技术领域
本发明涉及医药领域,特别是涉及一种辅助降血糖的保健食品及其制备方法。
背景技术
糖尿病是世界范围最常见的流行病之一,已经成为继心血管疾病、肿瘤之后的第三大严重威胁人类健康的疾病。患者一般表现为多饮、多食、多尿、体重减轻,还伴有脂肪、蛋白质代谢紊乱,易并发血管病变、酮症酸中毒。根据《中国居民营养与慢性病状况报告(2015年)》,2012年中国18岁及以上居民糖尿病患病率为9.7%,患者人数约1亿。近年来,随着人们生活水平的提高、人口老龄化和工作及生活方式的改变,高血糖的发病率呈上升趋势,且趋于年轻化。
目前,糖尿病的常规治疗药物主要为胰岛素促分泌剂、α-葡萄糖苷酶抑制剂、胰岛素增敏剂等化学药物及胰岛素。口服降糖药虽然可以有效控制患者血糖水平,但长期服用后会产生一定的副作用,有可能进一步加重器官的损害,甚至出现耐药性,且给药方式也有诸多不便。同时这些药物对糖尿病先期以及慢性并发症无理想的控制效果,容易引起低血糖或乳酸中毒,并且无法阻止胰岛β细胞的进一步坏死,导致胰岛素进一步依赖等副作用。保健食品功能明确,对人体没有任何急性、亚急性和慢性危害,因此急需研制服用方便、易于储存和携带的降血糖保健食品。
在长期的医疗实践中,我国传统医学积累了诸多宝贵经验,现代药理学研究表明中草药在防治糖尿病及各种慢性并发症方面有显著作用。专利申请“200310115012.0一种治疗糖尿病的药物及其制备方法”中公开了采用西洋参、黄芪、山药、生地黄、山茱萸、枸杞子、麦门冬、知母、天花粉、五味子、五倍子、葛根制成中药组合物,其具有明显的降血糖作用。专利申请“201210524373.X一种降血糖的中药组合物”中公开了由黑芝麻、龙眼肉、桑葚、生地黄、麦冬、天花粉、黄芪、枸杞子、葛根、红参、茯苓、牡丹皮、五味子、酸枣仁、柏子仁、合欢皮、石斛、枸杞子、女贞子、百合、川梀子组成的中药材,其可以改善糖尿病患者的症状。然而现有技术中中药组合物还存在诸多缺陷,如制备工艺复杂、原料昂贵、将血糖效果不明确或有待提高等问题。
发明内容
鉴于上述问题,本发明的目的在于提供一种辅助降血糖的保健食品及其制备方法。本发明的辅助降血糖的保健食品降血糖功能明确、安全无毒副作用、食用方便。
本发明为实现上述目的所采用的技术方案是:
一种辅助降血糖的保健食品,按照重量百分含量包括以下组分:
黄芪或黄芪提取物1-50%、葛根或葛根提取物1-50%、地黄或地黄提取物1-50%、知母或知母提取物0-25%、麦冬或麦冬提取物1-50%、枸杞子或枸杞子提取物0-50%、五味子或五味子提取1-50%、山药或山药提取物0-25%、桑叶或桑叶提取物0-25%、富铬酵母0.01-5%。
优选的,主要原料按照重量百分比含量包括:黄芪或黄芪提取物5-25%、葛根或葛根提取物2.5-12.5%、地黄或地黄提取物5-25%、知母或知母提取物1-10%、麦冬或麦冬提取物2.5-12.5%、枸杞子或枸杞子提取物2.5-12.50%、五味子或五味子提取1-10%、山药或山药提取物1-10%、桑叶或桑叶提取物1-10%、山药或山药提取物1-10%、富铬酵母0.01-4%。
选取了上述具有辅助降血糖功能的原料,根据传统中医理论和现代医学研究结果,其组方具有协同降血糖作用,无任何配伍禁忌,不仅能够降低血糖,还可以增强机体各项机能,保护胰岛组织,提高胰岛素的敏感性,延缓甚至阻止糖尿病并发症的发生。
黄芪为豆科植物蒙古黄芪或膜荚黄芪的干燥根,味甘,性微温。黄芪的药用历史悠久,是古代治疗消渴的重要药物之一,其具有补气升阳,利水消肿,生津养血的功效。现代药理学研究表明,黄芪水提物、黄芪多糖以及黄芪甲苷具有显著的降血糖作用、还可以保护胰岛β细胞、预防糖尿病并发症以及改善血液流变学指标。
葛根为豆科植物野葛的干燥根,辛甘而凉,具有鼓舞脾胃清阳之气上行、生津止渴之效。葛根富含黄酮类物质,其中的葛根素具有良好的降低空腹血糖、增加糖耐量、提高血清胰岛素水平、改善糖脂代谢等作用,功能明确。
地黄为玄参科植物地黄的干燥块根,味甘,性寒。生地黄苦寒清泄,味甘质润,具有清热凉血,养阴生津的功效。现代药理学研究表明,地黄中的寡糖能够调节糖尿病模型大鼠、小鼠的糖脂代谢,对肥胖糖尿病大鼠和糖尿病肾病大鼠均具有治疗作用。
知母为百合科植物知母的干燥根茎,味苦、甘,性寒。知母善清上中下三焦之热,擅长滋阴生津润燥,是治疗糖尿病的常用中药。研究发现知母具有多种药理活性,例如抗病原微生物、解热抗炎作用,抗血小板聚集作用以及降血糖作用。
麦冬为百合科植物麦冬的干燥块根,味甘微苦,性微寒。麦冬既养阴生津而润肺益胃,又清养心神而除烦安神,还可滋润肠燥而通便。药理研究表明麦冬具有增强免疫力、抗衰老、抗心脑血管疾病、抗炎、镇咳等药理作用,同时麦冬多糖具有显著的降血糖、改善胰岛素抵抗和肾脏保护作用。
枸杞子为茄科植物宁夏枸杞的干燥成熟果实,味甘,性平偏温。枸杞具有滋补肝肾、益精明目的功效。枸杞子应用广泛,是一味药食两用的中药,安全性良好。现代医学研究表明,枸杞多糖不仅可以降血糖、修复胰岛细胞、恢复胰岛功能,还可以保护视网膜,治疗视网膜病变。
五味子为木兰科植物五味子的干燥成熟果实,味酸,性温。具有收敛固涩,益气生津,补肾宁心的功效。因五味子具有敛阴固阴的作用,在该保健食品配方中可以增强其他药材的养阴生津作用。同时现代医学研究表明,五味子醇提物、水提物以及五味子煎剂均具有降血糖、降血脂、保护肾脏、修复胰岛β细胞的作用,其降糖效果显著。
山药为薯蓣科植物薯蓣的干燥根茎,味甘,性平。山药药力平和,兼涩敛之性,既平补气阴,为治气虚或气阴两虚之佳品,又滋阴益气而生津,为治肾阴虚及消渴所常用。同时山药也是营养价值很高的药食同源食品,具有增强免疫力、调整肠胃功能、降血糖、降血脂、抗衰老等功效。
桑叶为桑科植物桑的干燥叶,味甘、苦,性寒。桑叶中含有1-脱氧野尻霉素,是α-葡萄糖苷酶和葡萄糖淀粉酶的抑制剂,具有明显的抑制餐后血糖上升、减肥和增加胰岛素敏感性的作用。
富铬酵母是通过人工开发的方式,将酵母作为载体,培养出富集铬元素的可食用功能性酵母。铬是人体必需的微量元素之一,现代研究证实三价铬是葡萄糖耐量因子(GTF)的活性组成部分,主要通过GTF协同和增强胰岛素的作用,从而影响糖类、脂肪、蛋白质的代谢。富铬酵母具有食用安全,无毒性,人体吸收率高,补充铬的同时可补充B族维生素等诸多优点。
还包括填充剂和矫味剂,所述的填充剂选自预胶化淀粉、微晶纤维素、麦芽糊精、D-甘露糖醇、木糖醇、麦芽糖醇、异麦芽酮糖中的一种或几种,所述的矫味剂选自食用香精、柠檬酸中的一种或两种;还包括在制粒过程中使用的粘合剂和润湿剂,所述的粘合剂选自聚维酮、羟丙甲纤维素、羧甲基纤维素钠、淀粉浆中的一种或几种,所述的润湿剂选自纯化水、乙醇中的一种或两种。添加填充剂和矫味剂是为了增加物料的流动性以及调整颗粒剂的口味,添加粘合剂和润湿剂是为了颗粒剂或胶囊剂制粒的需求。其中填充剂和矫味剂、粘合剂和润湿剂可以根据配方需要和物料的性质,选择添加或不添加。
优选的,填充剂和矫味剂按照重量百分比含量包括:预胶化淀粉5-50%、微晶纤维素5-30%、麦芽糊精5-50%、D-甘露糖醇5-50%、木糖醇5-50%、麦芽糖醇1-20%、异麦芽酮糖1-20%、食用香精0.1-5%、柠檬酸0.1-5%;在制粒过程中使用的粘合剂和润湿剂选自纯化水1-40%、乙醇10-70%、聚维酮1-40%、羟丙甲纤维素1-40%、羧甲基纤维素钠1-40%、淀粉浆1-40%。
优选的,保健食品为颗粒剂或胶囊剂。
上述辅助降血糖保健食品的制备方法,包括以下步骤:
颗粒剂:
(1)粉碎过筛,将原料、需要添加的填充剂和矫味剂根据其粒度情况确定是否进行粉碎,如果原料、填充剂和矫味剂可以通过80-100目筛,则不需要粉碎,如果不能通过80-100目筛,则需要粉碎过筛,保证各原料、填充剂和矫味剂的粒度一致,保证后续混合均匀;
(2)称量,将原料、需要添加的填充剂和矫味剂进行精确称量,备用;
(3)混合搅拌,将称量好的各个物料按照配方进行混合搅拌;
(4)制粒,根据混合好的物料性质,选择适合的粘合剂和润湿剂,进行制粒,达到需要的粒度20-60目;
(5)将制好的颗粒进行包装,入库。
胶囊剂:
(1)粉碎过筛,将原料、需要添加的填充剂和矫味剂根据其粒度情况确定是否进行粉碎,如果原料、填充剂和矫味剂可以通过80-100目筛,则不需要粉碎,如果不能通过80-100目筛,则需要粉碎过筛,保证各原料、填充剂和矫味剂的粒度一致,保证后续混合均匀;
(2)称量,将原料、需要添加的填充剂和矫味剂进行精确称量,备用;
(3)混合搅拌,将称量好的各个物料按照配方进行混合搅拌;
根据配方组合后的物料流动性决定是否进行制粒,若流动性好,直接进入灌胶囊的步骤(5);若流动性不好,则需要先进行制粒的步骤(4),再进行灌胶囊的步骤(5);
(4)制粒,根据混合好的物料性质,选择适合的粘合剂和润湿剂,进行制粒,达到需要的粒度20-80目;
(5)灌胶囊,将混合好的物料或是制好的颗粒进行灌胶囊;
(6)将灌好的胶囊进行包装,入库。
本发明具有如下技术效果:
(1)本发明涉及的辅助降血糖保健食品配伍设计是根据传统中医理论及现代科研成果相结合来组方。配方中:黄芪、葛根补气升阳、生津止渴,地黄、知母清热滋阴,麦冬、枸杞子、山药生津止渴、五脏同补,五味子滋肾宁心,敛阴固阴,桑叶提取物中的1-脱氧野尻霉素是强效α-葡萄糖苷酶抑制剂,可显著降低糖尿病患者空腹血糖和餐后血糖,富铬酵母中富含三价铬,是葡萄糖耐受因子的重要组成部分,可以增加胰岛素活性,增强2型糖尿病患者胰岛素敏感性。该组方各原料之间无配伍禁忌,能够从多种途径合理调节血糖。
(2)本发明目标人群明确,对糖尿病患者具有标本兼治的作用,利用传统中医理论与现代医学相结合,共同降低血糖偏高人群的血糖指标,同时减轻糖尿病人群的并发症及服用西药降糖药物所带来的副作用。
(3)本发明颗粒剂和胶囊剂制备工艺简单,且产品食用方便,易于携带,适用范围广泛。
具体实施方式
以下具体实施例是对本发明提供的方法与技术方案的进一步说明,但不应理解成对本发明的限制。
实施例1:
选取6种主要原料黄芪提取物、葛根提取物、地黄提取物、麦冬提取物、五味子提取物、富铬酵母,添加麦芽糊精、D-甘露糖醇、麦芽糖醇为填充剂和矫味剂。黄芪、地黄、麦冬、五味子均为可用于保健食品的物品,葛根为药食同源的物质,各原料之间没有配伍禁忌,组方不存在安全性问题。
具体的配方组成为(按重量百分数):
黄芪提取物:10-20
葛根提取物:5-10
地黄提取物:10-20
麦冬提取物:5-10
五味子提取物:5-10
富铬酵母:0.1-2
麦芽糊精:20-40
D-甘露糖醇:10-30
麦芽糖醇:10-20
70%乙醇:混合物料重量的20%-50%。
生产加工方法如下:
(1)粉碎过筛:将上述原辅料粉碎过筛,100%通过80目筛,备用;
(2)称重:按配方比例称取上述原辅料;
(3)混合搅拌:将(2)中称量好的物料装入混合机内搅拌30min;
(4)制粒:根据(3)中混合好的物料性质,选择质量分数为70%的乙醇进行制粒,颗粒的粒度达到20-60目;
(5)包装:将(4)中制好的颗粒进行分装,入库。
实施例1所得到的颗粒剂进行了辅助降血糖功能动物试验:
一、试验方法:
(一)胰岛素损伤高血糖模型方法。
1实验动物与剂量:实验用ICR小鼠,清洁级,雄性,体重26±2g。实验设置低、中、高三个剂量组,分别为0.25、0.5、1.5g/kg体重,灌胃容量按0.1ml/10g体重计。
2、正常动物降糖实验:取20只小鼠,禁食4小时后,取小鼠尾血测血糖,按血糖水平分为1个对照组和1个高剂量组,每组10只,分别灌胃给予去离子水和高剂量样品,连续30天,各组动物禁食4小时后,测空腹血糖值,比较两组小鼠空腹血糖值。
3、高血糖模型降糖实验:
(1)胰岛损伤高血糖模型:120只小鼠随机取15只禁食4小时,测血糖,作为该批次动物基础血糖值。随后严格禁食24小时(自由饮水),小鼠尾静脉注射四氧嘧啶45mg/kg.BW,7天后禁食4小时,测血糖。筛检空腹血糖值为10-25mmol/L的小鼠52只,按血糖水平随机分为1个模型对照组和低、中、高剂量组。
(2)空腹血糖和糖耐量的测定:分别灌胃给予去离子水和不同剂量的样品,连续30天,各组动物禁食4小时后,测空腹血糖值,即0小时血糖值。各剂量组给予不同浓度样品,模型组对照组给予同体积去离子水,20分钟后各组灌胃给予葡萄糖2.0g/kg.BW,测定给葡萄糖后0.5、2小时的血糖值,比较各组小鼠实验前后空腹血糖值及血糖下降百分率;0.5、2.0小时血糖值及血糖曲线下面积。
血糖下降率(%)=(实验前血糖值-试验后血糖值)×100/实验前血糖值
血糖曲线下面积=(0h血糖+0.5h血糖)×0.5/2+(0.5h血糖+2h血糖)×1.5/2
(二)实验数据统计:实验数据均采用SPSS11.5统计软件统计。阴性对照组和高剂量组之间各指标比较采用t检验。除血糖下降率外,模型对照组和三个剂量组各指标采用均数±标准差表示,各组间比较经方差齐性检验方差齐后采用方差分析。血糖下降率采用四分位数(P25~P75)表示,血糖下降率各组之间的比较采用秩和检验。
二、实验结果
1、样品对正常及高血糖模型小鼠体重的影响

由表1可见,样品高剂量组(阴性)小鼠在试验初期、中期及终期分别和阴性对照组比较,差异均无显著性(P>0.05);样品低、中、高剂量组小鼠在试验各期分别和模型对照组比较,差异均无显著性(P>0.05)。
2、样品对正常及高血糖模型小鼠空腹血糖的影响

由表2可知,样品高剂量组(阴性)试验前后小鼠空腹血糖值和阴性对照组比较,差异无显著性(P>0.05);样品低、中、高剂量组小鼠试验前后空腹血糖值及血糖下降百分率分别和模型对照组比较,差异均无显著性(P>0.05)。
3、样品对高血糖模型小鼠糖耐量的影响

由表3可见,与模型对照组比较,样品中剂量组糖耐量2小时血糖值明显降低,差异有显著性(P<0.05)。
实验结果表明:以实施例1的辅助降血糖保健食品颗粒剂作为样品进行辅助降血糖动物试验,在本实验条件下,实验结果为阳性。
实施例2:
选取取7种主要原料黄芪提取物、葛根提取物、地黄提取物、麦冬提取物、枸杞子提取物、五味子提取物、富铬酵母,添加微晶纤维素、硬脂酸镁为填充剂。黄芪、地黄、麦冬、五味子均为可用于保健食品的物品,葛根、枸杞子为药食同源的中药,富铬酵母属于营养素物质,各原料之间没有配伍禁忌,组方不存在安全性问题。
具体的配方组成为(按重量百分数):
黄芪提取物:10-20
葛根提取物:5-10
地黄提取物:10-20
麦冬提取物:5-10
枸杞子提取物:5-10
五味子提取物:5-10
富铬酵母:0.1-2
微晶纤维素:20-40
硬脂酸镁0.5-1.5。
生产加工方法如下:
(1)粉碎过筛:将上述原辅料粉碎过筛,100%通过80目筛,备用;
(2)称重:按配方比例称取上述原辅料;
(3)混合搅拌:将(2)中称量好的物料装入混合机内搅拌30min;
(4)灌胶囊:将(3)中混合好的物料进行灌胶囊;
(5)包装:将(4)中灌好的胶囊进行分装,入库。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,可以对这些特征和实施例进行各种改变或等效替换。另外,在本发明的教导下,可以对这些特征和实施例进行修改以适应具体的情况及材料而补脱离本发明的精神和范围。因此,本发明不受此处所公开的具体实施例的限制,所有落入本申请的权利要求范围内的实施例都属于本发明的保护范围。
Claims (8)
1.一种辅助降血糖的保健食品,其特征在于,按照重量百分含量包括以下组分:黄芪或黄芪提取物1-50%、葛根或葛根提取物1-50%、地黄或地黄提取物1-50%、知母或知母提取物0-25%、麦冬或麦冬提取物1-50%、枸杞子或枸杞子提取物0-50%、五味子或五味子提取1-50%、山药或山药提取物0-25%、桑叶或桑叶提取物0-25%、富铬酵母0.01-5%。
2.如权利要求1所述的辅助降血糖的保健食品,其特征在于,原料按照重量百分比含量为:黄芪或黄芪提取物5-25%、葛根或葛根提取物2.5-12.5%、地黄或地黄提取物5-25%、知母或知母提取物1-10%、麦冬或麦冬提取物2.5-12.5%、枸杞子或枸杞子提取物2.5-12.50%、五味子或五味子提取1-10%、山药或山药提取物1-10%、桑叶或桑叶提取物1-10%、山药或山药提取物1-10%、富铬酵母0.01-4%。
3.如权利要求1所述的辅助降血糖的保健食品,其特征在于,还包括填充剂和矫味剂,所述的填充剂选自预胶化淀粉、微晶纤维素、麦芽糊精、D-甘露糖醇、木糖醇、麦芽糖醇、异麦芽酮糖中的一种或几种,所述的矫味剂选自食用香精、柠檬酸中的一种或两种。
4.如权利要求1所述的辅助降血糖的保健食品,其特征在于,还包括在制粒过程中使用的粘合剂和润湿剂,所述的粘合剂选自聚维酮、羟丙甲纤维素、羧甲基纤维素钠、淀粉浆中的一种或几种,所述的润湿剂选自纯化水、乙醇中的一种或两种。
5.如权利要求4所述的辅助降血糖的保健食品,其特征在于,所述的填充剂和矫味剂在原料中按照重量百分比含量为:预胶化淀粉5-50%、微晶纤维素5-30%、麦芽糊精5-50%、D-甘露糖醇5-50%、木糖醇5-50%、麦芽糖醇1-20%、异麦芽酮糖1-20%、食用香精0.1-5%、柠檬酸0.1-5%。
6.如权利要求5所述的辅助降血糖的保健食品,其特征在于,所述的粘合剂和润湿剂在原料中按照重量百分比含量为:纯化水1-40%、乙醇10-70%、聚维酮1-40%、羟丙甲纤维素1-40%、羧甲基纤维素钠1-40%、淀粉浆1-40%。
7.如权利要求6所述的辅助降血糖的保健食品,其特征在于,所述的辅助降血糖的保健食品为颗粒剂或胶囊剂。
8.如权利要求3-7中任一项所述的辅助降血糖的保健食品,其特征在于,包括如下步骤:
(1)可选地,粉碎过筛,将原料、填充剂和矫味剂过80-100目筛;
(2)称量,将原料、填充剂和矫味剂进行称量,备用;
(3)混合搅拌,将称量好的物料按照配方进行混合搅拌;
(4)可选地,添加粘合剂和润湿剂,进行制粒,达到需要的粒度20-60目;
(5)包装,入库或者进一步灌胶囊,将灌好的胶囊进行包装、入库。
