr at a ratio of 1:10 (total mass of radix Scutellariae, herba Violae, fructus forsythiae, radix Arnebiae, flos Chrysanthemi Indici, radix Paeoniae Rubra, radix Glycyrrhizae, herba Schizonepetae and radix Angelicae Dahuricae to water ratio of 1:10), filtering to obtain residue and filtrate; carrying out second water extraction on the obtained filter residue, wherein the feed-liquid ratio is 1:9 (the mass ratio of the filter residue to water is 1:9), and filtering to obtain filter residue and filtrate; combining the two filtrates to obtain an aqueous extract;
Step 2: concentrating the water extract until the relative density of the obtained concentrated solution reaches 1.12 (measured at 50 ℃), stopping concentrating, adding 95% ethanol water solution for alcohol precipitation for 48h, filtering to obtain filtrate, and recovering ethanol in the filtrate to obtain extract;

The same materials and components as in example 1 were prepared, with the only differences: the complex enzyme preparation adopted by the synergistic fermentation of the bacterial enzymes comprises hemicellulase, cellulase, laccase and alkaline protease, wherein the enzyme activity of the hemicellulase in the complex enzyme preparation is 10000U/g, the enzyme activity of the cellulase is 20000U/g, the enzyme activity of the laccase is 7000U/g, and the enzyme activity of the alkaline protease is 10000U/g.
Comparative example 4
The same materials and components as in example 1 were prepared, with the only differences: the composite microbial inoculum adopted by the synergistic fermentation of the microbial enzymes consists of bacillus licheniformis, bacillus subtilis and lactococcus lactis, wherein the effective viable count of the bacillus licheniformis, the bacillus subtilis and the lactococcus lactis in the composite microbial inoculum is 1:1:2, namely the effective viable count of the bacillus licheniformis, the bacillus subtilis and the lactococcus lactis in the composite microbial inoculum is 1X 10 8CFU/g、1×108 CFU/g and 2X 10 8 CFU/g respectively.
Comparative example 5

Concentrating the water extract until the relative density of the obtained concentrated solution reaches 1.12 (measured at 50 ℃), stopping concentrating, adding 95% ethanol water solution for alcohol precipitation for 48h, filtering to obtain filtrate, and recovering ethanol in the filtrate to obtain extract;
taking 110mL of purified water, boiling, adding sucrose, stirring to dissolve, continuously heating to 100 ℃, filtering (200-mesh filter cloth), adding a proper amount of hot water, and cooling to room temperature to 250mL to obtain single syrup;
Mixing the extract, simple syrup, potassium sorbate and Borneolum Syntheticum to obtain the mixture for removing toxic substances and cooling blood.
Experimental example 1 toxicity experiment
Test animals: 110 Kunming mice, about 50g in weight, about 10 male and female animals treated in examples 1-5 and comparative examples 1-5 respectively, were subjected to 2 mL/day of gavage, and had no obvious toxic or side effects on animals before and after administration by using distilled water of the same volume as the control group, and the animals were not dead or abnormally represented by continuous observation for 14 days. The test results show that: the detoxification and blood cooling mixture of examples 1-5 and comparative examples 1-5 has no obvious toxicity to experimental animals, which suggests that the detoxification and blood cooling mixture of examples 1-5 and comparative examples 1-5 is safe for clinical application.
Experimental example 2 clinical experiments
In order to prove the curative effect of the detoxification and blood cooling mixture provided by the invention, 420 subjects are tested, and the specific steps are as follows:

The rich sebaceous glands such as facial and upper trunk parts include acne, folliculitis papules, pustules, nodules, etc.

(2) The method is effective: compared with the prior treatment, the acne, pustule, nodule and papule on the affected part skin surface are obviously reduced and relieved, and the pustule and papule on the focus part are obviously reduced. After partial healing, there remains significant pigmentation or pits, hypertrophic scars and crab foot swelling.
(3) Invalidation: the affected skin lesions are not relieved or aggravated compared with those before treatment, or recur after healing.
After four weeks, counting the number of healed patients, the number of effective patients, the number of ineffective patients and the total effective rate, wherein the calculation formula of the total effective rate is total effective rate= (healed patients + effective patients)/total number of patients multiplied by 100%; the number of recurrent persons was counted in a follow-up visit after half a year, the recurrent rate was calculated, the calculation formula was recurrent rate=recurrent persons/(healed persons+effective persons) ×100%, and the investigation results are shown in table 2.

As can be seen from Table 2, the detoxification and blood cooling mixture prepared by adopting the examples 1-5 of the invention can remarkably improve the total effective rate and reduce the recurrence rate. The medicine has obvious advantages compared with positive medicines (tetracycline tablet).
From the above, the detoxification and blood cooling mixture prepared in examples 1-5 provided by the invention can effectively treat acne without toxic or side effects.

15-20 parts of radix scutellariae, 20-30 parts of dyers woad leaf, 15-20 parts of radix isatidis, 25-30 parts of honeysuckle, 10-15 parts of radix scrophulariae, 10-15 parts of garden burnet, 25-35 parts of herba violae, 20-30 parts of fructus forsythiae, 20-30 parts of lithospermum, 15-25 parts of wild chrysanthemum, 15-20 parts of radix paeoniae rubra, 10-15 parts of liquorice, 20-25 parts of herba schizonepetae and 10-15 parts of radix angelicae.
2. The use of the Chinese medicinal composition according to claim 1 in preparing a mixture for removing toxic substances and cooling blood.
3. 4. A method for preparing the detoxification and blood cooling mixture as claimed in claim 3, comprising the following steps:
Mixing Scutellariae radix, herba corydalis Bungeanae, fructus forsythiae, radix Arnebiae, flos Chrysanthemi Indici, radix Paeoniae Rubra, glycyrrhrizae radix, herba Schizonepetae and radix Angelicae Dahuricae, and performing first water extraction to obtain residue and first filtrate; carrying out second water extraction on the filter residues to obtain second filtrate; combining the first filtrate and the second filtrate to obtain an aqueous extract;
concentrating, precipitating with ethanol, filtering, collecting filtrate, and removing ethanol to obtain fluid extract;
Mixing folium Isatidis, radix Isatidis, flos Lonicerae, radix scrophulariae and radix Sangusorbae with water, and performing synergistic fermentation with bacteria and enzymes to obtain fermentation broth;
Mixing the fluid extract, the fermentation liquor, the sucrose, the potassium sorbate and the borneol to obtain the detoxification and blood cooling mixture.

The method according to claim 4, wherein the complex enzyme preparation used in the synergistic fermentation of the bacterial enzymes comprises xylanase, cellulase, laccase and alkaline protease; the adopted composite microbial inoculum comprises bacillus licheniformis, bacillus coagulans and lactococcus lactis.
