一、技术领域
本发明涉及一种富含肌醇-3-磷酸的豆乳及其生产技术,属于食品加工技术领域。
二、背景技术
大豆籽粒中富含蛋白质、不饱和脂肪酸、大豆皂苷、异黄酮等多种对人体有益的物质,同时还存在着植酸等抗营养因子,由此降低了大豆的营养价值。大豆籽粒发芽后,内源植酸酶被激活,植酸水解成肌醇-5-磷酸(IP5)、肌醇-4-磷酸(IP4)和肌醇-3-磷酸等低级磷酸肌醇。IP3等低级磷酸肌醇既有抗氧化与防癌症等生理功能,同时又有增补调控细胞代谢、保护心脏和降低血压的作用。大豆发芽后,蛋白质、糖类等贮藏性物质经内源酶水解与转化,形成的简单物质,更易被人体消化吸收。
专利(公开号103636801A,公开日2014年03月18日)公开了“一种枇杷豆乳及其制作方法”、专利(公开号103109936A,公开日2013年5月22日)公开了“香蕉豆乳及其加工工艺”、专利(公开号专利102599252A,公开日2012年7月25日)公开了“一种脱腥红枣豆乳粉的生产方法”,这些方式通过添加果汁或果酱的方法改善豆乳口味,但产品pH偏酸,影响豆乳中蛋白质的稳定性。专利(公开号105532894A,公开日2016年05月4日)公开了“一种低植酸豆乳粉的制备方法”,此方式通过浸泡、挤压膨化处理原料来降低产品中的植酸,与本发明采用喷淋发芽降低产品中植酸含量方式不同,并未涉及低对级磷酸肌醇的富集。论文《大豆芽菜植酸降解技术与低植酸豆乳研发》(仲磊.南京农业大学,2014),通过喷淋发芽大豆籽粒制成低植酸豆乳,但未突出对于功能性保健成分的富集,且培养液组分与本发明存在差异。该方法采用炼乳改善豆乳口感,但炼乳中蛋白质、矿物质和维生素A的含量皆少于奶粉,且炼乳过于甜腻。
本发明采用NaCl-乳酸钙溶液对豆乳原料进行喷淋发芽处理,激活多位点去磷酸化途径中的植酸酶,以此降低植酸含量,形成IP3等低级磷酸肌醇;同时激活磷脂酰肌醇特异性磷脂酶C途径(PI-PLC途径),在PI-PLC催化下形成IP3,以此富集IP3等低级磷酸肌醇,强化豆乳饮料的保健作用。与奶粉复配后,提高了产品中蛋白质含量、提升了产品的口感,因而本发明中富含IP3的豆乳是兼具营养与保健功能的新型饮料。
三、发明内容
技术问题:
本发明旨在提供一种富含肌醇-3-磷酸的豆乳及其生产技术,解决一般豆乳口感粗糙、豆腥味、苦涩味重等问题,同时以NaCl-乳酸钙溶液喷淋处理发芽大豆,使原料中富集IP3等低级磷酸肌醇,以此原料生产的豆乳兼具营养与保健功能。
技术方案:
本发明技术如下:用NaCl和乳酸钙混合水溶液喷淋发芽的豆类籽粒,其中的植酸在内源植酸酶作用下水解,生成对人体具有保健功能的IP3等低级磷酸肌醇;以此为原料,经制浆后,添加白砂糖、奶粉、黄原胶、海藻酸钠、羧甲基纤维素钠、聚甘油酸脂肪酸酯和单甘脂,再经均质、脱气、灭菌和无菌灌装,制得,具体包括以下步骤:
(1)喷淋发芽
A、原料挑选:选取颗粒饱满、发芽率高、未破损的大豆籽粒为原料,也可以用绿豆、红豆为原料;
B、消毒浸泡:按公知的方法对大豆籽粒进行消毒处理后,在清水中浸泡3~4h;
C、喷淋发芽:将浸泡过的大豆籽粒置于发芽机中,在公知的20~35℃温度下避光发芽1d。期间,每隔1~3h使用浓度为4~8mmol/L乳酸钙和1.6~3.2mmol/L NaCl混合水溶液喷淋2~5min,喷淋液流量为100~200mL/min。
(2)熟浆制备:发芽大豆洗净、去皮,按1∶5~1∶7比例(w/v)添加纯净水,按公知的方法用砂轮磨磨细,通过80目滤布提取豆浆,过滤后的豆浆通过管道送入储浆罐,于95℃煮浆,使浆液温度达到75℃,然后用250目筛网过滤,制得熟浆;
(3)混合调配:熟浆冷却后,按以下比例(v/w)添加白砂糖1%~5%、奶粉5%~10%、黄原胶0.02%~0.06%、海藻酸钠0.06%~0.14%、羧甲基纤维素钠0.07%~0.13%、聚甘油酸脂肪酸酯0.03%~0.07%和单甘脂0.05%~0.08%;
(4)均质、脱气:按公知的方法,将混合调配后的浆液在30~40MPa下均质2次,得到体态均匀的豆乳,在65~75℃、0.06~0.08MPa真空度下脱气;
(5)灭菌、灌装:按公知的方法用135℃对豆乳进行高温瞬时灭菌5~10s,板式热交换器冷却至室温,无菌灌装,制得的豆乳产品中肌醇-3-磷酸含量为20~150μg/100mL。
有益效果:
与现有工艺相比,本发明具有以下优点:
(1)本发明中豆类籽粒经NaCl-乳酸钙喷淋发芽富集低级磷酸肌醇,提高了豆乳的营养价值与保健功能,产品中低级磷酸肌醇的含量达20~150μg/100mL。
(2)采用本发明制得的产品营养价值高,口感细滑香醇,营养全面,稳定性好。
四、具体实施方案
实施例1
挑选颗粒饱满,无病虫害的大豆,用0.1~0.3%次氯酸钠水溶液对其消毒、用饮用水浸泡3~4h后,将籽粒置入发芽机中,在30℃下避光发芽1d,期间使用4~8mmol/L乳酸钙水溶液喷淋发芽大豆;发芽大豆洗净、去皮,按1∶5比例(w/v)添加纯净水,用砂轮磨磨细,通过80目滤布提取豆浆,过滤后的豆浆通过管道送入储浆罐,于95℃煮浆,使浆液温度达到75℃,然后用250目筛网过滤,制得大豆熟浆;在大豆熟浆中添加5%白砂糖、10%奶粉、0.02%黄原胶、0.14%海藻酸钠、0.07%羧酸基纤维素钠、0.07%聚甘油酸脂肪酸酯和0.05%单甘脂,然后在30~40MPa下均质2次,并在65~75℃、0.06~0.08MPa真空度下脱气;对均质、脱气后的物料在135℃下超高温瞬时灭菌5~10s,经板式热交换器冷却到室温后,无菌灌装。制得的产品中肌醇-3-磷酸含量为150μg/100mL。
实施例2
绿豆籽粒挑选、消毒、浸泡和喷淋发芽方法同实施例1,喷淋液使用1.6~3.2mmol/L NaCl水溶液;发芽绿豆洗净、去皮,按1∶6比例(w/v)添加纯净水,磨浆、滤浆、煮浆、过滤方法同实施例1,制得绿豆熟浆;在绿豆熟浆中添加3%白砂糖、8%奶粉、0.05%黄原胶、0.12%海藻酸钠、0.10%羧酸基纤维素钠、0.05%聚甘油酸脂肪酸酯和0.07%单甘脂,原浆配料的均质、脱气、灭菌、冷却、无菌灌装方法同实施例1。制得的产品中肌醇-3-磷酸含量为80μg/100mL。
实施例3
红豆籽粒挑选、消毒、浸泡和喷淋发芽方法同实施例1,喷淋液使用4~8mmol/L乳酸钙和1.6~3.2mmol/L NaCl混合水溶液;发芽红豆洗净、去皮,按1∶6比例(w/v)添加纯净水,磨浆、滤浆、煮浆、过滤方法同实施例1,制得红豆熟浆;在绿豆熟浆中添加5%白砂糖、10%奶粉、0.04%黄原胶、0.10%海藻酸钠、0.09%羧酸基纤维素钠、0.06%聚甘油酸脂肪酸酯和0.06%单甘脂,原浆配料的均质、脱气、灭菌、冷却、无菌灌装方法同实施例1。制得的产品中肌醇-3-磷酸含量为20μg/100mL。

Claims (2)
1.一种富含肌醇-3-磷酸的豆乳生产技术,其特征在于,用NaCl和乳酸钙混合水溶液喷淋发芽的豆类籽粒,其中的植酸在内源植酸酶作用下水解,生成对人体具有保健功能的IP3等低级磷酸肌醇;以此为原料,经制浆后,添加白砂糖、奶粉、黄原胶、海藻酸钠、羧甲基纤维素钠、聚甘油酸脂肪酸酯和单甘脂,再经均质、脱气、灭菌和无菌灌装,制得,具体包括以下步骤:
(1)喷淋发芽
A、原料挑选:选取颗粒饱满、发芽率高、未破损的大豆籽粒为原料;
B、消毒浸泡:按公知的方法对大豆籽粒进行消毒处理后,在清水中浸泡3~4h;
C、喷淋发芽:将浸泡过的大豆籽粒置于发芽机中,在公知的20~35℃温度下避光发芽1d;期间,每隔1~3h使用浓度为4~8mmol/L乳酸钙和1.6~3.2mmol/LNaCl混合水溶液喷淋2~5min,喷淋液流量为100~200mL/min;
(2)熟浆制备:发芽大豆洗净、去皮,按1∶5~1∶7比例(w/v)添加纯净水,按公知的方法用砂轮磨磨细,通过80目滤布提取豆浆,过滤后的豆浆通过管道送入储浆罐,于95℃煮浆,使浆液温度达到75℃,然后用250目筛网过滤,制得熟浆;
(3)混合调配:熟浆冷却后,按以下比例(v/w)添加白砂糖1%~5%、奶粉5%~10%、黄原胶0.02%~0.06%、海藻酸钠0.06%~0.14%、羧甲基纤维素钠0.07%~0.13%、聚甘油酸脂肪酸酯0.03%~0.07%和单甘脂0.05%~0.08%;
(4)均质、脱气:按公知的方法,将混合调配后的浆液在30~40MPa下均质2次,得到体态均匀的豆乳,在65~75℃、0.06~0.08MPa真空度下脱气;
(5)灭菌、灌装:按公知的方法用135℃对豆乳进行高温瞬时灭菌5~10s,板式热交换器冷却至室温,无菌灌装。
2.如权利要求1所述一种富含肌醇-3-磷酸的豆乳生产技术,其特征在于,豆类籽粒用大豆、绿豆、红豆为原料,制得的产品中肌醇-3-磷酸含量为20~150μg/100mL。
