Tribolium flour beetles produce defensive compounds, including quinones, putatively aimed at deterring predators and inhibiting microbes. Here we examine how effective the defensive secretions of Tribolium confusum and T. castaneum are at inhibiting growth of various microbes and how this varies with species, geographic strain, and sex of the beetles. We explore differences at both the kingdom and species level of common flour microbes in their susceptibility to defensive compounds. Beetle species and strains vary in their ability to inhibit microbial growth. In addition, microbes vary in their sensitivity to the beetles' defense compounds. The capability to suppress microbial growth is likely under stabilizing selection with optimum quinone production varying among populations and may be dependent on several environmental factors including temperature, humidity, and predators.

The genetic components responsible for qualitative and quantitative resistance of rice plants to three strains (CR4, CXO8, and CR6) of Xanthomonas oryzae pv. oryzae (Xoo) were investigated using a set of 315 recombinant inbred lines (RILs) from the cross Lemont (japonica) × Teqing (indica) and a complete linkage map with 182 well distributed RFLP markers. We mapped a major gene (Xa4) and ten quantitative trait loci (QTLs) which were largely responsible for segregation of the resistance phenotype in the RILs. The Teqing allele at the Xa4 locus, Xa4 T , acted as a dominant resistance gene against CR4 and CXO8. The breakdown of Xa4 T -associated resistance mediated by the mutant allele at the avrXa4 locus in the virulent strain CR6 results from significant changes in both gene action (lose of dominance) and the magnitude of gene effect (≈50% reduction). Nevertheless, Xa4 T still acted as a recessive QTL with a significant residual effect against CR6. The mutant alleles at the avrXa4 locus in CXO8 and CR6 that lead to a reduction in effect, or “breakdown”, of Xa4 T were apparently accompanied by corresponding penalties for their fitness. The quantitative component of resistance to Xoo in the RILs was largely due to a number of resistance QTLs. Most resistance QTLs mapped to genomic locations where major resistance genes and/or QTLs for resistance to Xoo, blast and sheath blight were identified in the same cross. Most QTLs showed consistent levels of resistance against all three Xoo strains. Our results suggest that a high level of durable resistance to Xoo may be achieved by the cumulative effects of multiple QTLs, including the residual effects of “defeated” major resistance genes.

A simple ultraviolet spectrophotometric method for the determination of simvastatin in methanol has been devised and compared with the existing pharmacopoeial RP-HPLC method for the estimation of the drug. Analytical parameters such as stability, selectivity, accuracy, and precision have been established for the method, using SIMS® tablets and human serum samples, and evaluated statistically to assess the application of the method. The method was validated under ICH and USP guidelines and was found to comprise the advantages for simplicity, stability, sensitivity, reproducibility, and accuracy for use as an alternative to existing nonspectrophotometric methods for the routine determination of the drug in pharmaceutical formulations and for pharmaceutical investigations involving simvastatin.

Two ionic liquids (ILs), namely (S,S)-1-butyl-3-(2′-hydroxy-cyclohexyl)-3H-imidazol-1-ium tetrafluoroborate and (S,S)-1-butyl-3-(2′-acetyl-cyclohexyl)-3H-imidazol-1-ium tetrafluoroborate have been employed as stationary phases in capillary gas chromatography. These new phases exhibit a column efficiency of 1,600 and 2,100 plates m−1 for IL 1 and IL 2, respectively, a wide operating temperature range and good thermal stability (bleeding temperature of 250 °C for IL 1 and 160 °C for IL 2). Inverse gas chromatography (GC) analyses were used to study the solvation properties of these ILs through a linear solvation energy model. The application of these ILs as new GC stationary phases was studied. These stationary phases exhibited unique selectivity for many organic substances, such as alkanes, ketones, esters, and aromatic compounds. The efficient separation of several mixtures containing compounds of different polarities and the good separation of fatty acid methyl esters (FAMEs) and cis/trans isomers indicate that these ILs may be applicable as a new type of GC stationary phases.

Aim  Bats communicate by emitting social calls, and these often elicit reactions in conspecifics. Many such vocalizations are species-specific so that unambiguous signals can be transmitted and interpreted by conspecifics. In species-rich assemblages, evolutionary pressures might prompt interspecific diversification of call structure so that communication with heterospecifics is avoided. In species-poor island communities, where no risk of miscommunication occurs, stabilizing selection should prevail and preserve call structure and function. Call structure in island bats might be inherited from colonizers from the mainland and be maintained with little change in the absence of selection from heterospecifics. To test this hypothesis we studied Pipistrellus maderensis, an insular taxon occurring on the Madeira Archipelago, the Canary Islands and the Azores. It is closely related to one of the most widespread European pipistrelles, Pipistrellus kuhlii. Pipistrellus maderensis most probably evolved from a common ancestor shared with P. kuhlii, or from founders of that taxon that colonized the islands. We hypothesized that on Madeira Island, where no risk of ambiguous communication with heterospecifics exists, the structure and function of social calls should have been preserved by stabilizing selection. Echolocation calls, subject to different selection pressures, may instead show more pronounced differences between P. maderensis and P. kuhlii.Location  Madeira Island (Portugal, Atlantic Ocean), central and southern Italy.Methods  We recorded social and echolocation calls from allopatric populations of the two pipistrelles and explored interspecific differences in time and frequency characteristics. We also conducted playback experiments by broadcasting recordings of social calls from P. kuhlii and P. maderensis (taken respectively in peninsular Italy and on Madeira) and monitoring the bats’ responses.Results  Social call structure showed a strong similarity between species, whereas echolocation calls were markedly different and exhibited a mean divergence of over 6 kHz in their frequency of maximum energy. On Madeira, P. maderensis significantly reduced flight activity when we broadcast P. kuhlii signals, as did P. kuhlii in Italy in response to P. maderensis calls.Main conclusions  Reliable interpretation of social calls provides benefits to both the signaller and the receiver because signals help to optimize food exploitation at foraging sites. In the absence of closely related species that can emit similar calls, this advantage may have acted as a strong evolutionary pressure, stabilizing social call structure in P. maderensis in insular ecosystems with limited foraging resources.

It is essential to use well defined and fully validated analytical methods to obtain reliable results that can be satisfactorily interpreted. This manuscript is intended to provide guiding principles for the evaluation of a method's overall performance. For this purpose, all of the variables of the method are considered, including sampling procedure, sample preparation, chromatographic separation, detection and data evaluation. The criteria considered are as follows: stability, selectivity, limits of quantification and of detection, accuracy, precision, linearity, recovery and ruggedness. A traditional focus of evolutionary paleoecology has been the reconstruction of the selective forces that have affected evolving lineages through time. If the history of those lineages is dominated by stasis and punctuation, however, this is at best an inadequate and at worst a misdirected research strategy for macroevolution, because long-term stasis implies that environmental factors may have less influence on evolving lineages than previously believed. Such reasoning has led some proponents of punctuated views to reject ecological interactions as predominant or even significant forces in evolution. This is not a necessary conclusion. It is possible to accept the empirical predominance of stasis in evolution and at the same time the importance of ecology in affecting the course of evolutionary trends within lineages. If stasis prevails, ecology matters in the evolution of lineages if either (1) stabilizing selection is an important cause of stasis or (2) ecological interactions play an important role in controlling the speciation process. Viewing allopatric speciation explicitly as a three-stage process (consisting of formation, persistence and differentiation of isolated populations) clarifies testing of the role of ecology in speciation and may redirect clade-specific evolutionary paleoecology towards more enlightening interaction with other areas of macroevolutionary study.
