Abstract
The invention provides a traditional Chinese medicine composition for treating furunculosis and a preparation method and application thereof, belonging to the technical field of traditional Chinese medicines and comprising the following raw materials in parts by weight: 20 to 30 parts of prepared rehmannia root, 20 to 35 parts of astragalus root, 10 to 20 parts of baical skullcap root, 10 to 20 parts of amur corktree bark, 5 to 15 parts of common burreed rhizome, 5 to 15 parts of zedoary, 15 to 25 parts of Chinese angelica and 5 to 10 parts of liquoric root. Meanwhile, the traditional Chinese medicine composition is non-toxic and harmless, high in safety and free of side effects.
Description
Technical Field
The invention relates to the technical field of traditional Chinese medicines, and in particular relates to a traditional Chinese medicine composition for treating furunculosis and a preparation method and application thereof.
Background
Furuncle is an acute, round, tender, localized hair follicle and deep infection around the follicle, and is called furunculosis due to multiple or repeated attacks, mainly caused by staphylococcus aureus infection. The traditional Chinese medicine considers that furuncle is caused by invasion of heat toxin into skin, belongs to pyocutaneous disease heat syndrome, and is most common on the head, the face, the neck, the back, the hip and the like. After furuncle, if the treatment is improper, the sore is too small, pus and diarrhea are not smooth, and sepsis is retained, or careless nursing is performed, scratch and bruise, and bruise and extrusion are generated, so that sepsis is caused to be side-blown, bacteria can enter the craniocerebra along with blood, and serious complications are caused. The patients with a large number of rashes which occur repeatedly and are not cured for a long time are called as furunculosis, although some furunculosis is treated, the recurrence is still inevitable, the furunculosis can be continuously generated in batches for months to years, particularly, children with poor nutrition and weak resistance have a lingering and frequent disease state, and the treatment difficulty is increased.
At present, the general methods for treating furunculosis comprise physical treatment, local treatment and drug treatment, wherein the physical treatment can be hot compress, ultraviolet therapy and the like, the local treatment can be iodine tincture external application, traditional Chinese medicine local external application and the like, the drug treatment can be antibiotic or multivalent staphylococcus bacterin treatment, but the deterioration of the furunculosis cannot be reversed after the furunculosis is suffered due to the reasons of bacterial drug resistance, insignificant curative effect of the physical or local treatment and the like, the treatment effect is poor, side effects can be caused, and various adverse consequences can be generated. Therefore, it is necessary to develop a Chinese medicinal composition for treating furunculosis with high efficiency.

The traditional Chinese medicine composition provided by the invention is used for treating the furunculosis, takes qi tonifying and blood circulation promoting, detoxifying and stasis removing, swelling subsiding and stasis removing as treatment principles, and has the effects of short treatment course, difficulty in relapse, remarkable curative effect, no side effect and the like.

Preferably, the traditional Chinese medicine composition comprises the following raw materials in parts by weight: 22 to 28 parts of prepared rehmannia root, 22 to 30 parts of astragalus root, 13 to 17 parts of baical skullcap root, 11 to 18 parts of amur corktree bark, 8 to 13 parts of common burreed rhizome, 6 to 14 parts of zedoary, 18 to 22 parts of Chinese angelica and 6 to 9 parts of liquoric root.
The invention also provides a preparation method of the traditional Chinese medicine composition, which comprises the following steps:
(1) Pulverizing the rhizoma sparganii, mixing the pulverized rhizoma sparganii with an ethanol solution, extracting, filtering, concentrating and drying to obtain rhizoma sparganii powder;
(2) Slicing Curcumae rhizoma, processing with vinegar, pulverizing, mixing with ethanol solution, performing supercritical extraction, separating, and drying to obtain Curcumae rhizoma powder;
(3) Mixing the rest materials, pulverizing, extracting with water, filtering, concentrating, and drying to obtain mixed powder;
(4) Mixing rhizoma Sparganii powder, curcumae rhizoma powder and the mixed powder, and pulverizing to obtain Chinese medicinal composition;
there is no order of limitation among (1), (2), and (3).
Preferably, in the step (1), the volume percentage of the ethanol solution is 30-90%, and the ratio of the water chestnut to the ethanol solution is 1:7-11.
Preferably, in the step (1), the extraction temperature is 50-70 ℃, the extraction time is 1-3 h, and the extraction times are 2-4.
Preferably, in the step (2), the vinegar processing of sliced curcuma zedoary comprises the following steps:
cutting the curcuma zedoary into 2-4 mm, mixing with rice vinegar, moistening, heating with slow fire until the rice vinegar is absorbed completely, and frying to be yellowish, wherein the mass ratio of the curcuma zedoary to the rice vinegar is 4-6:1.
Preferably, in the step (2), the material-liquid ratio of the curcuma zedoary to the ethanol solution is 1:4-8, the volume percentage of the ethanol solution is 5-10%, the pressure of the supercritical extraction is 24-28 Mpa, the extraction temperature is 45-50 ℃, and the extraction time is 1-2 hours.
Preferably, in the step (3), the mass-to-volume ratio of the residual raw material to water is 1:5-10, the extraction temperature is 90-100 ℃, the extraction time is 2-3 h, and the extraction times are 1-3 times.
Preferably, the pulverized particle size in step (1) is 20 to 40 mesh, the pulverized particle size in step (2) is 40 to 60 mesh, the pulverized particle size in step (3) is 40 to 60 mesh, and the pulverized particle size in step (4) is 100 to 150 mesh.
The invention also provides an application of the traditional Chinese medicine composition in preparing a medicament for treating furunculosis.
The invention has the following beneficial effects:
the traditional Chinese medicine composition for treating the furunculosis provided by the invention is simple in formula composition, adopts the rhizoma sparganii and the curcuma zedoary as monarch drugs, the prepared rehmannia root, the astragalus membranaceus and the angelica as ministerial drugs, the scutellaria baicalensis and the golden cypress as adjuvant drugs and the liquorice as conductant drugs, and is scientific and reasonable in compatibility, so that the traditional Chinese medicine composition has the effects of tonifying qi and activating blood, detoxifying and resolving masses, and diminishing swelling and removing blood stasis, achieves the purpose of synergy, remarkably improves the effective rate of treating the furunculosis, quickly cures the furunculosis, and is not easy to relapse. Meanwhile, the traditional Chinese medicine composition is non-toxic and harmless, high in safety and free of side effects.

The invention provides a traditional Chinese medicine composition for treating furunculosis, which comprises the following raw materials in parts by weight: 20 to 30 portions of prepared rehmannia root, 20 to 35 portions of astragalus root, 10 to 20 portions of baical skullcap root, 10 to 20 portions of amur corktree bark, 5 to 15 portions of common burreed rhizome, 5 to 15 portions of zedoary, 15 to 25 portions of Chinese angelica and 5 to 10 portions of liquoric root.
In the invention, the prepared rehmannia root is sweet in taste and slightly warm in nature, enters liver and kidney channels, has the effects of enriching blood and nourishing yin, and replenishing vital essence and marrow, and is used for treating blood deficiency and sallow complexion, palpitation and severe palpitation, metrorrhagia and metrostaxis, liver and kidney yin deficiency, soreness and weakness of waist and knees, bone steaming and tidal fever, night sweat and spermatorrhea, internal heat and thirst quenching, dizziness and tinnitus. The preferred weight part of the prepared rehmannia root is 22 to 28 parts, and more preferably 25 parts, and the source of the prepared rehmannia root is not particularly limited in the present invention, and products known in the art may be used.
In the invention, the astragalus membranaceus is sweet in taste and slightly warm, enters lung and spleen channels, has the effects of tonifying qi and invigorating yang, consolidating superficial resistance and arresting sweating, inducing diuresis to alleviate edema, promoting production of body fluid and nourishing blood, activating stagnancy and removing obstruction of qi, expelling toxin and pus, and healing sore and promoting tissue regeneration, is preferably 22-30 parts by weight, more preferably 25 parts by weight, has no special limitation on the source of the astragalus membranaceus, and can be prepared by adopting products known in the field.
In the invention, the scutellaria is bitter in taste and cold in nature, enters the channels of the lung, gallbladder, spleen, large intestine and small intestine, has the effects of clearing heat and drying dampness, purging fire and detoxifying and stopping bleeding, and is used for treating damp warmth, summer dampness, chest distress and nausea, damp-heat and fullness, diarrhea, jaundice, lung heat cough, high fever polydipsia, blood heat hematemesis and bleeding, carbuncle and sore toxicity. The preferable weight part of the scutellaria baicalensis is 13-17 parts, and the more preferable weight part is 15 parts. The source of the scutellaria baicalensis is not particularly limited, and products known in the field can be adopted.
In the invention, the phellodendron bark is bitter in taste and cold in nature, enters kidney and bladder channels, has the effects of clearing heat and drying dampness, purging fire and removing steam, detoxifying and treating sores, and is used for treating damp-heat diarrhea and dysentery, jaundice and dark urine, leukorrhagia and pruritus vulvae, heat stranguria and pain, bone steaming and fatigue heat, night sweat, pyocutaneous disease and pyogenic infections, and eczema. The phellodendron amurense is preferably 11 to 18 parts by weight, and more preferably 15 parts by weight. The source of the phellodendron amurense is not particularly limited in the invention, and products known in the field can be adopted.
In the invention, the rhizoma sparganii is pungent and bitter in taste and neutral in nature, enters liver and spleen channels, has the effects of breaking blood and promoting qi circulation, and removing food retention and relieving pain, and is used for treating abdominal masses, dysmenorrhea, blood stasis amenorrhea, chest stuffiness and pains, and food retention and distending pain. The weight of the trigone is preferably 8 to 13 parts, and more preferably 10 parts. The source of the rhizoma sparganii is not particularly limited in the invention, and products known in the field can be adopted.
In the invention, the curcuma zedoary is pungent and bitter in taste and warm in nature, enters liver and spleen channels, has the effects of promoting qi circulation, removing blood stasis, removing food retention, relieving pain and resisting bacteria, and is used for treating blood, qi and heart pain, food stagnation, abdominal distention pain, blood stasis amenorrhea, dysmenorrheal and traumatic injury. The preferable weight part of the zedoary is 6 to 14 parts, and the more preferable weight part is 10 parts. The source of the curcuma zedoary is not particularly limited, and products known in the art can be adopted.
In the invention, the angelica has sweet and pungent taste and warm nature, enters liver, heart and spleen channels, has the effects of enriching and activating blood, regulating menstruation and relieving pain, and relaxing bowel, and is used for treating blood deficiency chlorosis, irregular menstruation, blood deficiency, blood stagnation, traumatic injury, rheumatic arthralgia, carbuncle-abscess, pyocutaneous disease, intestinal dryness and constipation. The angelica is preferably 18 to 22 parts by weight, and more preferably 20 parts by weight. The source of the angelica is not particularly limited in the invention, and products known in the field can be adopted.
In the invention, the liquorice has flat nature and sweet taste, is in charge of heart, lung, spleen and stomach channels, has the effects of tonifying spleen and qi, clearing heat and removing toxicity, and harmonizing the medicines, is used for treating weakness of spleen and stomach, lassitude and hypodynamia, palpitation and shortness of breath, cough and excessive phlegm, abdominal and limb spasm and acute pain, carbuncle swelling and sore toxicity, and relieving drug toxicity and toxicity. The preferable weight part of the liquorice is 6 to 9 parts, and the more preferable weight part is 8 parts. The source of the liquorice is not particularly limited in the invention, and products known in the field can be adopted.

The invention also provides a preparation method of the traditional Chinese medicine composition, which comprises the following steps:
(1) Pulverizing the rhizoma sparganii, mixing the pulverized rhizoma sparganii with an ethanol solution, extracting, filtering, concentrating and drying to obtain rhizoma sparganii powder;
(2) Slicing Curcumae rhizoma, processing with vinegar, pulverizing, mixing with ethanol solution, performing supercritical extraction, separating, and drying to obtain Curcumae rhizoma powder;
(3) Mixing the rest raw materials, pulverizing, extracting with water, filtering, concentrating, and drying to obtain mixed powder;
(4) Mixing rhizoma Sparganii powder, curcumae rhizoma powder and the mixed powder, and pulverizing to obtain Chinese medicinal composition;
there is no order restriction among the above (1), (2), and (3).
In the invention, the common burreed rhizome is crushed and then mixed with ethanol solution for extraction, filtered, concentrated and dried to obtain the common burreed rhizome powder. The particle size of the crushed particles is preferably 20 to 40 meshes, more preferably 30 meshes, and the particle size of the invention can better ensure the quick dissolution of the active ingredients. The volume percentage of the ethanol solution is preferably 30 to 90 percent, more preferably 50 to 70 percent, and more preferably 65 percent, and the feed-liquid ratio of the mitsubishi to the ethanol solution is preferably 1:7 to 11, more preferably 1:8 to 10, and more preferably 1:9. The vinegar processing of the sliced curcuma zedoary preferably comprises the following steps: cutting the curcuma zedoary into 2-4 mm, mixing with the rice vinegar, moistening, heating with slow fire until the rice vinegar is absorbed completely, and frying to be yellowish, wherein the mass ratio of the curcuma zedoary to the rice vinegar is 4-6:1. The pulverized particle size of the present invention is preferably 40 to 60 mesh, and more preferably 50 mesh. The volume percentage of the ethanol solution is preferably 5-10%, more preferably 8-9%, the feed-liquid ratio of the curcuma zedoary to the ethanol solution is preferably 1:4-8, more preferably 1:5-7, the pressure of the supercritical extraction is preferably 24-28 Mpa, more preferably 25-26 Mpa, the extraction temperature is preferably 45-50 ℃, more preferably 47 ℃, and the extraction time is preferably 1-2 h, more preferably 1.5h. The invention can obviously improve the content of effective components of the curcuma zedoary, such as curcumenol and the like, through vinegar processing of the curcuma zedoary and supercritical extraction separation, thereby efficiently playing the roles of anticoagulation and antibiosis.
In the invention, the rest raw materials are mixed and crushed, mixed with water for extraction, filtered, concentrated and dried to obtain mixed powder. The pulverized particle size is preferably 40 to 60 mesh, and more preferably 50 mesh. The mass volume ratio of the residual raw materials to water is preferably 1:5-10, more preferably 1:8, the extraction temperature is preferably 90-100 ℃, more preferably 95 ℃, the extraction time is preferably 2-3 h, more preferably 2.5h, and the extraction times are preferably 1-3 times, more preferably 2 times. The invention can effectively ensure the dissolution of the effective components of the traditional Chinese medicine by the mode.
In the invention, the common burreed rhizome powder, the zedoary turmeric rhizome powder and the mixed powder are mixed and crushed to obtain the traditional Chinese medicine composition. The pulverized particle size is preferably 100 to 150 mesh, more preferably 110 to 140 mesh, and still more preferably 130 mesh. In the invention, the medicine takes the traditional Chinese medicine composition as a main component, and pharmaceutically acceptable carriers or auxiliary materials are added, wherein the pharmaceutically acceptable carriers or auxiliary materials comprise one or more of diluents, coloring agents, sweetening agents, coating agents, binding agents, absorbents, disintegrating agents, releasing agents, dispersing agents, wetting agents, cosolvent, buffering agents and surfactants. Example 1
A traditional Chinese medicine composition for treating furunculosis comprises the following raw materials by mass: 25g of prepared rehmannia root, 25g of astragalus root, 15g of scutellaria root, 15g of phellodendron bark, 10g of rhizoma sparganii, 10g of curcuma zedoary, 20g of angelica and 8g of liquorice;
grinding and crushing the rhizoma sparganii, sieving the crushed rhizoma sparganii with a 30-mesh sieve, mixing the crushed rhizoma sparganii with 100mL of a 65% ethanol solution, extracting the mixture at 60 ℃ for 2 hours, carrying out suction filtration, adding 100mL of the 65% ethanol solution into the filter residue, extracting the filter residue after the suction filtration for 2 hours at 60 ℃, extracting the filter residue again for 1 time in the above mode, combining the filter liquors after the suction filtration, concentrating the filter liquor under reduced pressure to 1/3 of the volume, and drying the filter liquor in a reduced-pressure drying box at 50 ℃ to constant weight to obtain the rhizoma sparganii powder;
cutting Curcumae rhizoma into 3mm pieces, mixing with 2g rice vinegar, moistening, heating with slow fire until the rice vinegar is absorbed completely, parching to slight yellow, pulverizing, sieving with 50 mesh sieve, mixing with 60mL 8% ethanol solution, performing supercritical extraction under 26Mpa at 47 deg.C for 1.5h, and naturally drying to obtain Curcumae rhizoma powder;
mixing the rest raw materials, pulverizing, sieving with 50 mesh sieve, mixing with 864mL of water, extracting at 95 deg.C for 2.5h, vacuum filtering, mixing the filter residue with 864mL of water, extracting at 95 deg.C for 2.5h, vacuum filtering, mixing filtrates, concentrating, and drying at 45 deg.C in a constant temperature drying oven to constant weight to obtain mixed powder;
mixing rhizoma Sparganii powder, curcumae rhizoma powder and the mixed powder, pulverizing, and sieving with 130 mesh sieve to obtain the Chinese medicinal composition.
Example 2
A traditional Chinese medicine composition for treating furunculosis comprises the following raw materials by mass: 20g of prepared rehmannia root, 35g of astragalus root, 10g of scutellaria root, 20g of phellodendron bark, 15g of common burreed rhizome, 5g of zedoary, 15g of angelica and 10g of liquorice;
grinding and crushing the rhizoma sparganii, sieving the crushed rhizoma sparganii with a 20-mesh sieve, mixing the crushed rhizoma sparganii with 165mL of 90% ethanol solution, extracting the mixture for 3 hours at 50 ℃, carrying out suction filtration, adding 165mL of 90% ethanol solution into the filter residue, extracting the filter residue for 3 hours at 50 ℃, extracting the filter residue after suction filtration for 2 times again in the above mode, combining the filter liquor after suction filtration, concentrating the filtrate under reduced pressure to 1/3 volume, and drying the filtrate in a reduced-pressure drying box at 50 ℃ to constant weight to obtain the rhizoma sparganii powder;
cutting Curcumae rhizoma into 2mm pieces, mixing with 1.25g rice vinegar, moistening, heating with slow fire until the rice vinegar is absorbed completely, parching to slight yellow, pulverizing, sieving with 60 mesh sieve, mixing with 20mL 10% ethanol solution, performing supercritical extraction at 24Mpa and 45 deg.C for 2 hr, and naturally drying to obtain Curcumae rhizoma powder;
mixing the rest raw materials, pulverizing, sieving with 40 mesh sieve, mixing with 1100mL water, extracting at 100 deg.C for 2h, filtering, concentrating, and drying at 45 deg.C in constant temperature drying oven to constant weight to obtain mixed powder;
mixing rhizoma Sparganii powder, curcumae rhizoma powder and the mixed powder, pulverizing, and sieving with 100 mesh sieve to obtain the Chinese medicinal composition.
Example 3
A traditional Chinese medicine composition for treating furunculosis comprises the following raw materials by mass: 30g of prepared rehmannia root, 20g of astragalus root, 20g of scutellaria root, 10g of phellodendron bark, 5g of rhizoma sparganii, 15g of curcuma zedoary, 25g of angelica and 5g of liquorice;
grinding and crushing the rhizoma sparganii, sieving the crushed rhizoma sparganii with a 40-mesh sieve, mixing the crushed rhizoma sparganii with 35mL of 30% ethanol solution, extracting for 1h at 70 ℃, carrying out suction filtration, adding 35mL of 30% ethanol solution into filter residue, extracting for 1h at 70 ℃, combining filtrates after suction filtration, concentrating under reduced pressure to 1/3 volume, and drying in a reduced-pressure drying oven at 50 ℃ to constant weight to obtain rhizoma sparganii powder;
cutting the curcuma zedoary into 4mm slices, mixing with 2.5g of rice vinegar, moistening, heating with slow fire until the rice vinegar is absorbed completely, frying to be yellowish, crushing, sieving with a 40-mesh sieve, mixing with 120mL of 5% ethanol solution, performing supercritical extraction separation at 28Mpa and 50 ℃ for 1h, and naturally drying to obtain curcuma zedoary powder;
mixing the rest raw materials, pulverizing, sieving with 60 mesh sieve, extracting with 550mL water, extracting at 90 deg.C for 3h, vacuum filtering, mixing the residue with 550mL water, extracting at 90 deg.C for 3h, extracting the filtered residue again for 1 time in the above manner, filtering, concentrating, and drying at 45 deg.C in a constant temperature drying oven to constant weight to obtain mixed powder;
mixing rhizoma Sparganii powder, curcumae rhizoma powder and the mixed powder, pulverizing, and sieving with 150 mesh sieve to obtain the Chinese medicinal composition.
Example 4
Inclusion criteria were: meets the diagnosis standard of repeated or acute suppurative folliculitis and perifollicular infection in Chinese clinical dermatology (3 rd edition), has a course of 1-2 weeks, and has no medicament for treating furunculosis by oral administration or external application within 2 weeks before treatment, or receives other treatments.
Exclusion criteria:

Clinical data:
160 patients with age of 6 months to 50 years and disease course of 1 week to 2 weeks are clinically diagnosed as furunculosis, wherein 80 men and 80 women are randomly divided into 2 groups by an envelope method, 80 patients in a treatment group and a control group respectively have no significant difference in the aspects of sex, age, disease course, skin severity degree score and the like (P is more than 0.05) and are comparable.
The treatment method comprises the following steps:
control group: the traditional Chinese medicine five-ingredient disinfection drink is orally taken, 15g of dandelion, honeysuckle, wild chrysanthemum and herba violae respectively and 10g of muskroot-like semiaquilegia root are decocted with water, 60mL of the decoction is orally taken each time, 3 times a day, and 7 days are continuously taken.
Treatment groups: the Chinese medicinal composition prepared in example 1 was administered with water 3 times daily for 7 consecutive days.
And (4) observation indexes are as follows: (1) symptoms, signs: the degree of skin lesions (erythema, nodules, pustules, and purulent secretions) and the degree of pain were scored on 4 scales, no (0 points), light (1 point), medium (2 points), heavy (3 points), respectively. The results in table 1 show that the effective rate of the traditional Chinese medicine composition for treating furunculosis is significantly higher than that of the control group, and that the traditional Chinese medicine composition has significant curative effect, and meanwhile, during the treatment period, the curative effect time (within 2 days) that the skin lesion begins to fade and the pain is significantly relieved by adopting the traditional Chinese medicine composition is earlier than that of the control group.
And (3) safety evaluation: the control group and the treatment group have no adverse reaction during and after the treatment period.
Recurrence of the disease: for two groups of recovery patients, the patients are followed for half a year, the treatment group has 3 relapses, the recurrence rate is 6.66 percent, the control group has 8 relapses, the recurrence rate is 26.67 percent, and the recurrence efficiency of the treatment group is obviously lower than that of the control group.

the preparation method of the traditional Chinese medicine composition comprises the following steps:
(1) Pulverizing the rhizoma sparganii, mixing the pulverized rhizoma sparganii with an ethanol solution, extracting, filtering, concentrating and drying to obtain rhizoma sparganii powder;
(2) Slicing Curcumae rhizoma, processing with vinegar, pulverizing, mixing with ethanol solution, performing supercritical extraction, separating, and drying to obtain Curcumae rhizoma powder;
(3) Mixing the rest materials, pulverizing, extracting with water, filtering, concentrating, and drying to obtain mixed powder;
(4) Mixing rhizoma Sparganii powder, curcumae rhizoma powder and the mixed powder, and pulverizing to obtain Chinese medicinal composition;
the (1), (2) and (3) are not limited in sequence;
in the step (1), the volume percentage of the ethanol solution is 30 to 90 percent, and the material-liquid ratio of the rhizoma sparganii to the ethanol solution is 1:7 to 11;
in the step (1), the extraction temperature is 50 to 70 ℃, the extraction time is 1 to 3h, and the extraction times are 2~4;
in the step (2), the vinegar processing of the rhizoma zedoariae slices comprises the following steps:
cutting the curcuma zedoary into 2-4 mm, mixing with rice vinegar, moistening, heating with slow fire after the rice vinegar is absorbed completely, and frying to be yellowish, wherein the mass ratio of the curcuma zedoary to the rice vinegar is 4~6:1;
in the step (2), the material-liquid ratio of the curcuma zedoary to the ethanol solution is 1:4~8, the volume percentage of the ethanol solution is 5 to 10%, the pressure of the supercritical extraction is 24 to 28Mpa, the extraction temperature is 45 to 50 ℃, and the extraction time is 1 to 2h.
