Abstract

Cellule nerveuse modifiée, son procédé de préparation et son application. La cellule nerveuse modifiée est dérivée d'une cellule souche pluripotente induite modifiée ; et la cellule souche pluripotente induite modifiée est une cellule souche pluripotente induite contenant des plasmides, les plasmides comprenant pTet-O-Ngn2-puro, Tet-O-FUW-EGFP, et FUdeltaGW-rtTA. Les cellules souches pluripotentes induites génétiquement modifiées sont rapidement préparées à grande échelle au moyen d'une transduction de vecteur viral, puis sont induites par un antibiotique et soumises à une culture de criblage et de différenciation pour obtenir une cellule nerveuse modifiée, et enfin, la cellule nerveuse modifiée est induite à l'aide d'une protéine beta-amyloïde de façon à obtenir une cellule ayant un phénotype de maladie d'Alzheimer. L'ensemble du processus de préparation est simple dans son fonctionnement et court dans sa durée, et une préparation à grande échelle peut être mise en œuvre. Ainsi, un matériau expérimental reproductible et aux propriétés uniformes est présenté pour le développement et la recherche de médicaments.
