Abstract
"PROCESSO PARA A DETEçãO DE COMPOSTOS QUE MODULAM OS EFEITOS DA PROTEìNA OBESA (OB)". Um processo para a deteção de um composto que imita, potencializa ou inibe o efeito fisiológico da proteína ob, cujo processo compreende: (a) para um composto que imita o efeito fisiológico da proteína ob, avalaição do efeito do composto sobre um elemento resposta ADN de sinal transdutor e ativador de transcrição (STAT) ativado com proteína ob acoplado a um gene repórter; ou (b) para um composto que potencializa ou inibe o efeito fisiológico da proteína ob, avaliação do efeito do composto sobre a resposta provida pela proteína ob sobre um elemento resposta ADN STAT ativado com proteína ob acoplado a um gene repórter; onde, o elemento resposta e o repórter sendo expressos em uma linha de célula responsiva a proteína ob, cuja linha de célula é selecionada da lista consistindo em: uma linha de célula derivada hipotalâmica; uma linha de célula derivada feocromocitoma; uma linha de célula derivada hematopoiética; uma linha de célula derivada pancreática; uma linha de célula derivada de fígado; uma linha de célula derivada preadipócito; uma linha de célula derivada de músculo esqueletal; e uma linha de célula derivada do ovário; ou o elemento resposta e o repórter são expressos em uma linha de célula, cuja linha de célula (a linha de célula engenheirada) é também transfectada com um polipeptídeo que é capaz de mediar a estimulação por proteína ob de um elemento resposta ADN STAT ativado com proteína ob e contém as apropriadas proteínas STAT."PROCESS FOR THE DETECTION OF COMPOUNDS THAT MODULATE THE EFFECTS OF OBESE PROTEIN (OB)". A process for the detection of a compound that mimics, enhances or inhibits the physiological effect of the ob protein, whose process comprises: (a) for a compound that mimics the physiological effect of the ob protein, evaluating the effect of the compound on a DNA response element transducer signal and transcription activator (STAT) activated with ob protein coupled to a reporter gene; or (b) for a compound that enhances or inhibits the physiological effect of the ob protein, evaluating the effect of the compound on the response provided by the ob protein on an STAT DNA response element activated with ob protein coupled to a reporter gene; where, the response element and the reporter being expressed in a cell line responsive to ob protein, whose cell line is selected from the list consisting of: a hypothalamic derived cell line; a pheochromocytoma-derived cell line; a hematopoietic derived cell line; a pancreatic derived cell line; a liver derived cell line; a preadipocyte derived cell line; a skeletal muscle-derived cell line; and a cell line derived from the ovary; or the response element and the reporter are expressed in a cell line, whose cell line (the engineered cell line) is also transfected with a polypeptide that is capable of mediating the ob protein stimulation of a DNA-activated STAT response element ob protein and contains the appropriate STAT proteins.
