Internal circuitry is energised whenever the incubator is connected to


mains power, and the mains power switch on the back of the incubator
is turned on, irrespective of whether or not the power STANDBY touch-
pad switch on the front panel has been pressed. Always turn off the mains
power switch on the back panel of the incubator and disconnect the unit
from mains power before fuse replacement, cord replacement, or
cleaning. Should any power cord or plug associated with the incubator
become cracked, frayed, broken or damaged it must be
replaced immediately.

To reduce the risk of electric shock, do not remove covers. Refer servicing
to qualified service personnel.

Protect the device from being splashed by liquid. Keep the power switch
dry. Should any liquid enter the device, discontinue use immediately.

2. ABOUT THE K-MINC-1000


2.1 Intended Use
The K-MINC-1000 is a microprocessor-controlled humidified, gassed
incubator intended for use in cell culture. The instrument is designed to
maintain temperature accurately at a user specified temperature within
the range of 350 to 400 Celsius, and to maintain accurate gas flow at a
user specified flow rate within the range of 15ml/min to 25ml/min. The
incubator utilises premixed gas to maintain optimum culture conditions
within the incubation chambers. All components in contact with the gas
flow, including external and internal gas lines, have undergone rigorous
testing to ensure a toxin free environment.

2.2 Contra-indication for Device Use


Should any electrical or mechanical fault occur while using the
K-MINC-1000, stop using the device until it has been checked by
authorised service personnel.

3.2 You Need to Supply

certificate of analysis, refer to §3.7.1.

l A source of sterile distilled water.
l A pressure manometer (to measure the K-MINC-1000 input gas
supply). This item is not required if the regulator has gas output
display gauges.
l A 14 mm (9/16") spanner.
l If a longer connecting hose is required, refer to §5.

3.7.3 Connection to Gas Bottle

The ferrules on the connecting hose have been pre-swaged before


shipping, so any ferrule and backnut on the bottle mounted regulator

It will be necessary to connect the hose to the regulator outlet fitting and
also to the K-MINC-1000 inlet fitting, thus the instructions below must be
performed on each end of the connecting hose.
To connect the hose to a fitting:
1. Ensure that the fitting body, tube end and ferrules are free of any
foreign materials.
2. Insert the tube adaptor with preswaged ferrules into the fitting body
until the front ferrule seats.
3. Tighten the back nut firmly by hand. Using a 14mm (9/16") spanner
tighten the nut slightly (usually 1/8 turn or less).

Check the seal is gas tight by covering with soapy water and looking for
the presence of bubbles. If bubbles are observed, disconnect and repeat
from step 1, tightening the nut further if no foreign material is found in
the fitting.

3.7.4 Series Connections of Devices

3.8 Humidification Flask


The K-MINC-1000 uses a disposable humidification flask, gas line and

2. Insert the flask into the appropriate position in the chamber.


The tubing exiting the flask should face the rear of the device.
Connect the filter with the Luer fitting to the gas outlet as shown
below. Rotate the filter and tubing anticlockwise 180° before
pressing onto the Luer fitting and rotating clockwise to lock the
filter in place. This will ensure no kinking or twisting of the
tubing occurs.

3. Insert the gas lines into the centre slots on each side of the
chamber as per the diagram below. Check to ensure that the gas
lines are not kinked and are seated correctly into the slots to
ensure they are not crimped when the lid is closed.

3.9 Activating the Device

3.10 Chamber Temperature Selection
When first turned on the incubator will default to a temperature of

3.11 Gas Flow Rate Selection


When first turned on the incubator will default to a gas flow of 15ml/min.
The front panel display will show actual gas flow in millilitres per minute
(ml/min).
To alter the gas flow rate settings depress the up or down arrow gas flow
selection touch-pad on the front panel. The gas flow will scroll up or
down in 5 ml/min increments.
When desired flow rate is reached remove finger from touch-pad. After
approximately one second the LED display will switch back to show the
actual flow rate and the new set flow rate will be saved.

3.12 Insertion of the Culture Dishes


The K-MINC-1000 has been designed to be used with NUNC® four-well

Four-well dishes or Petri dishes may be placed on the chamber base.
Ensure that they sit securely in the grooves designed to accept them.
There should be direct contact between the base of the dish and the
surface of the chamber base.
Use the magnetic backed white board on the chamber lids to record
culture dish contents with a marking pen. Pen markings can be removed
using an alcohol solution.

3.13 First Time Use


Leave the K-MINC-1000 to operate with both chambers at 37°C and gas
flow at 15ml/min for 24 hours to ensure any residual out-gassing of
components is complete.
Test each K-MINC-1000 chamber for pH maintenance using culture media
containing phenol red indicator (15µg/ml). Adjust the gas flow to 15
ml/min and place the culture media in culture wells in both incubation
chambers. After overnight incubation, observe that the phenol red
indicator is the correct colour (salmon pink).
If the unit is being used as an ICSI workstation a flow rate of 25ml/min is
advised. If the lids are opened infrequently the flow rate can be reduced
to 15 - 20ml/min.
The K-MINC-1000 is now successfully installed and commissioned.

3.14 Alarm Conditions


3.14.1 No Gas Flow or Gas Flow Out of Range
The gas flow display will show a "CO2" display if gas
flow differs from the set gas flow by greater than ±25%
(including no gas flow) for longer than a nominal
10 minutes.
The gas flow display will show an "Err" display and the
external alarm will be activated if gas flow differs from the
set gas flow by greater than 4 ml/min (including no gas
flow) for longer than 15 minutes. An "Err" display will take
precedence over a "CO2" display.

3.14.2 Temperature Out of Range


When the incubator has just been turned on or when the
set temperature has been adjusted, the temperature alarm
is deactivated for 40 minutes to allow the incubator to
reach stable conditions without constantly alarming.
After this time the temperature display will show "ERR" and
the external alarm will be activated if the set temperature
differs by greater than ±0.2°C, for longer than 2 minutes.

3.14.3 External Alarm


The K-MINC-1000 has the facility to connect to an external alarm monitor
that will alert staff of activated alarms out-of-hours. If an external alarm
is connected it will be triggered six minutes after the loss of mains power.
This external alarm is normally an "open circuit" and will "close" in the
event of the above. See §6 Technical Data for alarm contact rating. If the
power is restored within the limits of the trigger time the alarm would not
be activated.
Customer requirements for the external alarm monitor should be referred
to a company that specialises in this type of equipment.
3.14.4 Low Battery (power-fail external alarm)
In order for the mains power fail ‘external alarm’ feature described in
§3.14.3 to function, a battery has been fitted in the K-MINC-1000. This
battery is a long-life, no maintenance type which should offer many years
of service.
The K-MINC-1000 has an audible low battery warning that will sound
when the battery is almost exhausted and the incubator is operating.
Should this low battery alarm sound the K-MINC-1000 will continue to
operate, however the power-fail alarm may not function correctly. The
unit must be returned for servicing, as detailed in §4.6.
The alarm sound is a 0.1 second beep every 15 seconds.

4.3 Biannual Functionality Testing

are more than ±0.2ºC from the user set temperature for
more than two minutes, then contact your authorised service
agent. To test flow-rate, use an air calibrated gas flow meter
connected to the Luer lock outlet beneath the bacterial filter
as shown in §3.8. The gas flow meter should have no further
restriction on the outlet and be open to atmospheric
pressure. Start the gas flowing. The gas flow meter should
indicate twice the user set value ±15%. Twice the user set
value is observed because the display is a per chamber value.
An example would be to set the gas flow to 20ml/min, then
the expected result should lie between 34 to 46ml/min.
Check the flow rates over the user range of 15 to 25ml/min.
If measurements lie outside the expected ranges then contact
your authorised service agent. Gas flow rates for the K-MINC-
1000 are calibrated at 220C at sea level. Temperature and
atmospheric pressure must be taken into account when
calculating the expected gas flow. The table below shows
the flow obtained with altitude when the device has been set
at 15ml/min.

To test the external alarm contacts, turn the K-MINC-1000


off using the Mains Power Switch at the rear of the unit. The
contacts should close approximately 2 minutes later. Once
contacts close, turn the K-MINC-1000 on and allow to
operate for 1 hour before opening a chamber lid and misting
the chamber with cold water. The indicated temperature
should drop and the contacts should close approximately 2
minutes later. If the contacts fail to close then contact your
authorised service agent.

10. INSTALLATION AND SET-UP CHECKLIST
