与食品中掺入的功能性物质和/或药物有关的共同困难是活性随时间损失、食品制作工艺期间分解和/或产品通过生物体消化道期间的破坏。某些食品处理的严峻环境,像磨碎、混合、烘焙和挤压,能够在许多生物活性物质成为完成的食品之前将它们破坏。这对于对大多数类型的传统食品加工敏感的酶和维生素尤其如此。因此,食品工业一直在寻找保护生物活性化合物对抗加工、储存和胃通过期间分解的新的组合物和方法。
其它问题由需要的生物活性化合物与其它食品组分例如金属螯合剂、表面活性剂、吸湿性成分等之间的相互作用引起(Choe和Min,2006)。保护和提高生物活性物质的保留和恰当释放的一种方法是包囊。包囊也用于保护生物活性物质免受氧气、水和光,以及将所述物质转变成容易合并在各种食品中的自由流动粉末。这些年来已经做了各种各样的努力,以将生物活性剂包上或埋入许多不同类型的生物聚合物或合成聚合物,包括蛋白、碳水化合物和固体脂肪中(Nissim G.,2008)。
大多数包囊方法利用水溶性载体物质,例如蛋白、糖、改性淀粉和胶(PCT/US2004/004003,WO2004/082660)。典型的包囊方法包括喷雾干燥、空气悬浮包衣、喷雾冷却和激冷、共结晶和离心挤压。但是,因为包囊的生物活性物质在含水条件下氧化并随后降解,这些包囊类型不适合保护含有水或具有高水分活性的食品中的生物活性剂。因为水在食品制作过程和储存的某些阶段参与大多数食物的制备,包囊在水溶性聚合物中对于改善生物活性化合物的稳定性、或对于控制生物活性物质的保留和引导它们以程序化方式释放,适用性受到限制。
为了克服在潮湿环境中加工或储存期间活性损失的问题,有时使用固体脂肪的保护层对水溶性粒子进行脂肪包囊或顶涂。提出的固体脂肪包衣方法的实例包括:美国专利No.4,350,679,其公开了在软凝胶上施加巴西棕榈蜡包衣。如美国专利No.5,789,014所述,蜡质包衣的功能在于改善外壳强度和耐湿性,其中熔点在40℃与50℃之间的粉末或丸粒形式的蜡被加热到超过它的熔点并用于在流化床包衣设备中包衣。
美国专利申请公布No.2006/0051425公开了将活性成分在多层包衣中微囊化的方法。所述多层包衣由各种蜡和胶构成,在整个加工、配制和储存期间保护活性成分,并使得活性成分能够受控释放。美国专利申请公布No.2007/0042184公开了喷雾冷却包含活性成分的水性珠子,所述活性成分包囊在固体脂肪的疏水性外壳基质中或被其包囊。但是,这些类型微囊化的主要问题在于当在传统的食品制造过程期间添加水时,包衣容易破裂。使用脂肪包衣的另一个问题是在低于所述脂肪熔点的温度下加工时它的局限性。例如,该方法不适用于包括温度远远超过70℃的煮沸、烘焙、喷雾干燥或挤压的食品过程,因为包衣脂肪变得液化并丧失了它的保护性能。

发明内容
本文公开的发明允许活性成分高度荷载到显示出高度耐受热和剪切力微粒中,并且所述粒子在高水分活性环境中具有期望的高稳定度。这样的微粒还在肠的吸收性或其它适当的区域中显示出优良的释放动力学,这是特别需要的。
本发明的一个方面提供了包含生物活性物质、凝聚剂以及乳化剂和可食用固体脂肪的混合物的微粒。
本发明的另一方面提供包含生物活性物质、凝聚剂以及乳化剂和可食用固体脂肪的混合物的微粒,其中凝聚的生物活性物质在乳化剂和固体脂肪的双层包衣中包被。
本发明的又一个方面涉及包含生物活性剂和凝聚剂的混合物的微粒,以形成含有芯的生物活性剂,其中所述含有芯的生物活性剂包在第一和第二层中,并且其中第一层位于邻近生物活性剂芯并包含至少一种乳化剂,而第二层位于包含至少一种固体脂肪化合物的乳化剂层上。
本发明的再一个方面涉及制造自由流动的固体微粒的方法,包括:
将生物活性物质与凝聚剂混合;
形成生物活性物质/凝聚物微粒芯;
将所述芯与至少一种乳化剂接触,从而形成包裹所述芯的第一层;和
将所述第一层与在所述分层的芯的表面上固化的至少一种固体脂肪接触。
生物活性芯上沉积的层将生物活性凝聚物芯的尺寸增加了至少两倍,并更优选生物活性凝聚物芯的尺寸的约2至10倍。优选,所述微粒基本上是球形的,但也可应用其它几何形状,包括但不限于棒形、三角形、椭圆形和多面形。

本发明提供在食品加工和储存期间保护生物活性物质的组合物和产生所述组合物的方法。具体是,本发明提供包含乳化剂和固体脂肪双层包围由凝聚剂和生物活性物质构成的芯的微囊体。出乎意料地发现,保护在芯粒子中并用由富乳化剂内层和固体脂肪外层构成的双层包被的生物活性物质在亲水相和疏水相二者中非常稳定并保持完整。
本发明的一个方面是包含生物活性物质和凝聚剂的组合物,其中凝聚剂在组合物中的浓度是生物活性物质的约1至约50重量%。在优选的实施方案中,组合物中凝聚剂的浓度是生物活性物质的约2至约20重量%。多种生物活性物质适合用于本发明。一般而言,所述生物活性物质包含至少一种生物活性物质。多种凝聚剂适合用于本发明。一般而言,所述凝聚剂包含至少一种碳水化合物或蛋白质。所述碳水化合物可以是简单碳水化合物或由糖的长链构成的复合碳水化合物。所述蛋白质可以是任何适当的动物或植物来源的蛋白质(天然的、分离的、改性的或水解的)。
为了制备所述组合物,碳水化合物或蛋白质溶液与生物活性物质接触,以形成尺寸范围50-5000微米的凝聚的干燥微粒。然后通过本技术领域公知的技术例如热风干燥、喷雾干燥、冷冻干燥或真空蒸发来干燥凝聚的微粒。然后用由富乳化剂内层和富固体脂肪外层构成的双层包被所述凝聚的粒子。本发明的典型微粒呈现在图1中。
多种乳化剂适合用于本发明。在一种实施方案中,乳化剂可以是磷脂例如卵磷脂的混合物。卵磷脂的商业来源包括大豆、水稻、向日葵籽、蛋黄和乳脂。卵磷脂可以是去油和处理过的,致使它是基本上纯的磷脂混合物。卵磷脂也可以是改性的以使得所述磷脂更具水溶性。改性包括羟基化、乙酰化和酶处理,其中一个脂肪酸被磷脂酶除去并用羟基替换。
多种固体脂肪适合用于本发明。一般而言,固体脂肪可以是单纯的动物或植物固体油和脂肪和天然蜡。在优选的实施方案中,所述固体脂肪的熔点至少40℃并优选超过60℃。固体脂肪的最终熔点可以通过混合两种或多种不同熔点的脂肪来操纵。液体油可以通过加氢转化成固体脂肪,并可以单独或在与其它液体或固体脂质的混合物中使用,条件是它们在保存微粒或含有所述微粒的食品的温度下形成自由流动的粉末。
组合物也可以包含抗氧化剂以进一步稳定生物活性物质并防止其氧化。多种抗氧化剂适合用于本发明。抗氧化剂的浓度在凝聚的粒子中范围可以为约0.001%至约1重量%,在内层中范围为约0.01%至约10重量%。
在其他的实施方案中,双层包衣中也可以包含粘合剂和润滑剂。实例包括硬脂酸、硬脂酸镁、硬脂酸钙或其它金属性硬脂酸酯、滑石、蜡和甘油酯、轻质矿物油、PEG、甘油酰山嵛酸酯、胶态二氧化硅、玉米淀粉、聚乙二醇、烷基硫酸酯、苯甲酸钠、乙酸钠等等。
所述双层中乳化剂与固体脂肪的比率将根据所述乳化剂和所述固体脂肪的性质而变动。具体是,内层中乳化剂的浓度将是在保存微粒或含有所述微粒的食品的温度下保持所述内层坚实度所需的充分量。乳化剂的浓度通常在内层的约5至约50重量%的范围。在一种实施方案中,乳化剂的浓度可以在内层的约5至约30重量%的范围。在另一种实施方案中,乳化剂的浓度可以在内层的约5至约20重量%的范围。在优选的实施方案中,乳化剂的浓度可以在内层的约20至约40重量%的范围。
在具体的实施方案中,外层的熔化温度显著高于内层的熔化温度。具体地说,外层主要由熔化温度高的固体脂肪构成。在优选的实施方案中,外层由熔点约55℃至约85℃的氢化油、天然或烃蜡构成。
形成本发明的微粒通常首先凝聚生物活性物质,以将细小的微粒物质转变到允许顶涂的所需平均粒度。因为某些微粒物质展现出固有的胶粘性或粘合性特征,足以提供凝聚所需的粒子粘附力,在长期的实践中将物质与含有凝聚剂的溶液接触,所采取的方式要在搅拌粒子团时促进粘附。喷雾或雾化时通常引入凝聚剂溶液,然后干燥凝聚的产物以除去为了凝聚而添加的水分。
本发明中优选的凝聚剂是碳水化合物或蛋白质。碳水化合物的非限制性实例包括糖、淀粉、胶或其组合。蛋白质的非限制性实例包括明胶、乳蛋白、玉米蛋白和植物蛋白质或其组合。在一些实施方案中,混合物中可以添加至少一种抗氧化剂。在包含简单碳水化合物的实施方案中,可以形成包含约5至50%糖的溶液。在一种实施方案中,溶液中简单碳水化合物的浓度可以在约20至约30重量%的范围。在包含复合碳水化合物的实施方案中,可以形成包含约0.5至10%复合碳水化合物的溶液。在一种实施方案中,溶液中复合碳水化合物的浓度可以在约1至约5重量%的范围。在包含蛋白质的实施方案中,可以形成包含约1至5%蛋白质的溶液。在优选的实施方案中,凝聚性溶液是简单碳水化合物和复合碳水化合物的混合物,并可以进一步包括至少一种另外的抗氧化剂。
为了制造凝聚的微粒,凝聚剂溶液通常在喷雾或雾化时引入并允许与搅拌过的生物活性物质接触,以形成尺寸范围在50-5000微米的凝聚的微粒。然后通过本技术领域公知的技术例如风干、喷雾干燥、冷冻干燥或真空蒸发来干燥凝聚的微粒。生成的凝聚的微粒基本上是无水的。所述生物活性物质现在在碳水化合物和抗氧化剂复合物中部分稳定,但在与水接触后仍然可溶。
为了给本发明的组合物提供基本上耐水的环境并防热,使用由富乳剂的内层和富固体脂肪的外层构成的双层包衣。在一种实施方案中,内层中乳化剂的浓度将是在保存微粒或含有所述微粒的食品的温度下提供固体层的充分量。在一种实施方案中,所述内层是基本上无水的组合物并包含固体脂肪、乳化剂和抗氧化剂的混合物。在优选的实施方案中,内层包含氢化油或天然蜡、卵磷脂或单酸甘油酯、和生育酚或草本提取物。油混合物被加热到稍超过其熔点,并在恒定搅拌之下喷洒到凝聚的微粒上。涂敷内层包衣直到微粒总质量增加了它们初始重量的10%至25%。在一种优选实施方案中,涂敷内层包衣直到微粒总质量增加了它们初始重量的15%至20%。
在又一种实施方案中,提供基本上无水的外层。所述外层包衣包含高熔点的可食用固体脂肪。所述外层的熔化温度显著高于内层的熔化温度。在本发明的优选实施方案中,外层由熔点约60℃至约85℃的氢化油、天然或烃蜡构成。在特别优选的实施方案中,可食用固体脂肪与增塑剂(例如,硬脂酸、硬脂酸镁、硬脂酸钙或其它金属性硬脂酸盐、滑石、蜡和甘油酯、轻质矿物油、PEG、甘油酰山嵛酸酯、胶态二氧化硅、玉米淀粉和淀粉衍生物、聚乙二醇、烷基硫酸酯、苯甲酸钠、乙酸钠等)组合。固体脂肪混合物被加热到稍超过其熔点并在恒定搅拌之下均匀喷洒在包被了内层的凝聚微粒上。在内层的外面相继涂敷外层包衣,直到微粒的总质量增加了它们初始重量的20%至60%。在优选的实施方案中,涂敷外层包衣直到微粒的总质量增加了它们初始重量的30%至50%。
不受任何具体理论的束缚,所述双层包衣赋予微粒水不溶性和耐破裂性。图2证明了富乳化剂的内层包衣对粒子稳定性的作用。当所述微粒用于包括高水分活性的食品加工时,所述双层包衣充当水分的坚实屏障,从而保护和稳定了生物活性物质芯。它也维护和保护生物活性物质免受伴随面团制备和烘焙的热和剪切暴露。在这样的条件下,所述层将在生物活性物质芯(即碳水化合物复合物)上熔化和崩溃,因此仍然提供对热和水分的坚实防护。
可以使用几种方法产生本发明的微粒。凝聚方法可以包括喷雾干燥、锅包衣和旋转盘包囊(亦称旋转悬浮分离包囊)、超临界流体包囊、空气悬浮凝聚、流化床凝聚、喷雾冷却/激冷(包括基质粒化)、挤压、离心挤压、水凝胶喷雾捕集和本技术领域已知的其它凝聚方法。
本发明的一个方面中,可以相继使用相同的包囊法来产生本发明的微粒。这样的包囊方法可以利用旋转悬浮或空气悬浮工艺,其中生物活性物质首先在向上流动的空气流中旋转或悬浮的同时被凝聚,用加热的空气流干燥,然后涂敷双层包衣,开始施加富乳化剂的内层并相继施加最终高熔点的固体脂肪外层。旋转和空气悬浮包囊方法是本技术领域公知的(Gustavo等,2006;Shafiur R.M.2007)。
本发明的一个方面中,所述微粒可以用于任何食品,例如但不限于乳品或液体饮料食品(例如酸奶,奶酪,冰淇淋,搅打奶油,酸奶油,乳汁,豆浆,牛奶米糊、水果和植物饮料、营养饮料、能量饮料和液体婴儿配方食品)、干焙烤品(例如早餐谷类,炸土豆片,面包,蛋糕,馅饼,曲奇饼,饼干,格兰诺拉燕麦棒,营养棒,巧克力产品,营养增补剂和药物制剂)、和肉包括处理过的肉和人造肉产品。所述食品也可以是添加了生物活性物质微囊体的罐头食品。
在又一个实施方案中,本发明的微粒可以用于动物饲料产品。动物可以是伴侣动物、农业动物、或水生生物。所述饲料可以是成粒的、挤出的、喷雾包被的、顶涂的、或由任何其它方法形成。
因为在上述组合物、产品和方法中可以不背离本发明的范围产生各种变化,旨在以上描述和下面给出的实施例中包含的所有事件应该理解为说明性而非限制性意义。
实施例
以下实施例举例说明本发明的各种实施方案。
实施例1.流化床制备维生素A微粒
凝聚溶液通过将10g蔗糖和1g阿拉伯树胶溶解在100g温水中而制备(40-60℃)。向其添加50mg柠檬酸钠,并将溶液冷却到室温。在改良的流化床干燥器/成粒器/包衣器系统(Fluid Air-2型,最大工作容量2升),装备有空气速度可变和热控制可变的鼓风机,通过用双流体喷头在20psi的气压和约4ml/min的液体速率下将凝聚溶液呈细雾进行顶端喷雾约5分钟,来凝聚1000g细粉维生素A棕榈酸酯(BASF,FlorhamPark,NJ,粒度范围0.5微米至10微米)。由此产生的凝聚粒子在流化床干燥器中使用50℃至60℃范围的干燥温度风干至残余水份水平小于3%。然后涂敷双层包衣,开始是顶喷雾富乳化剂的内层,直到粒子的质量增加了它们原来质量的20%。所述内层由40%(w/w)大豆卵磷脂(Archer Daniels Midland Company,Decatur,IL)、55%(w/w)氢化大豆油(17 Stearine,Loders Croklan,Channahon,IL)和5%(w/w)迷迭香提取物(OxyLess,Naturex,Mamaroneck,NY)构成。然后相继涂敷富固体脂肪的外层,直到粒子质量增加了它们原来质量的40%。所述外层由100%氢化棕榈油(27硬脂,Loders Croklan,Channahon,IL)构成。氢化棕榈油的温度在整个喷雾过程期间保持在70℃,粒子温度保持在大约40℃。最终的步骤包括冷却微粒、收集和将所述微粒筛分至50与450微米之间的尺寸范围。图1显示了本发明的典型微粒和它们在油溶液中的保存稳定性。
也可以通过分批法生产微粒,其中收获干燥的凝聚微粒并筛分至需要的大小,然后返回流化床干燥器进行包衣。
实施例2.维生素A微粒的锅包衣制备方法
使用包衣锅例如通常用于制药工业的包衣锅来包被片剂,用实施例1描述的凝聚溶液来凝聚维生素A棕榈酸酯(BASF,Florham Park,NJ)。将锅设置为以60至75RPM旋转。双流体喷头与调节到15至25psi之间的热风供应相连接。卵磷脂(400g)和OxyLess(50g)在75℃下溶解在550g的17-硬脂中。熔融的卵磷脂/抗氧化剂/固体脂肪溶液以约6ml/min的速率被提供到喷嘴的液体侧。暖空气(40℃)被吹到锅里面,帮助促进均匀包衣。喷洒卵磷脂/抗氧化剂/固体脂肪溶液,直到沉积了15%的内层包衣。然后将包衣溶液改变为100%熔融的17-硬脂,并涂敷另外30%的外层包衣。然后使微粒冷却至室温并筛分,收集50与450μm之间的微粒部分。
也可以通过分批法生产微粒,收获干燥的凝聚粒子并筛分至需要的大小,然后返回至包衣锅进行包衣。
实施例3.维生素A、D3和K1混合物在微粒中的包囊
含有92%维生素A棕榈酸酯、0.5%维生素D3和7.5%维生素K1的维生素混合物(可从Sigma-Aldrich Co.,St.Louis,Missouri买到)如实施例1或2所述,在流化床干燥器或包衣锅中被凝聚和双层包被。凝聚溶液由10%(w/w)麦芽糊精、2%(w/w)羧甲基纤维素和0.5%(w/w)BHT构成。内层由40%(w/w)大豆卵磷脂、50%(w/w)的17-硬脂和10%(w/w)的OxyLess构成。外层由90%(w/w)的27-硬脂和10%(w/w)蜂蜡(Frank B.Ross Co.-Rahway,NJ)构成。
实施例4.铁矿物在微粒中的包囊
硫酸亚铁(可从Sigma-Aldrich Co.,St.Louis,Missouri买到)如实施例1或2所述,在流化床干燥器或包衣锅中被凝聚和双层包被。凝聚溶液由10%(w/w)麦芽糊精、2%(w/w)藻酸钠和0.5%(w/w)抗坏血酸(均来自Sigma-Aldrich Co.,St.Louis,Missouri)构成。内层由40%(w/w)单酸甘油酯(Cognis GmbH Manheim,德国)、50%(w/w)的17-硬脂和10%(w/w)α-生育酚乙酸酯(Sigma-Aldrich Co.,St.Louis,Missouri)构成。外层由90%(w/w)的27-硬脂和10%(w/w)蜂蜡(Frank B.Ross Co.-Rahway,NJ)构成。
实施例5.消化酶在微粒中的包囊

实施例7.复合维生素B在微粒中的包囊

实施例8.胡萝卜素从油中的微粒释放的动力学
为了证明胡萝卜素在本发明微粒中的保护,如实施例2所述包囊天然虾青素(Cyanotech,Kailua-Kona,HI)。微粒在含有大豆油的小瓶中以低(0.5g/10ml)和高(1g/10ml)浓度混合,并以150RPM摇动1或2.5小时和测量过滤的油的OD475。表1显示色素从未包衣和包衣的颗粒释放到油溶液。类胡萝卜素从未包衣的粒子完全释放到油中,但在双重包衣的粒子中完全保留。胡萝卜素只在短时暴露于高温(100℃)以后从双重包衣的颗粒中完全释放。

未包囊和包囊的胡萝卜素(虾青素)在大豆油中的释放动力学通过475nm处的光密度测量。包囊的粒子保留胡萝卜素比未包囊的颗粒好10倍。受热之后,胡萝卜素从微囊体完全释放。
实施例9.胡萝卜素微粒在消化液中的释放动力学和胃防护。
为了证明胡萝卜素在本发明微粒中的保护,如实施例2和6所述生产包衣和未包衣的天然虾青素颗粒。造粒溶液包含2%藻酸钠和10%麦芽糊精。微粒在含有模拟胃液(pH 2)和模拟肠液(pH 7.4)的小瓶中混合,在37℃温育2小时。图3显示了胡萝卜素释放到消化液中。虽然胡萝卜素在两种消化液中从未包衣颗粒释放,但它在胃液中被包含在双层包衣颗粒内,却在肠液中完全释放。该实验证明了本发明包囊系统的胃防护。
实施例10.维生素A的双层包衣颗粒在烘焙的曲奇饼中的稳定性
为了证明本发明的微粒对烘焙的曲奇饼中维生素A的保护,将维生素A棕榈酸酯(BASF,Florham Park,NJ)造粒,并如实施例3所述双层包衣。包衣和未包衣的颗粒以850IU/g的相等浓度在Betty Crocker曲奇饼面粉(General Mills Inc.,Minneapolis,MN)中混合,制备曲奇饼并按照厂家说明书在200℃烘焙20分钟。烘焙的曲奇饼中维生素A含量显示在表2中。含有未包衣颗粒的烘焙曲奇饼中维生素A的浓度比其烘焙前的初始浓度低30%,而在含有双层包衣颗粒的烘焙曲奇饼中,维生素A的浓度没有显示出维生素A损失,如表2所示。未包衣颗粒在烘焙之后损失了30%的维生素A,而双重包衣颗粒保留了100%的维生素A。

15.制造微粒的方法,制造自由流动的固体微粒的方法,包括:
将生物活性物质与凝聚剂混合;
形成生物活性物质/凝聚物微粒芯;
将所述芯与至少一种乳化剂接触,从而形成包裹所述芯的第一层;和
将所述第一层与在所述分层的芯的表面上固化的至少一种固体脂肪接触。
这样的方法的组成为:
16.权利要求15的方法,其中所述芯通过凝聚或喷雾干燥生物活性物质和凝聚剂的混合物并筛分所述粒子至需要的尺寸而产生。
17.权利要求15的方法,其中凝聚粒子的尺寸是50-5000微米。
18.权利要求15的方法,所述生物活性物质选自维生素、矿物质、蛋白质、酶、必需氨基酸、必需脂肪酸、以及它们的组合。
19.权利要求15的方法,其中所述凝聚剂选自碳水化合物、蛋白质、以及它们的组合。
20.权利要求15的方法,其中所述乳化剂选自单酸甘油酯和二酸甘油酯、磷脂、蛋或大豆卵磷脂、脂肪酸蔗糖酯、脂肪酸聚甘油酯、以及它们的组合。
21.权利要求15的方法,其中可食用固体脂肪选自氢化动物或植物油、蜡、及其混合物。
22.权利要求15的方法,其中所述微粒除了乳化剂以外,还包含抗氧化剂,所述抗氧化剂选自生育酚、抗坏血酸棕榈酸酯、硫辛酸、类胡萝卜素、植物营养素、草本提取物、以及它们的组合。
23.权利要求15的方法,其中所述双层的浓度是微粒的约1至约50重量%。
24.权利要求15的方法,其中第一层中乳化剂的浓度是内层的约5至约50重量%。
25.权利要求15的方法,其中第二层中固体脂肪的浓度是第二层的约70至约100重量%。
