Abstract

本发明涉及一种从山竹果皮中提取α-倒捻子素的方法。其实现步骤包括：(I)提取：(II)浓缩：将上述每次的提取液合并，减压浓缩，得浓缩液；(III)萃取：(IV)硅胶柱层析：(V)粗结晶：(VI)重结晶：将一次粗结晶加入1～5倍体积量的有机溶剂进行热溶；热溶完毕后，放置，降温至室温，进行重结晶，结晶时间为10～20小时；得将重结晶液过滤，离心，真空干燥，得98％的α-倒捻子素。本发明工艺简单，操作时间短。

Description

从山竹果皮中提取α-倒捻子素的方法
技术领域
本发明涉及一种从植物中提取多酚类化合物的方法，尤其涉及一种从山竹果皮中提取α-倒捻子素的方法。
背景技术
山竹(Garcinia mangostana L.)又称山竹子、莽吉柿、凤果，是藤黄科(Guttiferae)藤黄属(Garcinia)常绿乔木山竹的果实。山竹果皮中因含有α-倒捻子素，常被用来治疗腹痛、腹泻、痢疾、感染性刨伤、化脓、慢性溃疡、白带、淋病等疾病。近年来，山竹果皮中的α-倒捻子素(α-mangostin)成为人们的研究热点，α-倒捻子素的提取方法有水提取、醇提取，大孔吸附树脂分离法等。
2006年9月20号公告的中国专利200610016501.4公开了一种“山竹果皮提取物及其提取方法以及药物用途”，该方法将山竹果皮粉碎后仅用30％-95％乙醇提取，回收乙醇而得到α-倒捻子素，提取工艺粗糙，杂质残留较多，收率低，对原料造成极大的浪费，提取物质不明确。
2007年9月5号公告的中国专利200710090207.2公开了“一种山竹提取物及其制备方法及其应用”该方法以山竹为原料，收率较低，仅为50％的α-倒捻子素。经水或低碳醇提取、大孔吸附树脂层析，生产工艺复杂，操作时间长，生产效率低，该方法没有对α-倒捻子素进一步的分离，得到的仅仅是α-倒捻子素的粗产品，
发明内容
本发明的目的在于提供一种从山竹果皮中提取α-倒捻子素的方法，其解决了背景技术中工艺复杂、操作时间长、收率低的问题。
本发明的技术方案如下：
一种从山竹果皮中提取α-倒捻子素的方法，其实现步骤包括：
(I)提取：
(i)取山竹全果或果皮，清洗，破碎；
(ii)加入3～10倍体积量的有机溶剂，提取1～3次，温度为0℃～90℃，每次提取时间为1～2小时；
(iii)将每次提取液离心，过滤，收取；
(II)浓缩：将上述每次的提取液合并，减压浓缩，得浓缩液；
(III)萃取：
(i)向浓缩液中加入5倍体积量的萃取试剂氯仿或乙酸乙酯进行萃取，次数为1～3次，得萃取液；
(ii)萃取液浓缩至原体积的1/20；
(iii)浓缩后的萃取液与硅胶按体积比1∶2混合均匀，真空干燥得样品；
(IV)硅胶柱层析：
(i)将上述样品与硅胶按质量比1∶10～30装柱；
(ii)用混合试剂进行洗脱，分段收集有效成分，得洗脱液；
(V)粗结晶：
(i)将洗脱液浓缩至原体积的1/10～1/100，放置结晶20～30小时；(是否和时间有关)
(ii)将结晶液通过离心机过滤，得到一次粗结晶；
(VI)重结晶：
(i)将上述一次粗结晶加入1～5倍体积量的有机溶剂进行热溶；
(ii)热溶完毕后，放置，降温至室温，进行重结晶，结晶时间为10～20小时；得
(iii)将重结晶液过滤，离心，真空干燥，得98％的α-倒捻子素。
上述步骤(I)中有机溶剂采用低碳醇、丙酮或者乙酸乙酯为宜。
上述步骤(II)中浓缩温度是60℃～80℃，压力为0.06MPa～0.08MPa。
上述步骤(III)中萃取试剂采用乙酸乙酯最佳。
上述步骤(IV)混合试剂由两种试剂混合制得，所述混合试剂是将第一组试剂中正戊烷、石油醚、正己烷或氯仿，与第二组试剂中乙酸乙酯、丙酮、甲醇或乙醇，按体积比1～80∶1，混合制得。
上述步骤(IV)中混合试剂为氯仿和丙酮，按体积比20∶1混合制得。
上述步骤(IV)中采用120目的硅胶，120目的硅胶与步骤(III)所得样品按质量比20∶1装柱。
上述步骤(VI)的有机溶剂为65％的乙醇或者甲醇。
本发明具有以下优点：
1.工艺简单。使用硅胶柱层析，层析过程简单易操作。
2.操作时间短。硅胶柱层析后，进行分离时所用的混合试剂比例适当，洗脱时间短。
3.成本低。提取过程中使用的试剂简单，易得到，使得成本降低。
4.收率高。对洗脱液过滤后重结晶，进一步分离提取α-倒捻子素，提高了收率。

具体实施方式
本发明的实现工艺如下：
(I)提取：
(i)取山竹全果或果皮，清洗，破碎；
(ii)加入3～10倍体积量的有机溶剂，诸如低碳醇、丙酮或者乙酸乙酯，提取1～3次，温度为0℃～90℃，每次提取时间为1～2小时；
(iii)将每次提取液离心，过滤，收取；
(II)浓缩：将上述每次的提取液合并，减压浓缩，浓缩温度为60℃～80℃，压力为0.06MPa～0.08MPa，得浓缩液；
(III)萃取：
(i)向浓缩液中加入5倍体积量的萃取试剂氯仿或乙酸乙酯进行萃取，次数为1～3次，萃取试剂取乙酸乙酯最佳，得萃取液；
(ii)萃取液浓缩至原体积的1/20；
(iii)浓缩后的萃取液与硅胶按体积比1∶2混合均匀，真空干燥得样品；
(IV)硅胶柱层析：
(i)将上述样品与硅胶按质量比1∶10～30装柱；
(ii)用混合试剂进行洗脱，分段收集有效成分，得洗脱液；
所述步骤(IV)混合试剂由两种试剂混合制得，所述混合试剂是将第一组试剂中的正戊烷、石油醚、正己烷或氯仿，与第二组试剂中乙酸乙酯、丙酮、甲醇或乙醇，按体积比1～80∶1，混合制得。
(V)粗结晶：
(i)将洗脱液浓缩至原体积的1/10～1/100，放置结晶20～30小时；
(ii)将结晶液通过离心机过滤，得到一次粗结晶；
(VI)重结晶：
(i)将上述一次粗结晶加入1～5倍体积量的有机溶剂进行热溶；
(ii)热溶完毕后，放置，降温至室温，进行重结晶，结晶时间为10～20小时；得
(iii)将重结晶液过滤，离心，真空干燥，得98％的α-倒捻子素。
实施例1：
取山竹果皮100kg清洗、粉碎，加入5倍量体积的50％乙醇，在90℃下提取3次，每次2小时，并分别收取第一、二、三次提取料液，将每次提取液通过离心机离心，过滤，合并，在温度为60℃，压力为0.06MPa的条件下，减压浓缩至无醇味，得到浓缩液，加入5倍量体积的乙酸乙酯将浓缩液萃取3次，合并萃取液，浓缩至原体积的1/20，加入2倍体积量的硅胶拌样，得到拌样品，将拌样品与120目的硅胶按质量比1∶10装柱，用氯仿和乙酸乙酯按体积比20∶1混合后洗脱除杂，再按体积比10∶1混合后洗脱有效成分，并用薄层跟踪，收集有效成分的洗脱液。将洗脱液浓缩至原体积的1/30，放置结晶20小时，通过离心机过滤，收集1次粗结晶，用2倍量95％乙醇热溶1次粗结晶，放置结晶24小时，过滤，离心，真空干燥得到含量为98.45％的α-倒捻子素3.74kg，参见图1及表1。
实施例2：
取山竹果皮100kg清洗、粉碎，加入4倍量体积的75％乙醇，在80℃下提取2次，每次2小时，分别收取第一、二、三次提取料液，将每次提取液通过离心机离心，过滤，合并，在温度为60℃，压力为0.06MPa的条件下，减压浓缩至无醇味，得到浓缩液，加入5倍量体积的乙酸乙酯将浓缩液萃取3次，合并萃取液，浓缩至比重原体积的1/20，加入2倍体积量的硅胶，得到拌样品，将拌样品与120目的硅胶按质量比1∶10装柱，用氯仿和丙酮按体积比40∶1混合后洗脱除杂，再按体积比20∶1混合后洗脱有效成分，并用薄层跟踪，收集有效成分的洗脱液。.将洗脱液浓缩至原体积的1/40，放置结晶20小时，通过离心机过滤，收集1次粗结晶，用3倍量80％乙醇热溶1次粗结晶，放置结晶24小时，过滤，离心，真空干燥得到含量为99.37％的α-倒捻子素3.92kg。参见图2及表2。

仪  器：waters 515     压  力：1800psi
检测器：waters 2487    流  速：1ml/min；
流动相：80％甲醇-0.1％磷酸
检测波长：280nm

Claims (4)

1.一种从山竹果皮中提取α-倒捻子素的方法，其实现步骤包括：
(I)提取：
(i)取山竹全果或果皮，清洗，破碎；
(ii)加入3～10倍体积量的乙醇，提取1～3次，温度为0℃～90℃，每次提取时间为1～2小时；
(iii)将每次提取液离心，过滤，收取；
(II)浓缩：将上述每次的提取液合并，减压浓缩，得浓缩液；
(III)萃取：
(i)向浓缩液中加入5倍体积量的乙酸乙酯进行萃取，次数为1～3次，得萃取液；
(ii)萃取液浓缩至原体积的1/20；
(iii)浓缩后的萃取液与硅胶按体积比1∶2混合均匀，真空干燥得样品；
(IV)硅胶柱层析：
(i)将上述样品与硅胶按质量比1∶10～30装柱；
(ii)用氯仿和乙酸乙酯按体积比20∶1混合后洗脱除杂，再按体积比10∶1混合后洗脱有效成分，并用薄层跟踪，收集得到洗脱液；
或者用氯仿和丙酮按体积比40∶1混合后洗脱除杂，再按体积比20∶1混合后洗脱有效成分，并用薄层跟踪，收集得到洗脱液；
(V)粗结晶：
(i)将洗脱液浓缩至原体积的1/10～1/100，放置结晶20～30小时；
(ii)将结晶液通过离心机过滤，得到一次粗结晶；
(VI)重结晶：
(i)将上述一次粗结晶加入1～5倍体积量的乙醇进行热溶；
(ii)热溶完毕后，放置，降温至室温，进行重结晶，结晶时间为10～20小时得
(iii)将重结晶液过滤，离心，真空干燥，得98％的α-倒捻子素。
2.根据权利要求1所述的从山竹果皮中提取α-倒捻子素的方法，其特征在于：所述步骤(II)中浓缩温度为60℃～80℃，压力为0.06MPa ～0.08MPa。
3.根据权利要求2所述的从山竹果皮中提取α-倒捻子素的方法，其特征在于：所述步骤(IV)中混合试剂为氯仿和丙酮，按体积比20∶1混合制得。
4.根据权利要求3所述的从山竹果皮中提取α-倒捻子素的方法，其特征在于：所述步骤(IV)中采用120目的硅胶，120目的硅胶与步骤(III)所得样品按质量比20∶1装柱。
