Poecilia reticulata is a freshwater fish from the northeastern part of South America and spread widely to various countries in Asia and other continents. However, research about P. reticulate is limited even though it is a well-known fish species in Indonesia. The purpose of study was to identify the fish species of P. reticulata through DNA barcoding using the COI gene to determine the phylogenetic relationships among fish populations in East Java, Indonesia. In a present study, there were eight samples of P. reticulata from four different freshwater locations in East Java. Extraction, amplification, and sequencing of DNA samples were conducted to obtain the genetic data and construct a phylogenetic tree based on DNA sequences. BACKGROUND & OBJECTIVES The predatory potential of the larvivorous fishes can be affected by the presence of alternative preys. In the present study the predation pattern of the sewage dwelling Poecilia reticulata (Peters 1872) on the larvae of Culex quinquefasciatus Say 1823 (Diptera: Culicidae) was evaluated in the presence of alternative preys. METHODS The predation of Cx. quinquefasciatus larvae by different size groups of R. reticulata fishes was evaluated. In addition to this, the niche breadth (N) and diet breadth (B) were measured following Manly's selectivity index (Si) as an indicator of variation of such predation pattern in the presence of alternative prey types, like chironomid larvae and tubificid worms. In the branchial mitochondrion-rich (MR) cells of euryhaline teleosts, the Na+/K+/2Cl− cotransporter (NKCC) is an important membrane protein that maintains the internal Cl− concentration, and the branchial Na+/K+-ATPase (NKA) is crucial for providing the driving force for many other ion-transporting systems. Hence this study used the sailfin molly (Poecilia latipinna), an introduced aquarium fish in Taiwan, to reveal that the potential roles of NKCC and NKA in sailfin molly were correlated to fish survival rates upon salinity challenge. Higher levels of branchial NKCC were found in seawater (SW)-acclimated sailfin molly compared to freshwater (FW)-acclimated individuals. Transfer of the sailfin molly from SW to FW revealed that the expression of the NKCC and NKA proteins in the gills was retained over 7 days in order to maintain hypoosmoregulatory endurance. Meanwhile, their survival rates after transfer to SW varied with the duration of FW-exposure and decreased significantly when the SW-acclimated individuals were acclimated to FW for 21 days. Double immunofluorescence staining showed that in SW-acclimated sailfin molly, NKCC signals were expressed on the basolateral membrane of MR cells, whereas in FW-acclimated molly, they were expressed on the apical membrane. This study illustrated the correlation between the gradual reductions in expression of branchial NKCC and NKA (i.e., the hypoosmoregulatory endurance) and decreasing survival rates after hyperosmotic challenge in sailfin molly.

Genetic linkage maps of the guppy (Poecilia reticulata) were constructed from independent crosses between the Tuxedo strain and a feral line (Wildtype). Segregation patterns of random amplified polymorphic DNA (RAPD) markers and phenotypic markers were investigated in F2 offspring of Tuxedo ♂♂ × Wildtype ♀♀ and Wildtype ♂♂ × Tuxedo ♀♀ crosses. Among the 300 and 276 RAPD markers scored for the respective crosses, linkages were identified for 230 and 212, respectively. The Tuxedo ♂♂ × Wildtype ♀♀ and Wildtype ♂♂ × Tuxedo ♀♀ maps spanned 2100 Kosambi centiMorgans (cMK) and 1900 cMK, respectively, in 28 linkage groups. Average marker resolution was 10 cMK. Genome length was estimated at 4410 cMK and 4060 cMK for the respective crosses, with an average physical distance of 166 kbp/cMK. Several RAPD markers were closely linked to or mapped onto the loci for the sex-determining region (SdR), and the sex-linked black caudal-peduncle (Bcp) and red tail (Rdt) genes. These primary linkage maps are the initial step toward the construction of a composite high-density map to facilitate map-based cloning and marker-assisted selection of quantitative trait loci that are essential for the development of comprehensive breeding programs for the guppy.

Inbred lines are an important tool of genetic studies of all traits, including behavior. Independently derived strains of ornamental "designer" guppies are readily available and predicted to be inbred; however, little is known about actual levels of inbreeding in any of these strains or whether these lines differ in genetic traits that have not been under strong directional artificial selection. We genotyped five designer strains of guppies known to vary in their responses to predator cues and a wild reference population to determine whether designer strains show evidence of inbreeding and whether the strains differed from each other at five microsatellite loci. The designer strains exhibited lower allelic diversity and observed heterozygosity than the wild population. Observed heterozygosity departed significantly from expected heterozygosity for most markers in all five strains of designer guppies. Inbreeding coefficient (f) comparisons between the wild reference population and the designer strains show considerable inbreeding in the designer strains. F(is) values for the designer strains also provide evidence of inbreeding. Finally, F(st) values indicate that the designer strains differ significantly from each other and the wild population. We therefore concluded that designer guppies are inbred compared to wild populations and differ among strains, making them useful tools for genetic studies of behavioral or life history traits.

Traditional compounds used to treat fish diseases in aquaculture and the ornamental fish industry (such as formalin and malachite green) can be more toxic to the hosts than their parasites. With the reviviscence in the use of herbal products, various botanicals have been heralded as cures for particular pathogens, but the efficacy of these compounds for parasitic worms is questionable. Here, we tested a range of garlic (Allium sativum) products against a major aquarium pathogen, Gyrodactylus turnbulli, infecting the guppy (Poecilia reticulata). All garlic products significantly reduced parasite mean survival time in vitro, from 13 h to <1 h. In fully randomised trials, the number of parasites was also significantly reduced on infected fish exposed to garlic from different sources. Evidence on the effectiveness of low-cost, sustainable, biological vector-control tools for the Aedes mosquitoes is limited. Therefore, the purpose of this trial is to estimate the impact of guppy fish (guppies), in combination with the use of the larvicide pyriproxyfen (Sumilarv® 2MR), and Communication for Behavioral Impact (COMBI) activities to reduce entomological indices in Cambodia. In this cluster randomized controlled, superiority trial, 30 clusters comprising one or more villages each (with approximately 170 households) will be allocated, in a 1:1:1 ratio, to receive either (1) three interventions (guppies, Sumilarv® 2MR, and COMBI activities), (2) two interventions (guppies and COMBI activities), or (3) control (standard vector control). Likely one of the least appreciated guppy phenotypes to be found is Homozygous Asian Blau (Ab).  Such fish  are normally considered weak by-products of popular delta breedings involving blue iridophores and red color pigment;  Blue Topaz, Blue Grass, Half Black (HB) Neon quickly come to mind as examples.  Each strain is created by the removal of red color pigment, frequently an X-link, from phenotype.  It is still contained in genotype, only masked via epistasis.

41 Ambient exposure to a short synthetic peptide has enhanced fecundity (number of offspring) in 42 invertebrates and vertebrates, ostensibly by disinhibiting reproduction. In separate experiments 43 nematodes (Caenorhabditis elegans) and guppy fish (Poecilia reticulata) were exposed via 44 their aqueous environment to a dissolved synthetic hexamer peptide, IEPVFT (EPL036), at a 45 concentration of 1 μM. In the case of the worms peptide was added to their aqueous buffer daily 46 throughout the experiment (14 days); in the guppies peptide administration was on the first 15 47 alternate days in a 50-week experiment. Replicated ecological gradients are prime systems to study processes of molecular evolution underlying ecological divergence. Here, we investigated the repeated adaptation of the neotropical fish Poecilia mexicana to habitats containing toxic hydrogen sulphide (H2 S) and compared two population pairs of sulphide-adapted and ancestral fish by sequencing population pools of >200 individuals (Pool-Seq). We inferred the evolutionary processes shaping divergence and tested the hypothesis of increase of parallelism from SNPs to molecular pathways. Coalescence analyses showed that the divergence occurred in the face of substantial bidirectional gene flow. Population divergence involved many short, widely dispersed regions across the genome. Analyses of allele frequency spectra suggest that differentiation at most loci was driven by divergent selection, followed by a selection-mediated reduction of gene flow. Reconstructing allelic state changes suggested that selection acted mainly upon de novo mutations in the sulphide-adapted populations. Using a corrected Jaccard index to quantify parallel evolution, we found a negligible proportion of statistically significant parallel evolution of Jcorr  = 0.0032 at the level of SNPs, divergent genome regions (Jcorr  = 0.0061) and genes therein (Jcorr  = 0.0091). At the level of metabolic pathways, the overlap was Jcorr  = 0.2545, indicating increasing parallelism with increasing level of biological integration. The majority of pathways contained positively selected genes in both sulphide populations. Hence, adaptation to sulphidic habitats necessitated adjustments throughout the genome. The largely unique evolutionary trajectories may be explained by a high proportion of de novo mutations driving the divergence. Our findings favour Gould's view that evolution is often the unrepeatable result of stochastic events with highly contingent effects.

The interest in the ecological relevance of risk assessments and, thus, in in situ bioassays has been increasing in the last years. The present study developed a time- and cost-effective in situ bioassay, aiming at obtaining, in a short period of time and with a minimum of resources, a set of ecologically relevant toxicological information in a site-specific approach. Poecilia reticulata and Gambusia holbrooki were chosen as test species. Post-exposure feeding inhibition and the biomarkers acetylcholinesterase, lactate dehydrogenase and glutathione S-transferases were the endpoints tested. The battery of biomarkers as a whole was sensitive to the in situ exposure in an acid mine drainage impacted effluent, although responses varied between test species. Post-exposure feeding inhibition was the most sensitive endpoint, and its association with biomarker responses was discussed. The linkage between individual responses, such as feeding, and biomarkers suggested that, at least in this case, biomarkers can be relevant at higher levels of biological organization. Altogether, the proposed short-term in situ bioassay seems to be a promising tool, since it represents a reasonable compromise between sensitivity, time/cost-effectiveness and ecological relevance.

Variation in mating preferences coupled with selective predation may allow for the maintenance of alternative mating strategies. Males of the South American live-bearing fish Poecilia parae fall in one of five discrete morphs: red, yellow, blue, stripe-coloured tail (parae) and female mimic (immaculata). Field surveys indicate that the red and yellow morphs are the rarest and that their rarity is consistent across years. We explored the role of variable female mating preference and selective predation by visual predators in explaining the rarity of red and yellow males, and more generally, the maintenance of this extreme colour polymorphism. We presented wild-caught P. parae females and Aequidens tetramerus, the most common cichlid predator, with the five male colour morphs in separate trials to determine mating and prey preferences, respectively. We found that a large proportion of females shared a strong preference for the rare carotenoid-based red and yellow males, but a distinct group also preferred the blue and parae morphs. The cichlid predator strongly preferred red and yellow males as prey. Together, these results suggest that the interaction between premating sexual selection favouring and predation acting against the red and yellow morphs may explain their rarity in the wild. The trade-off between sexual and natural selection, accompanied by variation in female mating preferences, may therefore facilitate the maintenance of the striking colour polymorphism in P. parae.
