Recently, drug nanosuspensions have shown a potential for ophthalmic delivery. In this study, a hydrocortisone (HC) nanosuspension (NS) was developed using microfluidic nanoprecipitation as a recent, simple and cost-effective bottom-up technique of drug nanonization. For comparison, a second HC NS was prepared by top-down wet milling procedures. The produced nanosuspensions were characterized for particle size, shape and zeta potential. HC nanosuspensions of approximately 300 nm particle size were produced by adjusting experimental conditions of the two processing techniques. Results of X-ray diffraction and differential scanning calorimetry revealed that HC maintained the crystalline structure upon milling, while predominant amorphous particles were generated after precipitation. Ocular bioavailability of HC nanosuspensions was assessed in albino rabbits using HC solution as a control. A sustained drug action was maintained up to 9 h for the nanosuspensions compared to 5 h for the drug solution. The precipitated and milled NS achieved comparable AUC0–9h values of 28.06 ± 4.08 and 30.95 ± 2.2, respectively, that were significantly (P < 0.05) higher than that of HC solution (15.86 ± 2.7). After 2 months storage at room temperature, the milled HC NS showed good stability with no discernable changes in particle size, whereas the particle size of the precipitated HC NS increased to 440 nm.Changes in intraocular pressure (IOP) of Albino rabbits after administration of Hc solution and nanosuspensions produced by milling and precipitation.

Background Oxidized LDL (oxLDL) inhibits angiogenesis in part by downregulating endothelial basic fibroblast growth factor (bFGF). To determine the mechanism of the downregulation, we investigated the signal transduction pathway involving potential phospholipid mediators. Methods and Results Cultured bovine aortic endothelial cells were incubated with PBS (lipoprotein-free control), LDL, or copper oxLDL under serum-free conditions. At 24 hours, oxLDL (50 μg/mL) decreased bFGF mRNA (Northern blot), bFGF protein (Western blot and ELISA), and concomitant DNA synthesis, all by 40% to 50% compared with PBS. LDL had no effect. Pretreating the cells with 100 ng/mL pertussis toxin (PTX) for 18 hours before oxLDL exposure almost completely blocked the inhibitory effects of oxLDL. In this article, we explore a new fabrication process for a flexible, all polymer, active fluidic delivery system, incorporating a fusion of laser micromachining and microfabrication techniques as well as rapid prototyping technology. Here, we show selective fluidic delivery from isolated microchannels through an electrochemically driven pumping reaction, demonstrate the dispensing of dose volumes up to 5.5 μl, and evaluate the device’s performance in terms of its delivery speed and ejection efficiency. Finally, we move this work toward an implantable microfluidic drug delivery device by investigating the device’s biocompatibility through a statistical approach that overviews the viability of bovine aortic endothelial cells on polyimide and silicon substrates.

Several reports demonstrate that interleukin-1 alpha (IL-1 alpha) and interleukin-1 beta (IL-1 beta), although distantly related at the primary sequence level, bind to the same receptor. This raises the question of what separation, if any, there might be between their biologic activities. Receptor binding assays and cross-competition studies in three Epstein Barr virus (EBV) positive cell lines showed that a subset of IL-1 receptors may bind IL-1 alpha preferentially and that a separate subset may bind IL-1 beta preferentially. Crosslinking studies with 125I-IL-1 alpha or 125I-IL-1 beta demonstrated the expression of both relative molecular mass (Mr) 80,500 IL-1 receptor, known to be T cell-specific, and Mr 69,500 IL-1 receptor, recently detected on B-cell lineage only. Ehrlichia ruminantium (ER) is the causative agent of Heartwater, one of the most common tick-borne diseases affecting ruminants in African countries and West Indies. Although ER can be used as an inactivated vaccine for wild and domestic animals, there are currently no easy and reliable methods for the quantification of this obligate intracellular bacterium. This report describes the development of a SYBR Green I based real time PCR protocol for the quantification of ER for vaccine production purposes. The method was validated for four ER strains. The external-standard-based PCR protocol developed has a large dynamic quantitative range allowing accurate ER measurement in samples containing from 10(2) to 10(8) gene copies; the method is also reproducible and precise, with intra- and inter-assay coefficients below 5%. The detection limits were validated for samples collected from bovine aortic endothelial cell culture bulks, which are commonly used to produce the ER vaccine. In contrast to the methods based upon protein content, no interference from the host cells in ER quantification was observed. Furthermore, the extended applicability of the new technique was demonstrated by monitoring ER production in cell culture thus rendering it a valuable tool to ensure consistency between vaccine lots and to evaluate optimal vaccine dosage.
