O objetivo da presente pesquisa foi avaliar os graus de polimorfismos protéicos entre setenta e c... more O objetivo da presente pesquisa foi avaliar os graus de polimorfismos protéicos entre setenta e cinco linhagens de C. albicans isoladas da cavidade oral de crianças saudáveis provenientes de cinco categorias socioeconômicas e oito escolas (particulares e públicas) do município de Piracicaba, Estado de São Paulo, a fim de identificar subespécies de C. albicans e suas similaridades em grupos de populações infantis e estabelecer suas possíveis rotas de disseminação. Culturas celulares foram desenvolvidas em meio YEPD, coletadas por centrifugação e lavadas com solução salina gelada. As proteínas celulares totais foram extraídas por rompimento celular usando pérolas de vidro e submetidas à técnica de SDS-PAGE. Após a eletroforese, as bandas de proteínas foram coradas com Coomassie-blue e analisadas pelo conjunto de programas estatísticos NTSYS-pc versão 1.70. Matriz de similaridade e dendrograma foram gerados, pela aplicação do coeficiente de similaridade de Dice e do algoritmo UPGMA, respectivamente, os quais permitiram avaliar os graus de polimorfismo ou similaridade intra-específico, baseados nos padrões eletroforéticos de proteínas totais de isolados orais de C. albicans. Um total de 13 principais fenons (grupos) foi analisado de acordo com suas características homogêneas (categoria socioeconômica e/ou escola idênticas) e heterogêneas (categorias socioeconômicas e/ou escolas diferentes). Com relação ao aspecto epidemiológico socioeconômico, as composições dos grupos mostraram alta similaridade (0.788 < SD < 1.0) entre algumas linhagens de C. albicans isoladas de crianças saudáveis independentemente de suas camadas socioeconômicas (alta, média e baixa). Isolados de alta similaridade não foram encontrados nas cavidades orais de crianças saudáveis pertencentes às camadas sociais A e D, B e C, ou C e E. Isto pode ser explicado pela ausência de uma rota de disseminação entre estas crianças. Geograficamente, algumas crianças saudáveis entre escolas idênticas e diferentes (particulares e públicas) também são portadoras de linhagens semelhantes, mas tal similaridade não foi encontrada entre outros de determinadas escolas. Esses dados podem refletir uma rota de disseminação restrita destes microrganismos em alguns grupos de escolares saudáveis, a qual pode ser dependente da categoria socioeconômica ou local geográfico de cada criança. Em contraste à alta similaridade, a baixa similaridade ou alto grau de polimorfismo (0.499 < SD < 0.788) dos perfis protéicos foi demonstrado em 23 (30,6%) isolados orais de C. albicans. Considerando o aspecto epidemiológico social, 42,1%, 41,7%, 26,6%, 23,5% e 16,7% foram isolados de crianças provenientes das categorias socioeconômicas A, D, C, B e E, respectivamente, e geograficamente, 63,6%, 50%, 33,3%, 33,3%, 30%, 25% e 14,3% foram isolados de crianças provenientes das escolas LAE, MA, CS, AV, HF, FMC e MEP, respectivamente. Tais resultados sugerem um maior grau de polimorfismo entre algumas linhagens isoladas de crianças saudáveis independentemente de suas camadas sociais ou locais geográficos. Estudos complementares envolvendo escolares saudáveis e seus familiares, professores, serventes, hábitos nutricionais e de higiene deverão ser realizados a fim de estabelecer as fontes de tais padrões de colonização em grupos de populações de crianças saudáveis. Os perfis de proteínas totais obtidos por SDS-PAGE associados com análise numérica computadorizada podem proporcionar critérios adicionais para os estudos epidemiológicos e taxonômicos de C. albicans.
Material and methods: Fifty nine elders wearing complete dentures and living in retirement homes... more Material and methods: Fifty nine elders wearing complete dentures and living in retirement homes in Curitiba (southern Brazil), were divided into two groups: group #1, 26 patients with denture-induced stomatitis and group #2, 33 patients without denture-induced stomatitis. The two groups were evaluated in relation to the degree of denture-induced stomatitis, salivary fungal loads, and secretion of some histolytic enzymes.Results: Patients from group #1 showed higher degrees of colonisation by Candida albicans (p = 0.031). Candida krusei, Candida tropicalis, and Candida parapsilosis were also isolated, but there were no differences between the groups (p > 0.05). Secretory aspartyl protease (Sap) and chondroitinase did not show significant differences among the isolated Candida spp. in the two groups. Phospholipase secretion rates were higher among the strains of C. albicans from group #2 (p = 0.036). The same behaviour was not detected for non-albicans Candida species.Conclusions: The results could infer that differences in the secretion rates of candidal histolytic enzymes should not be imputed as imperative for the progress of denture-induced stomatitis.

An experimental analysis was made to quantify the adherence rates and the biofilm formation capac... more An experimental analysis was made to quantify the adherence rates and the biofilm formation capacity of Streptococcus mutans ATCC25175 and Candida albicans SC5314 on orthodontic material surfaces in the presence of cigarette smoke condensate (CSC).Metal brackets, bands, acrylic resin, and polyurethane elastic rings were coated with stimulated saliva and submitted to adhesion and biofilm formation tests with and without CSC in a dynamic system.The CSC increased the adhesion of S mutans ATCC25175 to the acquired pellicle (P <0.05) for bands (4.08 times), acrylic resin (2.89 times), and brackets (3.37 times) and reduced it in polyurethane elastic (2.66 times; P <0.05). S mutans ATCC25175 biofilm biomass was increased by CSC only on brackets (1.60 times; P <0.05). In the presence of CSC, the adhesion of C albicans SC5314 increased (P <0.05) on bands (1.81 times), brackets (9.61 times), elastics (29,133 times), and acrylic resin (177 times). Greater formation of C albicans SC5314 biofilm caused by CSC (P <0.05) was observed on acrylic resin (2.13 times) and brackets (2.32 times).The results indicated that cigarette tobacco smoke can interfere with the adhesion and biofilm formation of these microorganisms to various orthodontic materials.
The aim of this work was to evaluate the concentration of carbamide peroxide compounded at differ... more The aim of this work was to evaluate the concentration of carbamide peroxide compounded at different dispensing pharmacies. Immediate concentration analysis was made of bleaching gels dispensed by specialized pharmacies, and of a commercially available gel (control group) (n = 20). The carbamide peroxide concentration was determined by titration and the results were analyzed statistically by the Kruskal-Wallis test. The commercial bleaching agent (control group) and one of the gels from the pharmacies presented the best mean concentration values, close to 16%. In conclusion, the concentration of the manipulated and industrialized carbamide peroxide gels presented concentration values differing from 16%.
The production of Secretory Aspartyl Proteases (Sap) is an important virulence factor of Candida ... more The production of Secretory Aspartyl Proteases (Sap) is an important virulence factor of Candida albicans. Many studies have shown that a challenge with sub-inhibitory concentrations of antifungals lead species of Candida to the secretion of higher concentrations of Sap. Nevertheless, published studies only reported the secretion of such enzymes by cells growing in planktonic phase, with few mention of biofilms. The present study evaluated the alterations in the secretion of Sap by C. albicans grown in biofilms and exposed to sub-inhibitory concentrations of fluconazole. The MICs for fluconazole of seven clinical strains were determined for planktonic cells. Biofilm and planktonic cells were grown in the presence of ½ MIC, ¼ MIC, and no medication (control). The relative metabolic activity, indirectly related to cell loads, were estimated by the absorbance of reduced XTT and the Sap activity was evaluated by bovine albumin test. It was observed that 72 h-old biofilms under the influence of ½ MIC had fewer cells than ¼ MIC and control. The production of Sap was inversely proportional to the cell content, with higher secretion in ½ MIC, followed by ¼ MIC and control. Biofilms of C. albicans challenged by sub-MICs of fluconazole tend to secrete higher quantities of Sap.
2, 3-bis (2-methoxy-4-nitro-5-sulfophenyl)-5-[(phenylamino) carbonyl]-2H-tetrazolium hydroxide (X... more 2, 3-bis (2-methoxy-4-nitro-5-sulfophenyl)-5-[(phenylamino) carbonyl]-2H-tetrazolium hydroxide (XTT) reduction assay has been used to study Candida biofilm formation. However, considering that the XTT reduction assay is dependent on cell activity, its use for evaluating mature biofilms may lead to inaccuracies since biofilm bottom cell layers tend to be relatively quiescent at later stages of biofilm formation. The aim of this study was to improve XTT reduction assay by adding glucose supplements to the standard XTT formulation. Candida albicans ATCC 90028 was used to form 24-, 48- and 72-h biofilms. The oxidative activity at 90, 180 and 270 min of incubation was evaluated. The control consisted of standard XTT formulation without glucose supplements, and was modified by the addition of 50, 100 and 200 mM of glucose. The XTT assay with 200 mM glucose showed more accurate and consistent readings correlating with biofilm development at 24, 48 and 72 h. Biofilm growth yield after 180 min incubation, when evaluated with the 200 mM glucose supplemented XTT, produced the most consistent readings on repetitive testing. It may be concluded that glucose supplementation of XTT could minimize variation and produce more accurate data for the XTT assay.
Background: The virulence potential of Candida albicans strains enrolled in denture-related cand... more Background: The virulence potential of Candida albicans strains enrolled in denture-related candidosis still remains uncertain. Candida albicans cells with higher cell surface hydrophobicity (CSH) rates, so-called hydrophobic, present higher adhesion success in different host tissues than cells with lower rates, or even hydrophilic.Objective: The proposition of this study was to evaluate the differences in the CSH of strains isolated from denture users with and without denture-related candidosis.Material and methods: The strains were obtained from two paired groups of patients living a same retirement house. Fungal cells were submitted to CSH evaluation by the hydrocarbon partition test using xylene.Results: The measures revealed that the yeasts from patients with candidosis had CSH values ranging from 4.52% to 12.24%, with an average of 8.22 ± 2.92%. In the countergroup, the CSH ranged from 3.86% to 14.36%, with an average of 8.38 ± 3.76%. The difference between the groups were considered not relevant (p = 0.997).Conclusion: The results let to the inference that natural populations of C. albicans from patients with and without clinical manifestation denture-related candidosis do not differ one from the other regarding to CSH.
