Abstract
本发明提供一种厨房油污清洗剂,其包括月桂酰精氨酸乙酯(LAE)离子对衍生物。本发明还提供制备含有所述离子对衍生物的油污清洗剂的方法及其制备的油污清洗剂。本发明的油污清洗剂,其LAE离子对衍生物具有天然无毒、高效抑菌、易于降解、环境友好的特点。
Description
技术领域
本发明涉及一种清洁用品,尤其涉及一种厨房用油污清洁用品。更具体地是涉及月桂酰精氨酸乙酯及其衍生物作为厨房清洁用品的应用。
背景技术
厨房的油污是一个让人头疼的问题,不但清理麻烦,而且严重影响人们的生活环境。厨房油污的主要成分是食用油,这些油脂在高温下能发生氧化聚集反应,便和其他油污夹杂在一起。受到高温后蒸发,形成像油漆一样的粘性油垢。油垢越来越粘,甚至变干硬结,使清洗越发困难。
传统厨房油污清洗剂多为碱性清洗剂,例如,将漂白粉溶液与油污发生皂化反应来去除油污,或是使用氢氧化钠的乙醇溶液作为碱性清洗剂。然而,这种清洗剂对皮肤有腐蚀作用,且易于损坏厨房设备,因此受到一定限制。
改进型的厨房用油污清洗剂主要由三个部分组成:表面活性剂、碱性助剂、有机溶剂。其中,表面活性剂包括以阴离子表面活性剂与非离子表面活性剂进行复配能制备性能良好的厨房油污清洗剂,而碱性助剂及有机溶剂有助于去污力的提高。
例如,在阴离子表面活性剂与非离子表面活性剂的复配组分中,脂肪醇聚氧乙烯醚(AEO)4-6%:是非离子型表面活性剂中发展最快、用量最大的品种,亲油基和亲水基分别是由具有活泼氢的脂肪醇和环氧乙烷聚合制得,是环氧乙烷加成数不同的多种聚氧乙烯醚的混合物。作为非离子表面活性剂,起乳化,发泡、去污作用。是洗手液、洗衣液、沐浴露、洗衣粉、洗洁精、金属清洗剂的主要活性成分。
烷基苯磺酸钠(LAS)2-5%:一种人工合成的洗涤剂。有硬性和软性两类。易氧化,起泡力强,去污力高,易与各种助剂复配,成本较低,合成工艺成熟,应用领域广泛,是非常出色的阴离子表面活性剂。
脂肪醇聚氧乙烯醚硫酸钠(AES)2-4%:易溶于水,具有优良的去污、乳化、发泡性能和抗硬水性能,温和的洗涤性质不会损伤皮肤。广泛应用于香波、浴液、餐具洗涤剂、复合皂等洗涤化妆用品;用于纺织工业润湿剂、清洁剂等。
烷基醇酰胺(6501)1-3%是非离子型表面活性剂,作为增稠剂和泡沫稳定剂,大量用于洗涤剂、化妆品、柴油乳化以及塑料中的抗静电剂、金属加工清洗剂和防锈剂、纺织助剂和油田开采驱油用表面活性剂。
三乙醇胺1-3%:在液体洗涤剂中加入三乙醇胺,可改进油性污垢,特别是非极性皮脂的去除,同时,通过提高碱性可提高去污性能。并且有极好的其相容性。
增溶调理剂:甲苯磺酸钠1-3%。
防腐剂:苯甲酸钠0.4-0.5%:苯甲酸钠属于酸性防腐剂在酸性环境下防腐效果较好,是很常用的食品防腐剂,有防止变质发酸、延长保质期的效果。
有机溶剂:如乙醇2-4%。
然而,以上传统清洗剂只是针对普通油脂污渍,对于顽固性或严重性油污,尤其是已经滋生大量细菌的油污是无能为力的。因此,越来越多的厨房清洗剂里开始加入具有抑菌作用的清洗剂成分。
清洗剂中常用的抑菌成分有甲醛、戊二醛、季铵盐类化合物(如苯扎氯铵)、苯甲酸钠或二苯甲酸钠等。然而,这些抑菌成分或是强致癌剂(如甲醛),或对皮肤和黏膜具有刺激性的毒性(戊二醛),或其生物降解能力较低,易于在环境中蓄积(如季铵盐类化合物)等,并不能作为天然环保型清洗剂成分。
目前天然环保型的油污清洗剂的抑菌成分,多数成分为椰子油、椰子粉、海洋矿物元素、小苏打、食醋、茶叶等,虽然这些清洗剂天然无毒易降解,但或是需要配合抑菌成分而调整了去污能力强的成分,或是抑菌能力有限,或是自身难以长期保存活性,以及成本过高,因此仍然需要改进。
因此,目前需要天然环保型的油污清洗剂,该清洗剂在保留现有清洗剂的去污能力强时,还具有易于长期保存、质量稳定、杀菌效果好且天然无毒易降解的优点。
月桂酰精氨酸乙酯(Ethyl lauroylarginate,LAE)是一种由脂肪酸和二元氨基酸缩合而成的有机物,为白色吸湿性固体,在pH3~7范围内化学性质稳定,熔点50~58℃,该温度下247g的LAE可分散于1kg的水中,它在水和油中的分配系数大于10,即主要存在于水相中。研究发现,月桂酰精氨酸乙酯LAE具有抗菌能力强、生物毒性低、体内代谢效果好、与环境相容性高的特点。而其中最具代表性的特点是月桂酰精氨酸乙酯代谢无残留,相关研究显示月桂酰精氨酸乙酯在人体与动物体内可快速进行自然代谢,首先被水解成月桂酰精氨酸(LAS)和乙醇,之后LAS又被水解为天然存在的膳食成分月桂酸和精氨酸,月桂酸会被进一步代谢为二氧化碳和水,精氨酸则被代谢为鸟氨酸、尿素及二氧化碳。月桂酰精氨酸乙酯代谢过程中所产生的所有初级代谢产物及终产物都是无毒无害的,与人和动物日常摄取的食物在体内的代谢产物相同。
例如,中国专利申请CN201710056593、发明名称为“一种果蔬防腐保鲜剂及其制备方法和应用”公开了以月桂酰精氨酸乙酯盐酸盐和尼泊金甲酯钠为主要活性成分的组合物来作为果蔬防腐保鲜剂,能够有效抑制导致果蔬腐烂的细菌生长。然而,该发明申请中高浓度的尼泊金甲酯钠(2000μg/ml)的单独抑菌效果强于低浓度的LAE(1000μg/ml),这是因为其具有酚羟基结构,抗菌性能远远强于苯甲酸、山梨酸,因此在保证防腐性能的前提下,该方法明确指出使用尼泊金甲酯钠代替LAE,有助于降低防腐剂的用量成本。
中国专利申请CN201510748675、发明名称为“采用月桂酰精氨酸乙酯抑制酒精发酵污染微生物的方法”公开了采用月桂酰精氨酸乙酯抑制酒精发酵污染微生物的方法,该方法包括将LAE及其盐类化合物,以低于50μg/ml的浓度加入酿酒酵母的发酵液中,能有效抑制乳酸菌的生长,并控制其他污染微生物的生长。然而,该抑菌剂在一定程度上轻微影响酵母菌的生长,并导致酒精产量降低0.6%。
中国专利申请CN201610466729、发明名称为“一种温和的婴童洗发沐浴泡泡”公开了一种温和的婴童洗发沐浴泡泡,其针对婴童毛发及肤质的特点,选用椰油酰谷氨酸二钠、椰油酰胺丙基甜菜碱及磺酸羟丙酯月桂基葡糖苷交联聚合物钠复配作为表面活性剂体系,选用山茶籽油、α-葡聚糖寡糖/菊粉复配物作为调理成分,野菊花提取物和月桂酰精氨酸乙酯HCl复配作为防腐体系,该发明申请中各原料互相协作,清洁效果好,温和无刺激。
中国专利申请CN201280073013、发明名称为“协同作用的抗微生物剂”公开通过将有效量的N-α-长链烷酰基二元氨基酸烷基酯盐与甘油单脂肪酸酯组合提供协同作用的抗微生物组合物,产生更有效的抗微生物剂和食品防腐剂。同时,中国专利申请CN200810131638、发明名称为“杀微生物剂组合物”公开甲基异噻唑啉酮和LAE的组合物用于制备抗微生物剂和食品防腐剂的用途。然而,该方法涉及包括LAE在内的多种抑菌成分,并未研究LAE的单独抑菌作用。
中国专利申请CN201280027864、发明名称为“具有改进的耐水性的化妆防晒制剂或皮肤科防晒制剂”公开一种LAE用于制备化妆防晒制剂或皮肤科防晒制剂的用途,该制剂除了UV滤光剂之外还包括乳化剂聚甘油-10硬脂酸酯。
综上所述,虽然现有技术中已经开展对LAE的研究,但还未见将LAE用于制备天然环保无毒的厨房油污清洗剂的相关报道。
发明内容
针对现有技术存在的不足,本发明所要解决的技术问题是,提供一种天然环保型的油污清洗剂。
本发明原理之一在于,根据月桂酰精氨酸乙酯LAE具有抗菌能力强、生物毒性低、体内代谢效果好、与环境相容性高且常温下与其他化合物不发生反应的特点,首次提出将LAE成分作为油污清洗剂中的抑菌剂和防腐剂,将其与传统油污清洗剂的其他成分进行复配,从而获得一种天然环保型的油污清洗剂。其中,所述LAE成分作为主要或唯一的抑菌和防腐成分。
本发明原理之二在于,进一步对LAE进行改进,获得一种新型衍生物,即将LAE与有机酸盐发生缩合反应,从而获得LAE离子对化合物。该离子对化合物作为油污清洗剂中的抑菌剂和防腐剂成分,相对于LAE具有抑菌效果更好、且使用剂量更低的优点,从而更利于制备天然环保型的油污清洗剂。其中,所述LAE成分作为主要或唯一的抑菌和防腐成分。
因此,本发明第一个发明目的是提供月桂酰精氨酸乙酯LAE用于制备环保无毒的油污清洗剂的用途。
在一个实施方案中,所述LAE包括式(I)所示的月桂酰精氨酸乙酯化合物(LAE化合物)及其衍生物或其水合物或药学上可接受的盐。

在一个实施方案中,所述油污清洗剂包括基质和所述LAE离子对;所述LAE离子对在油污清洗剂中的质量百分比浓度为0.01-2%;优选地,为0.01-1%或0.1-1%、1-2%或1.5-2%;进一步优选地,为0.01-0.1%;进一步优选地,为0.05%。
所述油污清洗剂的基质按质量百分比包括10-30%的蔗糖脂肪酸酯、5-15%的丙二醇、5-10%乙酸、0-2%的乳酸、5-15%的葡萄糖酸钠、0-3%的焦磷酸钾,其他余量为水。
本发明第二个发明目的是提供作为抑菌剂或防腐剂的LAE离子对化合物用于制备环保无毒的油污清洗剂的用途,其中所述LAE离子对化合物具有如下式(III)所示结构式:
在一个实施方案中,其中,所述RCOO-有机酸或盐选自具有抗菌活性的水杨酸、甲酸、甲酸铵、甲酸钙、乙酸、双乙酸钠、丙酸、丙酸铵、丙酸钠、丙酸钙、丁酸、丁酸钠、乳酸、苯甲酸、苯甲酸钠、山梨酸、山梨酸钠、山梨酸钾、富马酸、柠檬酸、柠檬酸钾、柠檬酸钠、柠檬酸钙、酒石酸、苹果酸、磷酸、碳酸钠、草酸或碳酸。
在一个优选实施方案中,所述有机酸选自烟酸、酒石酸、草酸。
在一个实施方案中,所述油污清洗剂包括基质和所述LAE离子对化合物;所述LAE离子对化合物在油污清洗剂中的质量百分比浓度为0.01-2%;优选地,为0.01-1%或0.1-1%、1-2%或1.5-2%;进一步优选地,为0.01-0.1%;进一步优选地,为0.05%;

本发明第三个发明目的是提供制备含有上述LAE离子对化合物的环保无毒的油污清洗剂的方法,步骤包括:
(1)加热溶解式(II)所示的化合物,而后加入有机酸盐溶液;
(2)充分搅拌混匀,并在加热的条件下,反应得到LAE离子对化合物,所述反应如下反应式所示:
其中,RCOO-的有机酸或盐选自具有抗菌活性的水杨酸、甲酸、甲酸铵、甲酸钙、乙酸、双乙酸钠、丙酸、丙酸铵、丙酸钠、丙酸钙、丁酸、丁酸钠、乳酸、苯甲酸、苯甲酸钠、山梨酸、山梨酸钠、山梨酸钾、富马酸、柠檬酸、柠檬酸钾、柠檬酸钠、柠檬酸钙、酒石酸、苹果酸、磷酸、碳酸钠、草酸或碳酸;
(3)充分反应后,冷却室温,纯化后真空干燥,从而制备纯化的式(III)所示的月桂酰精氨酸乙酯有机酸离子对化合物;
(4)在容器中,在常温、常压下加入油污清洗剂基质,并加入上述LAE离子对化合物,通过泵循环充分搅拌后,得到所述环保无毒的油污清洗剂。
步骤(1)中,所述加热溶解的温度为50℃-100℃;优选地,为90℃。
步骤(2)中,所述反应的温度为50℃-100℃;优选地,为90℃。
步骤(2)中,所述反应的时间为1-3小时;优选地,为2小时。
步骤(3)中,所述真空干燥的条件为50℃-100℃;优选地,为60℃。

(3)充分反应后,冷却室温,纯化后真空干燥,从而制备式(III)所示的纯化的月桂酰精氨酸乙酯有机酸离子对化合物;
(4)在不锈钢制或惰性材质的容器中,在常温、常压下加入表面活性剂、碱性助剂、有机溶剂,并加入上述比例的LAE离子对化合物,通过泵循环充分搅拌后,分装得到所述环保无毒的油污清洗剂;
其中,所述油污清洗剂包括基质和所述LAE离子对化合物;其中,所述LAE离子对化合物在油污清洗剂中的质量百分比浓度为0.01-2%;优选地,为0.01-1%或0.1-1%、1-2%或1.5-2%;进一步优选地,为0.01-0.1%;进一步优选地,为0.05%;
所述油污清洗剂的基质按质量百分比包括10-30%的蔗糖脂肪酸酯、5-15%的丙二醇、5-10%乙酸、0-2%的乳酸、5-15%的葡萄糖酸钠、0-3%的焦磷酸钾,其他余量为水。

在另一实施方案中,所述RCOO-的有机酸盐的制备方法如下:将所述有机酸加入甲醇溶液中,并加入适量的NaOH,室温搅拌直至析出白色固体,抽滤并用甲醇分洗涤,得到所述RCOO-的有机酸盐。
本发明第四个目的是提供含有上述作为抑菌剂或防腐剂的LAE或其离子对化合物的厨房油污清洗剂。
本发明中,所述油污清洗剂的基质包括:3g蔗糖脂肪酸酯(单酯)、1.5g丙二醇、0.5g乙酸、0.5g葡萄糖酸钠、1g甘油、1mg LAE(终浓度100ppm)和5ml水;或,2g蔗糖脂肪酸酯(单酯),1.2g丙二醇,0.7g乙酸,1g葡萄糖酸钠,10mgLAE(终浓度100ppm)和5.1ml水;2.5g蔗糖脂肪酸酯(单酯),0.5g丙二醇,0.5g乙酸,1g葡萄糖酸钠,0.3g焦磷酸钾,0.1g乳酸,1mgLAE(终浓度100ppm)和5.1ml水;3g蔗糖脂肪酸酯(单酯)、1.5g丙二醇、0.5g乙酸、0.5g葡萄糖酸钠、1g甘油、1mg LAE乙酸离子对(终浓度100ppm)和5ml水;2g蔗糖脂肪酸酯(单酯),1.2g丙二醇,0.7g乙酸,1g葡萄糖酸钠,10mgLAE离子对(终浓度100ppm)和5.1ml水;2.5g蔗糖脂肪酸酯(单酯),0.5g丙二醇,0.5g乙酸,1g葡萄糖酸钠,0.3g焦磷酸钾,0.1g乳酸,1mgLAE离子对(终浓度100ppm)和5.1ml水。
术语和定义:
月桂酰精氨酸乙酯(Ethyl lauroylarginate,LAE)是一种由脂肪酸和二元氨基酸缩合而成的有机物,为白色吸湿性固体,在pH3~7范围内化学性质稳定,熔点50~58℃,该温度下247g的LAE可分散于1kg的水中,它在水和油中的分配系数大于10,即主要存在于水相中。研究发现,月桂酰精氨酸乙酯LAE具有抗菌能力强、生物毒性低、体内代谢效果好、与环境相容性高的特点。而其中最具代表性的特点是月桂酰精氨酸乙酯代谢无残留,相关研究显示月桂酰精氨酸乙酯在人体与动物体内可快速进行自然代谢,生成月桂酸和精氨酸,进一步被代谢为鸟氨酸、尿素、二氧化碳和水。月桂酰精氨酸乙酯代谢过程中所产生的所有初级代谢产物及终产物都是无毒无害的,与人和动物日常摄取的食物在体内的代谢产物相同。
本发明对LAE的衍生物进行改进,突破对于衍生物开发的传统思路,即不再局限于选择传统上适于LAE的酸、碱、盐/酯的合适形态,或对LEA进行酸、碱、盐或酯化基团的处理,而是创造性的选择一种能够增强LAE的抑菌协同效应、同时又能显著补充营养能量物质的酸根基团,并将二者非常规地通过分子间强烈的离子键结合成新的衍生物,即离子对化合物,从而显著地提高了LAE及其衍生物在作为油污清洗剂和抑菌剂中的用途。
本发明厨房油污清洁用品的优点是:
创造性使用LAE或LAE离子对化合物代替传统油污清洗剂中的抑菌剂和防腐剂,在保留传统清洗剂的成本低廉、制作工艺简单、去污能力强的优点的同时,还具有抑菌效果显著,且对人体无伤害,易于分解代谢,易于长期保存等优点。

结合以下具体实施例和附图,对本发明作进一步的详细说明,本发明的保护内容不局限于以下实施例。在不背离发明构思的精神和范围下,本领域技术人员能够想到的变化和优点都被包括在本发明中,并且以所附的权利要求书为保护范围。实施本发明的过程、条件、试剂、实验方法等,除以下专门提及的内容之外,均为本领域的普遍知识和公知常识,本发明没有特别限制内容。
实施例一:月桂酰精氨酸乙酯盐酸盐与烟酸合成离子对化合物的制备方法
将烟酸钠(购于梯希爱(上海)化成工业发展有限公司)2.0g溶于50mL水中,配制成烟酸钠盐水溶液(A);将月桂酰精氨酸乙酯盐酸盐6.8g溶于40mL水中,加热至90℃,直至月桂酰精氨酸乙酯盐酸盐全部溶解,制成月桂酰精氨酸乙酯盐酸盐水溶液(B);在90℃条件下将烟酸钠盐水溶液(A)缓慢加入到月桂酰精氨酸乙酯盐酸盐水溶液(B)中,不断搅拌,反应2小时,冷却至室温,过滤,用纯净水充分洗涤沉淀,沉淀60℃真空干燥,即得烟酸离子对化合物7.6g。
实施例二:月桂酰精氨酸乙酯烟酸离子对化合物分子式、分子量的分析
通过质谱、1H-NMR、13C-NMR波谱分析所得到的化合物分子式为:
1.质谱(ESI)分析
阳离子B+分子离子峰的m/z=385.3,参见图1;
质谱检测ESI+为124.2,参见图2。则ESI-为122.2,即阴离子A-分子离子峰的m/z=122.2。烟酸离子对化合物中阳离子的理论计算值为507.4,实测值与理论值吻合。
2.NMR分析
将月桂酰精氨酸乙酯盐酸盐(参见图3)、烟酸的1H-NMR(参见图4)和LAE烟酸离子对化合物的1H-NMR(参见图5)相比较。由于LAE离子对化合物在成盐过程中,在该离子对化合物中月桂酰精氨酸乙酯的峰形及化学位移变化不大,但烟酸上的所有氢都有位移变化,其波谱特征与原无机酸盐(即LAE盐酸盐)相比,酸碱两部分空间距离更加接近,产生影响,因此其与原LAE及其盐酸盐相比,产生相应变化,并不是简单的酸碱两部分的迭加,例如在纯净水洗涤沉淀时,溶解性已发生改变,这说明月桂酰精氨酸乙酯的所有氢核与烟酸之间产生了强相互作用,并通过强烈的离子键形成了稳定的单一化合物结构。
实施例三:月桂酰精氨酸乙酯盐酸盐与酒石酸合成离子对化合物的制备方法
将酒石酸(购于梯希爱(上海)化成工业发展有限公司)2.0g溶于50mL甲醇中,加入当量的NaOH,室温搅拌直至析出白色固体,抽滤并用30mL甲醇分三次洗涤,得到酒石酸钠盐。酒石酸钠盐溶于50mL水中,配制成酒石酸钠盐水溶液(A);将月桂酰精氨酸乙酯盐酸盐5.6g溶于40mL水中,加热至90℃,直至月桂酰精氨酸乙酯盐酸盐全部溶解,制成月桂酰精氨酸乙酯盐酸盐水溶液(B);在90℃条件下将酒石酸钠盐水溶液(A)缓慢加入到月桂酰精氨酸乙酯盐酸盐水溶液(B)中,不断搅拌,反应2小时,冷却至室温,过滤,用纯净水充分洗涤沉淀,沉淀60℃真空干燥,即得酒石酸离子对化合物6.3g。
实施例四:月桂酰精氨酸乙酯酒石酸离子对化合物分子量的分析
质谱(ESI)分析阳离子B+分子离子峰的m/z=385.3(参见图1)
阴离子A-分子离子峰的m/z=149.0(参见图6)
烟酸离子对化合物中阳离子的理论计算值为534.3,实测值与理论值吻合。
实施例五:月桂酰精氨酸乙酯盐酸盐与草酸合成离子对化合物的制备方法
将草酸(购于探索有限公司)1.0g溶于50mL甲醇中,加入当量的NaOH,室温搅拌直至析出白色固体,抽滤并用30mL甲醇分三次洗涤,得到草酸钠盐。草酸钠盐溶于50mL水中,配制成草酸钠盐水溶液(A);将月桂酰精氨酸乙酯盐酸盐4.7g溶于40mL水中,加热至90℃,直至月桂酰精氨酸乙酯盐酸盐全部溶解,制成月桂酰精氨酸乙酯盐酸盐水溶液(B);在90℃条件下将草酸钠盐水溶液(A)缓慢加入到月桂酰精氨酸乙酯盐酸盐水溶液(B)中,不断搅拌,反应2小时,冷却至室温,过滤,用纯净水充分洗涤沉淀,沉淀60℃真空干燥,即得草酸离子对化合物5.0g。
按照实施例二的方法,进行NMR分析和ESI分析,结果表明该离子对化合物的波谱特征不是简单的酸碱两部分的迭加,酸碱两部分空间距离接近,产生影响,其波谱数据与原LAE及其盐酸盐相比,产生相应变化,例如在纯净水洗涤沉淀时,溶解性已发生改变,这说明月桂酰精氨酸乙酯的所有氢核与草酸之间产生了强相互作用,并通过强烈的离子键形成了稳定的单一化合物结构。
实施例六:月桂酰精氨酸乙酯盐酸盐与碳酸合成离子对化合物的制备方法
将碳酸钠(购于探索有限公司)1.0g溶于50mL水中,配制成碳酸钠水溶液(A);将月桂酰精氨酸乙酯盐酸盐4.0g溶于40mL水中,加热至90℃,直至月桂酰精氨酸乙酯盐酸盐全部溶解,制成月桂酰精氨酸乙酯盐酸盐水溶液(B);在90℃条件下将碳酸钠水溶液(A)缓慢加入到月桂酰精氨酸乙酯盐酸盐水溶液(B)中,不断搅拌,反应2小时,冷却至室温,过滤,用纯净水充分洗涤沉淀,沉淀60℃真空干燥,即得碳酸离子对化合物4.0g。

实施例七:月桂酰精氨酸乙酯离子对化合物体外最小抑菌浓度(MIC)的测定
原理与目的:根据CLSI规定的微量肉汤稀释法,药物与细菌在96孔板内共孵育24h后,细菌生长被抑制的最小药物浓度为该药的最小抑菌浓度。
方法:将月桂酰精氨酸乙酯盐酸盐(LAE盐酸盐)及上述所制备的月桂酰精氨酸乙酯有机酸离子对分别用胰酪胨大豆肉汤培养基(TSB)二倍稀释成不同浓度,药物与细菌在96孔板里混合孵育,另设无细菌的空白对照培养基CK1和添加LAE(1000μg/ml)的培养基CK2以及不含药物的细菌正常生长对照培养基CK3。将96孔板放入37℃温箱中孵育24h后测定各孔625nm处的吸光光度值。与空白对照OD625值一致的孔视为细菌无明显生长。细菌无明显生长的药物最低浓度为LAE对细菌的最小抑菌浓度MIC(Minimal InhibitoryConcentration)。
所制备的多种LAE衍生物(离子对化合物)相对于原LAE化合物的抗菌活性的比较结果如下表1所示。

结果分析:
(1)离子对化合物大部分对大肠杆菌保持相同的抗菌活性,尤其是草酸离子对化合物的抗菌活性上升;
(2)离子对化合物大部分对金黄色葡萄球菌保持相同的抗菌活性,碳酸离子对化合物的抗菌活性下降,烟酸离子对化合物的抗菌活性显著上升;
结论:LAE衍生物的离子对化合物,不会对单一成分的原LAE的抗菌活性产生抑制作用,相反有益于抗菌活性。其中,烟酸离子对化合物对金黄色葡萄球菌产生了显著的抑菌效果。
实施例八:油污清洁剂的配方
称取3g蔗糖脂肪酸酯(单酯)、1.5g丙二醇、0.5g乙酸、0.5g葡萄糖酸钠、1g甘油、1mg LAE(终浓度100ppm)和5ml水。
将水用水浴加热到60℃,边搅拌边依次加入乙酸、丙二醇、甘油混匀,倒入蔗糖脂肪酸酯(单酯),葡萄糖酸钠和LAE搅拌至澄清冷却至室温获得淡黄色透明澄清蜂蜜状液体混合物,pH为6.85。冷却至4℃以下为乳白色膏状物,常温放置4年性状未改变。所有配料均为食品级。
实施例九:油污清洁剂的配方
称取2g蔗糖脂肪酸酯(单酯),1.2g丙二醇,0.7g乙酸,1g葡萄糖酸钠,10mgLAE(终浓度100ppm)和5.1ml水。
将水用水浴加热到60℃,边搅拌边依次加入乙酸、丙二醇混匀,倒入蔗糖脂肪酸酯(单酯)、葡萄糖酸钠和LAE搅拌至澄清,冷却至室温获得淡黄色透明澄清蜂蜜状液体混合物,pH为6.72。冷却至4℃以下为乳白色膏状物,常温放置4年性状未改变。所有配料均为食品级。
实施例十:油污清洁剂的配方
称取2.5g蔗糖脂肪酸酯(单酯),0.5g丙二醇,0.5g乙酸,1g葡萄糖酸钠,0.3g焦磷酸钾,0.1g乳酸,1mg LAE(终浓度100ppm)和5.1ml水。
将水用水浴加热到60℃,边搅拌边依次加入乙酸、丙二醇和乳酸混匀,倒入蔗糖脂肪酸酯(单酯)、LAE、葡萄糖酸钠和焦磷酸钾搅拌至澄清,冷却至室温获得淡黄色透明澄清蜂蜜状液体混合物,pH为6.65。冷却至15℃以下为乳白色膏状物,常温放置4年性状未改变。所有配料均为食品级。
实施例十一:油污清洁剂的配方
称取3g蔗糖脂肪酸酯(单酯)、1.5g丙二醇、0.5g乙酸、0.5g葡萄糖酸钠、1g甘油、1mg LAE乙酸离子对(终浓度100ppm)和5ml水。
将水用水浴加热到60℃,边搅拌边依次加入乙酸、丙二醇、甘油混匀,倒入蔗糖脂肪酸酯(单酯),葡萄糖酸钠和LAE乙酸离子对搅拌至澄清冷却至室温获得淡黄色透明澄清蜂蜜状液体混合物,pH为6.85。冷却至4℃以下为乳白色膏状物。所有配料均为食品级。
实施例十二:油污清洁剂的配方
称取2g蔗糖脂肪酸酯(单酯),1.2g丙二醇,0.7g乙酸,1g葡萄糖酸钠,10mgLAE乙酸离子对(终浓度100ppm)和5.1ml水。
将水用水浴加热到60℃,边搅拌边依次加入乙酸、丙二醇混匀,倒入LAE乙酸离子对、蔗糖脂肪酸酯(单酯)和葡萄糖酸钠搅拌至澄清,冷却至室温获得淡黄色透明澄清蜂蜜状液体混合物,pH为6.72。冷却至4℃以下为乳白色膏状物。所有配料均为食品级。
实施例十三:油污清洁剂的配方
称取2.5g蔗糖脂肪酸酯(单酯),0.5g丙二醇,0.5g乙酸,1g葡萄糖酸钠,0.3g焦磷酸钾,0.1g乳酸,1mg LAE乙酸离子对(终浓度100ppm)和5.1ml水。
将水用水浴加热到60℃,边搅拌边依次加入乙酸、丙二醇和乳酸混匀,倒入LAE乙酸离子对、蔗糖脂肪酸酯(单酯)、葡萄糖酸钠和焦磷酸钾搅拌至澄清,冷却至室温获得淡黄色透明澄清蜂蜜状液体混合物,pH为6.65。冷却至15℃以下为乳白色膏状物。所有配料均为食品级。
实施例十四:油污清洁剂的去污能力的比较
检测用实施例八至十三的清洗剂的去污力。并用德国进口Pril浓缩高效餐具洗洁精作为对照品。
1、油污垢的配制
配方:大豆油64.0g,无氨焦糖色素8.0g,小麦粉12.0g,猪油8.0g,牛油8.0g,单硬脂酸甘油酯2.4g。
在250ml烧杯中分别称取上述大豆油、猪油、牛油、单硬脂酸甘油酯,50度加热溶解,充分搅拌混匀后冷却至室温,在加入无氨焦糖色素,以1500r/min搅拌30min,搅拌均匀后加入小麦粉,在搅拌20min,室温成化24h,冷藏备用。
2、制备污片
选取8片预先称重(称准至0.0002g)的洁净试片,以约0.25g/片的量涂布油污垢,然后放置干燥的瓷板上,与200度高温干燥10min,然后至于45度的紫外恒温老化箱中,紫外照射1.5h。调整照射位置,再次照射1.5h,然后取出放置干燥器中陈化24h。而后对固定油污的试片进行称重(称准至0.0002g)。
3、清洗剂的清洗
将待测试的实施例八至十三的清洗剂,以及对照Pril浓缩高效餐具洗洁精的水溶液在50℃下超声30min,然后将上述7片试片分别悬挂在7个预先超声混匀的清洗剂烧杯中,完全浸泡10min,而后清洗5min。同时设无清洗剂的水溶液作为空白对照,并浸入第8片试片。
然后将清洗的试片置于120℃烘箱中干燥45min进行固定剩余的油污垢,并在干燥器中冷却30min后称量(称准至0.0002g),计算去污力。
4、结果计算
去污力计算公式为:f=(m1-m2)×100%/(m1-m0)

m2:污片清洗后的质量
结果如下表2所示。
实施例十五:滞留法比较油污清洁剂的抑菌能力的比较
1、检测原理
本试验通过模拟适合细菌生长、繁殖和可能产生感染的厨房油污台面下,使用随机性、双盲的、配对比较的方法检测油污台面清洗剂在一定时间内作用于清洁台面的滞留抑菌效果
2、试验阶段
(1)预处理
分别用实施例十四(表2)中的各种配方的清洗剂,清洗预设至少分为7个区域的油污台面,一共清洗3次,每次间隔至少1h。另将无菌水清洗的台面作为对照。
(2)接种细菌
将金黄色葡萄球菌连续转种3代,适当稀释菌悬液,使菌悬液浓度约为1×104-9×104cfu/ML。然后使用加样器取10μL上述菌悬液,接种于对应清洁后的台面区域,并用一次性无菌接种环,把菌悬液涂成一圆形,然后用无菌塑料杯封闭验区,使其与试验区边缘应有4mm-5mm的距离;
(3)回收存活细菌
室温接种后12h后,对接种的密封区域进行取样。将含0.1%AEO-9的磷酸盐缓冲液(0.03mol/L)1mL小心涂布在试验区中间部位,不要接触到封闭区的边缘。同时,用无菌微量移液器将涂布液吸取到无菌试管中。
(4)平皿接种与培养
对每一个取样进行平皿接种,以磷酸盐缓冲液(PBS)(0.03mol/L)对样品进行10倍系列稀释。选适当稀释度取0.1mL接种于含羊血5%的TSA平皿表面,用玻璃弯棒涂匀,在(35士2)℃的培养箱中培养(48士4)h,计数菌落数。
(5)抑菌率的计算
抑菌率(%)=(I-II)×100/I
其中,I:对照样品平均菌落数,即无菌水
II:试验样品平均菌落数
结果保留整数
结果如下表2所示。

结果分析:
去污力检测结果显示,实施例十二的油污清洁剂去污效果最好,LAE乙酸离子对相对于LAE单体有更强的去污能力。
抑菌效果检测结果显示,实施例十二的油污清洁剂去污效果最好,LAE乙酸离子对相对于LAE单体有更强的抑菌能力。
表2说明,LAE乙酸离子对相对于LAE单体不但有更强的去污能力,并且在规定的时间内具有更好的滞留抑菌效果。
另外,虽然两组试验中,0.1%剂量组的抑菌效果和去污力均是最高,并且从申请人在先提交的专利申请来看(发明名称:“月桂酰精氨酸乙酯衍生物和作为动物用抗菌剂的用途”、申请号:201810648982.3、申请日2019年6月22日)的试验数据来看,LAE及其衍生物一定范围内的剂量加大可提高抑菌效果,但相对于0.01%剂量组的抑菌效果并没有显著的提高,说明0.01%-0.1%的剂量范围已经满足生产需要。
此外,根据现行国家相关标准,在规定使用浓度规定作用时间,样品对大肠杆菌和金黄色葡萄球菌的抑制率分别达到50%,就可以认为清洗剂具备抑菌功能。如果提高LAE及其衍生物的添加量,虽然抑菌率相应提高,但过高抑菌率意味着较多的残留,并不有利于人体健康。即使如此,由于本发明的LAE及其衍生物的抑菌剂成分属于天然环保无毒的成分,因此高剂量的添加使用仍然比传统的化学品抑菌剂具有对人体友好亲和的优点。
因此,考虑到生产成本和实际生产需要,因此LAE及其离子对作为厨房油污清洗剂的活性成分的质量百分比浓度为0.01-1%或0.1-1%或1-2%或1.5-2%,优选有效浓度为0.01-0.1%、最优选0.05%时能有效防治发病,符合生产的需要。
Claims (7)
2.如权利要求1所述的用途,其特征在于,所述油污清洗剂包括基质和所述LAE离子对化合物;所述LAE离子对化合物在油污清洗剂中的质量百分比浓度为0.01-2%;所述油污清洗剂的基质按质量百分比包括10-30%的蔗糖脂肪酸酯、5-15%的丙二醇、5-10%乙酸、0-2%的乳酸、5-15%的葡萄糖酸钠、0-3%的焦磷酸钾,其他余量为水。
3.如权利要求1或2所述的用途,其特征在于,所述LAE离子对化合物在油污清洗剂中的质量百分比浓度为0.01-1%、0.1-1%或0.05%,其中,所述RCOO-的有机酸盐选自烟酸钠或草酸钠。
4.一种油污清洗剂的制备方法,其特征在于,包括以下步骤:
(1)加热溶解式(II)所示的化合物,而后加入RCOO-有机酸盐溶液;
(2)充分搅拌混匀,并在加热至90℃的条件下发生缩合反应得到LAE离子对化合物,所述缩合反应如下反应式所示:
其中,RCOO-有机酸盐选自具有抗菌活性的乙酸盐、烟酸盐或草酸盐;
(3)充分反应后,冷却室温,纯净水洗涤纯化后真空干燥,从而制备纯化的LAE离子对化合物;
(4)在容器中,在常温、常压下加入油污清洗剂的基质,并加入上述LAE离子对化合物,通过泵循环充分搅拌后,得到所述环保无毒的油污清洗剂;
其中,所述油污清洗剂包括基质和所述LAE离子对化合物;所述LAE离子对化合物在油污清洗剂中的质量百分比浓度为0.01-2%;所述油污清洗剂的基质按质量百分比包括10-30%的蔗糖脂肪酸酯、5-15%的丙二醇、5-10%乙酸、0-2%的乳酸、5-15%的葡萄糖酸钠、0-3%的焦磷酸钾,0-10%的甘油,其他余量为水。
5.如权利要求4所述的方法,其特征在于,所述RCOO-的有机酸盐选自烟酸钠或草酸钠。
6.如权利要求5所述的方法,其特征在于,所述RCOO-的有机酸盐的制备方法如下:将所述有机酸加入甲醇溶液中,并加入适量的NaOH,室温搅拌直至析出白色固体,抽滤并用甲醇分洗涤,得到所述RCOO-的有机酸盐。
7.如权利要求4-6之任一项所述方法制备得到的厨房油污清洗剂。
