技术领域
本发明属于中药制剂技术领域,具体地说,涉及一种防治儿童青少年近视的中药组合物眼膏及其制备方法。
背景技术
随着社会信息网络的快速发展,人们生活、学习、工作中用眼压力越来越大,儿童青少年学业繁重,用眼时间和强度较前增大很多,由此引起的儿童青少年近视眼发病越来越多。轻中度的近视眼,视远不清,眯眼看东西,戴眼镜,这些都给孩子们的生活、学习、成长、择业带来了诸多的不便;大于600度的高度近视尚伴有眼底的病理改变,如视网膜脱离、巩膜葡萄肿,使孩子视力严重受损,有失明危险,严重影响了儿童青少年的生活生命质量。
近视眼表现为近视力良好,视远处则模糊不清,常常眯眼看物,很容易眼疲劳。高度近视眼,眼珠较为突出,远视力严重下降。儿童青少年近视眼的发病原因主要是用眼过度,眼睛气耗津亏,经络不通,眼为肝之窍,眼伤则肝受损,久则亦伤肾,致肝肾两虚。
目前,防治近视眼的方法有戴眼镜,激光手术等。除治标不治本的弊病外,激光手术尚有对眼睛潜在的副作用。中医防治近视眼的方法有穴位针灸、眼贴眼膏、推拿按摩等。其中所用眼膏的中药配伍辨证不充分、不具体,组方笼统,用于包含近视眼在内的所有屈光不正患者。实际上,儿童青少年近视眼有自己特有的发病原因,应该给与单独的辨证论治,单独遣药组方。
目前,尚没有专门针对儿童青少年近视眼单独辨证论治的中药组方眼膏问世,本发明可填补这一空白。该眼膏发明组方独特,使用安全方便,可有效缓解儿童青少年近视的发生发展,助力孩子们的视力健康。
发明内容
本发明提供一种防治儿童青少年近视的中药组合物眼膏及其制备方法,该中药组合物眼膏针对儿童青少年近视眼的特有发病原因,辨证充分具体,中药遣方价廉,使用方便,安全有效。

一种防治儿童青少年近视的中药组合物眼膏的制备方法,包括以下步骤:
(1)中药组合物水煎过滤液:将黄芪、人参、当归、白芍、枸杞子、姜黄、艾草、野菊花、千里光、青皮、石菖蒲混合并置10倍质量的水中,煎煮2次,每次1h,合并煎出液,400目筛过滤,即得中药水煎过滤液;
(2)制作近视眼膏剂:将硬脂酸、单硬脂酸甘油酯、液体石蜡及白凡士林混合并组成油相,将中药水煎过滤液、三乙醇胺、甘油、吐温、尼泊金甲酯、山梨酸钾混合并组成水相,接着将油相、水相分别隔水加热至85℃,随后将油相缓缓倒入水相中并不断搅拌至膏体乳黄色、半固体状,然后将膏体冷却至55℃,随后加入珍珠粉、密陀僧粉,最后搅拌2h,直至膏体冷凝,即可。
本近视眼膏组方使用方法,每日3次,每次5分钟。眼周涂抹适量眼膏,食指自眉弓睛明穴始,沿上眼眶至眉稍瞳子髎穴,再到太阳穴,至下眶缘,至四白穴,至鼻内侧内眦侧结束,所经穴位处,食指按压收放停留,自体感觉酸胀痛为度。
相比于现有技术,本发明的有益效果为:
本发明中的君药为黄芪、人参,二者为补气血的良药,效果确切,作用缓和。儿童青少年为纯阳之体,阴液常不足,过度用眼极易耗气伤津,气血亏虚,经络迟滞,致使目昏不明,视远不清。黄芪、人参味甘,微温,可补气升阳,
生津养血,行滞通痹。针对儿童青少年用眼过度所致近视眼,对症施治。组方中二者为君药,用量最大。
用眼过度所致的视远物不清,气血亏虚,必然导致眼周经络不通,眼为肝之窍,眼周经络不通,必然伤肝,久之则肝肾两虚。组方中当归、白芍补血,
枸杞子补肝肾,姜黄、艾草温经通络,上述几味药为臣药,用量稍次于君药。
过度用眼所致眼周气滞津伤,肝肾两虚,往往表现有肝火上炎。组方中,野菊花,千里光,清热解毒,泻火平肝明目;珍珠粉,平肝潜阳,养肝阴,明目。青皮,行气破滞,长于舒肝郁。四者为佐药,用量少。石菖蒲,本品辛香走窜,苦燥温通,醒神益智,作用较为缓和,适宜于儿童青少年提神醒脑,是为使药,用量最小。
本发明中药组合物眼膏针对儿童青少年近视眼的特有发病原因,辨证充分具体,中药遣方价廉,使用方便,安全有效。

动物毒性试验
1.急性眼周皮肤过敏试验:
选用3月龄健康豚鼠10只,体重450—700g,雌雄各半。于给药前24h将豚鼠两眼间内上部位脱毛,面积约1cm*1cm。将实施例1近视眼膏剂均匀涂抹在脱毛区,用医用无刺激性胶布固定封闭24h,24h后去除胶布和眼膏,观察动物皮肤及全身中毒体征,观察期为7天,试验结束后拉颈法处死动物,进行大体病理观察。
试验结果:试验动物在受试期间无死亡,活动进食正常,全身皮肤未见异常。尸体大体尸检,心、肝、脾、肺、肾未见明显异常。
结论:豚鼠对受试眼膏无急性皮肤过敏反应
2.急性眼结膜刺激性试验
选用3月龄健康豚鼠6只,体重450—700g,雌雄各半。试验时将本申请中所得中药组合物水煎剂放置至常温,用无菌滴管吸取后滴入每只豚鼠左眼1滴,右眼做对照观察。滴入受试物后30min,1h,24h,48h,72h,观察豚鼠双眼结膜充血情况。
试验结果:滴入受试物后的各个时间点均未观察到眼部异常,观察期内,动物活动进食均正常,无动物死亡。
结论:该申请中所用中药组合物水煎液对豚鼠眼结膜无明显刺激性
近视眼膏的临床资料
1.病例来源
收集2021年11月至2022年4月,来自郑州市某眼科医院的近视青少年患者共240例。
2.诊断标准及分类
诊断标准参考《中华眼科学》中近视的诊断及分类标准拟定。近视诊断标准为眼在调节松弛的状态下,平行光线经眼的屈光系统后,在视网膜前形成焦点,远点移近,眼球轴长大于其屈光力的一种屈光不正。
近视程度分类:
1)轻度近视眼:<-3.0D的近视
2)中度近视眼:-3.0D~-6.0 D的近视
3)高度近视眼:>-6.0 D的近视
3.排除标准
1)与上述诊断及纳入标准不符合的
2)患者合并其它眼病,对疗效判定有影响者
3)患者有其它全身性疾病及精神病患者
4)正在接受其它治疗方法者
4.分组与治疗方法
将纳入观察的240例患者按照门诊就诊先后顺序,随机分为四组,全部病例均全程完成,无脱落病例。治疗前,各项检查结果两组间比较无统计学差异,资料具有可比性。其中,治疗1组60例,给予本发明实施例1近视眼膏治疗,观察期内,每天3次,眼周涂抹;治疗2组60例,给予本发明对比例1近视眼膏治疗,观察期内,每天3次,眼周涂抹;治疗3组60例,给予本发明对比例2近视眼膏治疗,观察期内,每天3次,眼周涂抹;治疗4组60例,给予博视润目型冷敷凝胶,观察期内,每天3次,眼周涂抹。2个月后,两组病例进行指标检测,观察疗效。
5.效果判断
使用效果分为有效和无效两种。
有效:原有自觉视不适感,如视物模糊、眼酸、眼胀、眼疼、头疼等消失;或者戴镜度数降低50度以上;或者裸眼远、近视力较前提高0.1以上。具备以上任何一条即为有效。
无效:仍存在自觉视不适感,如视物模糊、眼酸、眼胀、眼疼、头疼等;或者戴镜度数无改变;或者戴镜度数较前加深;或者裸眼远、近视力无改善;或者裸眼远、近视力较前下降0.1以上。
6.统计方法
采用SPSS17.0软件进行数据处理。各组内治疗的前后计量资料,采用配对t检验,组间比较采用方差分析,检验水准a=0.05,p<0.05则差异具有统计学意义。
7.观察结果

1.一种防治儿童青少年近视的中药组合物眼膏,其特征在于是由中药混合物和辅料制备而成,以重量份计:
中药混合物 44-82份,
辅料 441-579份;
所述的中药混合物的原料,以重量份计,为以下组分:
黄芪 7-11份,
人参 7-11份,
当归 4-8份,
白芍 4-8份,
枸杞子 4-8份,
姜黄 4-8份,
艾草 4-8份,
野菊花 2-4份,
千里光 2-4份,
青皮 2-4份,
石菖蒲 2-4份,
珍珠粉 2-4份;
所述的辅料,以重量份计,包括以下组分:
硬脂酸 90-110份,
单硬脂酸甘油酯 40-60份,
液体石蜡 40-60份,
白凡士林 90-110份,
三乙醇胺 40 -60份,
甘油 90 -110份,
吐温 5-15份,
尼泊金甲酯 3-7份,
山梨酸钾 3-7份,
密陀僧粉 20-40份。
2.根据权利要求1所述的防治儿童青少年近视的中药组合物眼膏,其特征在于是由中药混合物和辅料制备而成,以重量份计:
中药混合物 63份,
辅料 500份。
3.根据权利要求1所述的防治儿童青少年近视的中药组合物眼膏,其特征在于所述的中药混合物的原料,以重量份计,为以下组分:
黄芪 9份,
人参 9份,
当归 6份,
白芍 6份,
枸杞子 6份,
姜黄 6份,
艾草 6份,
野菊花 3份,
千里光 3份,
青皮 3份,
石菖蒲 3份,
珍珠粉 3份。
4.根据权利要求1所述的防治儿童青少年近视的中药组合物眼膏,其特征在于所述的辅料,以重量份计,包括以下组分:
硬脂酸 100份,
单硬脂酸甘油酯 50份,
液体石蜡 50份,
白凡士林 100份,
三乙醇胺 50份,
甘油 100份,
吐温 10份,
尼泊金甲酯 5份,
山梨酸钾 5份,
密陀僧粉 30份。
5.一种权利要求1-4任一项所述的防治儿童青少年近视的中药组合物眼膏的制备方法,其特征在于包括以下步骤:
(1)制备中药组合物水煎过滤液:将黄芪、人参、当归、白芍、枸杞子、姜黄、艾草、野菊花、千里光、青皮、石菖蒲混合并置10倍质量的水中,煎煮2次,每次1h,合并煎出液,400目筛过滤,即得中药水煎过滤液;
(2)制作近视眼膏剂:将硬脂酸、单硬脂酸甘油酯、液体石蜡及白凡士林混合并组成油相,将中药水煎过滤液、三乙醇胺、甘油、吐温、尼泊金甲酯、山梨酸钾混合并组成水相,接着将油相、水相分别隔水加热至85℃,随后将油相缓缓倒入水相中并不断搅拌至膏体乳黄色、半固体状,然后将膏体冷却至55℃,随后加入珍珠粉、密陀僧粉,最后搅拌2h,直至膏体冷凝,即可。
