    A few members of the bacterial genus Thermus have been shown to be incomplete denitrifiers, terminating with nitrite (NO2−) or nitrous oxide (N2O). However, the denitrification abilities of the genus as a whole remain poorly characterized. Here, we describe diverse denitrification phenotypes and genotypes of a collection of 24 strains representing ten species, all isolated from a variety of geothermal systems in China. Confirmed terminal products of nitrate reduction were nitrite or N2O, while nitric oxide (NO) was inferred as the terminal product in some strains. Most strains produced N2O; complete denitrification was not observed. Denitrification phenotypes were largely consistent with the presence of denitrification genes, and strains of the same species often had the same denitrification phenotypes and largely syntenous denitrification gene clusters.     The genus is an unique actinobacterial group that widely distributed in hypersaline environments. In this study, we investigated the growth conditions, transcriptome analysis, production and accumulation of ectoine by YIM 90087 under salt stress. The colony color of YIM 90087 changed from yellow to white under salt stress conditions. Accumulation of ectoine and hydroxyectoine in cells was an efficient way to regulate osmotic pressure. The ectoine synthesis was studied by transferring the related genes (, and ) to . Transcriptomic analysis showed that the pathways of ABC transporters (ko02010) and glycine, serine, and threonine metabolism (ko00260) played a vital role under salt stress environment.     Five bacterial strains isolated from water reservoirs of cooling systems were characterized using polyphasic taxonomic approaches. The isolates were observed to be Gram-negative, strictly aerobic and non-motile. Growth was enhanced in the presence of L-cysteine. The major fatty acids (>5 %) for strains were C10:0, C16:0, C16:0 3-OH, C18:0 3-OH and C18:1 9c. Ubiquinone-8 was detected as the respiratory quinone while the polar lipid profile consisted of phosphatidylethanolamine, diphosphatidylglycerol, phosphatidylglycerol, phosphatidylcholine, three unidentified phospholipids, two unidentified aminophospholipids, and three unidentified glycolipids.     To characterize a novel endoglucanase, Celal_2753, from the psychrophilic bacterium Cellulophaga algicola IC166(T). Celal_2753 was purified to homogeneity with a yield of 81 % and with a molecular weight of 40 kDa on SDS-PAGE. It had maximum hydrolytic activity towards carboxymethyl cellulose at 40 °C and pH 6. It showed 33 % of the maximum activity at 10 ºC. Its activity increased to 272-316 % in the presence of 0.25-2 M NaCl and KCl at 40 °C. Celal_2753 was stable in the presence of 10 % (v/v) Tween 20, 10 % (v/v) Triton X-100, 16 mM SDS, 6 M urea or 2 M guanidine hydrochloride. Celal_2753 that had been boiled for 5 min recovered 55 % of its initial activity by incubating at 30 °C for 60 min. Because of its cold-adapted, thermotolerant and denaturant-stable properties, endoglucanase Celal_2753 is promising in detergent industry and bioethanol production.

    A Gram-stain positve marine bacterium, designated NIO-1109T, was isolated from a marine sediment sample from the Chorao Island Goa, India. Phenotypic and chemotaxonomic characteristics and data from a phylogenetic analysis based on 16S rRNA gene sequences indicated that strain NIO-1109T was related to the genus Exiguobacterium. Strain NIO-1109T exhibited >98.0% gene sequence similarity with respect to Exiguobacterium acetylicum DSM 20416T (99.0%), and Exiguobacterium indicum IAM 15368T (98.6 %), other species were showing <98% similarity. The DNA-DNA hybridization relatedness between strain NIO-1109T, Exiguobacterium acetylicum DSM 20416T, and Exiguobacterium indicum LMG 23471T were less than 70% (33 and 37%, respectively).     A novel actinomycete strain, YIM 001T, was isolated from a soil sample collected from Hebei province, People's Republic of China. The strain was characterized by white to grey aerial mycelium. Long spore chains, borne on the aerial mycelium, were straight to Rectiflexibiles; the spore chains were composed of non-motile and coccoid spores with a warty surface. The cell wall of strain YIM 001T contained ll-diaminopimelic acid (A2pm) and traces of meso-A2pm. Whole-cell hydrolysates contained mainly glucose and small amounts of xylose, galactose and arabinose. The menaquinones were MK-9(H4) (4·6 %), MK-9(H6) (60 %), MK-9(H8) (30·7 %) and MK-9(H10) (4·7 %). Phosphatidylethanolamine was the diagnostic phospholipid.     A Gram-negative, non-motile, rod-shaped bacterium, designated strain AKS 1T, was isolated from a desert soil sample collected from Akesu, XinJiang Province, China. A taxonomic study, including phylogenetic analysis based on 16S rRNA gene sequences and phenotypic characteristics, was performed on the novel isolate. The predominant menaquinone of strain AKS 1T was MK-7. The major fatty acids included i-C15 : 0, ai-C17 : 1 B/i-C17 : 1 I and i-C17 : 0 3-OH. The G+C content of the DNA was 51.4 mol%. Based on the results of phenotypic and genotypic characteristics, strain AKS 1T should be assigned as representing a novel species of the genus Pontibacter, for which the name Pontibacter akesuensis is proposed.     A Gram-staining-negative, catalase-positive, carbaryl-degrading, non-spore-forming, non-motile, rod-shaped bacterium, designated strain X23T, was isolated from a wastewater treatment system.     A strain of thermotolerant actinomycete, designated YIM 75948T, was isolated from a soil sample in Yunnan province, China. The strain grew at 25–55 °C (optimum 37 °C). The substrate mycelium and aerial mycelium produced on Czapek’s agar were both pale yellow to white. The diagnostic diamino acid of the cell-wall peptidoglycan was meso-diaminopimelic acid, and the whole-cell sugars were mannose, ribose, glucose, galactose and arabinose. The major fatty acids were iso-C15 : 0, iso-C16 : 0 and C16 : 0 and the predominant respiratory quinone was MK-9(H4). The polar lipids consisted of phosphatidylethanolamine, phosphatidylethanolamine with hydroxy fatty acids, diphosphatidylglycerol, phosphatidylinositol and two unidentified phospholipids.     A mesophilic bacterium, designated strain YIM 31327T, was isolated from a forest soil sample collected from Yunnan Province, China, and was then investigated using a polyphasic approach. The strain grew optimally at 28–30 °C and pH 7·2. The cells were Gram-negative, short, rod-shaped, motile and non-spore-forming with flagella. The major ubiquinone was Q-8 and the cellular fatty acids were C16 : 0 and C12 : 0. The DNA G+C content of strain YIM 31327T was 62·8 mol%. Phylogenetic analysis revealed that strain YIM 31327T was a member of the β-Proteobacteria, being most closely related to Duganella zoogloeoides, with which it exhibited less than 96 % 16S rRNA gene sequence similarity. On the basis of the phenotypic and genotypic differences between strain YIM 31327T and D. zoogloeoides, a novel species, Duganella violaceinigra sp. nov., is proposed, with YIM 31327T (=CIP 108077T=KCTC 12193T) as the type strain.

    Two novel thermophilic, spore-forming bacterial strains, T-11T and E-112T, were isolated from hot springs in Tengchong and Eryuan counties of Yunnan province in south-west China. The strains were Gram-stain-positive rods, occurring singly or in chains. Growth of strain T-11T was observed between 30 and 75 °C (optimum 50 °C) and at pH 7–11 (optimum pH 8.5), while the temperature range for strain E-112T was 35–70 °C (optimum 55 °C) and the pH range was 7.0–11.0 (optimum pH 8.0). The DNA G+C contents of strains T-11T and E-112T were 41.1 and 42.6 mol%, respectively. On the basis of 16S rRNA gene sequence similarity, the two strains were shown to be related most closely to Anoxybacillus species. The chemotaxonomic characteristics [predominant isoprenoid quinone menaquinone 7 (MK-7); major fatty acids iso-C15 : 0 and iso-C17 : 0] also supported the affiliation of strains T-11T and E-112T to the genus Anoxybacillus.     A novel non-sporulating, non-motile, catalase- and oxidase-positive, strictly aerobic, Gram-negative, rod-shaped bacterial strain, designated DCA-1T, was isolated from activated sludge collected from a butachlor wastewater treatment facility. The strain was able to degrade about 85 % of 100 mg butachlor l−1 within 5 days of incubation. Growth occurred in the presence of 0–6 % (w/v) NaCl [optimum, 1 % (w/v) NaCl] and at pH 5.5–9.0 (optimum, pH 7.0) and 15–35 °C (optimum, 25–30 °C). Vesicular internal membrane structures and photoheterotrophic growth were not observed. The major respiratory quinone was ubiquinone 10 (Q-10) and the major cellular fatty acids were C18 : 1ω7c and 11-methyl C18 : 1ω7c. The genomic DNA G+C content of strain DCA-1T was 62.5 mol%.     The gene encoding esterase (CE1) from Bacillus pumilus ARA with a calculated molecular weight of 28.4 kDa was cloned, sequenced and efficiently expressed in Escherichia coli. The open reading frame of 747 nucleotides encoded a protein, which was classified as a carboxylesterase with an identity of 87 % to esterase from Bacillus subtilis 168. Recombinant CE1 was purified in a single step to electrophoretic homogeneity by IMAC (Ni2+). The enzyme displayed maximum activity toward p-nitrophenyl (pNP) acetate at 37–40 °C and pH 6.5–7.0. It was stable in the pH range from 6.5 to 8.0, and at temperature from 25 to 40 °C. Among four p-nitrophenyl esters tested, the best substrate was pNP acetate with Km and kcat values of 0.33 mM and 4.07 s−1, respectively. Amounts of 2 mM Ca2+ and Co2+ significantly increased the esterase activity to 190 and 121 %, respectively. These results suggest that CE1 has very attractive applications of increasing feed digestibility in animal nutrition in this moderate temperature range.

    A novel actinobacterial strain, designated NCCP-696T, was isolated from the Thal desert in Punjab, Pakistan, and characterized by using a polyphasic taxonomy approach. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain NCCP-696T belongs to the genus Nocardioides and showed the highest level of sequence similarity with respect to Nocardioides panacisoliGsoil 346T (98.2 %) and less than 96.4 % to the strains of other species of the genus Nocardioides. Cells of strain NCCP-696T were Gram-positive, aerobic, non-motile rods and formed cream-coloured colonies. The strain was positive for oxidase and catalase. Growth occurred at 20-42 °C (optimum 30-37 °C) at pH 5.5-9.0 (optimum pH 7.0) and with 0-4 % NaCl (optimum 0-2 %, w/v).     Copper sulfate (CuSO4) has been widely used as an algicide to control harmful cyanobacterial blooms (CyanoHABs) in freshwater lakes. However, there are increasing concerns about this application, mainly due to the general toxicity of CuSO4 to other aquatic species and its long-term persistence in the environment. This study reported the isolation and characterization of two natural algicidal compounds, i.e., tryptamine and tryptoline, from Streptomyces eurocidicus JXJ-0089. At a concentration of 5 μg/ml, both compounds showed higher algicidal efficiencies than CuSO4 on Microcystis sp. FACHB-905 and some other harmful cyanobacterial strains. Tryptamine and tryptoline treatments induced a degradation of chlorophyll and cell walls of cyanobacteria.     A novel Gram-staining negative, non-motile, rod-shaped and watermelon-red-pigmented aerobic bacterium strain, designated YIM 77969T, was isolated from a soil sample of Jiuxiang cave, a tourism cave located in Yiliang county, Yunnan province, south-west China. Phylogenetic analysis based on the 16S rRNA gene sequence indicated that strain YIM 77969T belongs to the genus Hymenobacter, and was closely related to Hymenobacter tibetensis XTM003T (96.58%), Hymenobacter gelipurpurascens Txg1T (96.02%) and Hymenobacter xinjiangensis X2-1gT (95.80%). Growth was found to occur from 5 to 35 °C, at pH 5.0-9.0 and in the presence of 0-1 % (w/v) NaCl concentration. The predominant menaquinone was MK-7.     A halotolerant, round-endospore-forming, aerobic, Gram-positive bacterium, designated BH724T, was isolated from a solar saltern at Taean in Korea. Cells of this strain were rod-shaped and found to be non-motile. Strain BH724T grew at salinities of 0–10 % (w/v) NaCl with an optimum of 3 % (w/v) NaCl and at temperatures of 15–50 °C with an optimum of 40 °C. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain BH724T belonged to the genus Bacillus and that Bacillus aquimaris TF-12T, Bacillus marisflavi TF-11T and Bacillus vietnamensis JCM 11124T were its closest neighbours, sharing 97.3, 97.2 and 97.0 % 16S rRNA gene sequence similarity, respectively.     Two novel actinobacteria isolates, designated YIM 70009T and YIM 70081T, were characterized in order to determine their taxonomic position. Cells of strains YIM 70009T and YIM 70081T were cocci, although only the latter were motile. The G+C contents of their DNAs were 64·0 and 64·5 mol%, respectively. On the basis of chemotaxonomic characteristics and 16S rRNA gene sequence analysis, the two isolates were classified in the genus Nesterenkonia. DNA–DNA hybridization and comparison of phenotypic characteristics revealed that strains YIM 70009T and YIM 70081T differed from each other and from known species. Therefore, it is proposed that they represent two separate novel species of the genus Nesterenkonia: Nesterenkonia sandarakina sp. nov.     The taxonomic position of two Gram-positive strains, YIM 70084T and YIM 70097T, isolated from hypersaline soils was determined by a polyphasic approach. Cells of strain YIM 70084T are motile cocci, whereas those of strain YIM 70097T are non-motile rods. The G+C contents of their DNA are 64·4 and 66·7 mol%. Both strains had chemotaxonomic markers typical of the genus Nesterenkonia and formed a coherent cluster with Nesterenkonia species in a phylogenetic inference based on 16S rDNA sequence analysis, exhibiting less than 97 % similarity to each other and to the other two type strains of the genus. Phylogenetic distinction and differences in the peptidoglycan type, composition of cell-wall sugars, phospholipid patterns, the major menaquinones and other phenotypic characteristics indicate that the strains under study represent two novel species, Nesterenkonia halotolerans sp. nov.     A Gram-positive, non-endospore-forming bacterium, designated strain SK 18T, was isolated from surface soil of a landfill site by dilution plating on trypticase soy broth agar. Preliminary characterization of strain SK 18T via biochemical tests, analysis of fatty acid methyl esters and partial 16S rRNA gene sequencing placed it within the genus Microbacterium . Analysis of the cell wall indicated that the peptidoglycan was of cross-linkage type B, containing the amino acids lysine and ornithine and with muramic acid in the N-glycolyl form. The polar lipids were phosphatidylglycerol, diphosphatidylglycerol, an unidentified phospholipid and an unidentified glycolipid. The major fatty acids of the cell membrane were anteiso-C17 : 0, anteiso-C15 : 0 and iso-C16 : 0. These data further strengthened placement of the strain within the genus Microbacterium .     A Gram-positive, aerobic, non-motile, mesophilic strain, djl-6T, able to degrade carbendazim, was isolated from a carbendazim-contaminated soil sample from Jiangsu province, China. The taxonomic position of this isolate was analysed by using a polyphasic approach.     A Gram-positive, non-spore-forming isolate, designated YIM 70212T, was isolated from a hypersaline soil sample collected from Qinghai, north-west China. Cells of the isolate were orange-pigmented, motile cocci with multiple flagella. A polyphasic taxonomic investigation was carried out on the isolate. The organism grew at 10–45 °C and pH 7.5–11.0, with optimum growth at 28 °C and pH 8.0–9.5. Strain YIM 70212T grew optimally in the presence of 10 % NaCl, KCl or MgCl2.6H2O and growth was observed in 1–25 % NaCl, KCl or MgCl2.6H2O. The peptidoglycan type was A1γ. Ribose and minor amounts of galactose were detected as the whole-cell sugars. MK-5 was the only menaquinone. The major cellular fatty acids were ai-C15 : 0 (52.4 %) and ai-C17 : 0 (26.5 %).     An actinomycete strain (0345M-7T) was isolated from a soil sample from Yilan county, Taiwan. The isolate displayed substrate mycelia, upon which were borne short spore chains. The spore chains were composed of non-motile, smooth-surfaced, oval spores. Strain 0345M-7T had meso-diaminopimelic acid in its peptidoglycan. Whole-cell sugars were galactose, glucose, arabinose and ribose. The only phospholipid found was phosphatidylethanolamine. The predominant menaquinone was MK-9(H4). Mycolic acids were not detected. Major cellular fatty acids were iso-C16 : 0 (38.1 %) and C17 : 1 (25.4 %). The DNA G+C content of strain 0345M-7T was 68.9 mol%. On the basis of phenotypic and genotypic data, it is proposed that strain 0345M-7T (=BCRC 16802T=KCTC 19116T) should be classified as the type strain of a novel species of the genus Amycolatopsis, Amycolatopsis taiwanensis sp. nov.

    An aerobic bacterium was isolated from saline soil located in Qinghai, north-west China. The bacterium, designated YIM 70157T, was investigated using a polyphasic taxonomic approach. The Gram reaction of the organism was positive. Comparative 16S rRNA gene sequence analysis demonstrated the isolate to be a member of the genus Marinococcus, the closest phylogenetic neighbour of the unknown bacterium being Marinococcus halophilus DSM 20408T with a similarity of 99·4 %. The peptidoglycan type of YIM 70157T was A1γ, with meso-diaminopimelic acid as diagnostic diamino acid. The major fatty acids were ai-C15 : 0, ai-C17 : 0 and i-C16 : 0. The menaquinones were MK-7 and MK-6. The phospholipids were diphosphatidylglycerol and phosphatidylinositol. The G+C content of total DNA was 48·5 mol%. On the basis of phylogenetic and phenotypic evidence and DNA–DNA hybridization data, this isolate should be classified as a novel species of Marinococcus, for which the name Marinococcus halotolerans sp....
