Highly efficient (>90%) protocols were developed for in vitro regeneration from de-embryonated cotyledon explants of peanut (Arachis hypogaea L.). Phytohormone combinations and concentrations, explant source and orientation, period of incubation and the response of genotypes were examined for optimization of the regeneration efficiency. Adventitious shoot primordia could be induced from de-embryonated cotyledon explants when (1) the proximal end of the explant was kept in contact with the shoot induction medium-I supplemented with 5 mg l−1 BAP + 2 mg l−1 2,4-D for 4 weeks, or (2) the distal end was kept in contact with shoot induction medium-II supplemented with only BAP at 20 mg l−1 for the first 2 weeks, followed by subculture in the same medium containing 15 mg l−1 BAP for the next 2 weeks. Orientation of placing the explant on the above media was critical for in vitro regeneration. The factors affecting the morphogenic responses like, repetitive organogenesis, shoot elongation, in vitro flowering and rhizogenesis were examined. Shoot bud formation was genotype independent. Histological studies showed multicellular origin of adventitious shoot primordia. The protocols gave healthy and fertile plants within 4 months.

The effects of two methods of cryopreservation involving chemical vitrification and air desiccation) were studied on isolated embryonic axes of A. hypogaea. Vitrification with PVS2 and desiccation in a laminar flow cabinet resulted in high levels (70-90%) of whole plant recovery after cryopreservation. A desiccation protocol based on 1h exposure of explants to the air flow was successfully applied to six wild species of section Extranervosae, resulting in recovery levels of 70-90% after liquid nitrogen treatment.

Peanut allergy is a major cause of food-induced severe anaphylactic reactions. To date, no medical care is available to prevent and treat peanut allergy and therefore hypoallergenic peanut varieties are of considerable health political and economic interest. Major allergens that induce IgE-responses in peanut-sensitive patients are Ara h 1, Ara h 2 and Ara h 3/4. In order to identify hypoallergenic peanuts, commercially locally available peanut varieties were screened for their allergen content. Ara h 1-deficient peanuts from Southeast Asia were identified by SDS-PAGE, immunoblotting, inhibition assays and ELISA. 2-D PAGE analyses demonstrated the different compositions of the tested extracts and revealed a number of variations of the allergen patterns of peanuts from different varieties. Mediator release experiments of these peanut extracts demonstrated similar allergenicities as compared with standard peanut extract. These results indicate that the allergenicity of peanuts with reduced Ara h 1 content might be compensated by the other allergens, and thus do not necessarily cause a reduction of allergenicity.

Since the early 1950s, numerous reports have been published suggesting that peanut milk and peanut milk based products can be prepared in a wide variety of ways. Emphasis has shifted from preparing inexpensive milk like beverages, very nutritious but somewhat lacking consumers appeal, to using the peanut milk or peanut protein isolates as an animal milk extender without changing flavor, to develop more attractive fermented products, and to precipitate proteins from the milk in order to get a curd called "tofu," and to produce cheese analogs. Great attention has been paid to the improvement of the stability, sensory properties, and shelf-life of the milk, using physical and chemical treatments. Many efforts have been deployed for supplementing the products. Thanks to recent advances, the removal of aflatoxin from peanut milk can be achieved using Flavobacterium aurantiacum as a biodegradater. It has also found application as liquid coffee whitener. However, despite all these developments and publications, there is still a need for much more diversified studies in order to definitely overcome the stability, the nutty flavor, and sensory problems always encountered when producing peanut milk and some peanut milk based products.

4. Tahap 4 ditandai dengan sintesis enzim amilase, maltase dan protease.

Praktikum pertumbuhan tanaman dilaksanakan pada hari Senin, tanggal 20 April 2015 sampai 04 Mei 2015, bertempat di Laboratorium Pendidikan IV, Jurusan Biologi, Fakultas Matematika dan Ilmu Pengetahuan Alam, Universitas Andalas. Praktikum ini bertujuan untuk meneliti laju tumbuh daun Arachis hypogea sejak dari embrio dalam biji sampai daun mencapai ukuran tetap dan untuk mengamati daerah tumbuh pada akar dan batang Phaseolus radiatus. Hasil yang didapatkan Laju pertumbuhan daun kecambah Arachis hypogea mengalami pertambahan setiap hari dengan rata-rata 0,83; 1,8; 1,83; 2,5; 2;6 cm. Laju pertambahan ini dapat disajikan dalam bentuk kurva huruf S (kurva sigmoid). Pada percobaan daerah tumbuh akar dan batang pada Phaseolus radiatus tumbuh batang mangalami rata –rata pertambahan panjang 0,64 mm tiap individu dan tumbuh akar mengalami rata-rata pertambahan panjang 0,1 mm tiap individu.

Objectives were to evaluate effects of forage species, stocking rate, and supplementation rate on performance and physiology of grazing lactating Holstein cows under intensive rotational stocking management during summer. Eight treatments were arranged in a 2 x 2 x 2 factorial design. Animals (n = 62) grazed pastures of Tifton 85 bermudagrass or Florigraze rhizoma peanut, a tropical legume. Low and high stocking rates were 7.5 and 10.0 cows/ha for bermudagrass and 5.0 and 7.5 cows/ha for rhizoma peanut. Within each forage-stocking rate combination, cows were fed supplement at 0.33 or 0.5 kg of supplement (as-fed basis)/kg daily milk production. Cows grazing rhizoma peanut pastures produced more milk (16.9 vs. 15.4 kg/d) but had higher rectal temperatures (39.4 vs. 39.1 degrees C). The rapid growth of tilapia culture has stimulated the expansion of tilapia feed production and a search for novel protein sources to replace fish meal. Vegetable or plant sources are promising alternatives and legumes are both naturally abundant and high in protein content. A study was carried out to evaluate the effects of replacing the protein from fish meal with peanut (Arachis hypogaea) leaf meal (PLM) in diets for male tilapia (Oreochromis niloticus, initial body weight 75.3 g). Four isonitrogenous (35% CP) and isocaloric (18.834 kJ g−1) diets were prepared to include 0% (CON), 10% (PLM10), 20% (PLM20) and 30% (PLM30) of PLM protein. Average fish weights at the end of the 126-day experiment were not statistically different among the fish fed CON, PLM10 and PLM20 diets. The PLM30 diet produced the poorest growth performance. Organic matter and protein contents of fish were similar in the CON, PLM10 and PLM20 diets. Carcass chemical composition showed a decrease in body fat content as PLM replacement levels increased. The high survival ratio in all dietary groups (>97%) suggests that PLM can be used in O. niloticus feeds for long periods without affecting fish growth performance or health.
