发明背景

发明概述

“可溶性碳酸盐”是指由碳酸或溶解的二氧化碳(其充分溶解并在所提供的浓度下与酸反应)所形成的任何盐。在水溶液中,碳酸根离子、碳酸氢根离子、二氧化碳和碳酸构成动态平衡。本文所用的术语“碳酸根”因此包括碳酸氢根(HCO3 -)和碳酸根(CO3 2-)形式及其混合物。因此,可溶性碳酸盐包括例如碳酸钾、碳酸氢钾、碳酸钠和碳酸氢钠。本发明因此包括组合物1.0,即一种泡腾可溶性固体(例如粉剂、粒剂或片剂),其包含:
a.酸源和可溶性碳酸盐;和
b.呈游离或盐形式的碱性氨基酸。
在一个实施方案中,碱性氨基酸(b)呈碳酸氢盐形式并因此可形成全部或部分的(a)的碳酸盐。本发明还包括下列组合物:
1.1组合物1.0,其中所述碱性氨基酸是精氨酸、赖氨酸、瓜氨酸、鸟氨酸、肌氨酸、组氨酸、二氨基丁酸、二氨基丙酸、其盐和/或其组合。
1.2组合物1.0或1.1,其中所述碱性氨基酸具有L-构型。

1.9前述组合物中的任一种,其中所述碱性氨基酸以相当于组合物总重量的约0.1%重量至约50%重量的量存在,碱性氨基酸的重量以游离碱形式计算。
1.10前述组合物中的任一种,其中所述酸源选自柠檬酸、苹果酸、酒石酸、己二酸、琥珀酸和富马酸、及其组合。
1.11前述组合物中的任一种,其中所述碳酸盐选自碳酸氢钠、碳酸氢钾、碳酸钠、精氨酸碳酸氢盐和碳酸钾。
1.12前述组合物中的任一种,所述组合物当溶于溶剂例如水中时产生二氧化碳。

1.14前述组合物中的任一种,所述组合物包含过碳酸、过硼酸、过硫酸、过焦磷酸(perpyrophosphate)和过一硫酸(monopersulfate)的碱金属盐。
1.15前述组合物中的任一种,所述组合物还包含氟化物或氟离子源。
1.16前述组合物中的任一种,所述组合物还包含防腐剂或抗微生物剂。
在本发明的一个实施方案中,提供组合物2.0,所述组合物包含呈片剂形式的组合物1.0-1.16中的任一种。
本发明也包括下列组合物:
2.1组合物2.0,所述组合物还包含润滑剂
2.2组合物2.0或2.1,所述组合物包含选自硬脂酸镁、苯甲酸钠、聚乙二醇、己二酸,及其组合的润滑剂,
2.3组合物2.0-2.2中的任一种,所述组合物还包含粘合剂,
2.4组合物2.0-2.3中的任一种,所述组合物包含选自葡萄糖、山梨醇、木糖醇、乳糖,及其组合的粘合剂。
本发明也包括方法3.0,即一种清洁牙科装置的方法,所述方法使用如上所述的组合物,以达到以下目的:
a.将精氨酸递送到口腔,
b.减少附着在装置上的生物膜和细菌,
c.和/或降低装置上致龋菌群的数量,
所述方法包括将本发明的组合物加入到含水溶剂中,将牙科器具浸泡在所得溶液中,其浸泡时间足以有效清洁所述牙科器具,然后将所述牙科器具在口腔内施用或使用。
所述方法还可用于促进装置使用者的口腔健康,以(i)减少或抑制龋齿的形成,(ii)减少、修复或抑制牙釉质的龋前缺损,例如经定量光导(light-induced)荧光(QLF)或龋齿的电学测量(ECM)所检测的,(iii)减少或抑制牙齿的去矿化并促进牙齿的再矿化,(iv)降低牙齿的超敏性,(v)减少或抑制牙龈炎,(vi)促进口腔内疮或伤口的愈合,(vii)降低产酸菌水平,(viii)提高精氨酸分解细菌的相对水平,(ix)抑制口腔内微生物生物膜的形成,(x)升高和/或维持牙菌斑pH水平为至少pH 5.5,(xi)减少牙菌斑的积聚,(xii)清洁牙齿和口腔,(xiii)使牙齿对致龋菌免疫,(xiv)减少侵蚀,(xv)增强系统健康,和/或(xvi)治疗或抑制口干症。
本发明的其它实施方案对于本领域技术人员来说将会是显而易见的。
发明详述
泡腾通常是由可溶性酸和可溶性碱在水中反应而产生二氧化碳或氧气所引起。这样的酸和碱是本领域众所周知的。这类酸包括可溶性有机酸,例如柠檬酸、苹果酸、酒石酸、己二酸、琥珀酸和富马酸。这类酸在加入到水中之前通常呈盐形式。碱包括氨基酸的碱性盐、以及水溶性碳酸盐和碳酸氢盐,例如碳酸氢钠、碳酸氢钾、碳酸钠和碳酸氢钾。这类碱在加入到水中之前通常呈盐形式。已经惊奇地发现,碱性氨基酸盐不仅可用作碱性盐,而且这类碱性氨基酸盐也可给口腔带来益处。
本发明的清洁用组合物包含清洁剂和氧化剂,例如产生氯的化合物(例如二氯代异氰尿酸酯),以及无氯漂白剂(包括过碳酸、过硼酸、过硫酸、过焦磷酸和过一硫酸的碱金属盐)。无水过硼酸盐例如过硼酸钠可用于通过释放氧气而引起泡腾,其除了引起泡腾之外,还有助于漂白牙科器具。过硼酸盐是本领域已知的,并包括过硼酸钙、过硼酸铵、过硼酸镁和无水过硼酸钾。过硼酸盐可占组合物总重量的约5%至约25%。单过氧邻苯二甲酸也可用作义齿清洁用组合物的漂白剂,而过硼酸盐或过一硫酸氢盐例如过一硫酸氢钾(potassium monopersulfate)可与单过氧邻苯二甲酸联用。一个优选的漂白剂是过一硫酸氢钾(potassium peroxymonosulphate),其可占组合物的约2%至约15%重量。其它清洁剂包括碱性次氯酸盐和碱性过氧化物,以及β-1,3-葡聚糖酶。
不想受到具体理论的束缚,据信口腔中的碱性氨基酸是由某些种类的细菌,例如血链球菌(S.sanguis)代谢的,这类细菌为不致龋的并与致龋菌,例如变性链球菌(S.mutans)竞争在牙齿上和口腔中的位置。精氨酸分解细菌(arginolytic bacteria)可利用精氨酸和其它碱性氨基酸,产生氨气,因而升高其环境pH,而致龋菌则代谢糖,产生趋于降低牙菌斑pH和使牙齿去矿化,最终生成牙洞的乳酸。据信使用本发明的组合物可导致精氨酸分解细菌相对增加和致龋菌相对减少,导致更高的牙菌斑pH。
可用于本发明的组合物和方法的碱性氨基酸不仅包括天然存在的碱性氨基酸例如精氨酸、赖氨酸和组氨酸,而且包括分子中具有羧基和氨基的任何碱性氨基酸,所述碱性氨基酸是水溶性的并提供pH约7或更大的水溶液。因此,碱性氨基酸包括但不限于精氨酸、赖氨酸、瓜氨酸、鸟氨酸、肌氨酸、组氨酸、二氨基丁酸、二氨基丙酸、其盐和/或其组合。在一个具体的实施方案中,碱性氨基酸选自精氨酸、瓜氨酸和鸟氨酸,优选精氨酸,例如l-精氨酸。
本发明的组合物是用于口腔的并任选可被咽下,所以用于本发明的盐在所提供的用量和浓度下,对这类使用而言应当是安全的。合适的盐包括本领域已知的为药学上可接受的盐的那些盐,通常认为在所提供的用量和浓度下是生理上可接受的。生理上可接受的盐包括由药学上可接受的无机或有机的酸或碱所衍生的那些,例如由形成生理上可接受的阴离子的酸所形成的酸加成盐(例如盐酸盐和氢溴酸盐),以及由形成生理上可接受的阳离子的碱所形成的碱加成盐,例如衍生自碱金属(例如钾和钠)的那些或衍生自碱土金属(例如钙和镁)的那些。可以用本领域已知的标准方法来获取生理上可接受的盐,所述方法例如通过使足够碱性的化合物例如胺与合适酸反应,生成生理上可接受的阴离子。一种优选的盐是碳酸氢盐,例如精氨酸碳酸氢盐。
在各种实施方案中,碱性氨基酸以组合物总重量的约0.1%重量至约50%重量,组合物总重量的约1%重量至约40%重量,例如组合物总重量的约1.5%重量、10%重量、20%重量或30%重量的量存在。
如果本发明的组合物可任选包括氟化物或氟离子源,例如当配制成溶于溶剂而用作漱口剂时。多种多样的产生氟离子的原料可用作本发明组合物中的可溶性氟化物的来源。合适的产生氟离子的原料的实例可参见Briner等的美国专利号3,535,421;Parran,Jr.等的美国专利号4,885,155和Widder等的美国专利号3,678,154,这些专利通过引用结合到本文中。代表性的氟离子源包括但不限于氟化亚锡、氟化钠、氟化钾、一氟磷酸钠、氟硅酸钠、氟硅酸铵、氟化胺、氟化铵,及其组合。在某些实施方案中,氟离子源包括氟化亚锡、氟化钠、一氟磷酸钠及其混合物。因此,这类泡腾粉剂也可含有氟离子源或供氟的成分,其用量足以提供约25ppm至约25,000ppm氟离子,通常至少约500ppm,例如约500至约2000ppm,例如约1000至约1600ppm,例如约1450ppm的氟离子。在一个实施方案中,可以约0.01%重量-约10%重量或在另一个实施方案中,以约0.03%重量-约5%重量或在另一个实施方案中,以约0.1%重量-约1%重量的组合物的水平,将氟离子源加至本发明组合物中。提供适当水平的氟离子的氟化物盐的重量显然随盐中的抗衡离子的重量而变化。
本发明的组合物也可包括防腐剂和抗微生物剂化合物,例如三氯生、草药提取物和精油(例如迷迭香提取物、麝香草酚、薄荷醇、桉油醇、水杨酸甲酯)、双胍(bisquanide)防腐剂(例如氯己定、阿来西定或奥替尼啶)、季铵化合物(例如西吡氯铵)、酚防腐剂、海克西定、聚维酮碘、地莫匹醇、5-正辛酰-3’-三氟甲基苯基水杨酰胺(salifluor)、金属离子(例如锌盐,例如柠檬酸锌)、血根碱、蜂胶和抗生素。当本发明的泡腾粉末剂要配制成溶解于溶剂而形成漱口剂时,这类防腐剂和抗微生物化合物是理想的。
本发明的组合物也可包括一种或多种矫味剂。本发明实践所用的矫味剂包括但不限于精油以及各种矫味用的醛、酯、醇和类似原料。精油的实例包括留兰香油、薄荷油、冬青油、黄樟油、丁香油、鼠尾草油、桉树油、马郁兰油、肉桂油、柠檬油、白柠檬油、葡萄柚油和甜橙油。有用的还有化学品,如薄荷醇、香芹酮和茴香脑。某些实施方案采用薄荷和留兰香油。产生泡腾现象的各种酸和碱也可以是矫味剂,例如柠檬酸和苹果酸。
将矫味剂以约0.1-约5%重量和约0.5-约1.5%重量的浓度掺入口服组合物中。在单次口腔护理组合物用量(即单次用量)中矫味剂的用量为约0.001-0.05%(重量),在另一个实施方案中为约0.005-0.015%(重量)
可将本发明的清洁用组合物压制成片剂形式,例如得到单次用量形式以添加到溶剂中。生产片剂的方法和常用的片剂组合物是本领域众所周知的。片剂通常含有本领域技术人员已知的粘合剂。优选地,粘合剂是可溶的,并包括例如葡萄糖、山梨醇、木糖醇和乳糖。优选地,粘合剂的用量能使片剂足够硬(能经受处理),足够软(当加入到溶剂中能崩解)和足够干燥(使其稳定)。
片剂也可包含有助于让片剂从压片机上脱出的润滑剂。这类润滑剂是本领域技术人员已知的,并包括硬脂酸镁、苯甲酸钠、聚乙二醇和己二酸。
Claims (25)
1.一种泡腾可溶性固体组合物,所述组合物包含(i)酸源和可溶性碳酸盐,和(ii)呈游离或盐形式的碱性氨基酸。
2.权利要求1的组合物,其中所述碱性氨基酸是精氨酸、赖氨酸、瓜氨酸、鸟氨酸、肌氨酸、组氨酸、二氨基丁酸、二氨基丙酸、其盐和/或其组合。
3.权利要求1或2的组合物,其中所述碱性氨基酸具有L-构型。
4.前述权利要求中任一项的组合物,其中所述碱性氨基酸是精氨酸。
5.前述权利要求中任一项的组合物,其中所述碱性氨基酸呈可溶性碳酸盐形式。
6.权利要求5的组合物,其中所述碱性氨基酸是精氨酸碳酸氢盐。
7.前述权利要求中任一项的组合物,其中所述可溶性碳酸盐包含呈碳酸盐形式的碱性氨基酸。
8.前述权利要求中任一项的组合物,其中所述碱性氨基酸以相当于所述组合物总重量的约0.1%重量至约50%重量的量存在,所述碱性氨基酸的重量以游离碱形式计算。
9.前述权利要求中任一项的组合物,其中所述酸源选自柠檬酸、苹果酸、酒石酸、己二酸、琥珀酸和富马酸。
10.前述权利要求中任一项的组合物,其中所述可溶性碳酸盐选自碳酸氢钠、碳酸氢钾、精氨酸碳酸氢盐、碳酸钠和碳酸钾。
11.前述权利要求中任一项的组合物,该组合物在溶于溶剂时产生二氧化碳。
12.前述权利要求中任一项的组合物,所述组合物包含漂白剂。
13.前述权利要求中任一项的组合物,所述组合物包含过碳酸、过硼酸、过硫酸、过焦磷酸和过一硫酸的碱金属盐。
14.前述权利要求中任一项的组合物,所述组合物包含氟化物或氟离子源。
15.前述权利要求中任一项的组合物,所述组合物包含防腐剂或抗微生物剂。
16.呈粉剂或粒剂形式的前述权利要求中任一项的组合物。
17.呈片剂形式的前述权利要求中任一项的组合物。
18.权利要求17的组合物,所述组合物包含粘合剂。
19.前述权利要求中任一项的组合物,所述组合物还包含表面活性剂。
20.紧接着的一项前述权利要求的组合物,其中所述表面活性剂选自十二烷基硫酸钠和十二烷基醚硫酸钠。
21.一种清洁牙科装置的方法,所述方法包括将权利要求1-20中任一项的组合物加入到含水溶剂中,将牙科装置浸泡到所得溶液中一段有效时间,以清洁所述牙科器具。
22.用于以下目的的权利要求21的方法:所述目的是在使用所述装置时将碱性氨基酸递送到口腔和/或抑制致龋菌在装置上的生长。
